Study on the variation of cellular physiology of Escherichia coli during high cell density cultivation using 2-dimensional gel electrophoresis

  • Physiological changes of Escherichia coli during the fed-batch fermentation process were characterized in this study. Overall cellular protein samples prepared at the different stage of fermentation were separated by 2-dimensional gel electrophoresis (2-DE), and differently expressed 15 proteins, Phosphotransferase enzyme I, GroEL, Trigger factor, ${\beta}$ subunit of ATP synthase, Transcriptional regulator KDGR, Phosphoglycerate mutase 1, Inorganic pyrophosphatase, Serine Hydroxymethyl-transferase, ${\alpha}$ subunit of RNA polymerase, Elongation factor Tu, Elongation factor Ts, Tyrosine-tRNA ligase, DnaK suppressor protein, Transcriptional elongation factor, 30S ribosomal protein S6 were identified using matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF MS). When bacterial cells grow to high cell density, and IPTG-inducible heterologous protein is produced, expression level of overall cellular proteins was decreased. According to their functions in the cell, identified proteins were classified into three groups, proteins involved in transport process, small-molecule metabolism, and synthesis and modification of macromolecules.

Temperature and Atmosphere Dependence of the Electrical Conduction of the Vacuum Evaporated Thin Metal Films on Glass Substrate (진공증착된 금속박막의 전기전도성에 대한 온도와 분위기 의존성)

  • Temperature and atmosphere dependence of electrical conduction of the metal Cu, Ag, Au films, vaccum evaporated on glass, was investigated. The structural changes of the metal films were examined by SEM and high temperature XRD. The electrical resistance slightly increased with initial temperature increase up to the inflection point and decreased to minimum value, after this rapidly increased with further temperature increased below minimum. These phenomena were caused by the thermally induced film failure as a result of the mass transport. The temperature for the film failure increased in the order of O2, Air, Vacuum, N2, Ar in Cu, Ag films and Air, Vacuum, N2, Ar in Au film. The increase of resistance at the lower temperature range was attributed to the lattice distortion by disordered crystal structure, while the decreasing resistance was attributed to the removal of structural defects and film densification.

  • Overhead cranes consist of trolley, girder, rope, objects, trolley motor, girder motor, and hoist motor. If objects are regarded as mass point, and the acceleration of hoisting motion for objects is neglected, analytical model of overhead cranes becomes a nonlinear model because the length of a rope changes. Equations of motion this model is derived of simultaneous differential equations fur motors and object. Positions of the model are controlled by optimal inputs which obtain from a nonlinear optimal control method. From the results of computer simulation, even if initial states of objects suing, it is founded that position of overhead cranes is controlled, and that swing of objects is suppressed.

  • Model for the dynamic simulation of dynamic behaviors of a solid oxide fuel cell (SOFC) is provided. This model is based upon (1) coupled mass and heat transfer characteristics and (2) important chemical reactions such as electrochemical and reforming reaction in high temperature fuel cells such as SOFC. It is found that the thermal inertia of solid materials in SOFC plays an important role to the dynamic behavior of cell temperature. Dynamic characteristics of cell voltage, power and chemical compositions with different levels of load changes are investigated.

  • Exergetic and thermoeconomic analyses were performed fer a 137-MW steam power plant. In these analyses, mass and energy conservation laws were applied to each component of the system. Quantitative balance of the exergy and exergetic cost for each component, and for the whole system was carefully considered. The exergo-economic model, which represented the productive structure of the system was used to visualize the cost formation process and the productive interaction between components. The computer program developed in this study can determine production costs of power plants, such as gas-and steam-turbines plants and gas-turbine cogeneration plants. The program can also be used to study plant characteristics, namely, thermodynamic performance and sensitivity to changes in process and/or component design variables.

  • An attempt was made to determine the effect of the volatile components of edible wild grass by cooking. We collect the volatile components of Agrimonia Japonica by dynamic\ulcorner head space method. Samples were analyzed by gas chromatography-mass spectrometry(GC-MS). Thirty-six components, including 16 hydrocarbons, 7 alcohols, 4 esters, 7 benzoid compounds, 1 aldehyde and 1 ketone were confirmed in raw samples. Also 6 hydrocarbons identified in cooked sample. Alpha-pinene and 3-he-xane-ol were regarded as the most abundant components in raw sample. By heating of Japonica, most of the volatile compounds were disappeared, but alpha-pinene was remained abundantly.

  • A vibration analysis for a rotating cantilever beam with the tapered cross section is presented in this study. The stiffness changes due to the stretching caused by centrifugal inertia forces when a tapered cantilever beam rotates about the axis perpendicular to its longitudinal axis. When the cross section of cantilever beam are assumed to decrease constantly, the mass and stiffness also change according to the variation of the thickness and width ratio of a tapered cantilever beam. Such phenomena result in variations of natural frequencies and mode shapes. Therefore it is important to the equations of motion in order to be obtained accurate predictions of these variations. The equations of motion of a rotating tapered cantilever beam are derived by using hybrid deformation variable modeling method and numerical results are obtained along with the angular velocity and the thickness and width ratio.

  • Nowadays, vehicle becoming necessity is increasing gradually and is main cause of environmental problem. The current automobile noise measurement test method such as ISO 362 which have been used many countries contribute seriously to enforce the regulation of the noise rigidly and actively study vehicle's noise, using that method. However, the amendment of ISO 362 was completed under the international agreement due to the changes in vehicle's techniques and road conditions. In this study, the difference between the current ISO 362(or ECE R51) and new ISO 362(or new ECE R51) is estimated by the test using the practical car producing domestically because the Korean manufacturer have to be interested in new test method.

The Effective Education of the Standard Pronunciations (효과적인 표준 발음 교육)

  • The purpose of this dissertation is to make the general korean speakers to learn the standard pronunciations. But it is in existence that the obstructions of the command of the standard pronunciations. They are the mistake in the education course on the korean pronunciations, the teacher's capability and the mass communications's duplicity. To overcome this obstructions, we must concentrate our efforts on the propagation of the standard pronunciations. To propagate of the standard pronunciations we can take a several method. These are the presentation of the pronunciation mistakes, audio-visual teaching, the presentation of the pronunciation principles and the use of the korean dictionary. The standard pronunciations are different from the pronunciations of the general korean speakers in many respects. So we can't make an accurate estimate of the pronunciation's changes. No one knows what will happen in the future about the korean pronunciations. But we must teach the standard pronunciations to the general korean speakers. The standard pronunciations are offically valid in the present time.

  • This paper provides the experimental verification of a non-destructive time domain approach to examine structural damage. Time histories of the vibration response of structure were used to identify the presence of damage. Damage in a structure cause changes in the physical coefficients of mass density, elastic modulus and damping coefficient. This paper examines the use of beam like structures with PVDF sensor and PZT actuator to perform identification of those physical parameters, and hence to detect the damage. Experimental results are presented from tests on cantilevered composite beams damaged at different location and with damage of different dimensions. It is demonstrated that the method can sense the presence of damage, and characterize the damage to a satisfactory precision.
