시설원예 작물의 생리장해 유발 토양요인구명 -I. 토마토, 배추, 무우 (Investigation of soil factors on physiological disorder of vegetable crops in vinyl house -(I). Tomato, Chinese cabbage and summer radish)

  • 예산읍내의 중요시설채소재배지 세부락을 중심으로 작부양식, 시비방법, 토양화학성, 작물의 영양상태와 생육상황을 조사하였다. 토마토의 시듦병 은 토양의 높은 EC와 질소함량 그리고 뿌리흑선충과 밀접하게 관련된것 같다. 칼슘결핍은 토양의 높은 K와 EC 그리고 토마토의 높은 철흡수에 기인하는것 같다. 여름배추는 80%의 생육억제를 보였는데 높은 EC(1.8mmho/cm) 때문이며 근류병(무사마귀명)을 보였는데 토양의 높은 인산함량(1,055 ppm) 때문인것 같다. 열무는 50%의 생육저하를 보였는데 높은 EC(1.6 mmho/cm), K 및 Mg 그리고 염기의 불균형때문으로 보인다. 다섯가지의 복합비료가 기비로 사용되고 질소칼리의 한가지 복비는 염화킬리 및 뇨소와 같이 추비로 사용하였다. 가축분의 다량사용과 매 적부시의 화학비료 다량 사용은 심토까지 과부화를 시켰다.

Strategies of Functional Food for Cancer Prevention in Human Beings

  • Functional food for prevention of chronic diseases is one of this century's key global challenges. Cancer is not only the first or second leading cause of death in China and other countries across the world, but also has diet as one of the most important modifiable risk factors. Major dietary factors now known to promote cancer development are polished grain foods and low intake of fresh vegetables, with general importance for an unhealthy lifestyle and obesity. The strategies of cancer prevention in human being are increased consumption of functional foods like whole grains (brown rice, barley, and buckwheat) and by-products, as well some vegetables (bitter melon, garlic, onions, broccoli, and cabbage) and mushrooms (boletes and Tricholoma matsutake). In addition some beverages (green tea and coffee) may be protective. Southwest China (especially Yunnan Province) is a geographical area where functional crop production is closely related to the origins of human evolution with implications for anticancer influence.

경북지역 오이에 발생하는 주요 바이러스 종류 및 발생실태 (Occurrence of Virus Diseases on Cucumber in Gyeongbuk Province)

  • 경상북도 오이 주산단지인 상주와 군위지역의 바이러스 병 발생을 조사한 결과 포장 발생율은 $55.0{\sim}88.6%$였으며, 상주지역이 군위보다 약간 높게 나타났다. 바이러스 발병 포장의 이병주율은 2006년 상주 지역에서 평균 25.6%로 매우 높게 나타났으며, 발생범위는 최소 1.5%에서 최대 90%로 농가별로 편차가 매우 심하였다. 포장에서 바이러스 증상으로 관찰되는 시료 217점을 RT-PCR을 이용하여 바이러스 종류를 분석한 결과 ZYMV가 85% 이상으로 가장 많이 검출되어 오이에 피해를 주는 주요 바이러스로 나타났다. ZYMV에 감염된 오이는 잎에 황화 및 심한 모자이크 증상이 나타났으며 일부 과일은 곡과 증상과 함께 표면이 울퉁불퉁하게 나타났다.

  • Numerous investigators have studied various activities of natural products and have found that they have not only nutritional effects, but also beneficial properties to cure various diseases and to maintain good health. Job's Tear(Yul-Moo) is a grass crop that has long been used in traditional medicine and as a nourishing food. Although its mechanism of action remains unclear, Job's Tear has been reported to exhibit anti-inflammatory, stomachic, anti-allergic, and anti-spastic effects and has been used in China for the treatment of warts, rheumatism, and neuralgia. Previous results in our laboratory demonstrated that the ethanol extract and the water extract of Job's Tear exerted an immune regulatory function on mice cells in vitro. The present study was performed to investigate the ex vivo effect of Job's Tear on immune function. Seven to eight weeks old mice(Balb/c) were fed chow diet ad libitum and water extract of Job's Tear was administered orally every other day for four weeks at two different concentrations(50 and 500mg/kg B.W.). Splenocytes proliferation with mitogen stimulation with Con A and LPS was enhanced at 50 mg/kg B.W. of Job's Tear compared to those of the control group. The results of this ex vivo study showed that proliferation of splenocytes and macrophage activation were seen in the mice orally administrated 50 mg/kg B.W. of Job's Tear water extracts. In conclusion, this study suggests that Job's Tear extracts may enhance immune function by regulating splenocyte proliferation and the cytokine prodution capacity of activated macrophages in mice.

Diagnosis of Alzheimer's disease through measurement of anti-beta amyloid antibody in patient's serum

  • Amyloid beta ($A{\beta}$) is believed one of the major pathogens of Alzheimer's disease (AD), and the reduction of $A{\beta}$ is considered a primary therapeutic target. Immunization with $A{\beta}$ can reduce $A{\beta}$ burden and pathological features in transgenic AD model mouse. This means $anti-A{\beta}$ autoantibodies may have a role in AD pathology. Recent findings suggest $anti-A{\beta}$ autoantibodies level decrease in AD patients. The early detection of AD is important for treatment of this disease. However, diagnosis on AD has only been possible through limited methods such as neuropsychological examination or MRI. To investigate whether it was possible to detect the presence and different levels of naturally occurring $anti-A{\beta}$ autoantibodies in the plasma of patients with AD and age-matched controls. An advanced ELISA was performed to detect levels of naturally occurring $anti-A{\beta}$ autoantibodies in the plasma. The level of $anti-A{\beta}$ auto-antibodies from patients with idiopathic Parkinson's disease or stroke and from normal controls were compared to that of AD patients. Our results showed a significantly lower $anti-A{\beta}$ autoantibodies level in AD compared to those with other neurological diseases or control. The level of $anti-A{\beta}$ autoantibodies in the serum may be used to diagnose the presence of AD.

Efficacy of Newer Molecules, Bioagents and Botanicals against Maydis Leaf Blight and Banded Leaf and Sheath Blight of Maize

  • Maize (Zea mays L.; 2N=20) is major staple food crop grown worldwide adapted to several biotic and abiotic stresses. Maydis leaf blight (MLB) and banded leaf and sheath blight (BLSB) are serious foliar fungal diseases may cause up to 40% and 100% grain yield loss, respectively. The present studies were undertaken to work out the efficacy of chemicals, botanicals and bioagents for the management of MLB and BLSB under field condition for two seasons Kharif 2014 and 2015. Five molecules (propiconazole 25 EC, hexaconazole 25 EC, carbendazim 50 WP, mancozeb 75 WP and carbedazim 12 WP + mancozeb 63 WP), two bioagents i.e. Trichoderma harzianum and T. viridae and three botanicals namely azadirachtin, sarpagandha and bel pathar were tested for their efficacy against MLB. Eight newer fungicides viz., difenconazole 250 SC, hexaconazole 5 EC, carbendazim 50WP, validamycin 3 L, tebuconazole 250 EC, trifloxystrobin 50 WG + tebuconazole 50 WG, azoxystrobin 250 EC and pencycuron 250 SC were evaluated against BLSB. Analysis revealed significant effects of propiconazole at 0.1%, carbendazim 12 WP + mancozeb 63 WP at 0.125% and sarpagandha leaves at 10% against MLB pathogen, whereas validamycin at 0.1% and trifloxystrobin 25 WG + tebuconazole 50 WG at 0.05% were found effective against BLSB. The slow rate of disease control virtually by the bioagents might have not shown instant effect on plant response to the yield enhancing components. The identified sources of management can be used further in strengthening the plant protection in maize against MLB and BLSB.

균근연구(菌根硏究)의 농림업(農林業)에의 응용(應用) (Application of Mycorrhizal Research to Agriculture and Forestry)

  • Recently mycorrhizal research has been one of the most fast-growing research areas in modern plant science and microbiology. The application potential of mycorrhizal techniques to agriculture and forestry is enormous in view of the ubiquitous nature of mycorrhizae and known benefits of mycorrhizae to host plants. Unfortunately, very few scientists in Korea are currently involved in mycorrhizal research. When a team of American plant pathologists visited Korea in September 1982 to participate in the Korea-U.S.A. Joint Seminar on Forest Diseases and Insect Pests, they were surprised by the principal author's statement that there was no single research project on mycorrhizae sponsored by Korean government or any scientific institutions. The author initiated a few years ago a research project on the ecology of tree mycorrhizae with a foreign financial support. Major areas of interest were survey of ectomycorrhizae in relation to soil fertility, taxonomic distribution of mycorrhizae among woody plants, identification of ectomycorrhizal fungi, and growth response of woody plants to artificial inoculation. In spite of the enormous application potential of mycorrhizae to agronomic plants, the subject of mycorrhizae has not been recognized by Korean agronomists, foresters or pathologists. The purpose of this review rather written in Korean is to introduce the techniques of mycorrhizal research to Korean scientists and to urge them to participate in challenging new scientific field which might bring us a remarkable increase in crop productivity and tree growth through manipulation of this unique symbiosis. In this review, following topics were discussed in the same order: introduction; brief history of mycorrhizal research; morphology and classification of mycorrhizae; distribution of mycorrhizae in plant kingdom and in soil profile; physiology of mycorrhizae (functions, mineral nutrition, mycorrhizal formation); interaction of mycorrhizae with soil-born plant pathogens. mycorrhizae in nitrogen-fixing plants; application of mycorrhizal techniques to nursery practices (isolation, culture, inoculation, and response); prospect in the future.

Cryopreservation of in vitro Grown Shoot Tips of Korean Potato Varieties by Droplet-vitrification

  • Potatoes are the world's 4th major food crop after maize, rice, and wheat and also are a staple food for 1.3 billion people. Due to their wide adaptability to various environmental conditions, their yeild capacity, and high commercial value, potatoes have contributed to global food security. Many potato germplasms are commonly preserved as whole plants in fields or in storage to maintain their particular genetic combinations. However, field maintenance is expensive and has the risk of potential losses from diseases, pests, plant ageing and climate change. Over the past four decades, meaningful efforts have been made toward the safe long-term conservation of potatoes through cryopreservation methods such as droplet-vitrification. In this study, we tested 4 Korean potato varieties('Golden Egg', 'Golden Ball', 'Ja-Young' and 'Ha-Ryeong') with the modified potato droplet -vitrification protocol. Potato shoot tips are precultured in a sucrose-enriched medium(0.3 and 0.7M for 7 and 17hrs, respectively) and submitted to a loading step with C4 solution for osmoprotection. The treated explants were dehydrated with Plant Vitrification Solution(PVS)2 which is 80% A3 solution in ice for 30 minutes. Thawing and unloading steps were performed with 0.8M sucrose solution for 30 sec(40℃) followed by 30min(25℃, room temperature). In a potato post-culture medium(MS+0.1 mg·L-1 GA3+0.1 mg·L-1 kinetin), we obtained a survival rates of post-thawed explants ranging 16.1-82.2%. The results suggest that modified and optimized protocols are required dependinig on every cultivar, genetic and ecological types. To achieve higher survival and regeneration rates, each step within the cryoprocedure must be carefully optimized.

국화재배지의 식물기생선충 분포조사 및 뿌리썩이선층의 ITS와 D3-28S rDNA 특성조사 (Occurrence of Plant Parasitic Nematodes in Chrysanthemum and ITS and D3-28S rDNA Characterization of Pratylenchus spp.)

  • 2005년 5월부터 6월까지 국내 주요 국화재배 포장에서 총 50개 토양시료를 채집하여 식물기생선충상을 조사하였다. 조사 결과, 뿌리썩이선충(Pratylenchus)의 포장발생률이 86%로 대부분의 포장에서 광범위하게 분포하였고, 밀도는 토양 200 cc와 뿌리 1g에서 평균 1,095마리가 검출되었다. 분자생물학적 동정을 위하여, '무안', '구미', '태안', '정읍', '마산' 등 5지역을 선정하고, 그 지역의 뿌리썩이선충을 대상으로 ITS와 D3-28S rDNA의 유전자 분석을 하였다. PCR 결과 증폭된 ITS는 '무안'의 경우 1 kb 크기로 나타났고, 그 외의 다른 지역의 뿌리썩이선충에서는 0.8 kb로서 약 200 bp가량의 차이가 있었다. D3-28S rDNA의 PCR 결과에서는 모든 지역에서 약 320 bp로 일정한 크기의 gene이 증폭되었으며, 증폭된 DNA는 sequencer를 이용하여 염기서열정보를 얻었다. 얻어진 각 isolate의 D3 염기서열 정보는 DNASTAR 분석 소프트웨어의 MegAlign 프로그램을 이용하여 분석 및 phylogenetic tree를 형성하였다. 분석 결과 '구미'와 '태안' isolate의 D3 염기서열은 100% 일치하였고, '정읍' isolate는 이들 isolate와 99.7%의 유사성을 나타내었다. 또한 이들 세 isolate는 Pratylenchus vulnus와 근연종임이 확인되었는데, '구미', '태안'은 96.7%, '정읍'은 96.3%의 유사성을 각 나타내었다. 한편, '마산' isolate는 P. penetrans와 100% 일치됨을 나타내었고, '무안' isolate는 P. brachyurus와 99.7%의 유사성을 나타냄으로써 높은 상관성을 나타내었다. 이와 같은 결과는 phylogenetic tree에서도 동일한 결과를 나타내고 있다.

Fine Mapping of the Rice Bph1 Gene, which Confers Resistance to the Brown Planthopper (Nilaparvata lugens Stal), and Development of STS Markers for Marker-assisted Selection

  • The brown planthopper (BPH) is a major insect pest in rice, and damages these plants by sucking phloem-sap and transmitting viral diseases. Many BPH resistance genes have been identified in indica varieties and wild rice accessions, but none has yet been cloned. In the present study we report fine mapping of the region containing the Bph1 locus, which enabled us to perform marker-aided selection (MAS). We used 273 F8 recombinant inbred lines (RILs) derived from a cross between Cheongcheongbyeo, an indica type variety harboring Bph1 from Mudgo, and Hwayeongbyeo, a BPH susceptible japonica variety. By random amplification of polymorphic DNA (RAPD) analysis using 656 random 10-mer primers, three RAPD markers (OPH09, OPA10 and OPA15) linked to Bph1 were identified and converted to SCAR (sequence characterized amplified region) markers. These markers were found to be contained in two BAC clones derived from chromosome 12: OPH09 on OSJNBa0011B18, and both OPA10 and OPA15 on OSJNBa0040E10. By sequence analysis of ten additional BAC clones evenly distributed between OSJNBa0011B18 and OSJNBa0040E10, we developed 15 STS markers. Of these, pBPH4 and pBPH14 flanked Bph1 at distances of 0.2 cM and 0.8 cM, respectively. The STS markers pBPH9, pBPH19, pBPH20, and pBPH21 co-segregated with Bph1. These markers were shown to be very useful for marker-assisted selection (MAS) in breeding populations of 32 F6 RILs from a cross between Andabyeo and IR71190, and 32 F5 RILs from a cross between Andabyeo and Suwon452.
