Anti-Proliferative Activity of Naturally Occurring Flavonoids on Cultured Human Tumor Cell Lines (천연 유리 Flavonoid 화합물들의 암세포성장 저해효과)

  • The flavonoids are a very large and important group of polyphenolic natural products, which are united by their derivatization from the heterocycle, flavone. They are distributed in higher plants and occur widely in the fruits and vegetables that make up the human diet. They exhibit a wide range of biological properties, including antitumor, antiinflammatory, hepatoprotective, antimicrobial, insecticidal and estrogenic activities. They are also major components of many plant drugs and it is possible that they contribute to the curative properties. For the purpose of developing anticancer agent of natural origin, we have evaluated forty four kinds of naturally occurring flavonoids for the inhibitory activity upon the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro.

Active Principles of the Methanol Extract of Korean Mistletoe Responsible for the Inhibitory Effect on the Proliferation of Human Tumor Cell Lines (한국산 겨우살이 전초의 Methanol 추출물로부터 암세포증식 저해성분의 분리)

  • A bioassay-guided fractionation of the whole extract of Viscum album var. coloratum Ohwi (Loranthaceae) led to the isolation of two triterpenoidal components; oleanolic acid (1) and ${\beta}-amyrin$ acetate (2), and a flavonoid, homoflavoyadorinin B (3) as well as large quantity of free fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble fraction and BuOH soluble fraction of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system), and HCT-15 (colon) in vitro, whereas the remaining water soluble fraction exhibited a poor inhibition. The intensive phytochemical investigation of the EtOAc soluble fraction and BuOH soluble fraction of the extract indicated that the oleanolic acid (1) and large amounts of free fatty acid mixtures might be attributed to the in vitro antitumoral property of the whole extract of Viscum album var. coloratum.

  • The voice has distinguishable acoustic properties of speaker as well as transmitting information. The speaker recognition is the method to figures out who speaks the words through acoustic differences between speakers. The speaker recognition is roughly divided two kinds of categories: speaker verification and identification. The speaker verification is the method which verifies speaker himself based on only one's voice. Otherwise, the speaker identification is the method to find speaker by searching most similar model in the database previously consisted of multiple subordinate sentences. This paper composes feature vector from extracting MFCC coefficients and uses the dynamic time warping algorithm to compare the similarity between features. In order to describe common characteristic based on phonological features of spoken words, two subordinate sentences for each speaker are used as the training data. Thus, it is possible to identify the speaker who didn't say the same word which is previously stored in the database.

  • Intraspecific fusion frequency between Filobasidium capsuligenum CBS6122-2 ade and met trp or met strains was $2.9\times 10^{-3}$ and $8.5\times 10^{-3}$, respectively. And intergeneric fusion frequency between Folobadidium capsuligenum CBS 4318 (cys his) and Saccharomyces cerevisiae 262-12-1 (hom3 thr1) or X2180-1A (ade thr) was $8.8\times 10^{-6}$ and $9.5\times 10^{-4}$, respectively. Nuclear fusion appears to occru in fusants between intraspecies and intergenera as strongly suggested by DNA content, nuclear staining, comparison of survival rate to UV light and mitotic segregation analysis. It was also found that $\alpha$-amylase and glucoamylase activity from intraspecific hybrids was 1.6-2.1 fold increased when compared with that from thier parents.

Feature Extraction Algorithm for Underwater Transient Signal Using Cepstral Coefficients Based on Wavelet Packet (웨이브렛 패킷 기반 캡스트럼 계수를 이용한 수중 천이신호 특징 추출 알고리즘)

  • In general, the number of underwater transient signals is very limited for research on automatic recognition. Data-dependent feature extraction is one of the most effective methods in this case. Therefore, we suggest WPCC (Wavelet packet ceptsral coefficient) as a feature extraction method. A wavelet packet best tree for each data set is formed using an entropy-based cost function. Then, every terminal node of the best trees is counted to build a common wavelet best tree. It corresponds to flexible and non-uniform filter bank reflecting characteristics for the data set. A GMM (Gaussian mixture model) is used to classify five classes of underwater transient data sets. The error rate of the WPCC is compared using MFCC (Mel-frequency ceptsral coefficients). The error rates of WPCC-db20, db40, and MFCC are 0.4%, 0%, and 0.4%, respectively, when the training data consist of six out of the nine pieces of data in each class. However, WPCC-db20 and db40 show rates of 2.98% and 1.20%, respectively, while MFCC shows a rate of 7.14% when the training data consists of only three pieces. This shows that WPCC is less sensitive to the number of training data pieces than MFCC. Thus, it could be a more appropriate method for underwater transient recognition. These results may be helpful to develop an automatic recognition system for an underwater transient signal.

Purification and Characterization of CDMHK, a Growth Inhibitory Molecule Against Cancer Cell Lines, from Myxobacterium sp. HK1 Isolated from Korean Soil

  • Myxobacterium sp. HK1, isolated from Korean soil, degrades cellulose, differentiates to fruiting body, and its 16s rDNA has $95\%$ similarity to Polyangium sp. An anticancer molecule, CDMHK, was identified from culture broth of Myxobacterium sp. HK1, and purified by Diaion HP20, Silica gel, Sephadex LH-20 chromatography, and preparative HPLC using an YMC OSD-A C18 column. The molecular structure and formula were determined to be $C_{l2}H_{l9}N_3O_2$ (M.W 237) by MS spectrometry, 300 MHz $^{1}H\;and\;^{13}C$ NMR. The CDMHK was not active against Escherichia coli, Staphylococcus aureus, and Candida albicans. However, this molecule inhibited the growth of various cancer cell lines. The $ED_{50}$ values of CDMHK were determined to be 0.147, 0.086, 0.18, 0.166, and 0.142 $\mu$g/ml against A549, SK-OV-3, SK-MEL-2, VF498, and HCTl5 cancer cell lines, respectively. In addition, the CDMHK was able to induce apoptosis of the CCRF-CEM cancer cell line, evidenced by DNA fragmentation assay and DAPI staining.

Expression, Purification, and Characterization of C-Terminal Amidated Glucagon in Streptomyces lividans

  • Glucagon, a peptide hormone produced by alpha-cells of Langerhans islets, is a physiological antagonist of insulin and stimulator of its secretion. In order to improve its bioactivity, we modified its structure at the C-terminus by amidation catalyzed by a recombinant amidase in bacterial cells. The human gene coding for glucagon-gly was PCR amplified using three overlapping primers and cloned together with a rat ${\alpha}$-amidase gene in plasmid pMGA. Both genes were expressed under control of the strong constitutive promoter of aph and secretion signal melC1 in Streptomyces lividans. With Phenyl-Sepharose 6 FF, Q-Sepharose FF, SP-Sepharose FF chromatographies and HPLC, the peptide was purified to about 93.4% purity. The molecular mass of the peptide is 3.494 kDa as analyzed by MALDI TOF, which agrees with the theoretical mass value of the C-terminal amidated glucagon. The N-terminal sequence of the peptide was also determined, confirming its identity with human glucagon at the N-terminal part. ELISA showed that the purified peptide amide is bioactive in reacting with glucagon antibodies.

Cytotoxic Effect of Flavonoids from the Roots of Glycyrrhiza uralensis on Human Cancer Cell Lines (감초(Glycyrrhiza uralensis Fisch.)로부터 분리된 flavonoid의 인체 암세포에 대한 세포독성)

  • The roots of Glycyrrhiza uralensis Fisch. were extracted with 30% aqueous ethanol (EtOH), and the concentrated extract was partitioned with n-hexane, chloroform ($CHCl_3$), ethyl acetate (EtOAc), n-butanol (n-BuOH), and $H_2O$, successively. From the $CHCl_3$ fraction, four flavonoids were isolated through the repeated silica gel ($SiO_2$), octadecyl silica gel (ODS), and Sephadex LH-20 column chromatographies (c.c.). According to the results of spectroscopic data including nuclear magnetic resonance spectrometry (NMR), electron ionization mass spectrometry (EI/MS), and infrared spectroscopy (IR), the chemical structures of the compounds were determined as glabrol (1), abyssinone II (2), glabridin (3), and isoliquiritigenin (4). The flavonoids were evaluated for cytotoxic effect against human cancer cell lines, HCT-116, HepG2, HeLa, SK-OV-3, SK-BR-3, MCF-7, and SK-MEL-5. Especially, glabrol (1) and glabridin (2) showed $IC_{50}$ values of lower than $25{\mu}M$.

Study on Antitumor Activity and Antimetastatic effect of Kamigumguesingihwan(KGSH) (가미금궤신기환(加味金櫃腎氣丸)의 항암(抗癌) 및 항전이(抗轉移) 효과(效果)에 관(關)한 연구(硏究))

  • To evaluate the antitumor activity and antimetastatic effects of Kamigumgusingihwan(KGSH) studies have ken done. The results were obtained as follows: 1. KGSH extracts exhibited a weak cytotoxicity against A549, SK-OV-3, B16-F10, and SK-MEL-2 cell lines. But exhibited potent cytotoxicity against P388 cell line in a dose-dependent manner. 2. The concentration inhibiting adhesion of A549, to complex extracellular matrix up to below 30% of control was recognized at $10^{-3}g/ml$ of KGSH 3. KGSH extracts showed a weak inhibitoty effect on DNA topo-isomerase I from calf thymus. 4. The T/C% was 137% in KGSH treated group in S-180 bearing ICR mice. 5. In pulmonary colonization assay, a number of colonies in the lungs were decreased significantly in KGSH treated group as compared with control group. 6. In hematological changes in B16-BL6 injected C57BL/6, numbers of WBC were decreased insignificantly in KGSH treated groups, and also those of platelet were increased insignificantly in KGSH treated groups as compared with control. 7. In CAM assay, KGSH extracts inhibited angiogenesis at $15{\mu}g/egg $concentration significantly as compared with control. Taken together these results, it is strongly demonstrated that KGSH significantly suppressed tumor metastasis by blocking cell adhesion to extracellular matrix. Therefore, KGSH is expected to be clinically a potent antimetastatic drug for the prevention and treatment of cancer.

KHG26792 Inhibits Melanin Synthesis in Mel-Ab Cells and a Skin Equivalent Model

  • The purpose of this study is to characterize the effects of KHG26792 (3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride), a potential skin whitening agent, on melanin synthesis and identify the underlying mechanism of action. Our data showed that KHG26792 significantly reduced melanin synthesis in a dose-dependent manner. Additionally, KHG26792 downregulated microphthalmia-associated transcription factor (MITF) and tyrosinase, the rate-limiting enzyme in melanogenesis, although tyrosinase was not inhibited directly. KHG26792 activated extracellular signal-regulated kinase (ERK), whereas an ERK pathway inhibitor, PD98059, rescued KHG26792-induced hypopigmentation. These results suggest that KHG26792 decreases melanin production via ERK activation. Moreover, the hypopigmentary effects of KHG26792 were confirmed in a pigmented skin equivalent model using Cervi cornus Colla (deer antler glue), in which the color of the pigmented artificial skin became lighter after treatment with KHG26792. In summary, our findings suggest that KHG26792 is a novel skin whitening agent.
