Twelve species of food microalgae were investigated to clarify the digestion index of Crassostrea gigas larvae using epifluorescence microscopy to choose an appropriate diet for artificial seed production in hatchery. An experiment was conducted using 1 (D shaped stage), 4 (Early umbo stage), 8 (umbo stage) and 12 (Full grown stage) days old larvae. larvae were stocked in 1 L flasks at 5 individuals/mL and fed $10{\times}10^4$ algal cells/mL of each species individually. Prior to larvae were fed for 3 h and then were observed under the microscope to detect ingestion; larvae were then sieved and replaced in 1 L flasks containing filtered seawater and were observed after 3, 5 and 8 h to analyse the digestion index. Values of digestion indices were specific for each alga. No evidence for the ingestion of Thalassiosira weissflogii was evident at all larval development stages tested. Digestion indices of others microalgae were 0.8-99.7% at 4 stage of larval development stages: Chlorella ellipsoidea (0.8-5.4%), Nannochloris oculata (1.4-5.0%), Isochrysis galbana (99.1-99.5%), Pavlova lutheri (99.1-99.5%), I. aff. galbana (99.4-99.5%), Cheatoceros calcitrans (0.0-99.2%), C. gracilis (0.0-99.7%), C. simplex (0.0-95.9%), Phaeodactylum tricornutum (0.0-99.6%), Tetraselmis tetrathele (0.0-99.7%) and Dunaliella tertiolecta (0.0-99.6%), respectively. Therefore, it is assumed that food microalgae showing the high digestion such as I. galbana should be supplied to the early umbo stage larvae, and then after the umbo larval stage, the mixed microalgae with diatoms and light green algae should be supplied to the full grown stage larvae to increase the digestion of their larvae.

In the present study, twenty eight marine algae species were evaluated for their antiproliferative effect on HT-29 human colon cancer cells. Among these, the methanolic extract of Symphyocladia latiuscula (SL Ex) showed the highest inhibitory activity on HT-29 cell growth. In this study, we examined the mechanism by which SL Ex inhibited the HT-29 cell growth. Cells were cultured with various concentrations of $(0{\sim}20{\mu}g/mL)$ SL Ex. The SL Ex substantially decreased the viable cell numbers and induced apoptosis of HT-29 cells in a dose-dependent manner Western blot analyses of total cell lysates revealed that SL Ex increased the levels of cleaved caspase-8, -9, -7, and -3, and poly (ADP-ribose) polymerase in HT-29 cells. In addition, SL Ex increased truncated Bid levels but moderately decreased Bax levels at only $20{\mu}g/mL$. Furthermore, SL Ex did not affect Bcl-2 protein levels but increased the levels of Fas in HT-29 cells. The present results indicate that SL Ex inhibits cell growth via inducing apoptosis in human colon cancer cells. The mechanism of apoptosis induction by SL Ex involves caspase-8 activation leading to changes in mitochondrial events and subsequent activation of the caspase-7/caspase-3 cascade. Our finding may lead to the development of new therapeutic strategies for the treatment of colon cancer.

The distributions of phytoplankton assemblages and environmental factors in Jinhae Bay and their relationships were investigated to estimate the potential limiting nutrient for phytoplankton growth and community structure. In situ algal bioassay experiments were also conducted to assess the species-specific characteristics in phytoplankton responses under different nutrient conditions (control, N(+) and P(+) treatment). During the study periods, bacillariophyceae and cryptophyceae occupied more than 90% of total phytoplankton assemblages. Phytoplankton standing crops in the inner part of Masan Bay were higher than that of Jinhae Bay. The DIN:DIP ratio, pH and transparency showed the significant positive correlation with phytoplankton biomass. According to cluster and multidimensiolnal scaling (MDS) analysis based on phytoplankton community data from each station, the bay was divided into three groups. The first group included stations from the south-western part of Jinhae bay where cryptophyta species were dominated. The second group was distinguished from inner stations in Masan Bay. These stations showed low transpancy and high DIN:DIP ratio. The other cluster included the stations from the eastern part and central part of Jinhae Bay, which was characterized by the high DSi:DIP ratio and dominant of diatom species. Phosphorous (P) was limited in Masan Bay due to significantly increases in the phytoplankton abundances. Based on stoichiometric limitation and algal bio-assay in Jinhae Bay, nitrogen (N) was a major limiting factor for phytoplankton production. However, silicate (Si) was not considered as limiting factor, since Si/DIN and Si/P ratio and absolute concentration of nutrient did not create any potential stoichiometric limitation in the bay. This implies that high Si availability in winter season contributes favorably to the maintenances of diatom species.

Marine algae have long been recognized as a health and beauty food, based on its anti-tumor, anti-inflammatory and anti-obesity activities. In this study, methanol extracts were prepared from 10 different phaeophyta, after which DPPH radical scavenging and cytoprotective activities of HT-22 cells against ${\beta}$-amyloid protein ($A{\beta}$), which has neurotoxic effects, were investigated. In DPPH experiments, Ecklonia cava and Ishige okamurai showed strong ROS scavenging activities, whereas eight other phaeophyta including Petalonia binghamiae (P. bin) showed weak ROS scavenging activities. To validate the cytoprotective effects of 10 different phaeophyta in $A{\beta}$-induced HT-22 cells, protein expression levels of APP, BACE1, iNOS, phosphorylated ERK1/2, phosphorylated p38 and phosphorylated JNK1/2 were determined along with MTT assay. In the MTT assay, P. bin showed the best effective cytoprotective activity at a concentrations of $25{\mu}g/mL$, whereas Sargassum confusum, Colpomenia sinuosa, Myelophycus simplex, and Sargassum hemiphyllum showed potential. Determination of protein expression levels related to $A{\beta}$-induced neurotoxicity in the five selected phaeophyta showed that P. bin inhibited BACE1 and iNOS expression in $A{\beta}$-induced HT-22 cells. These results indicate that the cytoprotective effects of P. bin are mediated by suppressing the pathways involving $A{\beta}$-induced ERK and p38 activation.
Growth optima and tolerance ranges of marine algae species nay vary greatly during the developmental stage. The establishment stage is clearly of great importance for each generation, affecting the performance of the adult population. The present study was aimed to determine the effect of various concentrations of inorganic nutrients and heavy metals on germination and germlings of Ulva pertusa Kjellman (Chlorophyta). Percent germination increased rapidly as irradiance level increased, reaching the maximum at 100 ${\mu}$mol m$\^$-2/s$\^$-1/ in both east and west seawater. Percent germination in the east seawater was higher than that in the west seawater at irradiances lower than 30 ${\mu}$mol m$\^$-2/s$\^$-1/, and there was no difference in percent germination between the two different seawaters at irradiances higher than 60 ${\mu}$mol m$\^$-2/s$\^$-1/. Germling growth increased in the both cases with increasing irradiances but no growth was found in the dark. Overall germling area was larger in the east seawater than in the west seawater. Number of cell increased with increasing irradiance and became light-saturated at 100 ${\mu}$mol m$\^$-2/s$\^$-1/ in the both cases. Germlings grown in the east seawater had more cells than those cultivated in the west seawater at irradiances lower than 60 ${\mu}$mol m$\^$-2/s$\^$-1/. In various combinations of nitrate and phosphate, percent germination increased with increasing nitrate concentrations irrespective of phosphate concentrations. At 3 days after inoculation, nitrate concentration of 2.5 ppm with all phosphate concentrations promoted germination. Area and number of cell of germlings increased with increasing nitrate concentrations in all phosphate concentrations. In various combinations of copper and lead, germination was delayed as copper concentrations increased in all lead concentrations. Percent germination was severely reduced at 1 ppm Cu$\^$2+/. Area and number of cell of germlings decreased with increasing copper concentrations in all lead concentrations. The highest copper concentration (1 ppm) clearly suppressed the germling growth in U. pertusa.
For the fundamental studies of radiation breeding in edible marine algae, the biological effects on conchospores of Porphyra species by gamma-irradiation were examined. Two varieties, Keun-cham-gim (Porphyra tenera Kjell. form tamatsuensis Miura) and Saga No.5, were chosen for this study, and their conchospores after r-irradiation($5.0{\sim}20.0$ KR) were cultured for 50 days. The results obtained were summarized as follows. 1. Gamma-irradiation in less than the dose of 20KR did not affect germination of conchospores, and almost all spores grew into two cells germ in 24 hours after irradiation, but withering germs were gradually increased in number according to higher exposure within 5 days old culture. 2. High irradiation caused the induction of giant cells, abnormal useless growth of hold-fast, lumpish thalli and callus-like lumpy tissues. 3. The liberation of neutral spores from young germs and carpospores from mature thalli were observed on the frond exposed at $10{\sim}20$ KR irradiation. All spores were normal in division and its size. 4. The best irradiation effect on growth of Keun-cham-gim was observed at 10 KR dose, whose growth-rates were $140\%$ in wet weight and $108\%$ in mean frond area, but only $48\%$ was recorded in wet weight at 20 KR exposure. Saga No.5 were in contrast with Keun-cham-gim, and their most growth-rate was $400\%$ in wet weight ($258\%$ in frond area) at 10 KR irradiation and the worst was $20\%$ at the dose of 20 KR. 5. The withering phenomenon to death by treatment of gamma-ray presented substantial difference between two varieties. Survival rate compared with control in Keun-cham-gim was $70.7\%$ at 20 KR, but that in Saga No.5 recorded $47.0\%$ at same dose. 6. Synthesizing the results of high and low r-irradiation, it was suggested tat high r-irradiation in more than 5.0 KR inhibited conspicuously the growth of germs derived from conchospores, and about half of them withered at 15.0 KR dose or more.

Sargassum macrocarpum is a widely distributed marine brown algae found in the North Pacific. The objective of this study was to evaluate the anti-inflammatory activity of an ethanol extract of S. macrocarpum (EESM). First, we investigated the anti-inflammatory activities of EESM in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. EESM treatment suppressed nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) production and inhibited the expressions of the inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the mRNA and protein levels. In addition, the expression of pro-inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and interleukin-1 beta ($IL-1{\beta}$), was decreased in a dose dependent manner. Investigation of the signaling pathways of nuclear factor kappa B ($NF-{\kappa}B$), phosphoinositide-3-kinase (PI3K)/Akt, and mitogen-activated protein kinases (MAPKs) revealed suppression of $NF-{\kappa}B$ translocation from the cytosol to nucleus by EESM treatment. The phosphorylation of the Akt and ERK proteins was also inhibited by EESM treatment. EESM treatment also stimulated the expression of the heme oxygenase-1 (HO-1) enzyme and its upstream transcription factor, nuclear factor-E2-related factor 2 (Nrf2). These results suggest that EESM has anti-inflammatory activity and could have potential uses in the field of nutraceuticals.

In order to understand environment condition and phytoplankton community before and after coastal upwelling, the influences of upwelling events on phytoplankton community were studied at 18 stations located the Southern part of East Sea, Korea from May to August 2013. The surface water masses showed low temperature and high salinity due to upwelling events at coastal stations (A1, B1, C1). Correlation between temperature and nutrients (DIP, r=-0.218, p<0.01; DIN, r=-0.306, p<0.01; silicate, r=-0.274, p<0.01) was significantly negative. This result could be explained that nutrients were supplied to surface water by the upwelling of bottom water. Phytoplankton communities were composed of 186 species. Phytoplankton abundance were relatively high in May (C1, $726{\times}10^3cells\;L^{-1}$) and July (A1, $539{\times}10^3cells\;L^{-1}$). Total chlorophyll a and micro-size fraction ($>20{\mu}m$) increased at coastal stations in July and August, while phytoplankton abundance and total chl. a was much low in June. Dominant species in June was Pseudo-nitzschia spp. of which the cell size was $309{\mu}m^3$. Cell size of Pseudo-nitzschia spp. was smaller than dominant species in other period. Therefore, the increase in total chloro-phyll a and the size of phytoplankton was resulted in the sufficient supply of nutrients. In contrast, these tendencies were not observed at outside stations. These results suggested that coastal upwelling was an important influencing factor to determine the species composition and standing stock of phytoplankton community in the coastal waters of the Southern part of East Sea, Korea.
In this study, we analyze seabottom conditions and characteristics integrated with topographic data, seafloor mosaic, underwater images and orthophoto(drone) of soft-hard bottom area around the Sib-Ri rock in the northern shore of the East Sea(Gyeongpo Beach, Gangneung). We obtained field survey data around the Sib-Ri rock(about 600 m × 600 m). The Sib-Ri rock is formed by two exposed rocks and surrounding reef. The artificial reef zone made by about 200 ~ 300 structures is shown the western area of the Sib-Ri rock. The underwater rock region is extended from the southwestern area of the exposed the Sib-Ri rock with 9 ~ 11 m depth range. The most broad rocky seabottom area is located in the southwestren area of the Sib-Ri rock with 10 ~ 13 m depth range. The study area were classified into 4 types of seabottom environment based on the analysis of bathymetric data, seafloor mosaics, composition of sediments and images(underwater and drone). The underwater rock zones(Type I) are the most distributed area around the Sib-Ri Rock(about 600 m × 600 m). The soft seabottom area made by sediments layer showed 2 types(Type II: gS(gravelly Sand), Type III: S(Sand)) in the areas between underwater rock zones and western part of the Sib-Ri rock(toward Gyeongpo Beach). The artificial reef zone with a lot of structures is located in the western part of the Sib-Ri rock. Marine algae(about 6 species), Phylum porifera(about 2 species), Phylum echinodermata(about 3 species), Phylum mollusca(about 3 species) and Phylum chordata(about 2 species) are dominant faunal group of underwater image analysis area(about 10 m × 10 m) in the northwestern part of the Sib-Ri rock. The habitat of Phylym mollusca(Lottia dorsuosa, Septifer virgatus) and Phylum arthropoda(Pollicipes mitella, Chthamalus challengeri hoek) appears in the intertidal zone of the Sib-Ri rock. And it is possible to estimate the range and distribution of the habitat based on the integrated study of orthphoto(drone) and bathymetry data. The integrated visualization and mapping techniques using seafloor mosaic images, sediments analysis, underwater images, orthophoto(drone) and topographic data can provide and contribute to figure out the seabottom conditions and characteristics in the shore of the East Sea.
