통영연안에 서식하는 보라성게(Anthocidaris crassispina)와 말똥성게(Hemicentrotus pulcherrimus)의 위 내용물과 서식지 해조군락 (Stomach Contents of the Sea Urchins, Anthocidaris crassispina and Hemicentrotus pulcherrimus and Characterization of the Marine Algal Community along the Tongyeong Coast of Korea)

  • The seaweed community and stomach contents of the dominant herbivorous sea urchins Anthocidaris crassispina and Hemicentrotus pulcherrimus were examined from November 2008 to October 2009, in coastal areas of Tongyeong, Korea. Thirty-five seaweeds including two green, nine brown, and 24 red algae, were found over the study period. Seventeen seaweeds (two green, two brown, and 13 red) and two invertebrates were found in the stomachs of A. crassispina. In H. pulcherrimus, stomach contents were two green, one brown, 11 red seaweeds, and two invertebrates. The shell diameter of A. crassispina and H. pulcherrimus ranged from 22.3 to 62.3 mm and 15.3 to 40.1 mm, respectively. Total body weights ranged from 48.5 to 86.7 g for A. crassispina and from 7.7 to 25.9 g for H. pulcherrimus. The total weight of stomach contents were 5.1 to 25.8 g in A. crassispina and 1.7 to 11.8 g in H. pulcherrimus. The range of gonad weight was 3.6 to 17.0 g in A. crassispina and 0.8 to 4.0 g in H. pulcherrimus. The gonad index (GI) for A. crassispina peaked in July and reached a minimum in December, whereas the GI for H. pulcherrimus was highest in February, and lowest in May.

볼락(Sebastes inermis)에서 분리된 비브리오속 세균의 동정 (Identification of Vibrio species isolated from dark-banded rockfish (Sebastes inermis))

  • We experienced mortality of dark-banded rockfish (Sebastes inermis) maintained in the rearing facility located in Gangneung, after transportation. Moribund fish showed various symptoms such as exophthalmia, skin ulcers, tail rots, gill rots, discoloration of liver with petechiae, yellowish fluid in intestines and ascites. Two different colonies were dominantly appeared after spreading the lesions on the agar plates and incubation. One isolate (SI_1) showed swarming movement on TSA, and formed yellow colonies on TCBS agar. The other (SI_2) showed no swarming motility and green colonies on TCBS agar. Both of them were Gram-negative. All of these results are similar with those of Genus Vibrio. They were identified as V. harveyi and V. gigantis by PCR with subsequent sequencing of 3 different genes (16 rDNA, recA, rpoA). V. harveyi is well-known as a serious pathogen of marine fish and invertebrates, while V. gigantis is known to be often isolated from marine invertebrates, but the pathogenicity is still unknown. We suspect V. harveyi as the cause of the mortality of dark-banded rockfish, but challenge experiments with these 2 Vibrio species are thought to be necessary to make a clear conclusion.

한국연안의 Vibrio vulnificus의 분포에 관한 연구 (Distribution of Vibrio vulnificus the Coast of South Korea)

  • 1986년 3월부터 10월 사이에 부산지역을 비롯하여 포항, 충무, 여수, 군산 및 시화만에서 해수 138개 해산물척추동물 102개 시료를 채식하여 시료별, 지역별로 V. vulnificus 분포실태를 조사한 결과는 다음과 같다. 1. 해수 138개 시료 중 31개에서 V. vulnificus가 검출되어 약 $22.5\%$의 검출율을 나타내었으며 외항수보다 내항수에서 검출율이 높았다. 2. 해산물척추동물 102개 시료중 16개중 V. vulnificus가 검출되어 $15.7\%$의 검출율을 나타내었으며 게와 피조개에서 그 검출율이 높았다. 3. 시기별로 수온이 높은 8월에서 10월 사이에 많이 검출되었으며 그 중 9기이 $40\%$의 가장 높은 검출율을 보였다. 4. 지역별로는 염분농도가 낮은 서해안의 군산이 검출율이 가장 높았으며 남해안에서는 부산지역의 광안리와 해운대에서 검출율이 높았다.

안정동위원소 비를 이용한 하구 갯벌에 있어서 저서 무척추 동물의 유기물 기원의 공간적 특성 (Spatial Characteristic in Food Sources for Benthic Invertebrates in an Estuary Tidal flat: Carbon and Nitrogen Stable Isotope Analyses)

  • 일본 나나키다 하구 갯벌에 있어서 탄소 질소 안정동위원소를 이용해 먹이 연쇄의 공간적 가변성에 대해 조사했다. 서로 다른 특징을 갖고 있는 갯벌에서 잠재적인 유기물(육상식물, 해양 입자성 유기물, 저서 부착 미세조류 및 하구 입자성 유기물), 퇴적유기물 및 저서 무척추 동물(Nuttallia olivacea and Nereidae)에 대한 샘플링을 행했다. 본 연구의 목적은 좁은 공간적 가변성에 따른 Nuttallia olivacea와 Nereidae의 먹이원을 결정하는 것이다. 육상식물(${\delta}^{13}C=-26.6{\pm}0.76$, ${\delta}^{15}N=2.7{\pm}0.31$) 과 하구 입자성 유기물(${\delta}^{13}C=-25.5{\pm}0.13$, ${\delta}^{15}N=5.2{\pm}0.46$)은 저서 부착 미소조류${\delta}^{13}C=-16.3$, ${\delta}^{15}N=6.2$)와 해양 입자성 유기물(${\delta}^{13}C=-19.6{\pm}0.08$, ${\delta}^{15}N=8.9{\pm}1.70$)의 탄소 질소 안정동위원소비 보다 낮았다. 퇴적물의 탄소 안정동위원소 비는 -27.4~-22.8‰ 나타냈으며, 하구에서 하천 방향으로 갈수록 낮은 탄소 안정동위 원소비를 나타냈다. 저서 무척추 동물의 탄소 질소 안정동위원소비는 각각 -22.8~-18.4‰, 8.1~11.9‰ 범위를 나타냈다. 이러한 결과와 더불어 혼합 모델을 이용해 저서 무척추 동물의 먹이원의 기여율을 추정한 결과 해양 입자성유기물과 저서 부착 미세조류의 기여율은 높았지만, 육상식물과 하구 입자성 유기물의 기여율은 비교적 낮았다. 이러한 저서 무척추 동물의 먹이 기여는 각 장소마다 계절 및 물리적 환경 요소의 영향을 받는다고 사료된다.

35종 해산 무척추동물의 일반성분조성과 스테롤함량 (Proximate Composition and Sterol Content of 35 Species of Marine Invertebrates)

  • 통영 연안해역에서 채취된 35종의 해산무척추동물의 일반성분 조성과 sterol 및 $\alpha$-토코페롤함량을 정량하여 종별로 비교하였다. 단백질함량은 복족류 ($17.2\pm4.30\%$ ), 갑각류 ($16.8\pm4.06\%$), 두족류 ($15.3\pm3.24\%$) 등 초식성 및 육식성의 무척추동물에 비교적 많았고, 2매패류 ($11.8\pm2.49\%$), 멍게류 ($8.20\pm1.00\%$) 등 plank-ton 식성의 경우에 적었으며, 특히 해삼류 ($3.94\pm1.13\%$)와 같은 니식자 (mud swallow)에서는 아주 적었다. 총지질함량은 $0.24\%(청삼)\~1.96\%(멍게)$의 범위로 비교적 적었다. 탄수화물함량은 멍게류 ($6.60\pm4.53\%$ ), 2매패류 ($3.15\pm1.82\%$), 복족류 ($3.02\pm1.61\%$)에서 비교적 많았고, 두족류 ($1.00\pm0.63\%$)와 갑각류 ($0.52\pm0.57\%$)에서는 아주 적어 보통 어류의 경우와 유사하였다. 또한 수분함량은 해삼류 ($92.0\pm1.94\%$)에서 가장 많았고, 복족류 ($77.0\pm1.95\%$)에서 가장 적었으며, 나머지는 $80\~82\%$로서 대체로 유사하였다. 수분함량과 단백질함량 및 지질함량, 그리고 단백질함량과 지질함량 사이에는 각각 y= -0.8716x+184.452 (r=-0.87, p<0.001), $y=128.52e^{-0.0601x}$ (r=-0.55, P<0.001), $y=6.3047e^{-0.0904x}$(r=-0.42, P<0.02)으로서 모두 역의 상관관계를 나타내었다. 한편 sterol 함량은 (청삼 56mg/100g edible portion)에서 가장 적었고, 새조개 (216mg/100g)에서 가장 많았다. 동물그룹별로 보면 두족류 (148 $\pm$30.0mg) > 2매패 (121$\pm$35.0mg) > 복족류 (118$\pm$20.0mg) > 갑각류 (116$\pm$31.0mg) > 멍게류 (78.0$\pm$2.00 mg)> 해삼류 (62.0$\pm$8.00 mg)의 순으로 sterol 함량이 많았다. $\alpha$-토코페롤함량은 갑각류에서 1$\~$2 mg/100g를 함유하였으나, 나머지 무척추동물에서는 흔적 또는 1 mg/100g 이하였다.

뉴칼레도니아의 해산 무척추동물에 공생하는 요각류(갑각강) (Copepods (Crustacea) Associated with Marine Invertebrates from New Caledonia)

  • 남태평양에 있는 뉴칼레도니아의 무척추동물에 공생하는 요각류를 기록하였다. 3신속 (Ruhtra, Pterioidicola, Pachysericola), 30신종이 포함되어 있으며, 이들은 천장입요각목 및 대롱입요각목 내의 다음과 같은 속에 해당된다: Anchimolgus속 (5신종), Jamescookina속 (2신종), Odontomologus속 (2신종), Schedomolgus속 (3신종), Scyphuliger속 (5신종), Critomolgus속 (3신종), Doridicola속 (1신종), Paramolgus속 (2신종), 신속 Pterioidicola (1신종), 과의 위치가 불분명한 신속 Ruhtra (1신종), Asteropontius속 (5신종). 뉴칼레도니아의 미기록종과 새로운 숙주에 대해서도 기록하였다. 지금까지 기록된 뉴칼레도니아산 공생성 요각류 251종과 이들의 무척추동물 숙주 163종의 목록도 제시하였다.

Mannan-binding lectin of the sea cucumbers Stichopus japonicus has common antigenic determinants with human serum mannan-binding lectin

  • The host defense system or immune system of all modern animals has their roots in very ancient organisms. After analyzing literature data concerning properties of invertebrates and vertebrates lectins we suggest that mechanism of mannans recognition may exist in marine invertebrates, as a universal mechanism for homeostasis maintenance and host defense, and mannan-binding lectins family of vertebrates has ancient precursor, as was shown for another S-type lectins family. We carried out the screening of mannan-binding type lectin among different species of echinoderms inhabiting in Piter the Grate Bay, the sea of Japan. As a result, the C-type lectins (SJL-32) specific for high mannose glycans was isolated from the coelomic plasma of the sea cucumbers Stichopus japonicus by ion-exchange chromatography on a DEAE-Toyopearl 650M, affinity chromatography on a mannan-Sepharose 6B and gel filtration on a Sephacryl S-200. SJL-32 is homodimer with molecular mass about 32 kDa on SDS-PAGE under non-reducing conditions. Protein part of the lectin has high conteins Asn, Glu, Ser. Hemagglutination of trypsin-treated O blood group human erythrocytes by SJL-32 was competitively inhibited by high-branched -D-mannan composed of -1,2 and -1,6 linked D-mannopyranose residues. In contrast, a variety of mono-, oligo-, and polysaccharides composed of residues of galactose and fucose showed absence or little inhibitory activities. The lectin activity strong depends on Ca2+ concentration, temperature and pH. Monospecific polyclonal antibodies were obtained to the lectin. As was shown by ELISA assay, antibodies to SJL-32 cross-reacted with human serum mannan-binding lectin. This data allows making conclusion about common antigenic determinants and structural homology of both lectins. In our opinion, SJL-32 belongs to evolutionary high conservative mannan-binding lectins (MBLs) family and takes part in the host defense against pathogenic microorganisms.

  • This is the third series of catalogs reporting on Korean species discovered through the 'Discovery of Korean Indigenous Species'. This catalog includes 22 species of invertebrates, excluding insects. The catalog includes the scientific name, an abridged list of synonyms, collection sites, distribution, diagnosis, and figures for each species. Additionally, we provide the newly assigned Korean name, specimen voucher, and, if available, mitochondrial CO1 or 16S gene sequences of the species listed. All species identified and documented here will be officially listed on the 'National Species List of Korea', a database maintained by the National Institute of Biological Resources(NIBR).

  • The oceans cover about $71\%$ of the Earth's crust and contain nearly 300,000 described species. Free-living bacteria in the sea and symbiotic bacteria of marine invertebrates are proving to be valuable sources of useful bioactive compounds. Marine sponges, in particular, which contain diverse communities of bacteria, produce many classes of compounds that are unique to the marine environment. Uncultured microorganisms are commonly believed to represent $99.9\%$ of the whole microbial community. They have been investigated for the possibility of isolating and over-expressing genes in viable microorganisms. Strict symbiotic species that have been adapted to the host are candidate unculturable species. With the enormous potential for discovery, development, and market value of marine derived compounds, supply of the products is a major limiting factor for further development.

  • Many people living in Asia countries, particularly Korea, Japan and China, have consumed very widely fresh seafood products, such as shrimps, oysters, mussels and other marine invertebrates and fishes, without any heating or cooking. A variety of Vibrio spp., including V. parahaemolyticus, V. cholera. V. vulnificus, and V. fluvialis, lives in these seafoods and cause great problems associated with human disease. A strong antimicrobial agent to effectively inhibit the growth of these pathogenic bacteria in in vivo or in vitro is urgently need for preventing fish and human diseases. (omitted)
