Chemokine Gene Expression in Mice during Orientia tsutsugamushi Infection

  • Orientia tsutsugamushi, an obligate intracellular bacterium, is the causative agent of scrub typhus which is histopathologically characterized by inflammatory manifestations. To understand the pathogenesis of scrub typhus, chemokine gene expression in mice after infection with O. tsutsugamushi was investigated. The mRNAs that were upregulated included macrophage inflammatory proteins 1${\alpha}$/${\beta}$ (MIP-1${\alpha}$/${\beta}$), MIP-2, monocyte chemoattractant protein 1, RANTES (regulated upon activation, normal T-cell expressed and secreted), and gamma-interferon-inducible protein 10. Peak expression of these chemokines was observed six days after infection. These responses returned to or approached baseline preinfection levels by eight days after infection. Chemokine profiles in infected mice were well correlated with the kinetics of inflammatory cell infiltration. Thus, O. tsutsugamushi appears to be a strong inducer of chemokines which may significantly contribute to the inflammation observed in scrub typhus by attracting and activating phagocytic leukocytes.

The Effect of Rancid perilla oil diet on the Immune Response in Mice (마우스에 있어서 부패들기름 식이가 면역반응에 미치는 영향)

  • The effect of rancid perilla oil on the immune response in mice was studied. ICR male mice were divided into 5 groups and were fed on the experimental diets for 4 weeks. Mice were sensitized and challenged with sheep red blood cell. Immune responses were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), Rosette forming cell and macrophage activity. Biochemical items were measured by serum protein and serum albumin. The weight of spleen, thymus and liver were measured. The rancid perilla oil diets decreased humoral and cellular immune responses, the number of peripheral circulating white blood cells and total protein and serum albumin. These results showed that the high rancid perilla oil diet decreased more humoral and cellular immune response, the number of peripheral circulating white blood cells, and total protein and serum albumin than the low rancid perilla oil diet did.

Increased production of human granulocyte-macrophage colony stimulating factor (hGM -CSF) by the addition of stabilizing polymer in plant suspension cultures

  • The effect of stabilizing polymer on hGM-CSF production was investigated in suspension cell cultures of transgenic tobacco. Secreted human GM -CSF from cell suspension cultures was detected in the medium at a maximum concentration of 180 ${\mu}g/L$ by ELISA. However, the secreted hGM -CSF was unstable in the medium, and rapidly degraded after day 5. In order to stabilize the secreted hGM-CSF, three stabilizing polymers were tested, polyethylene glycol, polyvinylpyrrolidone and gelatin. Gelatin was the most effective in stabilizing the secreted GM-CSF. Following the addition of 5% (w/v) gelatin, the maximum GM -CSF concentration reached 783 ${\mu}g/L$, a 4.6-fold increase over control.

  • Erythrosine used as a colouring agent in drugs, cosmetics and foods in Korea, was examined for its effects on murine immune system and methemoglobin formation. As immunotoxicologic assay parameters, we adopted circulating leukocytes and immunoorgan weights for pathotoxicology, IgM plaque forming cells and arthus reaction for humoral immunity, delayed hypersensitivity reaction of cell mediated immunity and carbon clearacnce for macrophage function. Erythrosine's effects were observed as follows; 1. Ery throsine showed no significant effects on circulating leulocyte counts and relative immunoorgan weight. 2. Erythrosine diminished IgM plaque forming cells. 3. Erythrosine decreased arthus reaction, in the dose dependent manner. 4. Erythrosine had no significant effect on delayed hypersensitivity. 5. Phagocytic and corrected phagocytic index were not affected. 6. Methemoglobin content was similar in the test and control groups.

Effect of Kamiinsamyangyoung-tang on Immune Response and Blood Formation

  • Kamiinsamyangyoung-tang(KIT) has been widely used to treat amenorrhea and bradymenorrea caused from vital energy and blood deficit. KIT was composed of Insamyangyoung-tang, Cervi Cornu Pantotrichum and Hominis Placenta. The aim of this study is to investigate effect of KIT on Immune response and Blood formation. We investigated thymocytes, splenocytes population, ${\gamma}$-interferon, IL-2, IL-4, NO and phagocytic activity. We found that KIT had no effect on the Th and Tc cell population of thymocytes, Th cell population of splenocytes and ${\gamma}$-interferon quantity was decreased. KIT decreased the formation of Nitric Oxide from abdominal macrophage, on the other hand, it had no influence on the quantity of IL-2, IL-4.

  • A case of necrotizing meningoencephalitis was described in a 3-year-old female pug dog. Significant histopathological changes were confined to the brain. Histopathologically, brain lesion of the pug dog was characterized by (1) severe lymphocyte, plasma cell and macrophage infiltration in the meninges and gray and white matter of the cerebral hemisphere, (2) perivascular mononuclear cell cuffing and (3) focal polioencephalomalacia in the gray matter of cerebrum. Replicate sections of the brain and lung were negative for fungal infection, canine distemper and toxoplasmosis on PAS stain and immunohistochemistry, respectively. Based on clinical signs, histopathological and immunohistochemical studies, this case was diagnosed as necrotizing meningoencephalitis in pug dog that had not been preriously described in Korea.

  • Tobacco cells transformed with hGM-CSF gene produced $162\;{\mu}g/L$ of hGM-CSF, a valuable therapeutic protein in seven days after inoculation. The protein concentration decreased after the maximum point. It is evidenced that the environment of cell culture was inappropriate for the stability of the protein(e.g., low osmolarity which can cause cell lysis).

  • To clarify the activating effects of Buthus martensi Karsch on immunological function, its effect on primary and secondary antibodies production in mice of various ages was investigated. Buthus martensi Karsch increased the number of both antibody producing cells(anti-IgM and anti-IgG producing plaque forming cells, PFC) and phagocytic activity of peritoneal macrophage. Futhermore, these phenomena were significantly increased with aging in mice. Buthus martensi Karsch also increased natural killer cell activity concerning to cancer immunology. These results suggest that Buthus martensi Karsch markedly increases the reduced activity in the elderly and activates the immune response in senescence mice.

Anti-Inflammatory Effect of Fermented Liriope platyphylla Extract in LPS-stimulated RAW 264.7 Macrophages

  • The present study was designed to evaluate the inhibitory effects of fermented Liriope platyphylla extract on the production of inflammation-related mediators (NO, ROS, NF-${\kappa}B$, iNOS and COX-2) and pro-inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, IL-6) in lipopolysaccharide-stimulated RAW 264.7 macrophages. Freeze-dried Liriope platyphylla was fermented by Saccharomyces cerevisiae and extracted with 70% ethanol. In lipopolysaccharide-stimulated macrophage cells, the treatment with fermented Liriope platyphylla extract decreased the generation of intracellular reactive oxygen species dose-dependently and increased antioxidant enzyme activities, including superoxide dismutase, catalase and glutathione peroxidase. Fermented Liriope platyphylla extract also inhibited NO production in lipopolysaccharide-stimulated RAW 264.7 cell. The expressions of NF-${\kappa}B$, iNOS, COX-2 and pro-inflammatory cytokines were inhibited by the treatment with fermented Liriope platyphylla extract. Thus, this study shows the fermented Liriope platyphylla extract could be effective at inhibiting the inflammation process.

The Study on the Effect of Gamiboatang (GMBT) on the Proliferation of Immune Function in Mice (가미보아탕(加味補兒湯)이 면역기능(免疫機能) 증진효과(增進效果)에 미치는 영향(影響))

  • This study was carried out to prove the effects of GMBT on the proliferation of splenic lymphocyte, active change of macrophage, T cell and B cell in continuous medication GMBT. The result were obtained as follows : 1. GMBT promote proliferation of splenic lymphocyte in vitro. 2. GMBT pretreated group was showed higher immune response than control group. 3. GMBT was not a rising effect in Con A but a rising effect in PHA. 4. GMBT treated group had highly producted more than 70% NO quality compared with control group. 5. GMBT was twice effect compared with control group in antibody production capacity of SRBC but meaningless m that of Anti-HBs Titer.
