Deubiquitinase USP35 as a novel mitotic regulator via maintenance of Aurora B stability

  • Aurora B is an important kinase involved in dynamic cellular events in mitosis. Aurora B activity is controlled by several post-translational modifications (PTMs). Among them, E3 ubiquitin ligase-mediated ubiquitination plays crucial roles in controlling the relocation and degradation of Aurora B. Aurora B, ubiquitinated by different E3 ligases, moves to the exact site for its mitotic function during metaphase-anaphase transition and is then degraded for cell cycle progression at the end of mitosis. However, how the stability of Aurora B is maintained until its degradation has been poorly understood. Recently, we have found that USP35 acts as a deubiquitinating enzyme (DUB) for Aurora B and affects its stability during cell division, thus being involved in the regulation of mitosis. In this review, we discuss the USP35-mediated deubiquitination of Aurora B and the regulation of mitotic progression by USP35.

Maintenance of cellular tetrahydrobiopterin homeostasis

  • Tetrahydrobiopterin (BH4) is a multifunctional cofactor of aromatic amino acid hydroxylases and nitric oxide synthase (NOS) as well as an intracellular antioxidant in animals. Through regulation of NOS activity BH4 plays a pivotal role not only in a variety of normal cellular functions but also in the pathogenesis of cardiovascular and neurodegenerative diseases, which develop under oxidative stress conditions. It appears that a balanced interplay between BH4 and NOS is crucial for cellular fate. If cellular BH4 homeostasis maintained by BH4 synthesis and regeneration fails to cope with increased oxidative stress, NOS is uncoupled to generate superoxide rather than NO and, in turn, exacerbates impaired BH4 homeostasis, thereby leading to cell death. The fundamental biochemical events involved in the BH4-NOS interplay are essentially the same, as revealed in mammalian endothelial, cardiac, and neuronal cells. This review summarizes information on the cellular BH4 homeostasis in mammals, focusing on its regulation under normal and oxidative stress conditions.

Economic Analysis of Plant Utilities Under Environment Factor (환경요소를 고려한 발전설비의 경제성 평가)

  • The purpose of this study is an economic analysis of power plant utilities by comparing electricity generating cost including environmental costs. Considering the enormous role of electricity in the national economy, it is very important to study the effect of environmental regulation on the electricity sector. Because power utilities need for large investment during construction, operation and maintenance, and also require much construction lead time. Economic analysis is the important process in the electric system expansion planning. This paper compares the costs of electricity generation including environmental costs between a coal-fired power plant and an LNG combined cycle power plants. With the simulation, this study surveys the sensitivity of fuel prices, interest rate and carbon tax. In each case, this sensitivity can help to decide which utility is economically better in environmental regulation circumstance.

  • Demarcation point is technical regulation for preventing conflict and improving the efficiency of the maintenance by discriminating facility area between telecommunication operator and telecommunication user. Since current demarcation point is defined on the basis of the telephone network, it is necessary to examine the applicability to various connection methods for the introduction of new services and facilities. This paper is to discuss for the domestic and international regulations of the demarcation point, and matters to be taken into account.

Lactate: a multifunctional signaling molecule

  • Since its discovery in 1780, lactate has long been misunderstood as a waste by-product of anaerobic glycolysis with multiple deleterious effects. Owing to the lactate shuttle concept introduced in the early 1980s, a paradigm shift began to occur. Increasing evidence indicates that lactate is a coordinator of whole-body metabolism. Lactate is not only a readily accessible fuel that is shuttled throughout the body but also a metabolic buffer that bridges glycolysis and oxidative phosphorylation between cells and intracellular compartments. Lactate also acts as a multifunctional signaling molecule through receptors expressed in various cells and tissues, resulting in diverse biological consequences including decreased lipolysis, immune regulation, anti-inflammation, wound healing, and enhanced exercise performance in association with the gut microbiome. Furthermore, lactate contributes to epigenetic gene regulation by lactylating lysine residues of histones, accounting for its key role in immune modulation and maintenance of homeostasis.

  • RNA metabolism plays a central role in regulating of T cell-mediated immunity. RNA processing, modifications, and regulations of RNA decay influence the tight and rapid regulation of gene expression during T cell phase transition. Thymic selection, quiescence maintenance, activation, differentiation, and effector functions of T cells are dependent on selective RNA modulations. Recent technical improvements have unveiled the complex crosstalk between RNAs and T cells. Moreover, resting T cells contain large amounts of untranslated mRNAs, implying that the regulation of RNA metabolism might be a key step in controlling gene expression. Considering the immunological significance of T cells for disease treatment, an understanding of RNA metabolism in T cells could provide new directions in harnessing T cells for therapeutic implications.

  • Telomerase plays a pivotal role in the pathology of aging and cancer by maintaining genome integrity, controlling cell proliferation, and regulating tissue homeostasis. Telomerase is essentially composed of an RNA component, Telomerase RNA or TERC, which serves as a template for telomeric DNA synthesis, and a catalytic subunit, telomerase reverse transcriptase (TERT). The canonical function of TERT is the synthesis of telomeric DNA repeats, and the maintenance of telomere length. However, accumulating evidence indicates that TERT may also have some fundamental functions that are independent of its enzymatic activity. Among these telomere-independent activities of hTERT, the role of hTERT in gene transcription has been investigated in detail. Transcriptional regulation is a fundamental process in biological systems. Several studies have shown a direct involvement of hTERT in gene transcription. This mini-review will focus on the role of hTERT in gene transcription regulation, and discuss its possible mechanisms.

Emerging role of RUNX3 in the regulation of tumor microenvironment

  • A number of genes have been therapeutically targeted to relieve cancer, but cancer relapse is still a growing issue. The concept that the surrounding tumor environment is critical for the progression of cancer may foster an answer to the issue of cancer malignancy. Runt domain transcription factors (RUNX1, 2, and 3) are evolutionarily conserved and have been intensively studied for their roles in normal development and pathological conditions. During tumor growth, a hypoxic microenvironment and infiltration of the tumor by immune cells are common phenomena. In this review, we briefly introduce the consequences of hypoxia and immune cell infiltration into the tumor microenvironment with a focus on RUNX3 as a critical regulator. Furthermore, based on our current knowledge of the functional role of RUNX3 in hypoxia and immune cell maintenance, a probable therapeutic intervention is suggested for the effective management of tumor growth and malignancy.

  • A nationwide questionnaire survey was conducted to investigate the actual conditions of local authorities in operating and maintaining sewerage systems especially for urban flooding. The questionnaire includes the incidences of flooding since 1980, the existence of any guidance for preventing flood damage and the design frequency currently used for sewers in each local authority. Among 160 local authorities responded to the survey, 95(59.3%) has experienced flooding more than once since 1980 while only 36(37.9%) of the 95 has prepared operation/maintenance guidelines for flooding. Most of the respondents have applied the 10 year design frequency for sewer design and 57(35.6%) of them agreed to adjust the 10 year design frequency regulation.

  • The purpose of this study is to provide an remodification method for elementary school complex facilities in Seoul Metropolitan city. This study analyzes the questionnaire survey for 568 entire elementary schools and the questionnaire survey for school facility complex project operated 44 elementary schools. Then the result of this study was deduced from the comparison between the 568 entire survey data and 44 survey data in terms of the cognition of school facility complex project, the required type of facility and the problems of complex facility maintenance and operation. Four major findings were revealed as the result of research. First, there is a very high agreement with the school facility complex projects. Second, it was need that the process of planning and design was remodified to enhance participation of parents of students and local residents for selecting an appropriate complex facility. Third, education environment and student safety should be protected from any other school facility complex environments. Fourth, an agreement and regulation is needed for more effective maintenance and operation of school complex facilities.
