Identification of Actinomycins by High Performance Liquid Chromatography and Fast Atom Bombardment Mass Spectrometry

  • An acinomycin complex isolated from culture broth of a soil microorganism, SNUS 9305-011 has been examined by High performance liquid chromatography (HPLC). From the analysis of the fractions obtained by column chromatography of the ethyl acetate extract, three actinomycin components are confimed . The HPLC analysis is carried out with a CN-bonded nucleosil column. Comparison of the retention times of the components with those of actinomycin D, C complex, $X_{o{\beta}$, and V and suggests that they are different actinomycins. FBA mass spectra fo the coponents also shows different molecular ions from those of standards and other reported actionbmycins. The present work has demonstrated that actinomycin components can be separated by a CN-bonded HPLC column, and that ocmparison of their HPLC chormatograms with authentic smaples and information on their molecular ions can be successfully employed for indentification of actionmycins.

Model Indentification and Discrete-Time Sliding Mode Control of Electro-Hydraulic Systems (전기-유압 서보 시스템의 모델규명 및 이산시간 슬라이딩 모드 제어)

  • This paper describes the model identification and the discrete-time sliding mode control of electro-hydraulic servo systems which are composed of servo valves, double-rod cylinder and load mass. The controlled plant is identified as a 3th-order discrete-time ARMAX model obtained from the prediction error algorithm, where a nominal model and modeling errors are zuantitatively constructed. The discrete sliding mode controller for 3th-order ARMAX model is designed in discrete-time domain, where all states are observed from Kalman filter. The discrete sliding mode controller has better tracking performance than that obtained from continuous-time sliding mode controller, in experiment.

Indentification of Some Metabolites of Carbinoxamine in Rat Urine (흰쥐 뇨에서의 Carbinoxamine의 대사체 확인)

  • The metabolic profile of carbinoxamine, 2-[(4-chlorophenyl)-2-pyridinyi-methoxy] N, N-dimethylethanamine, was determined in rat urine. Urinary extracts obtained with or without enzyme hydrolysis were derivatized with MSTFA/TMSCI (N-methyl-N-trimethylsilyl trifluoroacetamide/Trimethylchlorosilane) and analyzed by GC/MSD. In rat urine, which obtained after oral treatment with carbinoxamine maleate, chlorobenzolyl pyridine, (4-chlorophenyl)-2-pyridinyl methanol : carbinol, 2-[(4-chlorophenyl)-2-pyridinylmethoxy]-N-methylethanamine : norcarbinoxamine, 2-[(4-chlorophenyl)2-pyridinylmethoxy]ethanamine : bis-norcarbinoxamine and parent carbinoxamine were detected in free form. Norcarbinoxamine and bisnorcarbinoxamine were also detected in conjugated form(acetylation). These data suggest that in the rat, hydroxylation of either the benzyl or pyridinyl ring can occur during carbinoxamine elimination. O-demethylation and subsequent conjugation represents the primary pathway of carbinoxamine elimination in the rat.

  • This paper presents the theoretical development and qualitative evaluation of a new concept in the mathematical modeling of dynamic structures. We use both test data and analytical approximations to identify the parameters of an incomplete model. The model has the capability of predicting the response of the points of interest on the structure over the frequency range of interest and can be used to predict the changes in natural frequencies and normal modes due to structural changes. The theory was tested by running simulated tests on a relatively simple structure, identifying the parameters of the incomplete model, and using this model to predict the effects on frequency and mode shapes of several mass and stiffness changes. The conditions of the tests were varied by selecting different numbers of points of measurement, varying the frequency range, and by including assumed measurement error. It is recommended that the theoretical development be continued and that applications to more complex structures be carried out in order to develop a better understanding of the limitations and capabilities of the method. A successful, more definitive sevaluation could lead to immediate practical applications.

Rapid Separation and Indentification Method of Tea Catechins (녹차 중 카테킨류의 신속 분리 및 동정법)

  • The tea tannins, epigallocatechin, epigallocatechin gallate, were successfully separated by a Sephadex LH-20 column by the acetone based gradient elution. Each fractions was collected by monitoring at 280nm. Purified fractions were directly characterized by fast atom bombardment mass spectrometry. Epigallocatechin and epigallocatechin gallate were identified and shown as low as 70% purity in the reversed phase column. This revised method is more advantageous than known methods in purity and rapidity.
