Cytotoxicity of Daucus Carota L. on Various Cancer Cells (인체 암세포에 대한 당근 추출 성분의 세포독성효과)

  • We investigated the cytotoxic effects of roots and seeds Daucus carota L. on HepG2, HeLa, MCF7, SW626, C6 and NB41A3 cell lines by the MTT assay. Among extracts, the ethylacetate partition layer (DCMEA) of root of Daucus carota L. showed the strongest cytotoxic effects on HepG2, HeLa, C6 and NB41A3 cell lines. On the other hand, methanol(DCM), n-ethylacetate(DCMEA) and n-butanol(DCMB) extract of the seeds of Daucus carota L. also showed significant cytotoxic activities for all six cell lines. $\beta$-carotene, a well-known main component of Daucus carota L. was also tested for its cytotoxic effect. However, in all six cell lines, $\beta$-carotene falied to show significant cytotoxicity. Therefore, the anticancer effect of DCMEA of root fo Daucus carota L. and DCM, DCMH, DCMEA and DCMB extracts of seeds may be caused by components other than $\beta$-carotene. Futher studies are under way to isolate the compounds responsible for the significant cytotoxic activity.

Antioxygenic and Peroxide-decomposing Activities of Antarctic Krill Lipids (Krill 지질(脂質)의 항산화성(抗酸化性)과 과산화물(過酸化物) 분해작용(分解作用))

  • The oxidation rate of krill lipids is very slow and no peroxides are accumulated even after long storage. By means of various chromatographic techniques and mass spectrophotometry, the primary antioxidant has been identified as ${\alpha}$-tocopherol. The phospholipid fractions did not show any antioxidative activity but peroxide-decomposing properties of total lipids depended upon the phospholipid contents. The peroxide-decomposing activities of phospholipids were due to the presence of polar materials generated during the storage. The most peroxide-decomposing fractions of oxidized krill lecithin by DEAE-cellulose column chromatography was low-molecule fraction (mean molecular weight: 182) and high-molecule fraction (mean molecular weight: 1942) was the next. The separation of peroxide-decomposing properties from low-molecule fraction was achieved by partitioning between chloroform and methanol/water. The methanol/water fraction showed strong peroxide-decomposing activities and main component of this fraction was assumed hydroxyamine compounds derived from choline.

  • The electronic absorption spectra of five Schiff bases derived from 2-hydroxy-1-naphthaldehyde with glycine, alanine, leucine, valine and phenylalanine have been measured in various solvents. The observed bands were assigned to the proper electronic transitions and compared with the predicted transitions at the semiempirical level of theory. The calculated equilibrium constants are in agreement with the experimental results, predicting the existence of all studied compounds predominantly or completely in keto-imine tautomerism. On the other hand, a correlation between $\nu\;(cm^{-1})$ (main frequency of each compound) and the well known solvent parameters $E_T$ (30), $\varepsilon_T$, $\pi^*$ been made. Furthermore, the acid dissociation constants, $pK_a$, were determined by using three different spectrophotometric methods.

  • White birch bark is being called baekhwapi usually and is used urination, travail, removal of fever in Oriental medicine. Betulin had been known as main chemistry ingredient of white birch bark. So, in an experiment that see betulin and connection inorganic compounds separation and enforced to recognize antioxidative activity and anti HIV. Betulin's yield separated vast quantity for the first time in Korea by 6.9%, and 3,4-dihydrobenzoic acid in ethylacetate soluble as experiment result.   • Low-salted and fermented squid product, squid jeotkal was prepared with the addition of 10% salt and fermented for 50 day at 1$0^{\circ}C$. During fementation of squid, sensory evaluation and changes of volatile components were examined. Volatile flavor components in raw squid and low-salted squid jeotkal were extracted using a rotary evaporating system. The volatile concentrates were identified by GC and GC-MS. Major volatile components of raw squid were methional and 2-methyl-2-propanol. However, alcohols such as propanol, isoamyl alcohol, methionol and phenylethyl alcohol increased during the period of fermentation. The model reaction using microorganism was carried out, in order to confirm formation mechanism ofvolatile flavor compounds of the squid during fermentation. The main volatile components of Pseudomonas sp. D2 model system were isoamyl alcohol and acetoin. Those of Staphylococcus xylosus model system were isoamyl alcohol and phenylacetaldehyde.

Treatment Characteristics of Biological Aerated Filter Process Using the Upflow and Downflow System (상향류 및 하향류 생물막여과공정의 처리특성에 관한 연구)

  • This paper presents the test results of biological aerated filtration(BAF) process to replace activated sludge process by enhancing treated effluent quality and reducing the costs. In BAF process both BOD and SS compounds in wastewater are degraded and removed by biological reaction and filtration. Upflow BAF with expanded polypropylene media and downflow BAF with ceramic media were used to investigate the effects of hydraulic and organic loads on effluent quality. As a result, in BAF processes which has different media, upflow BAF reactor shows 5% higher efficiency than downflow BAF and this phenomena caused by backwashing methods and operational conditions. The results of influence factors analyzed by Factor Analysis Method in BOD and SS treatment efficiency are the size of media, hight of media bed and type of media. The quantitative effects of media size are 5.73% in TBOD, 5.78% in SBOD and 7.65% in TSS, so we confirmed the main factor is media size.

  • In-pipe formation of THM in water distribution systems was simulated by using the continuously and easily measurable parameters such as water temperature, residual chlorine and soluble organic compounds. The concentration of miscellaneous organics which could be the precuror of THM, was measured and represented as the absorbance of ultraviolet at wave length 260 nm. As the results, the developed equation in this study showed a more reliability on the change of THM than the normally regressed equation. In addition, the simulation was successfully fitted in the actual water treatment and distribution systems. Of the THM components, dibromochloromethane was the main cause dropping the overall reliability in the simulation.

Forsythiae Fructus and Its Active Component, Arctigenin, Provide Neuroprotection by Inhibiting Neuroinflammation

  • In this study, we found that Forsythiae fructus (FF) and one of its main compounds, arctigenin, significantly inhibited nitric oxide production in lipopolysaccharide (LPS)-stimulated BV-2 microglial cells. Arctigenin also suppressed the expression of inducible nitric oxide synthase and cyclooxygenase-2, and inhibited the activation of extracellular signal-regulated kinase, c-Jun N-terminal kinase and p38. Moreover, it also reduced levels of proinflammatory cytokines, interleukin $1{\beta}$, tumor necrosis factor ${\alpha}$ and prostaglandin E2, and inhibited neuronal death in LPS-treated organotypic hippocampal cultures. Therefore, we suggest that arctigenin may confer a neuroprotective effect via the inhibition of neuroinflammation.

  • Electrochemical reduction of nitrobenzene and substituted nitrobenzenes on lead electrode was studied by galvano-static measurements and cyclic voltammetry in basic ethanol-water solvents. Nitroso compounds or hydroxylamines were detected as the main reduction product depending on the potential. Mechanisms of production and further reduction of substituted and unsubstituted nitrosobenzenes are deduced from Tafel slope, pH dependence and reaction order. The reduction of most of the substituted nitrobenzenes to corresponding nitrosobenzene derivatives seemed to follow the reaction mechanism of nitrobenzene reduction with a few exceptions. A Hammett type relationship between the magnitude of the reduction current and the kinds of substituents was found with the ${\rho}$ value of 0.54.

X-Ray Absorption Spectroscopy: A Complementary Tool for Structural and Electronic Characterization of Solids

  • The purpose of this paper is to show that X-ray absorption spectroscopy (XANES and EXAFS) is a powerful technique for characterizing both crystalline and amorphous solids from structural (local order) and electronic point of view. The principle of this technique is briefly described by showing the main factors which must be considered for recording and fitting the experimental results. Some non-trivial examples have been selected for demonstrating that XAS spectroscopy is the only technique for bringing a definitive answer as for example: the determination of the local distortion of the $NiO_6$ octahedra in the $Li_{1-z}Ni_{1+z}O_2$ layered oxides and the evidence of the presence of copper pairs in the NASICON-type phosphate $CuZr_2 (PO_4)_3$. Are also reported some significant examples for which XAS spectroscopy is decisive with other characterization methods as (i) Raman spectroscopy for glasses (ii) Mossbauer spectroscopy for $LiNi_{1+z-t}Fe_To_2$ oxides (iii) magnetic measurements for Ce-based intermetallic compounds.
