Structural Conservation and Food Habit-related Liver Expression of Uncoupling Protein 2 Gene in Five Major Chinese Carps

  • The full-length cDNA of grass carp (Ctenopharyngodon idellus) and silver carp (Hypophthalmichthys molitrix) uncoupling protein 2 (UCP2) was obtained from liver. The grass carp UCP2 cDNA was determined to be 1152 bp in length with an open reading frame that encodes 310 amino acids. Five introns (Intron 3, 4, 5, 6 and 7) in the translated region, and partial sequence of Intron 2 in the untranslated region of grass carp UCP2 gene were also obtained. Gene structure comparison between grass carp and mammalian (human and mouse) UCP2 gene shows that, the UCP2 gene structure of grass carp is much similar to that of human and mouse. Partial UCP2 cDNA sequences of bighead carp (Aristichthys nobilis) and mud carp (Cirrhinus molitorella), were further determined. Together with the common carp (Cyprinus carpio) UCP2 sequence from GenBank (AJ243486), multiple alignment result shows that the nucleotide and amino acid sequences of the UCP2 gene, were highly conserved among the five major Chinese carps that belong to four subfamilies. Using beta-actin as control, the ratio UCP2/beta-actin mRNA (%) was determined to be $149.4{\pm}15.6$ (common carp), $127.4{\pm}22.1$ (mud carp), $96.7{\pm}12.7$ (silver carp), $94.1{\pm}26.8$ (bighead carp) and $63.7{\pm}16.2$ (grass carp). The relative liver UCP2 expression of the five major Chinese carps, shows a close relationship with their food habit: benthos and detrituseating fish (common carp and mud carp) > planktivorious fish (silver carp and bighead carp) > herbivorious fish (grass carp). We suggest that liver UCP2 might be important for Chinese carps to detoxify cyanotoxins and bacteria in debris and plankton food.

A comparative study of ectoparasites occurrence between grass carp and silver carp in guilan province culture ponds, Iran

  • Parasitic infection is among the most common problems for carp cultivation. They are also important for the principal entrance of other hazardous infections as well. This study was carried out for determining of parasitic fauna of two major carp known as silver and grass carp with the comparison of prevalence value and intensity rate of parasites among them, alongside the relationship between the biometric characteristics and host sex with the infection level. For this purpose, a total of 94 fish samples were caught randomly using a fishing net, from Guilan ponds during spring and summer of the year 2018 and transported alive to the laboratory. Upon arriving, the biometric characteristics and genus of each carp were measured individually. Specimens were then acquired from the skin, gills, and eyes of the carp and examined according to standard parasitology methods. Recovered parasites were observed under a light microscope and then fixed for identification. As the result, the occurrence and intensity in the higher length group were comparatively greater than the lower one. Also, the prevalence and intensity of total parasites in male carp were higher than in females. In this research, Dactylogyrus hypophthalmichthys and Dactylogyrus aristhichtys were observed in silver carp and Dactylogyrus lamellatus was detected in grass carp. In the paper below, we found that the host specificity varies in different species of Dactylogyrus isolated from grass carp and silver carp.

An Enzyme-Linked Immunosorbent Assay for Carp (Cyprinus carpio) Vitellogenin and Assay for Oestrogenic Chemicals

  • The egg-yolk precursor vitellogenin(VTG) is secreted by the liver of female and male fish, in response to estrogenic compound. Carp(Cryprinus carpio) vitellogenin of one major protein is 160kDa, two minor proteins is 110kDa and 170kDa. We were induced vitellogenin by inject of 17-estradiol and purified in a two step procedure by separating it from plasma protein precipitated by 35% saturated ${(NH_4)}_2SO_4$ and then from the remainder by Mono-Q chromatography. We found major carp(Cyprinus carpio) vitellogenin band at 160kDa. Effect of different concentration of oestrogen on vitellogenin synthesis in carp(Cyprinus carpio) exposed for 4 weeks. Show differential effects on vitellogenin synthesis 7 days after treatment. Plasma vitellogenin was measured 3 times for each by an enzyme linked immunosorbent assay(ELISA). ELISA was developed for the detection of the egg yolk precursor vitellogenin in plasma of carp(Cyprinus carpio). The ELISAS performance was optimized and characterized.

Cytogenetic Characteristics of Cyprinidae between Diploid and Spontaneous Triploid in Major River of Korea

  • This study investigated cytogenetic and hematological and histological characteristics between diploid and spontaneous triploid on Cyprinidae (Crucian carp, Carassius auratus; crucian carp, C. cuvieri and common carp, Cyprinus carpio) in four major rivers of Korea. In our results, DNA contents of triploid Cyprinidae were 50% more than those of diploid Cyprinidae. Also, erythrocyte size of triploid Cyprinidae was 50% larger than those of diploid Cyprinidae. In all sampling rivers, sex ratios of C. auratus were biased to female, and especially, triploid groups of C. auratus were all female groups (p<0.05). In principle, sex ratios of C. cuvieri and common carp were equivalent between male and female.

우리나라 하천에 서식하는 붕어와 주변 퇴적토에 축적된 HBCDs의 분포 특성 (Distribution characteristics of hexabromocyclododecanes in crucian carp and sediment from the major rivers)

  • 본 연구에서는 우리나라 주요 하천에 서식하는 붕어와 주변 퇴적토에 축적된 HBCDs(hexabromocyclododecanes)의 농도를 분석하고 분포특성을 조사하였다. 조사한 15개 지점에서 구한 모든 붕어 근육 및 퇴적토 시료에서 HBCDs가 검출되었는데 이는 HBCDs가 광범위하게 오염되어 있음을 나타내는 것이다. 붕어 근육과 퇴적토에 축적된 HBCDs의 총 농도(${\Sigma}HBCDs$, ${\alpha}$-, ${\beta}$- 및 ${\gamma}$-HBCD 농도를 모두 합한 값)는 각각 1.3~19 ng/g (lipid wt)과 0.17~30 ng/g (dry wt) 범위로 나타났다. 지금까지 일반적으로 알려진 다른 연구에서와 마찬가지로 붕어에서는 ${\alpha}$-이성질체가 그리고 퇴적토에서는 ${\gamma}$-이성질체가 각각 88%와 60%로 가장 우세하게 축적된 것으로 나타났다. 각 하천의 하류에서 채집한 붕어 근육에 축적된 ${\Sigma}HBCDs$가 상류에서의 시료보다 훨씬 높은 수준으로 검출되었다. 또한 하천 하류에 위치하지만 고립된 지점에서의 붕어 근육에 축적된 ${\Sigma}HBCDs$는 하천의 다른 지점에서 채집한 붕어 근육에 축적된 값보다 크게 낮은 값으로 나타났다. 이는 하천의 주변 환경이 HBCDs 오염에 기여하고 있는 것을 나타내는 것으로 보인다.

Inclusion of Pangasiid Catfish in Polycultures of Major Indian Carps (Catla, Rohu and Mrigal) Increases Yield and Economic Gain

  • An experiment was conducted in twelve 0.02-ha rain-fed earthen ponds for 18 weeks to evaluate the effects of including pangasiid catfish (Pangasius hypophthalmus) in polycultures of major Indian carps, catla (Catla catla), rohu (Labeo rohita), and mrigal (Cirrhinus cirrhosus), on yield and economic gain. Treatment 1 ($T_1$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal per ha, treatment 2 ($T_2$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal+2,500 pangasiid catfish per ha, treatment 3 ($T_3$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal+5,000 pangasiid catfish per ha, and treatment 4 ($T_4$) consisted of 5,000 catla+2,500 rohu+2,500 mrigal+7,500 pangasiid catfish per ha. The growth performance of fishes was evaluated by calculating specific growth rate, daily weight gain, and percent weight gain. Inclusion of pangasiid catfish in the carp polyculture resulted in significant increases in the growth and yield of catla and rohu. Catla and rohu grew most in $T_4$, with 7,500/ha of pangasiid catfish. The presence of pangasiid catfish did not influence the growth of mrigal. The growth of pangasiid catfish varied with stocking density, and they grew best at 5,000/ha ($T_3$). The carp and the combined fish yields were significantly higher in the carp polycultures with pangasiid catfish than with carps alone. The total carp and the combined fish yields were highest in $T_4$, in which the density of pangasiid catfish was greatest. However, the total carp and the combined fish yields in $T_3$ and $T_4$ were not significantly different. Profit differed significantly among treatments. Economic gain was highest in $T_3$, with 5,000/ha of pangasiid catfish, and lowest in $T_1$. The inclusion of pangasiid catfish in major carp polycultures results in higher fish yields and greater economic gain.

Carbofuran 의 잉어(Cyprinus carpio)체내 대사 (In Vivo Metabolic studies on Carbofuran Degradation in carp(Cyprinus carpio L.))

  • 잉어체내에서 $^{14}C-carbofuran$의 흡수, 분포, 대사 및 배설시험을 이해 carbofuran을 40ppb 수준으로 처리하였다. 약제처리 12시간 후에 적출된 잉어의 조직(간, 신장, 창자, 쓸개)과 혈액에서 방사능$(^{14}C)$의 검출율이 가장 높았다. 잉어의 간과 신장에서 carbofuran은 3-ketocarbofuran과 3-hydroxycarbofuran으로 대사되었고 주요 대사산물로는 3-hydroxycarbofuran이었다. 약제처리 후 조직에서 그의 분포율이 가장 높은 시간대의 잉어를 공시수에 분양하여 배설시험을 수행한 결과 carbofuran은 분양 3시간 후에 배설율이 현저히 증가하였고 검출된 대사산물로는 3-ketocarbofuran과 3-hydroxycarbofuran으로 이 시간 동안의 배설시험에서 이들 대사산물의 검출정도는 각각 32.3%와 52.8%였고 미확인 대사산물은 2.6%이었다.

  • This paper describes about the behavior of the lseraeli carp (Cyprinus cartio. body length 43cm) and the Red seabream (Chrysophrys major. body length 35cm) aquacultured by telemetry techniques. The telemetry system consists of a pinger of 50kHz, three omni-directional hydrophones and ultrasonic receivers, a single board computer for the signal processing, two RF transceivers for the data communication, and a personnel computer. Those fish tagged the pingers were tracked by the LBL method, and its location was calculated by the hyperbolic method. The average speed of the lsraeli carp was about 1.0 times of the body length in a farming water tank (24$\times$24$\times$1m), and the Red seabream 1.3 times of the body length in a farming raft (10$\times$10$\times$5m). A variation of the speed of the lsraeli carp was higher than the other one.

  • This study was undertaken to understand development stage of ovary and changes of hormones concentration from February to May in the Israeli carp, Cyprinus carpio. The results obtained in these experiments are as follows ; 1. Serum LH levels began to increase sharphy in March, coinciding with the onset of rapid ovarian development. 2. LH levels were well correlated with changes in gonadosomatic index. 3. Dramatic increase in gonadosomatic index occurred during the months of March. 4. Ripe stage(Stage Ⅴ) rapidly increase in March. 5. Early perinucleolus oocyte radpily develop into late perinucleolus oocytes in March. 6. The vitellogenic phase begins as these late perinucleolus oocytes become transformed into early maturing oocytes through the accumulation of yolk. 7. The cytoplasm completely fills with yolk as oocytes reach the late maturing stage. 8. Changes in the microscopic appearances of the ovaries were well correlated with changes in both gonadosomatic index and macroscopic appearance. 9. It is concluded from these observations that LH plays a major role in sexual maturation of the Israeli carp.

주요 하천 및 호수에 서식하는 붕어 (Carassius auratus and Carassius cuvieri) 근육에 축적된 2,3,7,8-치환 PCDDs, PCDFs 및 DL-PCBs 분석 (Analysis of 2,3,7,8-substituted PCDDs, PCDFs, and DL-PCBs in muscle of crucian carp (Carassius auratus and Carassius cuvieri) from major rivers and lakes)

  • 우리나라 주요 하천수계의 대표적인 지점 한 곳씩 선정하여 이곳에 서식하는 붕어에 대한 PCDDs, PCDFs 및 DL-PCBs의 체내 축적 정도를 분석하고 분포특성을 살펴보았다. 회수율은 PCDDs 및 PCDFs는 50.6%~88.3%, DL-PCBs는 52.3%~93.2%이었으며, 붕어 체내에 축적된 PCDDs, PCDFs 및 DL-PCBs를 모두 합한 ${\sum}$dioxins은 평균 0.39 pg TEQ $g^{-1}$ wet wt., 중앙값 0.14 pg TEQ/g wet wt., 그리고 농도 범위는 0.047-1.0 pg TEQ/g wet wt.으로 나타났다. PCDDs 및 PCDFs는 일부 시료에서 검출된 반면, DL-PCBs는모든 시료에서 검출되었으며 ${\sum}$dioxins에 대한 기여도가 83.6%로서 가장 높았다. 다음으로 PCDFs와 PCDDs가 각각 ${\sum}$dioxins의 12.7%와 3.7%를 기여하고 있었다.
