Dispersion, Speciation and Adsorption Treatment of Heavy Metals in the Vicinity of the Shi-Heung Cu-Pb-Zn Mine (시흥 Cu-Pb-Zn 광산 주변에서의 중금속원소들의 분산 및 존재형태와 흡착처리)

  • In order to investigate the dispersion patterns and speciations of Cu, Pb, Zn and Cd in soils, stream sediments and stream waters, geochemical studies of soil, stream sediment and stream water samples collected in the vicinity of the Shi-Heung Cu-Pb-Zn mine was carried out Cation exchange capacity measurement, size analysis, X-ray diffraction analysis and batch test were performed to select applicable soil for adsorption treatment The average content of Cu, Pb, Zn and Cd in soils collected from tailings and ore dressing plant is 1084 ppm, 2292 ppm, 3512 ppm and 29.2 ppm, respectively, and therefore, tailings and ore dressing plant site may be the major contamination sources in this study area. The mean content of Cu, Pb, Zn and Cd in stream sediments is extremely high up to 794 ppm, 1633 ppm, 2946 ppm and 25.2 ppm, respectively. Tailing particles and heavy metal ions are dispersed along the tributary system. Results from the sequential extraction analysis indicate; (1) most of Cu is bound to organic matters and sulphides, (2) fraction of Pb is mainly bound to Fe and Mn oxides. Most of Zn is largely bound to Fe and Mn oxides and residual fraction. Ion exchangeable fraction of Cd is relatively higher than those of Cu, Pb and Zn. Batch test on soils collected from the kaolinite and/or pyrophyllite mines and from the control areas was carried out to select an applicable soil samples for adsorption treatment The sample, S10, collected from the control area 2 (clay content 33.2%) shows the highest $K_d$ (distribution coefficient). Organic content in soils and several clay minerals shows relatively good correlation with $K_d$. It means that applicable soils for adsorption treatment of heavy metals show high organic and clay content.

Mechanistic insight into the progressive retinal atrophy disease in dogs via pathway-based genome-wide association analysis

  • The retinal degenerative disease, progressive retinal atrophy (PRA) is a major reason of vision impairment in canine population. Canine PRA signifies an inherently dissimilar category of retinal dystrophies which has solid resemblances to human retinis pigmentosa. Even though much is known about the biology of PRA, the knowledge about the intricate connection among genetic loci, genes and pathways associated to this disease in dogs are still remain unknown. Therefore, we have performed a genome wide association study (GWAS) to identify susceptibility single nucleotide polymorphisms (SNPs) of PRA. The GWAS was performed using a case-control based association analysis method on PRA dataset of 129 dogs and 135,553 markers. Further, the gene-set and pathway analysis were conducted in this study. A total of 1,114 markers associations with PRA trait at p < 0.01 were extracted and mapped to 640 unique genes, and then selected significant (p < 0.05) enriched 35 gene ontology (GO) terms and 5 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways contain these genes. In particular, apoptosis process, homophilic cell adhesion, calcium ion binding, and endoplasmic reticulum GO terms as well as pathways related to focal adhesion, cyclic guanosine monophosphate)-protein kinase G signaling, and axon guidance were more likely associated to the PRA disease in dogs. These data could provide new insight for further research on identification of potential genes and causative pathways for PRA in dogs.

Analysis of Off-flavor Generated from a Polyethylene Terephthalate Water Bottles and Caps by Using an Electronic Nose (MS-전자코를 사용한 페트(polyethylene terephthalate) 생수병과 뚜껑의 이취 이행 분석)

  • The objective of the study was to investigate the off-flavor generated from PET water bottles and their caps by using a mass spectrometry-based electronic nose. The ion fragment data obtained from the electronic nose were used for discriminant function analysis (DFA). In the case of increased concentrations of the contamination of water, the off-flavor pattern depended on the discriminant function second score instead of the discriminant function first score. To identify the cause of off-flavor in polyethylene terephthalate (PET) bottled water, the PET bottle and its cap were analyzed by DFA. The results showed that the cap generated more volatile compounds than the bottle or mineral water did. The substances causing the off-flavor were predicted to be 2,4-di-tert-butylphenol (2,4-DTBP), nonanal, and decanal when the main peak of the mass spectrum was compared with the major ion fragments of the electronic nose. Thus, using this method, we could determine whether the PET water bottle was contaminated and whether the off-flavor resulted from contamination of the bottle cap.

Interaction Between Groundwater and Stream Water Induced by the Artificial Weir on the Streambed (하상 인공구조물에 의해 유도되는 지하수-하천수 시스템의 상호작용)

  • This study investigated the interaction between groundwater and stream water systems, which is caused by the artificial weir on streambed, enforcing external stresses on the groundwater system. The study area is in Nami Natural Recreation Woods located in Chungcheongnam-do Geumsan-gun Nami-myeon Geoncheon-ri. In this study both of hydrophysical methods (hydraulic head) and hyrdochemical investigations (pH, EC, major ion analysis) were applied. In order to identify the relationship between each of study results, cross-correlation analysis is performed. From results of hydrophysical methods, water level fluctuation at BH-14, installed by the weir, shows the double-recession pattern much more frequently and much higher amplitudes than the fluctuation at each of other monitoring wells. Using the results by hydrochemical investigations, hydrochemical properties at BH-14 is similar to the hydrochemical characteristics in stream water. To analyze the interrelationships between the results from each of applied methods, cross-correlation analysis was applied. Results from the correlation analyses, water levels at BH-14 and stream weir showed the highest cross-correlation in hydrophysical aspects. On the other hand, the correlation between stream weir and bridge was the highest in hydrochemical aspects. The difference between the results from each of methods is due that the hydrophysical response at BH-14, such as water level, is induced by the pressure propagation-not with mass transfer, but the hydrochemical interaction, caused by mass transport, takes much more times. In conclusion impermeable artificial weir on streambed changes the interfacial condition between the stream and surrounding aquifers. The induced water flux into the groundwater system during flood period make water level at BH-14 increase instantly and groundwater quality higly similar to the quality of stream water. Referred similarities in both of water level and water quality at BH-14 become much higher when water level at weir grow higher.

The wintertime distribution of ionic components in Seoul and Busan: Comparative analysis between fine and coarse particles (겨울철 서울과 부산지역의 분진 중 이온분포의 특성 연구: 미세와 조대영역간의 비교)

  • Using our PM2.5 and PM10 concentration data obtained from Seoul and Busan during winter 2002, we conducted comparative analysis on the role of inorganic ions in constituting airborne particles in two distinctive urban areas. Whereas the mass concentration of fine particle was more significant in Seoul, no such pattern was found in Busan. In addition, when the major components were compared between different particle fractions and between different sites, clear pattern was apparent between those. Although the major components of fine particles were generally compatible each other ($NH{_4}^+$, $NO{_3}^-$, and NSSS), those of coarse fractions were clearly distinguished. Although anthropogenic signatures were still important in Seoul, the influence of oceanic sources was clear in coarse fraction of Busan ($Cl^-$ and $Na^+$). Detailed statistical analysis of our data consistently supports the importance of different source processes between particle modes and source processes.

Development of an Automated and Continuous Analysis System for PM2.5 and Chemical Characterization of the PM2.5 in the Atmosphere at Seoul (자동연속측정시스템 개발 및 이 시스템을 이용한 서울 대기 중 PM2.5의 화학적 조성과 특성에 관한 연구)

  • An automated analysis system for water soluble constituents in $PM_{2.5}$ has been developed. The system consists of a high capacity multi tube diffusion scrubber (MTDS), a low temperature particle impactor (LTPI), and two ion (anion and cation) chromatography (IC) systems. Atmospheric particles have been collected by passing sample air through a thermostated MTDS followed by a LTPI. This system allows simultaneous measurements of soluble ions in $PM_{2.5}$ at 30 minutes interval. At the air sampling flow rate of 1.0L/min, the detection limits of the overall system are in the order of tens of $ng/m^3$. This system has been successfully used for the measurement of particulate components of Seoul air from April 2003 to January 2004. $SO_4^{2-},\;NO_3^-,\;NH_4^+,\;NO_2^-,\;Cl^-,\;Na^+,\;K^+,\;Ca^{2+},\;and\;Mg^{2+}$ are the major ionic species for $PM_{2.5}$ at Seoul. Among them, $SO_4^{2-},\;NO_3^-\;and\;NH_4^+$ are the most abundant ions, contributed up to $86\%$ of the total and the concentrations were higher than those in any other urban sites in the world except for Chinese cities. There are high pollutant episodes which contribute about $15\~20\%$ of annual average values of the major ions. During the episode, the all parcels were transported from the asian continent and $PM_{2.5}$ were significantly neutralized. This suggests that aged and long range transported pollutants caused the high pollutant episodes. They showed a distinct daily and seasonal variations:they showed a peak in the early morning caused by the night-time accumulation of particulate matters. Atmospheric reactions including gas-to-particle reactions and inter-particle reactions and meteorological parameters including relative humidity and ambient temperature were described with related to the $PM_{2.5}$ 5 concentrations. All of the ionic species showed higher concentrations during the spring than those for summer and winter.

Solution structure and functional analysis of HelaTx1: the first toxin member of the κ-KTx5 subfamily

  • Scorpion venom comprises a cocktail of toxins that have proven to be useful molecular tools for studying the pharmacological properties of membrane ion channels. HelaTx1, a short peptide neurotoxin isolated recently from the venom of the scorpion Heterometrus laoticus, is a 25 amino acid peptide with two disulfide bonds that shares low sequence homology with other scorpion toxins. HelaTx1 effectively decreases the amplitude of the K+ currents of voltage-gated Kv1.1 and Kv1.6 channels expressed in Xenopus oocytes, and was identified as the first toxin member of the κ-KTx5 subfamily, based on a sequence comparison and phylogenetic analysis. In the present study, we report the NMR solution structure of HelaTx1, and the major interaction points for its binding to voltage-gated Kv1.1 channels. The NMR results indicate that HelaTx1 adopts a helix-loop-helix fold linked by two disulfide bonds without any β-sheets, resembling the molecular folding of other cysteine-stabilized helix-loop-helix (Cs α/α) scorpion toxins such as κ-hefutoxin, HeTx, and OmTx, as well as conotoxin pl14a. A series of alanine-scanning analogs revealed a broad surface on the toxin molecule largely comprising positively-charged residues that is crucial for interaction with voltage-gated Kv1.1 channels. Interestingly, the functional dyad, a key molecular determinant for activity against voltage-gated potassium channels in other toxins, is not present in HelaTx1.

Mineralogical and Geochemical Studies of Uranium Deposits of the Okchon Group in Southwestern District off Taejon, Korea (대전서남지대(大田西南地帶)에 있어서의 옥천대(沃川帶) 우라늄광상(鑛床)에 대(對)한 광물학적(鑛物學的) 및 지화학적(地化學的) 연구(硏究))

  • Uraniferous black slates of the Okchon sequence occur in Koesan (northeast) through Miwon-Boun (middle) to the southwest off Taejon (southwest) within the Okchon fold belt. The Uraniferous balck slates in the southwest off Taejon are particularly well developed in Chubu (northeast) and Moksso-ri (middle) areas whereas they are less developed in Jinsan (southwest) area. The uraniferous beds range from less than a meter to 40 meters in thickness and range from less than 0.02% $U_3O_8$ (cut-off-grade) to 0.05% $U_3O_8$ in the southwestern district off Taejon. Electron microprobe analysis of uranium-minerals found in graphitic slate samples enables to estimate their major compositions semi-quantitatively so that uraninite, ferro-uranophane and chlopinite are tentatively identified. Uranium-minerals are closely associated with carbon and metal sulfides. Correlation analysis of trace element concentrations revealed that U and F.C., and U and Mo are lineary correlative respectively and their correlation coefficients are positively high whereas those of U and V, U and Mn, and U and Zr are negatively low, implying that uranium mineralization has been closely related with concentrations of carbon and molybdenum. Stable isotope analyses of pyrite sulfur range widely from +11.5% to -23.3% in ${\delta}^{34}S$ values whereas those of graphite carbon fall within a narrow range between -23.3% and -28.9% in ${\delta}^{13}C$ values. The wide range of ${\delta}^{34}S$ values suggests that the sulfur could be of meteoric origin rather than of igneous source. The narrow range of ${\delta}^{13}C$ values, which are close to those of coal, indicates that the graphite is organic carbon in origin. Therefore, it is concluded that the uranium mineralization in the Okchon sequence took place primarily in sedimentary environment rich in organic matter and sulfide ion, both of which served as the reducing agents to convert soluble uranyl complex to insoluble uranium dioxide.

Studies on Pollution Characteristics and Sources of Precipitation in Jeiu Island

  • The pH, electric conductivity. and the major ionic components were analyzed for the precipitation samples collected at 1100 Site of Mt. Halla and Jeju city. The quality of analytical data was verified by the comparison of ion balances, conductivities and acid fractions, all of which correlation coefficients were over 0.952. The ionic strengths lower than 10$^{-4}$ M were found in 57 and 28% at 1100 Site and Jeju city respectively. The precipitation in Jeju city was influenced more by the oceanic effect than those in 1100 Site. The acidification of precipitation was caused mostly by S $O_4$$^{2-}$and N $O_3$$^{[-10]}$ in both areas, and the organic acids have contributed to the acidity with only 7~8%. The neutralization factors by N $H_3$ were about 44 and 47% at the 1100 site and the Jeju city, respectively, whereas those by CaC $O_3$were 21 and 24%, and the free acidity were about 38 and 28% at two sites. From the investigation of seawater and soil enrichment factors, the S $O_4$$^{2-}$, N $O_3$$^{[-10]}$ and N $E_4$$^{+}$ were immigrated by other sources rather than from the seawater or soil origins. but not in the case of $Mg^{2+}$, C $l^{[-10]}$ , N $a^{+}$, and $K^{+}$. Factor analysis has shown that the precipitation at the 1100 site had been influenced mostly by anthropogenic sources, followed by soil and sea-water sources. On the other hand, the precipitation at the Jeju city was mainly influenced by oceanic sources, followed by anthropogenic and soil sources.urces.

Characteristics of Optimized Analytical Method of Hydrogen Cyanide in Cigarette Mainstream Smoke by Using Continuous Flow Analyzer(CFA) (Continuous Flow Analyzer(CFA)를 이용한 담배 주류연 중 Hydrogen Cyanide(HCN)의 최적 분석방법 구명)

  • Hydrogen cyanide (HCN) is present in both the particulate and vapor phase of cigarette mainstream smoke. It is one of the 44 harmful substances on Hoffmann's list and is known to be a major ciliatoxic agent in cigarette mainstream smoke. Typically the determination of HCN in cigarette mainstream smoke has been done through colorimetric and electrochemical techniques, such as UV-spectrophotometry (UV), continuous flow analyzer (CFA), ion chromatography (IC) and capillary GC-ECD. In particular, CFA commonly has been using analysis hydrogen cyanide in cigarette smoke and the basic principle is pyridine-pyrazolone reaction. In this study, the more optimized analytical method is suggested isonicotinic acid-pyrazolone reaction method than previous pyridine-pyrazolone reaction method, a commonly used method for the determination of cyanide in water and air, by CFA. Sample collection was optimized by trapping particulate and vapor phase of smoke separately. The optimum NaOH concentration of the trapping solution was shown to be 0.2 M. HCN was stable up to 6 hours in this concentration but only 3 hours in 0.1 M solution. The sensitivity of this method was fairly good and it might be used in analysis of HCN in cigarette mainstream smoke.
