Hepatocyte Growth Factor-mediated Regulation of OCT4 in human Mesenchymal Stem Cells

  • Mesenchymal stem cells (MSCs) hold great promise as a source of stem cells for therapy, but several limitations remain. We previously proposed that human embryonic stem cell-derived MSCs (hE-MSCs) expressing higher hepatocyte growth factor (HGF) levels were better alternatives, exhibiting greater expandability in vitro and greater therapeutic capacity in vivo. In this study, we aimed to examine the regulation of OCT4 expression in stem cells and to elucidate its underlying mechanism of transcriptional regulation of OCT4. We detected higher expression of OCT4, a stemness-associated gene in hE-MSCs than in human bone marrow-derived MSCs (hBM-MSCs). To determine the underlying regulatory mechanism of OCT4 expression in human MSCs (hMSCs), ELISA was performed using cell culture supernatants of hMSCs. Unlike fibroblast growth factor 2 or vascular endothelial growth factor, HGF was strongly expressed in hE-MSCs, also HGF treatment significantly increased OCT4 expression in hBM-MSC. Moreover, senescence-associated heterochromatin foci were decreased in HGF-treated hBM-MSCs compared with those in the HGF non-treated group. HGF increased Rb phosphorylation, and we confirmed the increased binding of E2F1 to the OCT4 promoter region at -233 from the transcription start point in the presence of HGF. Taken together, these results suggest that HGF-mediated regulation of OCT4 via E2F1 can help enhance the lifespan of hBM-MSCs during in vitro expansion.

  • In this paper, an accelerated life test procedure for a vacuum cleaner motor is proposed. We investigate the failure mechanism of the motor and select some accelerating factors and determine the orifice size as a key accelerating factor. Three stress levels of orifice size are tested and the failure data with censored data are analyzed. The modified accelerating test will reduce the test time in design phase by using the accelerating factor.

  • Objectives : The aim of this study was to understand electro-physiological peculiarity in meridian and acupoints, and understand acupuncture therapy mechanism as an electro-physiological viewpoints. Methods : I reviewed the articles on the electro-physiological peculiarity of the meridian and acupoints Results and Conclusions : It has been reported that meridian and acupoints have high-electrical conductivity and row-electrical resistance. On this scientific basis, to understand the mechanism of acupuncture therapy, I made some hypotheses. At first, there is electro-property in meridian and acupoint. The second, energy flowing in meridian is related with electro-property. The third, there is electronic interaction between practitioner of acupuncture therapy and patient receiving acupuncture therapy. The forth, acupuncture effects which may be expressed by the electro-charge capacity is transfered between practitioner and patient via acupuncture. Electro-charge induced via acupuncture in practitioner may be an important factor that initiate the electro-charge changes in meridian and acupoint of patient.

  • Quadruped robot is very useful mechanism for a various area. Recently, home entertainment and military robots adapted quadruped platform and useful function have been introduced. Our goal is the development of quadruped robot locomotion for any type of ground included to sloping one and irregular terrain. This paper, as a first step, deals with design and construction of quadruped robot walking on the flat ground. The most important factor of quadruped robot is stability of locomotion. At first, we introduce the developed quadruped robot based on dynamic simulation and experimental study of general gait algorithm. Finally, propose unique locomotion proper to our mechanism. Future work of this study is the performance test and analysis on the ground of various conditions and proposes the improved mechanism and gait algorithm.

  • This paper proposes a procedure for designing an accelerated test using SMAT(Stress, (failure) Mechanism and Test) model describing the relation among stress, failure mode/mechanism and test method. In SMAT model the stresses to be applied are derived from the environmental factor analysis, the relative importance of those stresses can be estimated using AHP(Analytic Hierarchy Process) and failure mode/mechanism and test method are derived from the fields failure information and FMEA(Failure Mode and Effect Analysis). By applying the procedure we can make a selection of major factors to cause the failure of assembly and design the accelerated test using DOE(Design of Experiments) The procedure is illustrated with an qualification test case study of washing machine shaft assembly in "A" electric appliance company.

  • Agrobacterium-mediated genetic transformation has been widely used for the production of genetically modified transgenic plants to obtain specific desired traits. Most of the molecular mechanisms that underlie the transformation steps have been well elucidated over the years. However, a few steps, such as nuclear targeting, T-DNA integration, and Agrobacterium-plant proteins involved remain largely obscure and are still under extensive studies. This review describes the major steps involved in the molecular mechanism of Agrobacterium-mediated transformation and provides insight in the recent developments in studies on the Agrobacterium-mediated genetic transformation system. Some factors affecting the transformation efficiency are also briefly discussed.

  • The present work have been developed the interpretation processor including the behavior of material failure and the separation phenomena under transient dynamic loading (the operation of explosive bolt) using AUTODYN V4.3, SoildWork 2003 and TrueGrid V2.1 programs. It has been demonstrated that the interpretation in ridge-cut explosive bolt under dynamic loading condition should be necessary to the appropriate failure model and the basic stress of bolt failure is the principal stress. The use of this interpretation processor developing the present work could be extensively helped to design the shape and the amount of explosives in the explosive bolt having a complex geometry. It is also proved that the interpretation processor approach is an accurate and effective analysis technique to evaluate the separation mechanism in explosive bolts.

  • The present experiment was carried out to examine the mechanism of cytotoxicity of Chromium in CHO cells. Chromium induced chromosomal aberrations in a dose-dependent manner. The most frequent type of aberration was chromatid deletions and chromosome type exchanges were also observed. Ultrafiltrates of culture media from CHO cells treated with Chromium induced sister chromatid exchanges(SCE) in CHO cells and Chromium induced lipid peroxidation. It was suggested that indirect effect through formation of clastogenic factor(CF) as well as direct effect on DNA might contribute to the cytotoxicity of Chromium.

  • The shock performance of hard disk drives has been a serious issue for portable computers and AV application HDD. Focusing on the motion of an actuator, we investigated non-operational shock mechanism and studied several parameters that affect the shock performance by experimental analysis. It was found that there are two important factors fort the actuator to endure high shock revel. One is a shock transmissibility and the other is a beating between the arm blade and the suspension. To generalize the shock transmissibility, the concept of shock response spectrum was introduced. The shock response spectrum of the actuator system was obtained experimentally and compared with that of an analytical single degree of freedom model. It was found that there was a good agreement. The first bending natural frequency of the arm blade was found to be the most important factor for the low shock transmissibility. By applying the shock response spectrum and avoiding the beating, we could design an actuator of high shock performance.

A study on the mechanism for reduction of lead-induced toxicity in nervous system by aloe vera (Aloe vera의 연 유도성 신경계 독성 저감 기전에 관한 연구)

  • Effects of water extract of aloe vera on lead-induced neurotoxicity were investigated in sciatic nerve isolated from rat. The mechanism on toxicity reduction by measuring activities of axonal enzymes, metabolism of myo-inositol in nerve, lead concentration in several organs and so on were further examimed. In the lead-treated rats, the transport rate of axonal enzyme, such as acetyl cholinesterase and choline acetyltransferase, was reduced by from 50% to 30% respectively. Reduction in myo-inositol concentration and $Na^+/K^+$ ATPase activity were also observed in sciatic nerve from lead-treated rat. However, the aloe extract administration significantly eliminated the impairment and maintained myo-inositol concentration to about 85% of normal level. Also aloe extract promoted the excretion rate of lead which is accumulated in blood, sciatic nerve and kidney. These results suggest that lead-induced neurotoxicity was significantly reduced by administration of aloe extract and the mechanism might be partly increase in kidney excretion rate of lead and parity normalization of $Na^+/K^+$ ATPase activity which is critical factor in order to keep nerve maintaining normal myo-inositol level.
