Plant Production from Desiccated Somatic Embryos of Acanthopanax chiisanensis (지리오가피 (Acanthopanax chiisanensis) 체세포배의 건조처리를 통한 식물체 증식)

  • An efficient method of plant regeneration from Acanthopanax chiisanensis somatic embryos was developed. Cotyledonary somatic embryos were obtained in liquid Murashige and Skoog (MS) medium from embryogenic cell suspension cultures. They were desiccated for 0 to 72 hr and then cultured on MS medium containing NAA, BA, GA$_3$, (0-0.5mg/L). The highest multiple shoots formation (100%) was obtained from 72 hr desiccated somatic embryos on ifs medium with 0.5mg/L NAA+0.5mg/L BA or 0.5 mg/L NAA+0.5mg/L BA+0.5mg/L GA$_3$ after 6 weeks culture. Plant conversion from multiple shoots was not high. The highest plant conversion from multiple shoots was obtained on 1/3MS medium with 1.0mg/L GA$_3$. Converted plantlets were transferred to ex vitro condition and the highest survival rate (70%) of the plantlets was obtained on plastic pots containing vermiculite and sand. These results indicate that micropropagation procedure can be applied for an efficient mass propagation of Acanthopanax chiisanensis.

Mass Propagation by In Vitro Culture of Bupleurum latissimum Nakai (섬시호(Bupleurum latissimum Nakai)의 조직배양을 통한 대량생산)

  • This study was carried out to establish the micropropagation system of Bupleurum latissimum Nakai that is a Korean native endangered species. Callus were induced from the leaf, petiole and floral bud and the percentage of callus formation was highest in the floral bud on the MS medium containing 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D. Especially, callus induced from floral bud was formed 77.8% and the percentage of shoot formation was 42.6% on the MS medium containing 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$ $L^{-1}$ TDZ. For simultaneously callus formation and shoot regeneration, 1/2 MS medium was more effective than MS medium. The percentage callus formation, shoot regeneration and rooting were 46.3%, 13.0%, 13.0% in 1/2 MS medium, respectively. Soot regeneration from callus was good in 1/2 MS medium supplemented with 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$ $L^{-1}$ BA where percentage of shoot regeneration was 74.1 %, and the number of shoot per explant was 2.4. The percentage of rooting was lowest (57.8%) in control while it was highest (97.8%) in 1.5 mg ${\cdot}$ $L^{-1}$ NAA. In acclimatization of regenerated plantlets, the percentage of survived plantlets was highest (86.1%), and plant height, root length and fresh weight were good in the soil for horticulture.

Paralytic Shellfish Poisoning (PSP) Analysis using Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS를 이용한 마비성패류독소 분석조건 검토)

  • The AOAC Mouse Bioassay method (MBA) has been widely used for routine monitoring of paralytic shellfish poisoning (PSP) for more than 50 years. However, this method has low sensitivity and experiences interference from other components in the extract. Also, ethical issues have been raised against the continued use of this live-mouse assay. To establish an alternative method for PSP analysis, we attempted to develop PSP analysis conditions using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The LC-MS/MS analysis of reference material showed very reasonable accuracy, and the analysis time was just 15 min. However, the recovery rate of toxin spike samples using the LC-MS/MS analysis was 59.4-91.0%. We also attempted to remove the matrix effect using shellfish extracts, but recoveries of C1 and C2 did not improve. A comparison between the results of MBA and LC-MS/MS analysis revealed good correlations, with values of 0.8878 and 0.9211 for oyster and mussel matrices, respectively.

Method Development for Determination of Trichothecene Mycotoxins in Nuts by LC-MS/MS (LC-MS/MS를 이용한 견과류 중 트리코테센계 곰팡이 독소 10종 동시분석법 개발)

  • This study presents a method validation for extraction and quantitative analysis of trichothecene mycotoxins in nuts based on quick, easy, cheap, effective, rugged, and safe (QuEChERS) approach for extraction and enhanced matrix removal (EMR)-lipid-disperive-SPE (d-SPE) cleanup method, with detection and quantification by high-performance liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) in positive- and negative-ion modes. Linearity, precision, and accuracy were validated for LC-MS/MS methods. Results obtained with LC-MS/MS were linear, with correlation coefficient ($R^2$) of 0.998. Limits of detection and quantification for mycotoxins were $0.41-3.57{\mu}g/kg$ and $1.23-10.82{\mu}g/kg$, respectively. Intra- and inter-day precisions (RSD, %) were 0.40-8.44% and 1.93-12.46%, respectively. Results indicated to be rapidly and accurately identifying trichothecene mycotoxins and may be used as a suitable safety management method for nuts and nuts related commodities.

In Vitro Propagation of Cindium officinale Makino Through Shoot Tip Culture (천궁의 경정배양을 통한 기내번식)

  • This experiment was conducted to identify the optimal in vitro propagation condition of Cnidii rhizoma (Cnidium officinale Makino). It was effective to reduce contamination and improve regeneration of shoot when shoot tips taken in July were cultured in 1/2 strength Murashige and Skoog medium supplemented with 500 mg/L carbenicillin disodium 1.0 mg/L BA and 1.0mg/L $GA_3$followed by surface sterilization of explant source in solution of 1% sodium hypochlorite for 20 minutes. When shoot tips were 쳐cultured in 1/2 strength MS medium with 0.5 mg/L BA and 60 g/L sucrose, shoot elogation and subsequent multiplication of the formed shoot were favorable than in other media. Regenerants were well rooted in 1/2 strength MS medium containing 3.0 mg/L NAA.

Determination of phenols in water by derivatization and GC/MS (유도체화와 GC/MS를 이용한 물중의 페놀류 분석)

  • Phenol compounds in water were simultaneously analyzed by extractive derivatization. Aqueous solution was controlled to pH 13 with sodium hydroxide, and then $500{\mu}L$ of benzoyl chloride was added to the solution. The solution was shaken for 15 minutes and extracted with diethyl ether and then analyzed using GC/MS-SIM. The calibration curves of phenolic compounds in the range of 0.05 to $5.0{\mu}g/mL$ showed a good linearity having the correlation coefficient of $r^2=0.9915$ and standard deviation of ${\leq}8.5%$, respectively. The recoveries by this method ware 58.4-114.0%.

  • The aim of this study was to investigate the bacteriophage removal in various clay minerals and clay-amended soils. Batch experiments in kaolinite, montmorillonite, and bentonite showed that kaolinite was far more effective at the MS2 removal than montmorillonite and bentonite. In kaolinite, the log removal increased from 0.046 to 2.18, with an increase in the adsorbent dose from 0.3 to $50g\;L^{-1}$, whereas the log removals in montmorillonite and bentonite increased from 0.007 to 0.40 and from 0.012 to 0.59, respectively. The MS2 removal in kaolinite-amended silt loam soils was examined at three different soil-to-solution (STS) ratios. Results indicated that the log removal of MS2 increased with an increase in the kaolinite content and the STS ratio. At the STS ratio of 1:10, the log removal of MS2 increased from 2.33 to 2.80 with an increase in the kaolinite content from 0% to 10% in kaolinite-amended soils. The log removals of MS2 at the STS ratios of 1:2 and 1:1 increased from 2.84 to 3.47 and from 3.46 to 4.76, respectively, with an increase in the kaolinite content from 0% to 10%. Results also indicated that the log removals of PhiX174 and $Q{\beta}$ in kaolinite-amended soils were similar to each other, but they were far lower than those of MS2 at all the kaolinite contents. The log removal of PhiX174 increased from 0.16 to 0.32, whereas the log removal of $Q{\beta}$ changed from 0.17 to 0.22 with an increase in the kaolinite content from 0% to 10%.

  • This work proposes a 14b 150MS/s 0.13um CMOS ADC for SDR systems requiring simultaneously high resolution, low power, and small size at high speed. The proposed ADC employs a calibration-free four-step pipeline architecture optimizing the scaling factor for the input trans-conductance of amplifiers and the sampling capacitance in each stage to minimize thermal noise effects and power consumption at the target resolution and sampling rate. A signal- insensitive 3-D fully symmetric layout achieves a 14b level resolution by reducing a capacitor mismatch of three MDACs. The proposed supply- and temperature- insensitive current and voltage references with on-chip RC filters minimizing the effect of switching noise are implemented with off-chip C filters. The prototype ADC in a 0.13um 1P8M CMOS technology demonstrates a measured DNL and INL within 0.81LSB and 2.83LSB, at 14b, respectively. The ADC shows a maximum SNDR of 64dB and 61dB and a maximum SFDR of 71dB and 70dB at 120MS/s and 150MS/s, respectively. The ADC with an active die area of $2.0mm^2$ consumes 140mW at 150MS/s and 1.2V.

In Vitro Propagation of Lilium Asiatic Hybrid 'Hae Hwa' via the Formation of Shoot Clusters (신초 Cluster 형성에 의한 Lilium Asiatic Hybrid 'Hae Hwa'의 기내번식)

  • This experiment was conducted to micropropagate bulblets via shoot cluster formation and massproduce normal bulblets from the sections of proliferated shoot clusters in Lilium asiatic hybrid 'Hae Hwa'. The induction of shoot clusters from the culture of bulblet sections was more effective than that of bulb scales on MS medium with 1.0 mg/L BA and 0.5 mg/L IAA. Proliferation of shoot clusters from the formed shoot cluster sections was the most favorable on medium containing 5.0 mg/L BA and 0.5 mg/L IAA. The formation and the growth of bulblets from shoot cluster sections were achieved effectively on medium with 60∼90 g/L sucrose. The leaves derived from shoot clusters grew vigorously but the bulblets from shoot clusters grew very poor in 5L air-lift bioreactor culture. By the addition of 30 mL fresh liquid medium containing doulble strength MS salts, 250 g/L sucrose and 5 g/L activated charcoal after 8 weeks in the shoot cluster culture on MS medium with 5.0 mg/L BA and 0.5 mg/L IAA, the number of bulblets was increased in light condition, but the growth of bulblets was not affected by light. Bulblet production was possible with the bulblet product at 53 to 68 mg in fresh weight by liquid medium addition after the proliferation of shoot cluster.

A Study on the Characteristics of Echolocation Signals of the Common Dolphin, Delphinus Delphis (참돌고래의 반향정위 신호특성에 관한 연구)

  • The characteristics of echolocation signals of the Common Dolphin, Delphinus Delphis was observed by the hydrophone in order to detect exactly distribution and migration on whales and dolphins in Korean Coastal waters. It's observation was carried out at the position of 13 mm off Gam-Po of Korean east-southern sea at 3rd-5th. April and 13th-15th. October, 1999. The results obtained are summarized as follows: (1) The frequency range of ship's noise and ambient noise in the observed station was 0.5-0.3 kHz, that ones could be influenced to the behavior of common dolphins which carry out echolocation using low-frequency. (2) The common dolphin was radiated single click of 8.6 ms and double click of 4.8 ms pulse width during these observation (3) The high click frequencies of common dolphin were 5.10 kHz, 7.22 kHz, 10.60 kHz with the click pulse width of 4.0 ms, 2.6 ms, 1.0 ms, respectively. In case of low-frequency 1-2 kHz, that is, 1.12 kHz, 1.38 kHz, 1.82 kHz, pulse width were 22.4 ms, 2.05 ms, 11.9 ms, respectively and they showed a tendency using triple click signal. (4) The pulse width, pulse recurrence interval and frequency range of the observed echolocation signals were 2.4-8.4 ms, 9.0-40.0 ms, 0.60-10.63 kHz respectively, and frequency spectrum level was 100-125 dB for single, double, triple click signals.
