Ascidian soft tunic syndrome (AsSTS) caused by Azumiobodo hoyamushi (A. hoyamushi) is a serious aquaculture problem that results in mass mortality of ascidians. Accordingly, the early and accurate detection of A. hoyamushi would contribute substantially to disease management and prevention of transmission. Recently, the loop-mediated isothermal amplification (LAMP) method was adopted for clinical diagnosis of a range of infectious diseases. Here, the authors describe a rapid and efficient LAMP-based method targeting the 18S rDNA gene for detection of A. hoyamushi using ascidian DNA for the diagnosis of AsSTS. A. hoyamushi LAMP assay amplified the DNA of 0.01 parasites per reaction and detected A. hoyamushi in 10 ng of ascidian DNA. To validate A. hoyamushi 18S rDNA LAMP assays, AsSTS-suspected and non-diseased ascidians were examined by microscopy, PCR, and by using the LAMP assay. When PCR was used as a gold standard, the LAMP assay showed good agreement in terms of sensitivity, positive predictive value (PPV), and negative predictive value (NPV). In the present study, a LAMP assay based on directly heat-treated samples was found to be as efficient as DNA extraction using a commercial kit for detecting A. hoyamushi. Taken together, this study shows the devised A. hoyamushi LAMP assay could be used to diagnose AsSTS in a straightforward, sensitive, and specific manner, that it could be used for forecasting, surveillance, and quarantine of AsSTS.
The apicomplexan parasite, Perkinsus sp., has been reported as a causative pathogen responsible for the mass mortality of the Manila clams, Ruditapes philippinarum in European countries as well as in Korea. Prevalence, infection intensity, histopathologic features and zoosporulation of Perkinsus found in the Manila clams distributed along the coast of Cheju Island were investigated in this study. Clams were collected from nine different beaches along the Cheju Island from May to July in 2000. Ray's Fluid Thioglycollate Medium (RFTM) and Choi's NaOH lysis methods were applied in the diagnosis and quantification of the Perkinsus infection. Prevalence, percentage infection of the parasite examined was 100% in Pyosun, 70% in Sungsan, 63% in Kumneong, 33% in Jongdalri, 21% in Iho, 17% in Moslpo, and 14% in Seogwipo. No Perkinsus was found in the clams collected from Kimnyong and Yongmeo-ri. Infection intensity as a number of Perkinsus cells per gram tissue wet weight (twwt), was 98,430 cells/g twwt in Pyosun, 78,553 cells/g twwt in Sungsan, 18,980 cells/g twwt in Kumneong, 4,290 cells/g twwt in Jongdalri, 1,527 cells/g twwt in Iho, 1,069 cells/g twwt in Moslpo, and 853 cells/g twwt in Seogwipo. Histological preparation of the infected tissues revealed trophozoites of Perkinsus sp., ranged from 5 to 10 ${\mu}{\textrm}{m}$, in diameter mostly distributed in the digestive gland and the gill filaments. Zoospores were discharged from the hypnospore via discharging tube about 2 days after incubated in filtered and aerated seawater. In general, the prevalence and infection intensity of Perkinsus in Cheju Island were much lower than that reported from the western and southern coast of Korea.

To investigate the effect of large patch infection on oxidative stress induction, growth, $H_2O_2$ concentration and lipid peroxidation were compared between pathogen-infected and healthy (control) zoysiagrass. The sampling for leaves and roots were carried out every 2 days for a period of 6 days. Pathogen-infection increased root mortality by 30% compared to control. Dry mass was not significantly affected by pathogen-infection until day 4, but significant decreases in both leaves (-14%) and roots (-20%) were observed at day 6. The $H_2O_2$ concentration in pathogen-infected leaves rapidly increased within the first 2 days(+28%) and then slightly decreased. The increase of $H_2O_2$ in pathogen-infected roots was distinct, showing 1.7-fold higher level than control at day 6. The extent of lipid peroxidation caused by pathogen-infection continuously increased for the first 4 days. This was then stagnated until day 6. In roots, the significant increase of lipid peroxidation was observed only at day 2. These results indicate that large patch-infection induces oxidative stress, and that the oxidative stress responsive pattern was plant organ specific.
Purpose: Intracorporeal anastomosis is the most difficult procedure during pure single-incision distal gastrectomy (SIDG) that affects its generalization. We introduced unaided delta-shaped anastomosis (uDelta), a novel anastomosis technique, for gastroduodenostomy after pure SIDG, and compared the results with those of previously reported Roux-en-Y anastomosis (RY). Materials and Methods: Between March 2014 and March 2015, SIDG with D1+ lymph node dissection was performed for early gastric cancer through a 2.5-cm transumbilical incision without any additional port. uDelta was performed by the operator alone, without any intracorporeal assistance. Results: uDelta was performed on 11 patents, and uncut RY was performed on 5-patients without open or multiport conversion. R0 resection was performed in all cases. No significant differences were observed in mean age and body mass index between patients who underwent uDelta or RY. Mean operation times were $214.5{\pm}36.2$ minutes for uDelta and $240.8{\pm}65.9$ minutes for RY, which was not significantly different. Reconstruction time for uDelta was shorter than that for RY, with marginal statistical significance ($26.1{\pm}8.3$ minutes vs. $38.0{\pm}9.1$ minutes, P=0.05). There were no intraoperative transfusions, 30-day mortality, or anastomosis-related complications in either group. Average length of hospital stay was $8.2{\pm}1.9$ days in the uDelta group and $7.2{\pm}0.8$ days in the RY group (P=0.320). Conclusions: After carefully considering indications, uDelta can be a feasible and can be a reproducible reconstruction method after SIDG in early gastric cancer.
The genus Cordyceps known as an insect parasite forms a sclerotium in insect bodies and then produces perithecia on the single or multiple stromata produced from sclerotium. Collected Cordyceps were identified into 5 species: Cordyceps militaris, C. nutans, Cordyceps sphecocephala, Isaria japonica, and Torrubiella sp. The fruit bodies of Cordyceps in petri-dish cover were fixed by tape and put the lid on water agar plates to isolate these collected Cordyceps. The germinated spores were transferred from water agar to Potato dextrose agar(PDA) after six hours. Mycelial growth of C. nutans and C. militaris was the most successful on Hamada media and was also good on Complete media and PDA. Mannose as a carbon source was good for two species and Glutamic acid as a nitrogen source was satisfactory to C. militaris and Asparagine gave a good result to C. nutans. C. militaris and C. nutans showed similar mycelial growth rate on the media that contained thiamine-HCI, biotine or nicotinic acid as a vitamine. When conidia of C. nutans were inoculated to insects, mortality was high in Artogeia napi L, Hemiptera, Plutella xylostella and 50% in Orthoptera, 12% in Acantholyda posticalise M, but not Agelastica coerulea B. in Aphididae, C. nutans was collected from only Hemiptera in nature, but killing effect on other insects was proved. Mycelial growth and fruit-body formation were good on the media that consist of rice powder 5g, wheat flour 5g, water 100ml, but formed fruit-body was not complete stromata but a mass of conidia according to results of observing microscope.

Temperature is one of the most important criteria considered in species preference for aquaculture. Acute drop in temperature during winter is a cause of disease and mass mortality in farmed fish. This study was carried out the low water temperature tolerance, oxygen consumption, hematologic and histological responses to use as basic data for the management of fish farming which frequently cause death due to winter water temperature drop. Low-lethal water temperature for 4 days of file fish Thamnaconus modestus (4day-LT50) was 6.97℃ (6.69~7.27℃). Oxygen consumption rate decreased with decreasing water temperature, showing a significant difference between water temperatures. SOD activity increased significantly at 6℃ experimental group than control group (10℃) (p<0.05), but CAT did not show any significant difference between experimental temperatures (p>0.05). Cortisol increased with decreasing experimental water temperature compared to control group. Histological changes in the liver include decreased blood vessels in the blood vessels, proliferation of acid cells, condensation of the nucleus, atrophy of pancreatic exocrine gland cells, and enzyme source granules.

Protozoan parasites belonging to the genus Perkinsus elicit severe inflammatory responses and are associated with mass mortality of commercially important marine shellfish worldwide. In the present study, we examined the external features of P. olseni zoospores in detail using light and scanning electron microscopy. Our study showed that the zoospores have an oval body with a long anterior flagellum and a short posterior flagellum. The anterior flagellum has a unilateral array of mastigonemes. Mean body dimensions were $3.37{\pm}0.33{\mu}m{\times}1.72{\pm}0.22{\mu}m$. The average length of the anterior and posterior flagella was $16.34{\pm}1.52{\mu}m$ and $8.25{\pm}1.39{\mu}m$, respectively. Zoospores of P. olseni found in Korean waters have shorter and narrower bodies, longer anterior flagella, and shorter posterior flagella than zoospores of Perkinsus spp. found in the mollusks of North America and Europe.
To investigate bioactive materials for development of natural conservative agent on organic cultural heritage, essential oil from Asarum sieboldii was screened for antifungal and insecticidal activity against 4 wood rotting fungi and adult of Lasioderma serricorne. Antifungal activity of essential oil was tested by using paper disc soaking method. Antifungal activity expressed as $IC_{50}$ value showed $1.50{\sim}2.84{\mu}l/disc$ range and the most significant antifungal activity was observed in Lentinus lepideus. The insecticidal activity of essential oil was examined by topical application method against L. serricorne adults. 50% and 100% of essential oil gave 98.3% and 100% mortality for 24 hours, respectively. The major components of the essential oil were methyl eugenol (56.32%), eucarvone (11.53%), safrole (5.79%), ${\delta}$-3-carene (2.09%), which were identified by gas chromatography-mass spectrometry. From these results, essential oil from A. sieboldii could be useful for conservation of organic cultural heritage against biological deterioration by insect and wood rotting fungi.
Background : Solitary fibrous tumors of the pleura (SFTP) is one of rare neoplasms that originated from submesothelial mesenchyme. Clinical course or extent of surgical resection is not well known. Material and Method: We retrospectively reviewed all the clinical records of the patients who had undergone surgical resection of benign and malignant SFTP, Result: Twenty two (male female 14 : 8) patients were enrolled and mean age was 50.2(range 25∼83). Number of symptomatic patients at the time of diagnosis was 13 (59%) and the most common symptom was dyspnea. Operative approach was carried out through thoracotomy (n=14) or video-assisted thoracic surgery (n=8). Mass excision was performed in 12 cases and en bloc resection including adjacent structure in 10 cases. In all cases complete resection was performed. There was no operative mortality. Malignant SFTP were 11 cases and benign SFTP 11 cases. Local recurrences (n=2) or distant metastasis (n=6) occurred only in malignant SFTP. Conclusion: Number of symptomatic patients, on bloc resection, and recurrence was more in malignant SFTP. Although complete surgical resection is known as treatment of choice for SFTP, further study should be performed about systemic therapeutic modalities pre- or postoperatively to control recurrence and metastasis.

Hemodynamically unstable pelvic fractures show a remarkably high mortality rate of 40% to 60%. However, their standard of care remains controversial. We report here a case of a 78-year-old woman who was admitted to the Emergency Department with pelvic pain following a fall. Based on pelvic radiography, she was diagnosed with an unstable pelvic fracture. Her blood pressure was 60/40 mmHg, and owing to her unstable vital signs, emergency angiography was performed without computed tomography (CT). Both internal iliac arteries were embolized without sub-branch selection for prompt control of pelvic bleeding. Following embolization, her vital signs were stabilized. Subsequent CT revealed free intra-abdominal air, suggesting bowel perforation had occurred and necessitating emergency laparotomy. An approximately 1 cm-sized free perforation of the small intestine was identified intraoperatively, and primary closure was performed. A retroperitoneal hematoma identified intraoperatively was not explored further because it was a non-expanding and non-pulsatile mass. The patient was admitted to the Intensive Care Unit and transferred to the general ward on postoperative day 3. In this case, the hemodynamically stable pelvic fracture with bowel perforation was successfully and safely treated by prompt angioembolization without conducting CT.
