Corrosion Behavior of Hastelloy C-276 for Carbon-anode-based Oxide Reduction Applications

  • The corrosion behavior of Hastelloy C-276 was investigated to identify its applicability for carbon-anode-based oxide reduction (OR), in which Cl2 and O2 are simultaneously evolved at the anode. Under a 30 mL·min-1 Cl2 + 170 mL·min-1 Ar flow, the corrosion rate was less than 1 g·m-2·h-1 up to 500℃, whereas the rate increased exponentially from 500 to 700℃. The effects of the Cl2-O2 composition on the corrosion rate at flow rates of 30 mL·min-1 Cl2, 20 mL·min-1 Cl2 + 10 mL·min-1 O2, and 10 mL·min-1 Cl2 + 20 mL·min-1 O2 with a constant 170 mL·min-1 Ar flow rate at 600℃ was analyzed. Based on the data from an 8 h reaction, the fastest corrosion rate was observed for the 20 mL·min-1 Cl2 + 10 mL·min-1 O2 case, followed by 30 mL·min-1 Cl2 and 10 mL·min-1 Cl2 + 20 mL·min-1 O2. The effects of the chlorine flow rate on the corrosion rate were negligible within the 5-30 mL·min-1 range. A surface morphology analysis revealed the formation of vertical scratches in specimens that reacted under the Cl2-O2 mixed gas condition.

Anticancer Activity of Glycyrrhiza cultivar Extracts in Breast Cancer Cells

  • Several studies report the anticancer effect of Glycyrrhiza glabra (G. glabra), Glycyrrhiza uralensis (G. uralensis) and their compounds. However, the anticancer effect of Glycyrrhiza cultivar roots are limited. In this study, we compared the anticancer effect of Glycyrrhiza cultivar (Wongam and Shinwongam) extracts with G. glabra and G. uralensis extracts in breast cancer cell lines. Freeze dried Glycyrrhiza root extracts were dissolved in cell culture media at 2 mg/mL and filtered by $0.2{\mu}m$ filter. Glycyrrhiza root extracts were serially diluted at the concentrations of $10{\mu}g/mL$, $100{\mu}g/mL$, $200{\mu}g/mL$, $400{\mu}g/mL$, $800{\mu}g/mL$, $1000{\mu}g/mL$ and $2000{\mu}g/mL$. MCF-7 and MDA-MB-231 breast cancer cells were treated with different concentrations of Glycyrrhiza root extracts and the cell viability was measured using MTT assay. In MCF-7 cells, G. glabra showed no significant difference with Wongam and showed significant difference with Shinwongam at $1000{\mu}g/mL$ (G. glabra 101.2% and Shinwongam 82.68%) and $2000{\mu}g/mL$ (G. glabra 83.07% and Shinwongam 54.05%). G. uralensis showed significant difference with Wongam at $2000{\mu}g/mL$ (G. uralensis 66.48% and Wongam 95.02%) and showed no significant difference with Shinwongam. In MDA-MB-231 cells, G. glabra showed no significant difference with both Wongam and Shinwongam. G. uralensis showed significant difference with Wongam at $2000{\mu}g/mL$ (G. uralensis 72.59% and Wongam 93.47%) and showed no significant difference with Shinwongam. In conclusion, the current study demonstrated that G, glabra and G. uralensis compared with Wongam, and Shinwongam at low concentrations ($10{\mu}g/mL{\sim}800{\mu}g/mL$) display similar cytotoxic potency.

  • We define the spherical non-commutative torus $L_{\omega}$/ as the crossed product obtained by an iteration of l crossed products by actions of, the first action on C( $S^{2n+l}$). Assume the fibres are isomorphic to the tensor product of a completely irrational non-commutative torus $A_{p}$ with a matrix algebra $M_{m}$ ( ) (m > 1). We prove that $L_{\omega}$/ $M_{p}$ (C) is not isomorphic to C(Prim( $L_{\omega}$/)) $A_{p}$ $M_{mp}$ (C), and that the tensor product of $L_{\omega}$/ with a UHF-algebra $M_{p{\infty}}$ of type $p^{\infty}$ is isomorphic to C(Prim( $L_{\omega}$/)) $A_{p}$ $M_{m}$ (C) $M_{p{\infty}}$ if and only if the set of prime factors of m is a subset of the set of prime factors of p. Furthermore, it is shown that the tensor product of $L_{\omega}$/, with the C*-algebra K(H) of compact operators on a separable Hilbert space H is not isomorphic to C(Prim( $L_{\omega}$/)) $A_{p}$ $M_{m}$ (C) K(H) if Prim( $L_{\omega}$/) is homeomorphic to $L^{k}$ (n)$\times$ $T^{l'}$ for k and l' non-negative integers (k > 1), where $L^{k}$ (n) is the lens space.$T^{l'}$ for k and l' non-negative integers (k > 1), where $L^{k}$ (n) is the lens space.e.

A karyotype analysis of Lactuca (Asteraceae) in Korea (한국산 왕고들빼기속(Lactuca)의 핵형분석)

  • The chromosome morphology of two Korean Lactuca (L. indica, L. triangulata) is reported herein. The chromosome number and karyotype of a naturalized plant, L. scariola are reported for the first time. The basic chromosome number was x = 9. Polyploid forms were not recorded. The karyotypes of L. indica, L. scariola, and L. triangulata were 2 n = 18 = 2 m+ 7 sm, 2 n = 18 = 1 m + 6 sm+ 2 st, 2 n = 18 = 2 m + 5 sm+ 2 st, respectively. Both L. indica and L. triangulata had satellites at the ends of their short arms. The haploid genome lengths of L. indica, L.scariola, and L. triangulata were $56.3{\mu}m$, $35.3{\mu}m$, and $72.5{\mu}m$ respectively. Each chromosome length of naturalized L. scariola was $2.7-5.2{\mu}m$; the smallest among Korean Lactuca. The chromosome lengths of L. indica and L. triangulata were $4.7-7.6{\mu}m$ and $2.9-7.9{\mu}m$, respectively. The karyotype of L. scariola differed from that of L.indica and L.triangulata both of which belong to sect. Tuberosae. Therefore, L. scariola is thought to belong to sect. Lactuca subsect. Lactuca.

  • Paper electrophoretic separation of octahedrally bonded (Ruc $l_{6}$ )$^{3-}$ has been carried out by using the specially designed migration apparatus. The supporting electrolyte solutions are as follows: 0.1M-HCl $O_4$, 0.05 M-HCl+0.09M-KCl, 0.1M-HCl, 5$\times$10$^{-3}$ M-NTA, 0.01M-HCl, 0.01M-HCl $O_4$, 0.01M-citric acid, 0.01M-K $H_2$P $O_4$+0.01M-N $a_2$HP $O_4$, 0.05M-borax, 0.025M-N $a_2$C $O_3$+0.025M-NaHC $O_3$, 0.01M-N $a_3$P $O_4$, 0.01M-NaOH and 0.1 M-NaOH. The (Ruc $l_{6}$ )$^{3-}$ appears in 2 to 4 peaks and is found in several chemical species such as (RuCl ($H_2O$)$_{5}$ )$^{2+}$, cis and trans (RuC $l_2$($H_2O$)$_4$)$^{1+}$, (RuC $l_3$($H_2O$)$_3$)$^{0}$ , (RuC $l_4$($H_2O$)$_2$)$^{1-}$, (RuC $l_{5}$ ($H_2O$))$^{2-}$ and (RuC $l_{6}$ )$^{3-}$. The retention value has been found to be highest in the 0.025M-N $a_2$C $O_3$+0.025M-NaHC $O_3$ electrolyte solution.n.

Effects of Protoberberine Alkaloids on L-DOPA-Induced Cytotoxicity in PC12 Cells (Protoberberine 알칼로이드가 PC12 세포중의 L-DOPA 유도 세포독성 작용에 미치는 영향)

  • Previously, protoberberine alkaloids such as berberine and palmatine have been found to lower dopamine content in PC12 cells (Shin et at., 2000). In this study, the effects of berberine and palmatine on L-DOPA-induced increase in dopamine level and cytotoxicity in PC12 cells were investigated. Treatment of PC12 with L-DOPA at concentration ranges of 20∼50 $\mu$M increased dopamine content and the increase in dopamine levels by L-DOPA was inhibited by 10∼40 $\mu$M berberine and 10∼80 $\mu$M palmatine, which the concentration ranges did not show a cytotoxicity. However, berberine and palmatine at concentrations higher than 50 $\mu$M and 100 $\mu$M caused a cytotoxicity, respectively. In addition, berberine (10∼20 $\mu$M) and palmatine (10∼50 $\mu$M) at non-cytotoxic concentration ranges aggravated L-DOPA-induced cytotoxicity in PC12 cells (L-DOPA concentration ranges, 20∼50 $\mu$M). The L-DOPA-induced cytotoxicity was also significantly potentiated by berberine (50 $\mu$M) and palmatine (100 $\mu$M) with cytotoxic ranges. These data demonstrate that berberine and palmatine inhibit L-DOPA-induced increase in dopamine content and stimulate L-DOPA-induced neurotoxicity. Therefore, the possibility that the long-term L-DOPA treated patients with berberine and palmatine could be checked the adverse symptoms.

Effect of Houttuynia cordata Extracts on Tyrosinase Gene Expression (어성초(Houttuynia cordata) 추출물이 Tyrosinase유전자 발현에 미치는 효과 -연구노트-)

  • The methanol extract of Houttuynia cordata repressed the expression of tyrosinase gene of Bl6 mouse melanoma cell containing tyrosinase promoter. The extract repressed expression of tyrosinase gene by about 13$\%$ and 45$\%$ at 10$\mu$g/mL and 100 $\mu$g/mL, respectively. In the MTT assay, the same extract exhibited very low cytotoxicity at 1$\mu$g/mL, 10$\mu$g/mL, and 100 $\mu$g/mL, respectively. The fractions of ethyl acetate, butyl alcohol, and water did not showed the repressive effect on the expression of tyrosinase gone, but methylene chloride fraction layer repressed highly at 10$\mu$g/mL and 100$\mu$g/mL.

Effects of Gadolinium Contrast agent on Bone Mineral Density Measurement using Dual Energy X-ray Absorptiometry (가돌리늄조영제가 이중에너지 X-선 흡수법을 이용한 골밀도검사에 미치는 영향)

  • Radiographic contrast agents are used for diagnostic purposes and are one of the factors affecting measured values in bone density tests. They are absorbed into tissues and have an effect of increasing the measured values of bone density, so they are avoided as much as possible before performing a bone density test. MRI contrast agents, which have different physical properties and mechanisms of action than radiographic contrast agents, are based on gadolinium, a metal element. They have radiopacity characteristics, so MRI are generally performed prior to examination using radiation. The purpose of this study was to investigate the effects of MRI contrast agents on bone mineral density examination using dual energy X-ray absorption. Two types of gadolinium based MRI contrast agents were injected into an acrylic water tank for each volume, and the humanoid spine phantom was inserted and the BMD and T-score from (L1-L4) were analyzed by scanning a total of 30 times, 5 times for each injection type. The average value of the measured total (L1-L4) bone density for each of the two contrast agents was 0.952±0.052, 0.957±0.050, and 0.956±0.05g/㎠, respectively, for the Gadoterate Meglumine component 0mL, 7.5mL and 15mL, when the gadobutrol components were 0mL, 5mL, and 10mL, there was no statistically significant difference at all sites at 0.953±0.001, 0.954±0.001, and 0.945±0.001g/㎠, respectively(p>0.05). The average value of total T-score was -0.46±0.05, -0.4±0, -0.42±0.04 when the Gadoterate Meglumine component was 0mL, 7.5mL and 15mL, respectively. When the Gadobutrol ingredients were 0mL, 5mL and 10mL, there was no statistically significant difference in all areas, with -0.46±0.05, -0.46±0.05, and 0.5±0.00, respectively. In this experiment, the MRI contrast agent was found to have no effect on bone density tests, using the dual-energy X-ray absorption method. There is a limitation in that physical conditions such as kidney and health conditions etc. were not taken into consideration, so further clinical research is expected to be conducted in the future.

The Effect of Bacillus-Fermented Scutellariae Radix Acupuncture Solution on Interleukin Production in Mouse Macrophage Stimulated by Lipopolysaccharide (바실러스균 발효황금약침액이 Lipopolysaccharide로 활성화된 마우스 대식세포의 인터루킨 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of Bacillus-fermented Scutellariae Radix acupuncture solution (SB) on interleukin(IL) production in mouse macrophage stimulatedby lipopolysaccaride(LPS). Methods : Productions of interleukins were measured y High-throughput Multiplex Bead based Assay with Bio-plex Suspension Array System based on $xMAP^{(R)}$(multi-analyte profiling beads) technology. To begin with, cell culture supernatant was obtained after treatment with LPS(1 ${\mu}g/mL$) and SB for 24 hour. Then, it was incubated with the antibody-conj${\mu}g$ated beads for 30 minutes. And detection antibody was added and incubated for 30 minutes. After incubating for 30 minutes, Strepavidin-conjugated Phycoerythrin(SAPE) was then added. Incubating for another 30 minutes, the level of SAPE fluorescence was analyzed on Bio-plex Suspension Array System. Results : The results of the experiment are as follows. SB significantly inhibited the LPS-induced production of IL-3($9.15{\pm}0.35$ pg/mL) by $6.92{\pm}0.05,\;7.21{\pm}0.11,\;6.96{\pm}0.33,\;and\;7.45{\pm}0.74$ pg/mL at the concentration of 25, 50, 100, and 200 ${\mu}g/mL$ in mouse macrophage RAW 264.7 cells (p<0.05). SB significantly inhibited the LPS-induced production of IL-5($7.30{\pm}0.48$ pg/mL) by $6.50{\pm}0.29,\;6.30{\pm}0.25,\;6.30{\pm}0.25,\;and\;5.80{\pm}0.25$ pg/mL at the concentration of 25, 50 100, and 200 ${\mg}g/mL$ in RAW 264.7 cells (p<0.05). SB significantly inhibited the LPS-induced productiion of IL-9($17.26{\pm}0.19$ pg/mL) by $15.01{\pm}0.43$ pg/mL at the concentration of 25 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). SB significantly inhibited the LPS-induced productioh of IL-13($187.80{\pm}2.90$ pg/mL) by $152.80{\pm}4.25,\;172.80{\pm}3.97,\;162.10{\pm}6.67,\;and\;165.30{\pm}11.80$ pg/mL at the concentration fo 25, 50, 100, and 200 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). SB significantly inhibited the LPS-induced production of IL-17($18.30{\pm}0.95$ pg/mL) by $13.30{\pm}1.25,\;13.80{\pm}1.11,\;13.30{\pm}0.75,\;and\;14.00{\pm}1.08$ pg/mL at the concentration of 25, 50 100, and 200 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). SB significantly inhibited the LPS-induced production of IL-23($43.90{\pm}0.83$ pg/mL by $39.50{\pm}1.26,\;38.00{\pm}1.78,\;and\;39.60{\pm}2.49$ pg/mL at the concentration of 25, 100, and 200 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). Conclusions : These results suggest that SB has anti-inflammatory activity related with its inhibition of IL-3, IL-5, IL-13, IL-17, and IL-23 production in macrophages.

Overproduction of Bacterial Trypsin in Streptomyces - Optimization for Streptomyces griseus Trypsin Production by Recombinant Streptomyces (미생물을 이용한 트립신 과대 생산 연구 - Streptomyces용 숙주-벡터계를 이용한 트립신 유전자의 대량발현 최적화 -)

  • The expression vector (pWHM3-TR1R2) for sprT gene encoding Streptomyces griseus trypsin (SGT) followed by two regulatory genes, sgtR1 and sgtR2, was introduced into Streptomyces lividans TK24 and Streptomyces griseus IFO 13350. Various media with different compositions were used to maximize the productivity of SGT in the recombinant trains. he SGT productivity was best when the transformant of S. lividans TK24 was cultivated in R2YE medium (0.74 unit/mL) at 5 days of cultivation. C5/L (0.66 unit/mL) medium also gave a good productivity, but Livid (0.08 unit/mL) and NDSK (0.06 unit/mL) yielded poor productivities. S. griseus IFO 13350/pWHM3-TR1R2 produced SGT by 1.518 unit/mL (C5/L), 1.284unit/mL (R2YE),0.932 unit/mL (NDSK), and 0.295 unit/mL (Livid) at 7 days of cultivation, which was much higher than those from S. lividans TK24/TR1R2. The SGT protein was purified from the culture broth of S. griseus IFO 13350/pWHM3-TR1R2 in C5/L to homogeneity via ammonium sulfate fractionation, and CM-sepharose and SP-sepharose column chromatographies. The specific activity of purified SGT was 69,252 unit/mg, and the final purification fold and recovery yield were 6.5 and 1.4%, respectively.
