Caveolin-1 inhibits membrane-type 1 matrix metalloproteinase activity

  • Membrane-type 1 matrix metalloproteinase (MT1-MMP) is a zinc-dependent proteinase found in cholesterol-rich lipid rafts on the plasma membrane. MT1-MMP hydrolyzes extracellular matrix (ECM) proteins, activates pro-matrix metalloproteinase-2 (proMMP-2) and plays an important role in ECM remodeling, cancer cell migration and metastasis. The role of caveolin-1, an integral protein of caveolae, in the activation of MT1-MMP remains largely unknown. Here, we show that the expression of caveolin-1 attenuates the activation of proMMP-2, reduces proteolytic cleavage of ECM and inhibits cell migration. We utilized the cytoplasmic tail domain deletion (${\Delta}CT$) or the E240A mutant of MT1-MMP. Co-expression of caveolin-1 with the wild-type or the ${\Delta}CT$ MT1-MMP decreased the proMMP-2 activation and inhibited collagen degradation and cell migration. Caveolin-1 had no effect on the catalytically inert E240A MT1-MMP. Our findings suggest that caveolin-1 is essential in the down-regulation of MT1-MMP activity by promoting internalization from the cell surface.

Inhibitory Effect of Siderophore Purified from Burkholderia sp. CAS-5 on the Matrix Metalloproteinase-2 (Gelatinase A) (Burkholderia sp. CAS-5 균으로 부터 생산된 시드로포어의 Matrix metalloproteinase-2(Gelatinase A) 억제 활성)

  • Matrix metalloproteinase-2 is known to be involved in pathological processes such as tumor invasion or rheumatoid arthritis. A soil microorganism producing siderophore under low iron stress $(up\;to\;5\;{\mu}m\;of\;iron)$ was identified as Burkholderia sp. Hydroxamate type siderophore produced by Burkholderia sp. CAS-5 was partially purified. MMP inhibitory activity of siderophore was confirmed by gelatin zymography. The $Zn^{2+}-chelating$ activity of siderophore correlated with the inhibition of MMP-2 activity.

Tissue engineering of dental pulp on type I collagen

  • The purpose of this study was to regenerate human dental pulp tissues similar to native pulp tissues. Using the mixture of type I collagen solution, primary cells collected from the different tissues (pulp, gingiva, and skin) and NIH 3T3 ($1{\;}{\times}{\;}10^5{\;}cells/ml/well$) were cultured at 12-well plate at $37^{\circ}C$ for 14 days. Standardized photographs were taken with digital camera during 14 days and the diameter of the contracted collagen gel matrix was measured and statistically analyzed with student t-test. As one of the pulp tissue engineering, normal human dental pulp tissue and collagen gel matrix cultured with dental pulp cells for 14 days were fixed and stained with Hematoxyline & Eosin. According to this study, the results were as follows: 1. The contraction of collagen gel matrix cultured with pulp cells for 14 days was significantly higher than other fibroblasts (gingiva, skin) (p < 0.05), 2. The diameter of collagen gel matrix cultured with pulp cells was reduced to 70.4% after 7 days, and 57.1% after 14 days. 3. The collagen gel without any cells did not contract, whereas the collagen gel cultured with gingiva and skin showed mild contraction after 14 days (88.1% and 87.6% respectively). 4. The contraction of the collagen gel cultured with NIH 3T3 cells after 14 days was higher than those cultured with gingival and skin fibroblasts, but it was not statistically significant (72.1%, p > 0.05). 5. The collagen gel matrix cultured with pulp cells for 14 days showed similar shape with native pulp tissue without blood vessels. This approach may provide a means of engineering a variety of other oral tissue as well and these cell behaviors may provide information needed to establish pulp tissue engineering protocols.

The calculation method of the traffic using incidence matrix in vehicle network tunnels (네트워크 도로터널에서 근접행렬을 이용한 교통량 계산 방법)

  • In order to design the ventilation in the road tunnel, it is necessary to know the ratio of average annual daily traffic by vehicle type. In general, the road tunnels are onedirectional tunnel, so the traffic of each vehicle type does not change along the tunnel. On the other hand, in the case of network road tunnels, since the connections in the tunnels are complex, the traffic of vehicle-type varies depending on the network composition of tunnels. In the studying the easy method for calculating the ratio of vehicle type for the network road tunnel are proposed with using incidence matrix.

In vitro Angiogenic Activity of Aloe vera Gel on Calf Pulmonary Artery Endothelial (CPAE) Cells

  • Angiogenic activity of Aloe vera gel was investigated by in vitro assay. We obtained the most active fraction from dichloromethane extract of Aloe vera gel by partitioning between hexane and 90% aqueous methanol. The most active fraction (F3) increased the proliferation of calf pulmonary artery endothelial (CPAE) cells. In addition, F3 fraction induced CPAE cells to invade type I collagen gel and form capillary-like tube through in vitro angiogenesis assay, and increased the invasion of CPAE cells into matrigel through in vitro invasion assay. Furthermore, the effect on the MRNA expression of proteolytic enzymes which are key participants in the regulation of extracellular matrix degradation was investigated by northern blot analysis. F3 fraction enhanced mRNA expression of urokinase-type plasminogen activator (u-PA), matrix metalloproteinase-2 (MMP-2), and membrane-type MMP (MT-MMP) in CPAE cells whereas the expression of plasminogen activator inhibitory (PAl-1) mRNA was not changed.

  • Let HEI, HEII, HEIII and HEIV be the first, second, third and fourth type Loo-Keng Hua domain respectively, 𝜑 a holomorphic self-map of HEI, HEII, HEIII, or HEIV and u ∈ H(𝓜) the space of all holomorphic functions on 𝓜 ∈ {HEI, HEII, HEIII, HEIV}. In this paper, motivated by the well known Hua's matrix inequality, first some inequalities for the points in the Bers-type spaces of the Loo-Keng Hua domains are obtained, and then the boundedness and compactness of the weighted composition operators W𝜑,u : f ↦ u · f ◦ 𝜑 on Bers-type spaces of these domains are characterized.

James-Stein Type Estimators Shrinking towards Projection Vector When the Norm is Restricted to an Interval

  • Consider the problem of estimating a $p{\times}1$ mean vector ${\theta}(p-q{\geq}3)$, $q=rank(P_V)$ with a projection matrix $P_v$ under the quadratic loss, based on a sample $X_1$, $X_2$, ${\cdots}$, $X_n$. We find a James-Stein type decision rule which shrinks towards projection vector when the underlying distribution is that of a variance mixture of normals and when the norm ${\parallel}{\theta}-P_V{\theta}{\parallel}$ is restricted to a known interval, where $P_V$ is an idempotent and projection matrix and rank $(P_V)=q$. In this case, we characterize a minimal complete class within the class of James-Stein type decision rules. We also characterize the subclass of James-Stein type decision rules that dominate the sample mean.

Cloning of a matrix metalloproteinase cDNA from Scylliorhinus torazame (두툽상어 matrix metalloproteinase 유전자 cDNA의 클로닝)

  • Matrix metalloproteinases(MMPs) are a group of zinc enzymes responsible for degradation of the matrix components such as collagen and proteoglycans in normal embryogenesis and remodeling and in many disease processes such as arthritis, cancer, periodontitis, and osteprocess. Genetically distince MMPs have been characterized and their genes have been cloned thus far from a variaty of species but not from fishes. In this stydy, a mmp cDNA was cloned by using RT-PCR(reverse transcriptase dependent polymerase chain reaction) from Scylliorhinus toraxzame(shark), agroup of cartilaginous fish, abundant in the coast of Pusan, Korea. It has 74% base homologue with membrane type matrix matalloproteinase-3 genes(mt3-mmps) from human, rat and chick, and also shows more than 90% residue homologue with them. In addition, it has cysteine switch domain, zinc binding domain(HExGH motif), propeptide cleavage site, and RRKR motif, which are present in MMPs. This result indicates that cDNA fragment cloned here may be mt3-mmp or its analogous gejne cDNA fragment of Scylliorhinus torzame.

  • Parallel optical arithmetic techniques have been developed using the correlation property of optical phase conjugate wave generated by degenerated four wave-mixing. In this paper, conventional rectangular-type coded pattern used for optical logic system is replaced by circular one for effective beam coupling in a photorefractive $BaTiO_3$ material. By adequately adjusting the distance between circular-type pixels of the input pattern and grouping the correlated output, optical binary half addition/subtraction, binary multiplication and, matrix-matrix computation are demonstrated.

  • In this paper, robust adaptive control algorithms which can be applied to unknown uncertain systems are suggested. Transform matrix for dividing states into "uncontrolled" states and "controlled" states and general searching procedure for the transform matrix which assign arbitrary n-1 eigen values for the uncontrolled subsystem of n-th orther single-input single-output systems of which state variables can be observable are also studied and utilized for the design of new-type controllers. We drived new-type control laws by using adaptive control theory and variable structure system and its stability is proved by using Lyapunov stability theory.
