Recently, as the interest in the accelerated global warming and the food shortage problem is increased, the concerns for microalgae as photosynthetic microorganisms are also increased. Specially, photosynthetic microalgae, Spriulina platensis have been an attractive source for $CO_2$ gas fixations and for a vast array of valuable nutritious compounds. In this paper, to culture the microalgal Spirulina platensis NIES 39 in a batch culture with high mass, optimal conditions for the culture temperature, initial pH, light intensity and concentration of carbon and nitrogen, were tested. At the most favorable culture condition, $35^{\circ}C$, initial pH 9.5, 4500 lux and carbon and nitrogen concentration of 16.8 g/L $NaHCO_3$ and 2.5 g/L $NaNO_3$, the excellent yields of 2.10 g/L biomass and 29.53 mg/L chlorophyll were obtained.
The objective of this study is to promote germination for mass cultivation of Adenophora triphylla. After the seed soaking for I day in BA and $GA_3$ solution, seed germination was effectively promoted in the treatment of 500mg/L $GA_3$ and the highest germination rate was 94%.The average germination days were 1.5days earlier than those of non-treated seeds. In a chemicals treatment, seed germination rate was 54% in 1% $KNO_3$ treatment but KOH treatment was no significantly effected. The durations of chilling treatment for breaking of dormancy were longer than 2weeks. The optimal temperature for germination was $25^{\circ}C$. The promotion of seed germination is presumed to be due to the breaking of dormancy by $GA_3$, chilling treatment rather than seed coat maceration by KOH or $KNO_3$ treatments.

The purpose of this study is to investigate the volatile compounds of Korean traditional gochujang from various districts. The volatiles from each traditional gochujang are being extracted by simultaneous steam distillation extraction (SDE), and analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). Twenty compounds are identified as major volatile components which include 8 esters, 4 alcohols and 4 acids. The most traditional gochujang possesses more volatile components rather than commercial gochujang products. Most acids come from fatty acids and the alcohols derive from the oxidative degradation of linolenic acid. The most abundant volatile compounds for both traditional and commercial gochujang include 10 compounds such as 2-methyl-1-propanol, hexanal, 2-methyl-1-butanol, octanoic acid ethyl ester, as well as the various type of acids and esters. They represent most of the total GC peak areas, respectively. From the results, the characteristics of the flavors for traditional gochujang from each district are not clear but have shown various components than the commercial products.
The aim of this study is to examine the physiological characteristics of an agarophyte Agarophyton vermiculophyllum (Ohmi) Gurgel, J.N. Norris et Fredericq in the early life stage of tetrasporophytes (2n) and gametophytes (n) to select appropriate seedlings for mariculture. Growth experiments were carried out at the combinations of four temperatures (20, 25, 30, and 35℃) and three light intensity levels (20, 60, and 100 µmol photons m-2 s-1) in the two ontogenetic stages: discoid holdfasts and erect sporelings. Holdfast areas and sporeling lengths of tetrasporophytes and gametophytes were estimated after 14 days in culture. Relative growth rates (RGRs) for holdfast areas were 7.08-28.38% day-1 for tetrasporophytes and 11.58-23.67% day-1 for gametophytes. At 35℃, holdfasts of tetrasporophytes survived with RGRs of 7.08-23.28% day-1 but those of gametophytes died. Maximal holdfast growth of tetrasporophytes occurred at 30℃ and 100 µmol photons m-2 s-1, which were different from gametophytes (25℃ and 100 µmol photons m-2 s-1). RGRs of tetrasporophytic sporelings were 2.93-11.11% day-1 and were between 0.78-10.82% day-1 for gametophytes. Maximal growth of A. vermiculophyllum sporelings occurred at 25℃ and 60 µmol photons m-2 s-1 for tetrasporophytes, and at 20℃ and 100 µmol photons m-2 s-1 for gametophytes. In conclusion, the present results indicate that carpospores could be used as resources of spore-seedling methods having genetic diversity for mass field cultivation because tetrasporophytes showed higher-temperature tolerance and faster-growing ability than gametophytes of A. vermiculophyllum in the discoid holdfast and sporeling stages.
A strain named as HJ35 was isolated from the skin of sixty-five men and fourteen women for acne therapy, in order to find an effective antimicrobial agent against Propionibacterium acnes. Isolate HJ35 was identified as Enterococcus faecium based on 16 rDNA sequence and produced enterocin HJ35 having antimicrobial activities against most lactic acid bacteria, Enterococcus spp., Staphylococcus aureus, S. epidermidis, Clostridium perfringens, some bacilli, Micrococcus flavus, Listeria monocytogenes, L. ivanovii, Escherichia coli, Pseudomonas fluorescens and Propionibacterium acnes, in the modified well diffusion method. Especially, enterocin HJ35 showed a bactericidal activity against Propionibacterium acnes P1. The antimicrobial activity of enterocin HJ35 was disappeared completely with the use of protease XIV. But enterocin HJ35 activity is very stable at high temperature (up to $100^{\circ}C$ for 30 min), in wide range of pH (3.0${\~}$9.0), and by treatment with organic solvents. The apparent molecular mass of enterocin HJ35 was estimated to be approximately 4${\~}$4.5 kDa on detection of its bactericidal activity after SDS-PAGE. In batch fermentation of E. faecium HJ35, enterocin HJ35 was produced at the midlog growth phase, and its maximum production was obtained up to 2,300 AU/mL at the late stationary phase. By employing fed-batch fermentation, the enhanced production of enterocin HJ35 was achieved up to 12,800 AU/mL by feeding with 10 g/L glucose or 6 g/L lactate.
Growth kinetics of mammalian cell, Chinese Hamster Ovalry(CHO) was investigated to effectively produce pharmaceutically important Erythropoeitin under perfusion chemostat conditions. Perfusion rate, D is correlated with total viable is to be an essential factor in controlling growth kinetic parameters under this kind of operations. It is also found that the measurement of oxygen uptake rates is a relatively accurate method to understand cell growth, in case that the traditional cell count method is no longer useful due to heavy cell clumpings. True growth yield, Ymax and maintenance coefficient, me associated with mammalian cell growth were estimated as $2.86{\times}10^8$ cells/ g of glucose and 0.0063 g of glucose/ cells/ hr, respectively.
The possibility of using Chinese cabbage juice as a substrate for the production of Candida utilis cell mass was explored. Dry cell weight production and cell yield coefficient were 1.35-1.45 g/100 ml undiluted juice and 47-50%, respectively, when C. utilis was grown by shake flask culture at $30^{\circ}C$ for 24 hr on more than three-fold diluted Chinese cabbage juice to make the final sugar content be equal to or less than 1.0%. Supplementation of glucose(2%), $KH_2PO_4(0.2%)$ and $(NH_4)_2SO_4(0.2%)$ to three-fold diluted Chinese cabbage juice did not enhance the dry cell weight yield or the protein content of the yeast cell, while supplementation of yeast extract(0.2%) and peptone(0.2%) increased dry cell weight production and protein content but not as much as the amount of each nutrient added. It was found that Chinese cabbage juice was an excellent substrate for the cultivation of C. utilis.
This study was conducted to investigate the application effect of Lysobacter antibioticus HS124 (gelatinase and chitinase producing microorganism; GCM) for the improvement of soybean yield and control of stink bug. Our results showed that the yield of GCM-treated soybean increased by 17.0, 20.3, 19.0, and 25.6% in the experimental field sites of Muan-gun, Sunchang-gun, Gwangju 1, and Gwangju 2, respectively, compared to the yield of the soybean obtained by conventional practice treatment (CPT); however in the Gimjae-si, the yield decreased by 10.6%. Results in both pods and seeds $plant^{-1}$ were significantly increased in Gwangju 2 by the GCM cultured broth treatment (GCMT). Ratio of 3 seeds $pod^{-1}$ in Sunchang-gun was statistically significant between GCMT and CPT, however, the result in the other field showed no significance. Germination rate was only statistically improved by GCMT in Gwangju 2 field site. GCMT reduced the appearance of stink bug in all experimental field sites except in Gimjae-si. The soybean seed damage by stink bug was no significance in all of treatments. Therefore, GCMT could improve the productivity of soybean and also control the infestation of stink bug.
The purpose of this study is to construct the main system of simulator for the environment control of agricultural production facilities. The model describing the system was based on the energy and mass balance in an unsteady - state situation. The model consist of the three major parts : the main model, the light model, and the environmental control model, and each part was separated to be developed individually. The main model which is the core of this system includes the thermal model, the soil model, the ventilation model, the cultivation model, and the carbon dioxide model. And also the environmental control model includes the thermal curtain model, the heater/cooler model and the underground heat exchanger model. The equations used in this model were written in analog programming methods using PCSMP The simulator was evaluated through comparison between simulated and measured temperatures controlled during daytime and night. The results showed good agreements between the predicted and measured temperatures.
A Gram-type negative bacteria that can utilize crude oil as the sole source of carbon and energy was isolated from an activated sludge of a local sewage treatment plant and identified tentatively as belonging to the genus Acinetobacter. The isolate could degrade n-alkanes and unidentified hydrocarbons in crude oil and utilize n-alkanes, hydrophobic substrates, as sole carbon and energy sources. n-Alkanes from tridecane (Cl3) to triacontane (C30) in crude oil were degraded simultaneously with no difference in degradation characteristics between the two close odd and even numbered alkanes in carbon numbers. The linear growth of the isolate and the degradation characteristics of Pr-alkanes suggested that the transport of substrates from the oil phase to the site where the substrates undergo the initial oxidation in microorganism might be the rate limiting in the biodegradation process of crude oil constituents. The remainder fraction of substrates after cultivation was considered to reflect the hydrocarbon inclusions in the cell mass, characteristics in Acinetobacter species, and to control the transport of substrates from crude oil phase. On the basis of the results, the isolate was considered to play an important role in the degradation study of hydrophobic environmental pollutants.
