Differentially Expressed Genes in Marine Medaka Fish (Oryzias javanicus) Exposed to Cadmium

  • To screen the differentially expressed genes in cadmuim-exposed marine medaka fish (Oryzias javanicus), a candidate marine test fish for ecological toxicity, the differential display polymerase chain reaction (DD-PCR) was carried out, since the genome-wide gene expression data are not available in this fish species yet. A total of 35 clones were isolated from cadmium-exposed fish and their nucleotide sequences were analyzed. The differentially expressed gene candidates were categorized to response to stimulus (3); ion binding (3); DNA binding (1); protein binding (6); carbohydrate binding (1); metabolic process (4); biological regulation (3); cellular process (2); protein synthesis (2); catalytic activity (2); sense of sight (1); immune (1); neurohormone (1); signaling activity (1); electron carrier activity (1) and others (3). For real-time quantitative RT-PCR, we selected catalase, glucose-6-phosphate dehydrogenase, heat shock protein 70, and metallothionein and confirmed that cadmium exposure enhanced induction of these four genes.

Accumulation of Butyltin Compounds in Shellfish and Fish from Korean Coastal Areas

  • Butyltins (BTs) were measured in 2 species of shellfish and 16 species of fish collected along the Korean coast. Tributyltin (TBT) was detected in the mussels and oysters of sentinel organisms (20 to 940 ng Sn/g dry wt), indicating widespread contamination of TBT in the Korean coast. The elevated concentrations of TBT in the shellfish found in the sites near harbors or shipyards suggested that antifouling paints are probable major sources of butyltins in these areas. The TBT compound was detected in 12 out of 16 fish samples. The concentrations of TBT in fish muscles wee between 7 and 151 ng Sn/g dry wt, while the level in whole body of anchovy was very high (793 ng Sn/g dry wt). Exposure doses to Korean people via consumption of these marine products wee evaluated. The results suggested that the environmental levels of TBT were below the level of concern.

Bioaccumulation of Pb and Cd in Blue Mussel (Mytilus edulis) and Oliver Flounder (Paralichthys olivaceus) Exposed to Rearing Media (노출실험을 통한 진주담치(Mytilus edulis) 넙치(Paralichthys olivaceus)의 Pb 및 Cd 축적에 관한 연구)

  • The bioaccumulation of Pb and Cd dissolved in seawater was assessed measuring the concentrations recorded within blue mussel (Mytilus edulis) and oliver flounder (Paralichthys olivaceus) after two weeks exposure period. The Pb and Cd concentration within the whole body of two testing organisms increased according to the exposure concentrations, and the such tendency was clear specially from the mussel. Maximum metal concentration reached $5,260({\pm}70)\;{\mu}g/g$ for Pb reared under 5.0 mg/L Pb, $1,040({\pm}40)\;{\mu}g/g$ for Cd reared under 1.0 mg/L Cd in the mussel, and indicated that the bioaccumulation of Pb and Cd was directly related to the rearing medium concentrations. Bioconcentration factors (BCF) reached very high values for Pb (maximum value: $12,100{\pm}1,400$) in the mussel reared under lowest Pb concentration (0.01 mg/L). The BCF value for Cd in the mussel were also far higher at exposure to low Cd concentration than high Cd concentration. At higher external concentrations, the BCF for Cd and Pb declined. This demonstrated the ability of two testing organisms to rapidly uptake heavy metals particularly when exposed to low external concentration. The mean Pb concentration was slightly higher in the gill of mussel than in the digestive gland, while Cd showed a higher level in the digestive gland than in the gill.

Corrosion evaluation of a newly developed high-strength steel in marine environments

  • This study aims to evaluate the corrosion behavior of a newly developed high-strength steel in marine environments. Metals used in seawater are easily deteriorated because of the presence of corrosive species such as chloride ions in it. Seawater causes much higher corrosion than fresh water. Thus, the corrosion of steel in marine environment has been recognized as a crucial problem in designing structures which cannot be cathodically protected. In this study, the corrosion resistance of a newly developed high-strength steel was evaluated. Four different specimens were tested to confirm the corrosion resistance. The exposure corrosion test was carried out by exposing the specimens to different marine environments such as atmospheric, tidal, splash, and submerged zones for two years. The specimens taken out from each location were cleaned ultrasonically and chemically prior to the evaluation of their corrosion resistance by the weight loss method. Finally, the pitting depth of the specimens was also measured to evaluate their pitting corrosion. The conditions used for the corrosion test were similar to the environmental conditions. The corrosion test results revealed that the corrosion rate and pitting corrosion of the newly developed high-strength steel was lower than that of the other carbon steels.

  • The noise levels on board ship recognized at Europe in the early 1970s and the noise regulations on the ship began to put in a statutory form. After that, in 1982 'International Code on Noise Levels on Board Ships' adopted by IMO and it became a standards to the newly built ship and it remains up to recently. Especially, the ship engine room, which have huge main engine and various kinds of subsidiary machines, is under an extremely loud condition and so the worker who works in it is easy to lose his hearing. Recently, each nation regulates the allowable noise exposure time by law to protect the industrial employee from the occupational hardness of hearing. In our country, the allowable noise exposure time is regulated by the labor. standard law but the international provisions regulated by IMO have been applied in case of the ship engine room. In this paper, the cabin's noise levels of cargo-passenger ships plies south-west coast line were investigated.

  • Seawater batteries are next-generation secondary batteries that use seawater as a cathode. They utilize marine resources to provide competitive prices, high eco-friendliness, and a structure suitable for marine applications. Based on these advantages, pouch types and prismatic types have been studied and developed assuming natural seawater exposure. However, because of the electrical characteristics of the secondary battery, its capacity and internal resistance vary depending on the use environment. These characteristics are not only utilized for predicting the life of a battery but also have a direct effect on the capacity and power suitable for a specific situation. Therefore, the internal resistance was analyzed in this study by measuring the capacity depending on the seawater battery use environment and the state-of-charge-open-circuit-voltage measurement method.

Effects of periodic air-exposure and nutrients on the competition of Ascophyllum nodosum and Fucus vesiculosus germlings

  • To examine the relationship between competition and environmental factors, the germlings of Ascophyllum nodosum (L) Le Jolis and Fucus vesiculosus L. were cultured in monocultures and mixtures of the two species under two different exposure and nutrient levels. Both intra- and inter-specific competition were examined in comparison of the mortality and growth of germlings in monocultures and mixtures of the two species. The mortality of germlings increased with increasing density and emergence periods both in the monoculture and mixtures of the two species, and the mortality of Ascophyllum was significantly higher than that of Fucus both in submerged and emerged treatments. The growth of germlings of both species reduced with increasing density but F. vesiculosus always grew faster than Ascophyllum. The values of log output ratio were more than 0.1, indicating that Fucus 'won' in the competitive battles with Ascophyllum under two nutrient- and air-exposure levels. Log output ratio was greater in high than in low nutrients, indicating that the growth of Fucus is more enhanced than that of Ascophyllum in high nutrients. In the present study, the outcome of inter specific competition between germlings of Fucus vesiculosus and Ascophyllum nodosum was slightly altered by duration of emergence and nutrient concentration, but not to such an extent as to change the outcome.

Seasonal Changes in the Carbon and Nitrogen Contents of Zostera marina Populations in the Intertidal and Subtidal Zones of the Seomjin Estuary, Korea (섬진강 하구의 조간대와 조하대에 자생하는 거머리말(Zostera marina)의 탄소와 질소 성분의 계절변화)

  • To study the difference in growth characteristics between intertidal and subtidal eelgrass Zostera marina populations, environmental factors and changes in tissue carbon (C) and nitrogen (N) contents in eelgrass from the lower intertidal and upper subtidal zones were surveyed monthly in the Seomjin Estuary from May 2003 to December 2004. Water temperatures, water column nutrient concentrations, sediment pore water, and ammonium and phosphate concentrations showed no significant differences between intertidal and subtidal zones. Sediment pore water nitrate+nitrite concentrations were slightly higher in the intertidal zone than in the subtidal zone. Average monthly exposure times in the intertidal and subtidal zones were $42.4{\pm}5.8h\;month^{-1}$ and $15.3{\pm}3.5h\;month^{-1}$, respectively. Eelgrass leaf N content decreased from March to August and increased from September to February in the subtidal zone, whereas these trends were not observed in the intertidal zone. Eelgrass rhizome C content increased from April to September and decreased from October to March in the subtidal zone, whereas those tendencies were not recorded in the intertidal zone. Eelgrass rhizome C:N ratios showed a strong reverse trend to leaf C:N ratios in the subtidal zone, but these tendencies were not observed in the intertidal zone. The effects of exposure appeared to cause no seasonal trend in eelgrass tissue C and N content or C:N ratios in the intertidal zone in the Seomjin Estuary.

Optimal Coating Materials and Methods to Enhance Interfacial Bonding with Steel Pipes in Marine Environments (해양 환경 노출 코팅 강관의 부착성능 확보를 위한 최적 코팅 및 공법연구)

  • This study reviews previous research on various coating materials and methods designed to improve the adhesion and durability of steel pipes exposed to real-world marine environments, with the goal of identifying optimal solutions. The results concerning the adhesion and corrosion resistance of the pipes were categorized and analyzed based on exposure tests in extreme marine conditions to evaluate their stability. Furthermore, a detailed microstructural analysis of the coatings applied to the pipes was performed to assess surface damage, including peeling and delamination. The findings confirmed that coatings effectively prevent corrosion and provide sufficient adhesion and durability. Based on these results, we have proposed suitable coating materials, types, and application methods for steel pipes used in marine environments and offered recommendations for future exposure tests.

Persistence of Marine Vibrio vulnificus in Oysters within Environmental Parameters (환경 변수에 대한 생굴에서의 해양 Vibrio vulnificus의 인내성)

  • This project studies marine Vibrio vulnificus in oysters in the marine environment and attempts to correlate this bacteria's presence within various environmental parameters; we design this study to determine how different storage temperatures affect the survival of V. vuinficus in oysters and whether V. vulnificus is able to persist in oysters after exposure to UV light-disinfected seawater. Experimental depuration systems consist of aquaria containing temperature-controlled seawater treated with UV light and 0.2 ㎛ pore size filtration. Results showed that depuration at temperatures higher than 25℃ caused V. vuinificus counts to increase in oysters. Throughout the process, depuration water contained high concentrations of U vuinificus indicating"that the disinfection properties of UV radiation and 0.2 ㎛ pore size filtration were less than 어e release of V. vuinificus into seawater, In contrast, when depuration seawater was maintained at 10℃, the numbers of V. vuinificus were very little and multiplication in oysters was inhibited.
