해양생물의 특징을 활용한 일러스트레이션 디자인 연구 -해양생물 색채와 형태를 중심으로- (A study on Illustration Design using the characteristics of Marine Life -Centered on the colors and forms of Marine life-)

  • 현대의 일러스트레이션 디자인은 독창성을 필요로 하는데, 해양생물의 형태와 색채는 일러스트레이션 창작에 풍부한 소재를 제공한다. 일러스트레이션은 디자인적 창조 가치가 매우 높다. 본 연구는 해양생물을 거울삼아 색채학적 지식과 시각적 형태설계를 통해 이를 밝히고, 일러스트레이션 디자인과 해양생물을 주요 연구대상으로 하였다. 본 연구목적은 ① 해양생물의 전형적 특징을 색과 형태 중심으로 정리, ② 이를 해양생물을 대표하는 그래픽 기호로 도출하여, ③ 일러스트레이션으로 접목할 수 있음을 해양생물 일러스트레이션 제작으로 사례를 제시하였다. 연구를 통해서 결론을 도출하고, 해양생물과 육지생물을 구분하는 전형적인 특징 중 하나로 곡선적인 특징을 부각시켰다. 열대어 색채 현상에 대한 분류 연구와 비교분석을 통해 도출한 열대어류의 색채 특성을 바탕으로 해수 열대어의 색채를 추출 및 가공하여 일러스트레이션 디자인에 활용할 수 있다.

Isolation and Identification of Cytotoxic and Biological Active Toxin from the Puffer Fish Arothron stellatus

  • This study is to investigate the biological, biochemical and cytotoxic effects of puffer fish (Arothron stellatus) toxin extracts under in-vitro condition. Extracted toxins from various organs of puffer fish were purified by using active charcoal column, and Bio-gel-P2 column chromatography. The lethality of toxin was tested in crabs, which consists of neurotoxic compounds. The degree of the brine shrimp lethality assay was found directly proportional to the concentration of the toxin extracts, which was well supported by hemolytic assay. The experimental results suggested that the gonad was found higher toxins than the liver and muscles. The mortality rate of brine shrimp nauplii was increased with the raise of concentrations of toxin level. Among the different doses and time dependent cytotoxic effect of human cervical carcinoma (HeLa) cells were showed $4.0{\mu}g/mL$ of toxin, which was effectively inhibited cancer cell proliferation. HPLC and TLC analysis was revealed that the A. stellatus toxin contains tetrodotoxin (TTX), related compounds 4-epi TTX and anhydro-TTX. The present results suggested that the A. stellatus contain TTX as a major and anh-TTX as a minor toxin. It could be the potential candidate in the field of anticancer drug discovery against human cervical cancer cells. The present data is confirming that the puffer fish toxin as an interesting source of novel bioactive natural compounds with potent applications in pharmacology.

Starvation-induced Physiological Responses and RNA/DNA Ratios in Rock Bream, Oplegnathus fasciatus, and Olive Flounder, Paralichthys olivaceus

  • In a 12-week experiment, the rock bream, Oplegnathus fasciatus, and olive flounder, Paralichthys olivaceus, were investigated to determine the effects of starvation on their physiological parameters. The protein and DNA contents of the starved fish were significantly higher than the initial values and those of the fed fish. The RNA contents and RNA/DNA ratios of the fed fish were significantly higher than those of the other groups (P<0.05). The hematocrit, hemoglobin, red blood cells (RBC), and mean corpuscular volume (MCV) of the fed rock bream were significantly higher than at baseline (P<0.05), whereas the mean corpuscular hemoglobin concentration (MCHC) of the fed fish was lower than at baseline (P<0.05). The hematocrit, hemoglobin, RBC, and MCHC of the starved group were significantly lower than the baseline values, whereas the MCV of the starved group was significantly higher than the baseline value (P<0.05). No significant difference in alanine aminotransferase was observed between the fed fish and baseline, whereas the starved fish value was significantly higher than the baseline value (P>0.05). There were no significant differences in cortisol levels. However, the glucose level in the fed group was significantly higher than the baseline level and that in the starved group was significantly lower than the baseline level (P<0.05).

넙치 배합사료에 고추냉이 추출물, 키토산 및 김 분말 첨가 효과 (Effects of Supplemental Dietary Wasabi Extract, Chitosan and Pophyra on Growth and Body Composition of Juvenile Flounder, Paralichthys olivaceus)

  • Two feeding trials were conducted to investigate the effects of several dietary additives on growth and feed utilization of juvenile flounder. In the first experiment, three replicate groups of juveniles (average weight 1.5 g) were fed diets with or without wasabi extract and chitosan for 7 weeks. Survival rate, weight gain, protein efficiency ratio, hepatosomatic index and condition factor were not affected by the different dietary additives (P>0.05). Feed efficiency of fish fed the wasabi extract diet was significantly higher than that of fish fed the control diet (P<0.05). Daily feed intake of fish fed the wasabi extract diet was significantly lower than that of fish fed the other diets (P<0.05). In the second experiment, three replicate groups of juveniles (average weight 1.4 g) were fed diets with or without Pophyra powder for 7 weeks. Survival rate and weight gain were not significantly affected by dietary Pophyra powder (P>0.05). Feed efficiency and protein efficiency ratio of fish fed the Pophyra diet were significantly lower than those of fish fed the control diet (P<0.05). The results of these experiments suggest that feed efficiency of juvenile flounder may be improved by dietary supplementation with wasabi extract.

Performance of a Recirculating Aquarium System for the Culture and Holding of Marine Fish

  • To supply fresh and quality quarantined seafood in live seafood specialty restaurants, facilities for short-term culture or holding of live marine fish and shellfish are a necessity. In this study, the performance of a simple recirculating aquarium system for the culture and holding of marine fish was evaluated. The aquarium system consisted of a culture tank, a foam fractionator for solids removal, and a Styrofoam bead filter for nitrification and solids trapping. In the first trial, the aquarium was stocked with a total of 12 kg Korean rockfish, which were fed approximately $0.5\%$ of the total fish body weight daily. During the 2-month culture period, total ammonia nitrogen (TAN) and nitrite nitrogen $(NO_2-N)$ concentrations remained below 1mg/L and 2mg/L, respectively. The chemical oxygen demand (COD) fluctuated between 13.6 and 31.2 mg/L on selected sampling days. The total suspended solids (TSS) removed by the foam fractionator was between 2.7 and 4.6g daily. The Styrofoam bead filter not only reduced TAN and $NO_2-N$ in the culture tank water, but also trapped solids equivalent to 8.3-26.7\% of the weight of feed supplied. In Trial 2, 30kg of live fish were held in the aquarium without feeding for a 24-hour period and the water quality parameters were monitored. TAN and $NO_2-N$ concentrations first increased and then decreased to around 0.3mg/L. These results demonstrate that the recirculating aquarium system is a functional option for the short-term culture or holding of marine fish.

사료 내 면실박 및 대두박 첨가가 넙치의 번식 조직학에 미치는 영향 (Effects of Dietary Supplementation of Cottonseed and Soybean meal on Reproductive Histology of Olive Flounder, Paralichthys olivaceus)

  • The gossypol existed in cottonseed meal is a well known antispermatogenic agent which can impair reproductive performances of male fish as well as mammals. Two feeding experiments were conducted to examine a toxic effect of dietary supplementation of cottonseed meal on reproduction in juvenile olive flounder (the first experiment) for 19 weeks and growing olive flounder (the second experiment) for 26 weeks. After each feeding study, females and males were sampled for histological examination in gonads and liver to verify any negative effects by the dietary supplementation of cottonseed and soybean meal on reproduction. After two feeding trial, the gonad somatic index (GSI) of male and female (from the first feeding trial) were not significantly different among all the dietary treatments. The GSI values of female (from the second feeding trial) were not significantly different among all the dietary treatments. However, males fed cottonseed and soybean meal containing diets exhibited significantly lower GSI than that fed the control diet after the second feeding trial. Histological examination of gonads and liver of fish fed cottonseed and soybean meal did not show any negative effects compared to those of fish ffd the control diet. Hepatosomatic index of fish in the first and second feeding trials were not significantly different among all the dietary treatments. The findings in this study suggest that dietary supplementation of cottonseed and soybean meal up to 40% fish meal replacement might not deteriorate the gametogenesis of juvenile and growing olive flounder. However, the supplementation in diets over 30% fish meal replacement might reduce GSI of male in growing olive flounder.

  • The effects of dietary antioxidant [saltwort (SW), leek (LK), and dandelion (DD)] supplementation on the growth, body composition, serum chemistry, and challenge test results of juvenile rockfish, Sebastes schlegelii, were determined. In an experiment, 320 fish were randomly distributed into eight, 50-L flow-through tanks (40 fish per tank). Four experimental diets were prepared: the control diet (Con) with synthetic antioxidant: and diets with SW, LK, or DD from natural sources. Each diet was randomly assigned to duplicate tanks of fish, which were hand-fed to satiation twice daily for 8 weeks. After the 8-week feeding trial, 20 fish from each tank were artificially infected with Streptococcus iniae and monitored for 6 days. The dietary additives did not affect survival, weight gain, specific growth rate, feed efficiency (FE), protein efficiency ratio (PER), or plasma chemistry. However, the cumulative mortality of fish fed the SW, LK and DD diets was lower than that of fish fed the control diet beginning 4 days after infection. In conclusion, dietary supplementation with SW, LK, and DD did not affect the growth, FE, PER, or plasma chemistry of rockfish. However, dietary inclusion of SW, LK, and DD lowered the mortality of rockfish following infection with S. iniae.

남해안에 출현하는 눈볼대 (Doederleinia berycoides)의 식성 (Feeding Habits of the Blackthroat Seaperch Doederleinia berycoides in the Southern Sea of Korea)

  • The feeding habits of the blackthroat seaperch Doederleinia berycoides were studied using 650 specimens collected in the Southern Sea of Korea, from January to December, 2005. The size (standard length, SL) of the specimens ranged from 6.3 to 35.2 cm. D. berycoides is a piscivore that consumes mainly teleosts such as Engraulis japonicus, Acropoma japonicum, and Myctophum nitidulum. Of the fish species Engraulis japonicus was the preferred prey. D. berycoides also eats small quantities of shrimps, amphipods, euphausiids, mysids, and crabs. D. berycoides showed ontogenetic changes in its feeding habits. Smaller individuals (5-10 cm SL) fed mainly on shrimp. The proportion of shrimp decreased with increasing fish size, and this decrease paralleled the increased consumption of fish. Fish accounted for almost all of the stomach contents of individuals larger than 15 cm SL. Cluster analysis based on the percentage dry weight (%DW) identified two size groups: group A (<15 cm SL) consumed primarily shrimp and fish; and group B (15< cm SL) preyed exclusively on fish. There was little seasonal change in the diet of D. berycoides, but shrimp constituted 41.8% of the summer diet in %W.

  • This study was conducted to examine a mixture of plant protein sources as a fish meal (FM) substitute. Two feeding trials were carried out using similar dietary formulations but different FM levels. In Experiments 1 and 2, the basal diets were formulated to contain 65% and 60% of FM, respectively. The other five diets were formulated replacing FM by 10, 15, 20, 25 and 30% with a mixture of soybean meal, wheat gluten and soy protein concentrate. Three synthetic amino acids (lysine, threonine and methionine) were added to the test diet. Groups of fish in experiment 1 ($6.76{\pm}0.03g$) and experiment 2 ($32.5{\pm}0.1g$) were fed one of the experimental diets for 7 and 9 weeks, respectively. Each experiment was carried out in triplicate. There were no significant differences among groups in terms of growth performance, feed utilization, survival or hematological parameters in either experiment. The results indicated that a mixture of soybean meal, wheat gluten and soy protein concentrate, supplemented with three synthetic amino acid, can replace fish meal by up to 30% in diets for juvenile olive flounder.

  • This study was conducted to investigate the effect of extruded pellet (EP) size on the growth performance of olive flounder Paralichthys olivaceus at the juvenile (Exp-I; $62.2{\pm}0.05$), growing (Exp-II; $150{\pm}1$) and sub-adult (Exp-III; $299{\pm}1$) stages. In Exp-I (3, 4, 5 and 6 mm diets), Exp-II (4, 9, 11 and 13 mm diets) and Exp-III (9, 11, 13 and 16 mm diets), triplicate groups of fish were fed four EP diets of different pellet sizes. In Exp-I, larger pellet sizes were significantly better than the smallest pellet size (3 mm). In Exp-II, significantly improved feed and protein efficiency were observed with increasing pellet size. The feed conversion ratios of the fish fed the 4 and 9 mm diets were significantly lower than those of fish fed 11 and 13 mm diets. The dietary digestibility of protein and energy was higher in smaller EPs than in larger sizes. In Exp-III, the growth performance of fish was not affected by the pellet size. However, feed intake was significantly affected, increasing with increasing pellet size. Therefore, the optimum pellet sizes for olive flounder under the conditions in this study were 5 mm for 60-150 g fish and 9 mm for 150-500 g fish.
