Effect of Cultural Media and Conditions on the Mycelial Growth of Pholiota adiposa (배지와 배양조건이 검은비늘버섯의 균사생장에 미치는 영향)

  • To elucidate the optimal conditions of mycelial growth of Pholiota adiposa, the effect of a kinds of media, incubation temperature and shaking culture were investigated. Mushroom complete medium was selected as superior media for mycelial growth in this fungus. The optimal temperature for mycelial growth of this fungus shows at 25℃. Shaking culture could be obtain the mycelia in two or three times than the stationary culture.

Interorder Protoplast Fusion between Pleurotus ostreatus and Ganoderma applanatum (느타리버섯과 잔나비걸상버섯과의 이목간(異目間) 원형질체(原形質體) 융합(融合))

  • Interorder heterokaryons were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Pleurotus ostreatus in agaricales and Ganoderma applanaturm in aphyllophorales. When transferred to MMM plates, all fusion colonies exhibited an extremely growth rate. During three times subcultivation on MCM the growth rate of fusants showed faster little by little. Seventy-five % fusion products of potoplasts showed mixed morphologies between those of P. ostreatus and G. applannatum in the first subcultivation on MCM and MGM. The phenotype of these fusants changed similar those of P. osteatus type after three times subcultivation on MCM. However, phenotype of 25% stable strains did not change on subcultivation. Hyphae of all fusion products did not form true clamp connection. All these types did not produce primordia. A comparrison of interorder somatic hybrids was made using isozyme analysis of esterase, malate dehydrogenase and peroxidase. In most cases the enzyme patterns of G. applanatum were not distinct, however, fusant showed non-parental bands.

  • During the development of flight software, the processor emulator and satellite simulator are essential tools for software development and verification, which can be substituted for the actual hardware. LEO satellites being developed by KARI recently use the MCM-ERC32SC processor for on-board computer (OBC). For the flight software (FSW) development and testing, the software-based spacecraft simulator was developed using TSIM-ERC32 processor emulator from Aeroflex Gaisler. It is needed to get rid of the constraints and dependencies of TSIM-ERC32 processor emulator and to obtain high performance processor emulator to develop full satellite simulator. This paper presents the development of the ERC32 emulator based on open source dynamic translator, QEMU, as the first step. And it describes the software development and testing/debugging on the developed emulator.

Viral Effects of a dsRNA Mycovirus (PoV-ASI2792) on the Vegetative Growth of the Edible Mushroom Pleurotus ostreatus

  • A double-stranded RNA (dsRNA) mycovirus was detected in malformed fruiting bodies of Pleurotus ostreatus strain ASI2792, one of bottle cultivated commercial strains of the edible oyster mushroom. The partial RNA-dependent RNA polymerase (RdRp) gene of the P. ostreatus ASI2792 mycovirus (PoV-ASI2792) was cloned, and a cDNA sequences alignment revealed that the sequence was identical to the RdRp gene of a known PoSV found in the P. ostreatus strain. To investigate the symptoms of PoV-ASI2792 infection by comparing the isogenic virus-free P. ostreatus strains with a virus-infected strain, isogenic virus-cured P. ostreatus strains were obtained by the mycelial fragmentation method for virus curing. The absence of virus was verified with gel electrophoresis after dsRNA-specific virus purification and Northern blot analysis using a partial RdRp cDNA of PoV-ASI2792. The growth rate and mycelial dry weight of virus-infected P. ostreatus strain with PoV-ASI2792 mycovirus were compared to those of three virus-free isogenic strains on 10 different media. The virus-cured strains showed distinctly higher mycelial growth rates and dry weights on all kinds of experimental culture media, with at least a 2.2-fold higher mycelial growth rate on mushroom complete media (MCM) and Hamada media, and a 2.7-fold higher mycelial dry weight on MCM and yeast-malt-glucose agar media than those of the virus-infected strain. These results suggest that the infection of PoV mycovirus has a deleterious effect on the vegetative growth of P. ostreatus.

  • The results of examining characteristics of mycelial growth and culture condition for determining the condition of artificially culturing Fomitopsis pinicola are as follows. 1) Mycelial growth and density of F. pinicola. were the highest in the medium of PIDA(Pine Dextrose Agar;66.3mm/10d) followed by the order of GDA, PDA, CDA, PODA, ODA, YM, MCM, MEA(pH 4.7), CHA, and MEA(pH 4.7). 2) Optimal temperature for the mycelial growth and density of F. pinicola were shown to be $30^{\circ}C$, but the mycelia were dead at $40^{\circ}C$. the mycelial growth and density of KNAC9005 strains was the highest at $30^{\circ}C$(66.3mm/10d) followed by the order of 25, 20, 15, 35, 10, and $5^{\circ}C$. 3) Optimal pH for the mycelial growth and density of $40^{\circ}C$ was revealed to be 6.0(88.4mm/10d). above or below pH 6.0, the mycelial growth and density were shown to be retarded. 4) Optimal carbon, nitrogen and organic acid sources for the spawn growth of $40^{\circ}C$ were maltose(331mg/25ml/15d), peptone(347mg/25ml/15d), and glutamic acid(357mg/25ml/15d), respectively. Optimal level of biotin was 370mg/15d and optimal C/N ratio was 40.

Cultivation technique using plastic container and selection the superior strain of nameko mushroom (Pholiota nameko) (맛버섯(Pholiota nameko) 우량균주 선발 및 병 재배법)

  • These experiment was conducted to find the superior strain selection, cultivation technique and optimum environmental condition of nameko mushroom culture using plastic container. The results was following as Mycelium of Pholiota nameko grown well at MCM and Hamada media, and its media acidity was pH 6~7. The optimum temperature condition for growing mycelium was $25^{\circ}C$. Under $15^{\circ}C$ and above $30^{\circ}C$ of temperature condition, mycelium growing speed was delayed remarkably. Among the 29 strains of nameko mushroom, the most productive strains was JNM19007, JNM19026, JNM19027 and JNM19028. The optimum media composition rate for produce fruitbody was pine sawdust 80% + wheat bran 20%. In this condition, the average fruitbody amount was 188g per 1,100cc container. The optimum post-culturing period was 50 days and mushroom sprout appeared 7 days after old mycelia removed. The suitable temperature was $12^{\circ}C$ for induce sprout, growing period was $16^{\circ}C$ and the optimum relative humidity was 95% in all culturing periods.

Analysis of genetic characteristics and development of substrate for cultivation in brown strains of Flammulina velutipes (갈색팽이버섯의 유전적 특성검정 및 배지재료 개발)

  • Analysis of genetic characteristics of Flammulina velutipes showed that strains have a range of 85% in genetic distribution diagram. According to this result, we divided these strains into five groups. In experiment of determinating the optimum media and condition in cultivating F. velutipes, we found the optimum temperature and pH range for hypha growth were $25^{\circ}C$ and 6.0 to 7.0, respectively. in addition, the best media for growth of that in plate was MCM (Mushroom Complete Media) which have a growth length from 68 to 83 mm. In vivo test, we observed that fast growth and good density of hyphae in mixture media of douglas fir sawdust, cotton seed meal and beet pulp (6:2:2 V/V). Also when we cultivated F. velutipes in this media, we harvested high yield of fruiting body.

Favorable Condition of Culture and Sclerotial Formation by Inonotus obliquus (차가버섯(Inonotus obliquus)의 배양조건과 균핵 형성)

  • Altogether twenty isolates were collected from Wakayama, Japan. The optimum mycelial growth of I. obliquus was observed in BMYA and MCM. The optimum temperature and pH for the mycelial growth were $25^{\circ}C\;and\;6.0{\sim}7.0$, respectively. The optimum carbon and nitrogen sources were dextrose and yeast extract, respectively. Similarly, the optimum mineral salt was $K_{2}HPO_{4}$. The optimum number of mycelial discs for the mycelial growth was $6{\sim}7$ per 100 ml. Similarly, the optimum culture period was $21{\sim}22$ days in liquid broth. The optimum brown rice : water ratio was 1 : 1. No difference in mycelial growth was observed in all the four types of tree stumps used.

  • Coriolus versicolor, is one of the most popular medicinal mushrooms due its various biologically active components. This study was conducted to obtain basic information regarding the mycelial culture conditions of C. versicolor. Based on the culture, and MCM media were suitable for the mycelial growth of the mushroom. The optimum carbon and nitrogen sources were dextrin and yeast extract, respectively, and the optimum C/N ratio was 10 to 2 when 2% glucose was used. Other minor components required for optimal growth included thiamine-HCl and biotin as vitamins, succinic acid, lactic acid and citric acid as organic acids, as well as $MgSO_4{\cdot}7H_2O$ as mineral salts.
