Cytokine Gene Expression of Peritoneal Tissues in Response to Mixed Infection of Bacteroides fragilis and Escherichia coli (Bacteroides fragilis와 대장균의 혼합 감염에 의한 복강 조직의 Proinflammatory Cytokine 유전자 발현 조절)

  • Bacteroides fragilis and Escherichia coli, normal colonic inhabitants, are the most frequently isolated bacteria in infected tissues, particularly in intraabdominal abscesses. This study was designed to determine whether enteric bacteria may alter the B. fragilis-induced expression of pro inflammatory cytokines in mouse peritoneal tissue (MPT). After C57BL/6 mice were inoculated with abscess-forming mixture containing B. fragilis in the presence or absence of E. coli, RNA was extracted from MPT. Expression of interleukin (IL)-$1{\alpha}$ and tumor necrosis factor $(TNF){\alpha}$ mRNA was assessed using RT-PCR and standard RNA. Each cytokine protein was also measured by ELISA. The co-inoculation of E. coli into mouse peritoneal cavity advanced the onset of abscess development by B. fragilis infection. When mouse was co-infected with E. coli and B. fragilis intraperitoneally, there was a synergistic increase in the expression of IL-$1{\alpha}$ and $TNF{\alpha}$ mRNA in MPT and this was paralleled by increased cytokine protein secretion. Mixed inoculation of heat-killed E. coli and B. fragilis did not cause a synergistic increase in those cytokine mRNA expression. These results suggest that enteric bacteria may significantly affect proinflammatory cytokine signal produced by host peritoneal cavity in response to B. fragilis infection.

  • The genus Martensia (Delesseriaceae, Rhodophyta) is characterized by thalli composed of one to several blades that consist of proximal membranous sections and distal latticework. Nerves or veins are absent in the membranous sections. The life cycle of Martensia is accomplished by isomorphic alternation of generations. The gametophytes of Martensia are dioecious, and the male and female gametangial plants are morphologically similar. The type species of Martensia is M. elegans Hering. In this study, nine species were confirmed to occur in the subtidal regions of Jeju Island, Korea: M. albida sp. nov., M. australis Harvey, M. bibarii Y. Lee, M. elegans Hering, M. flammifolia sp. nov, M. fragilis Harvey, M. jejuensis Y. Lee, M. palmata Y. Lee, and M. projecta Y. Lee. Three of these, M. australis, M. fragilis, and M. elegans, are new records in the flora of Korea. The results of molecular analyses of the internal transcribed spacer (ITS) 1 region in the nrDNA showed that M. elegans is identical to M. australis, and M. fragilis coincides with M. bibarii. It may be a less effective tool for the species discrimination in Martensia.

Dientamoeba fragilis Infection in Patients with Digestive and Non-Digestive Symptoms: A Case-Control Study

  • In most developing countries, Dientamoeba fragilis infection is an obscure protozoan infection. We aimed to determine a frequency and clinical importance of D. fragilis infection in Taif, Saudi Arabia. A 1-year case control study included patients with gastrointestinal (cases, n=114) or non-gastrointestinal symptoms (controls, n=90). The fecal samples were examined with the classical parasitological methods for intestinal protozoa, and by real time PCR for D. fragilis. The infection by D. fragilis was detected in 5.8% by PCR and in 4.4% patients by microscopy. The infection was identified more in control group (n=9) than in cases (n=3); a sole infection in 11 patients and mixed with Giardia in 1 patient. The other enteric parasites detected were Blastocystis sp. (8.3%), Giardia sp. (5.3%), Cryptosporidium sp. (2.9%), Entamoeba histolytica (1.4%), Entamoeba coli (0.9%) and Hymenolepis nana (0.4%). Our results tend to reinforce the need to increase awareness of D. fragilis infection in Saudi Arabia.

  • The cell of Kluyveromyces fragilis yeast, which is worthy of an algal substitute, was disrupted by a chemical treatment to increase the digestion of filter-feeders that yeasts are fed to. The optimum conditions of the chemical treatment were obtained by incubating yeasts at 3$0^{\circ}C$ for one hour after treated by 1 M of Na$_{2}$-EDTA that was dissolved in 0.2 M of Tris-buffer and by 0.3 m of 2-mercaptoethanol. The percentage of protop[last production was about 30%. The percentage could be doubled by the pretreatment of three times of 30 seconds sonication.

Production of Oligosaccharide by Beta-galactosidase of Kluyveromycces fragilis (Kluyveromyces fragilis의 Beta- galactosidase에 의한 Oligosaccharide의 생성)

  • Kluyveromyces fragilis로부터 $\beta$-galactosidase의 생성조건과 조효소액의 성질 그리고 당 전이 반응에 의한 oligosaccharide 생성을 조사한 결과 다음과 같은 결과를 얻었다. \circled1 Peptone-Yeast extract 배지에 6% lactose를 첨가하였을 때 최대 효소생산을 보였다. \circled2 0.05 M potassium phosphate buffer (pH 7.0)에서 3% toluene를 첨가하여 37$^{\circ}C$ 5시간 배양하였을 때 K. fragilis로부터 효소가 최대로 추출되었다. \circled3 효소의 최적온도는 4$0^{\circ}C$이고 4$0^{\circ}C$ 이상의 열처리에서는 효소가 파괴되었으며, pH6-7에서는 상당히 안정하였다. \circled4 ONPG 기질로 사용하였을 때 Km 값은 2.5mM이었다. \circled5 당 전이 반응의 결과, 7개의 oligosaccharide가 생성되었다. 이상의 실험결과로 볼 때, 본 실험에 사용한 K. fragilis SKD 7001은 Beta-galactosidase의 생산을 위해서 이용 가치가 인정되었으며, 특히, 이 효소의 활성이 중성 pH에서 강하고 안정한 상태를 보이는 것은 시유나 원료우유의 lactose를 중성에서 해야 한다는 점을 고려할 때, 실용적 가치가 있다고 본다. 또, 이 효소의 작용과정에서 생성되는 oligosacchride는 장내 bifidus균의 생육을 촉진시키는 효과가 인정되고 있기 때문에 이용가치를 더욱 높여주는 것으로 생각된다.

The Reaction Conditions of $\beta$-Galactosidases from Aspergillus oryzae, Bovine Liver, and Saccharomyces fragilis to Asialofetuin (Asialofetuin에 대한 Aspergillus oryzae, bovine liver Saccharomyces fragilis 유래 $\beta$-galactosidase의 반응 조건)

  • The enzymatic properties of $\beta$-galactosidases from Aspergillus oryzae, bovine liver and Saccharomyces pragilis have been studied using enzyme-linked lectin assay based on the RC $A_{120}$ and BS-II lectins which specifically bind to terminal galactose and GlcNAc residue, respectively. Asialofetuin, a monomeric glycoprotein with approximately 48 kDa in molecular weight, was used as a substrate. This glycoprotein contains three N-linked triantennary complex type carbohydrate chains with each of which terminating in Ga1$\beta$P1 longrightarrow4G1cNAc (74%). Their optimal pHs were 3.5 and 6.5 (A. oryzae), and 3.5~5.5 (bovine liver and S. fragilis) at 37$^{\circ}C$ during 24 hrs, and the effective concentrations were 0.9, 2.9, and 1.7 mg/ml, respectively The enzyme from A oryzae requires 100 mM N $a^{+}$ or $K^{+}$, while the enzyme from bovine liver requires $Ba^{2+}$ for activity. However all of the three $\beta$-galactosidases were inactivated by SDS and C $u^{2+}$. These results indicate that the hydrolysis of glycoprotein such as asialofetuin depends on the reaction conditions of $\beta$-galactosidases and some metal ions. ions.

Effects of Artemisia capillaris Extracts on Intestinal Microflora In vitro and In vivo (In vitro 및 In vivo에서 인진쑥 추출물이 장내미생물에 미치는 영향)

  • This study was designed to investigate the effect of Artemisia capillaris extracts on the intestinal microflora. In agar diffusion method, the solvent fractions of Artemisia capillaris showed growth inhibition against the intestinal microflora. In particular, the chloroform fraction of Artemisia capillaris had strong antibacterial activity against Clostridium perfringens, Clostridium difficile, Eubacterium limosum, and Bacteroides fragilis, but did not show antibacterial activity against Bifidobacterium bifidum and Lactobacillus acidophilus. Most chloroform fraction of Artemisia capillaris inhibitory activities were not reduced by heat treatment or pH variation against C. perfringens, C. difficile, E. limosum, and B. fragilis. MICs of the chloroform fraction were 1.25 mg/mL against C. perfringens, E. limosum and B. fragilis and 2.5 mg/mL against C. difficile. MBCs of chloroform fraction were 5 mg/mL against C. perfringens, E. limosum and 2.5 mg/mL against C. difficile, B. fragilis. The ethyl acetate fraction of Artemisia capillaris showed $3.08{\pm}0.03$ mg/10 mg total polyphenol and $1.91{\pm}0.03$ mg/10 mg total flavonoid contents. In vivo tests were performed to investigate the influence of Artemisia capillaris extract on the intestinal microflora in rats. The results showed the possibilities of utilizing Artemisia capillaris extracts as a functional food component to control intestinal microflora.

  • In this study, we investigated the antimicrobial activity of herbs related with treatments of intestinal diseases against intestinal pathogens under anaerobic broth system. The water extract of Saussurea lappa Clarke and Myristica fragrans Houtt. showed no growth inhibition against tested pathogens(Eubacterium limonsum ATCC 10825, Escherichia coli ATCC 25922, Bacteroides fragilis KCTC 5013, Clostridium perfringens STCC 3627, Staphylococcus aureus KFCC 11764 및 Salmonella typhimurium ATCC 14028). All tested pathogens were not inhibited in broth containing 100$\mu\textrm{g}$/$m\ell$ of Areca catachu L. Water extract but its extract strongly inhibited the growth of Eubacterium limonsum STCC 10825, Bacteroides fragilis KCTC 5013, Clostridium perfringens ATCC 3627 and Salmonella typhimurium ATCC 14028 at 1,000 to 2,000$\mu\textrm{g}$/$m\ell$ of concentration. Escherichia coli ATCC 25922 and Staphylococcus aureus KFCC 11764 hardly grew in broth containing 2,000$\mu\textrm{g}$/$m\ell$ of Terminalia chebula Retz. water extract.

Growth Characteristics and Physiological Functionality of Yeasts in Pear Marc Extracts

  • Kluyveromyces fragilis KCTC 7260 and Saccharomyces cerevisiae KCTC 7904, which both grew well in pear marc extract, were selected and their growth profiles and physiological functionalities were determined. Both of the selected yeasts established maximal growth by 20 hr of cultivation at $30^{\circ}C$ in pear marc extract. The cell-free extracts showed high antihypertensive angiotensin I-converting enzyme inhibitory activity of 68.9% and 52.1%, respectively. The extracts also displayed 9.2 U/mL and 12.0 U/mL of protease activity, respectively.

  • 1987년 6월부터 1990년 5월까지 진해만의 저서동물 군집에서 우점하는 아기반투명조개(Theora fragilis A. Adams)에 대한 개체군 생태학적 연구를 수행하였다. 조사결과, 이 종의 주된 분포 구역은 진해만의 입구 정점이었으나 겨울철에는 내만의 안쪽으로 확장되었다. 이 종은 여름철과 겨울철의 2회에 걸쳐 치패의 착저가 관찰되었으나 주 착저시기는 여름철이었다. 진해만 입구의 정점에서는 이러한 대량착저로 인해 그 밀도가 2,116개체/m$^{[-992]}$ 에 달하였다. 그러나 착저한 치패의 초기 감모로 인해 가을철의 밀도는 급격히 낮아졌다. 한편, 이 종의 분포는 퇴적물의 유기물 함량과 깊은 관계를 보여서 3.0-5.0%(평균 4.54$\pm$1.42%), 퇴적물의 평균 입도 8.97($\pm$0.62)$\varphi$사이에서 상대적으로 높은 밀도를 보였다. 그러나 퇴적물의유기물 함량의 증가와 함께 이 종의 밀도가 대체로 증가하지만 5.0% 이상에서는 다시 감소하는 현상을 보였다. 이 종은 여름철 저층수의 빈산소 수괴가 사라진 후에는 다시 진해만의 안 쪽 내만정점에서 높은 밀도를 나타냈다. 따라서 진해만에서의 본 종의 분포는 전반적으로 퇴적물의 유기물 함량. 퇴적물의 입도 조성 및 저층수의 용존 산소량 등에 의해 결정된다고 판단된다.
