Effects of Storage Duration on Physicochemical and Antioxidant Properties of Tomato (Lycopersicon esculentum Mill.)

  • This study explored the physicochemical and nutritional changes associated with storage duration of fresh tomatoes. Fruits of the 'TY Megaton' and 'Yureka' tomato cultivars were harvested at the pink stage and stored at $12^{\circ}C$ for 20 days. During storage, firmness, weight loss, skin color (Hunter L, a, b, a / b values), soluble solids content (SSC), titratable acidity (TA), pH, antioxidant contents (lycopene, ascorbic acid, and total phenolics) and antioxidant activity were evaluated. Firmness was above the minimum marketable limit and fresh weight loss was below maximum acceptable weight loss after 3 weeks of storage, and no deleterious effect on antioxidant contents or activities were observed. Significant differences in SSC, TA, and pH were seen between varieties, but not between fruits stored for different durations. In both varieties, Hunter a values increased more than five-fold after 8 days of storage; this correlated with a more than four-fold accumulation of lycopene after two weeks of storage. The antioxidant activity of tomatoes was highest at the beginning of the storage period, likely because of the effective DPPH - reducing power of ascorbic acid and total phenolics. Antioxidant activity increased after 12 days of storage because of increasing lycopene content. Hence, this study indicates that pink - stage tomatoes may be stored at $12^{\circ}C$ for up to 3 weeks without affecting marketability or nutritional value.

Growth, Yield, and Leaf-macronutrient Content of Grafted Cherry Tomatoes as Influenced by Rootstocks in Semi-forcing Hydroponics (반촉성 수경재배시 대목에 따른 방울토마토 접목묘의 생육, 수량 및 엽 내 양분 함량)

  • There are many different types of cultivation in tomatoes for year-round production. One of them, semi-forcing cultivation is characterized by growing seedlings in winter season. If grafted seedlings are used in winter season that energy cost can be reduced, because they have tolerance to cold stress. This study was conducted to analyze the rootstock performance by measuring the growth, yield, and leaf-macronutrient content of cherry tomatoes grown in semi-forcing hydroponics. Three domestic rootstocks 'HSF4', '21LM', '21A701', and a control cultivar 'B-blocking' were grafted onto jujube-shaped cherry tomato (Lycopersicon esculentum L.) commercial cultivar 'Nonari'. The total yield per plant with grafted cherry tomato '21A701' was 3,387g, which was 11%, 22% and 24% higher than the yield with 'B-blocking', non-grafted one and 'HSF4'. The stem diameter of '21A701' was thick with 8.26mm, whereas non-grafted one was thin with 7.23mm at 160 days after transplanting. The flowering position of '21LM' was 34% and 47% higher than the flowering position of 'B-blocking' and non-grafted one at 153 days after transplanting. The NO3-N concentration in petiole sap of '21LM' was the highest with 1,746mg·L-1 and non-grafted one and 'HSF4' were the lowest with 1,252mg·L-1 and 1,245mg·L-1 at 167 days after transplanting. The results indicated that rootstock/scion combinations in cherry tomatoes can affect the plant growth, yield, and the concentration of different NO3-N in leaves at the late growth stage. Both '21A701' and '21LM' have vigorous root system, which influence the growth and yield increased.

Functional Properties of the Lycopene Cultivar of Cherry Tomato (Lycopersicon esculentum var. cerasiforme) (방울토마토 (Lycopersicon esculentum var. cerasiforme) 라이코펜 품종의 기능적 특성)

  • This study was carried out to investigate the effectiveness of the Lycopene cultivar of cherry tomatoes as a functional food and food material by measuring the total polyphenol and flavonoid content, anti-oxidative and anticancer activity. The contents of polyphenol and flavonoid were $12.28{\pm}1.78mg$ and $3.89{\pm}0.54mg$ per one g of dried cherry tomatoes respectively. The anti-oxidative activity of the cherry tomato was verified by measuring ${\alpha}$-${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radical scavenging activity (DSA), 2,2'-azinobis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity (ASA) and ferric reducing antioxidant power (FRAP). 50% of radical scavenging concentrations ($IC_{50}$) of DSA and ASA were $328.64{\pm}4.190{\mu}g/mL$ and $350.61{\pm}3.300{\mu}g/mL$ respectively. FRAP value was $26.92{\pm}0.68{\mu}mol$ $Fe^{2+}/g$. The effects of the cherry tomato extract on the growth of a normal lung cell (Hel299), lung cancer cell (A549), cervical cancer cell (HeLa) and a liver cancer cell (HepG2) were investigated using MTT assay. The cherry tomato extract showed a significantly strong growth inhibition effects against A549 cell and $IC_{50}$ was $375.46{\pm}33.670{\mu}g/mL$. The extract also inhibited growths of HeLa and HepG2 cells weakly. In this study we found that Lycopene cultivar of cherry tomato had anti-oxidative activity and strong inhibition effect against lung cancer cells. These results indicate that the Lycopene cultivar of cherry tomato would be a functional food and food material.

Purification and Characteristics of Endo-Polygalacturonase from Korean Tomato (한국산 토마토의 Endo-Polygalacturonase 정제 및 성질)

  • Endo-polygalacturonase was purified from tomato, Lycopersicon esculentum L. The purification procedures included gel filtration on Sephadex G-150 and DEAE-cellulose ion exchange chromatography. Yield of the enzyme purification was 12.74 %. Purified enzyme was confirmed as a active single band by the SDS-polyacrylamide gel electrophoresis. When the purified enzyme was applied to SDS-PAGE, the molecular weight was estimated about 50,000. The optimum pH for the enzyme activity was 5.0 and the range of its stability to the pH was 4.0 to 5.0. The optimum temperature was $50^{\circ}C$, while the enzyme was abruptly inactivated above $50^{\circ}C$. From the result of the study on the effects of metals ion, it was found that $Ag^+$, $Zn^{++}$ and $Mg^{++}$ increased on the enzyme activity. In contrast, $Ba^{++}$, $Hg^{++}$, $Pb^{++}$, $Ca^{++}$, $Mn{++}$, $Cu^{++}$, $Fe^{+++}$, $Na^+$ and $K^+$ decreased it. the reaction catalyzed by this enzyme followed typical Michaelis-Menten kinetics with the Km value of $1.43{\times}10^{-1}\;mol/l$.

Effects of Plant Growth Promoting Rhizobacteria on the Growth of Hydroponicelly Grown Tomato Plants, Lycopersicon esculentum Mill. cv. 'Seokwang' (植物生長促進 根圈細菌이 養液栽培 토마토의 生長에 미치는 影響)

  • This study was conducted to clarify the plant growth promoting effects of the various rhizobacteria on the growth of hydroponically grown tomatoes in rockwool, perlite and cocopeat cultures. Strains in terms of $Azospirilham\;sp.(4.5{\times}10^7cells/g),\;Rhodopseudomonas\;sp.(5.8{\times}10^5cells/g),\;Pseudomonas\;sp.(6.1{\times}10^6cells/g$), fusant of $Bacillus\;sp.\;and\;Corynebacterium\;glutamicum(9.1{\times}10^5cells/g$) was bacterialized into the root zone of tomatoes before sowing. Overall growth of tomato plants was promoted by bacterialization of the various rhizobacteria. Strains which showed the highest plan growth promoting effects of hydroponically grown tomatoes was Azospirillum sp., and optimum cultural substrates for the plant growth promotion by rhizobactera were in the order of cocopeat > perlite = rockwool cultures.

Plant Growth Substances Produced by Methylobacterium spp. and Their Effect on Tomato (Lycopersicon esculentum L.) and Red Pepper (Capsicum annuum L.) Growth

  • Bacteria from the Methylobacterium genus, called pink-pigmented facultative methylotrophic bacteria (PPFMs), are common inhabitants of plants, potentially dominating the phyllosphere population, and are also encountered in the rhizosphere, seeds, and other parts of plants, being versatile in nature. The consistent success of the Methylobacterium plant association relies on methylotrophy, the ability to utilize the one-carbon compound methanol emitted by plants. However, the efficiency of Methylobacterium in plant growth promotion could be better exploited and thus has attracted increasing interest in recent years. Accordingly, the present study investigated the inoculation effects of Methylobacterium sp. strains CBMB20 and CBMB 110 on seed imbibition to tomato and red pepper on the growth and accumulation of phytohormone levels under gnotobiotic conditions. Seeds treated with the Methylobacterium strains showed a significant increase in root length when compared with either the uninoculated control or Methylobacterium extorquens $miaA^-$ knockout mutanttreated seeds. Extracts of the plant samples were used for indole-3-acetic acid (IAA), trans-zeatin riboside (t-ZR), and dihydrozeatin riboside (DHZR) assays by immunoanalysis. The treatment with Methylobacterium sp. CBMB20 or CBMB 110 produced significant increases in the accumulation of IAA and the cytokinins t-ZR and DHZR in the red pepper extracts, whereas no IAA was detected in the tomato extracts, although the cytokinin concentrations were significantly increased. Therefore, this study proved that the versatility of Methylobacterium as a plant-growth promoting bacteria could be better exploited.

Improvement of Shelf-life and Quality in Fresh-Cut Tomato Slices:

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury. Effect of Pig Slurry Fertigation on Soil Chemical Properties and Yield of Tomato (Lycopersicon esculentum Mill.) (돈분 액비 관비가 토마토의 수량 및 토양화학성에 미치는 영향)

  • This study was conducted to evaluate fertigation effects of pig slurry (PS) and chemical fertilizer (CF) in tomato by analyzing the growth and yield, nutrient content and uptake, nutrient use efficiency, and soil characteristics in greenhouse cultivation. The treatments compared were; no-fertilizer, two different levels of PS (26 mg $L^{-1}$ and 52 mg $L^{-1}$), and a control treatment of chemical fertilizer. There was no significant difference in growth and yield between PS and CF treatments. however, yield reduction was observed in PS 26 mg $L^{-1}$ treatment. The N-utilization efficiency in CF treatment was similar to that of PS 52 mg $L^{-1}$ treatment. Nutrient utilization efficiency decreased in order of potassium (K), nitrogen (N), phosphate (P) with 29.2~43.3% in K, 15.8~36.7% in N, and 3.0~6.3% in P. In soil chemical characteristics, soil pH in PS treatment was higher than in CF treatment. In contrast, nitrate content in soil was higher in CF treatment than in PS treatment. The content of exchangeable K in soil was higher in PS and CF 52 mg $L^{-1}$ treatments. There was no significant difference in exchangeable Ca and Mg among those treatments. Therefore, it can be concluded that chemical fertilizers can be substituted by PS based on soil chemical analysis in tomato fertigation culture.

Fruit Set and Yield Enhancement in Tomato (Lycopersicon esculentum Mill.) Using Gibberellic Acid and 2,4-Dichlorophenoxy Acetic Acid Spray (지베렐린과 2,4-D 처리를 이용한 토마토 착과율 및 수확량 증가)

  • High temperature stress in summer season at plastic house is a limiting factor for tomato fruit set and yield. This study was performed to assess the effects of gibberellic acid ($GA_3$) and 2,4-Dichlophenoxy acetic acid (2,4-D) spray on fruit set, yield, and quality of tomato cv. 'Adoration'(Enza Zaden Co., Ltd.) under the plastic house in Hwacheon, 2011. Four concentrations (0-, 5-, 10- and $15mg{\cdot}L^{-1}$) of $GA_3$, and three concentrations (0-, 5- and $10mg{\cdot}L^{-1}$) 2,4-D were sprayed in early flowering of tomato in the plastic house, and fruit set, yield and quality characters were observed. The results showed that spray of $10mg{\cdot}L^{-1}$ $GA_3$ significantly increased the fruit set by 14.2% than unsprayed. The spray of $GA_3$ significantly increased the marketable fruit number, fruit weight, and yield. The spray of 2,4-D on blossoms significantly affected the fruit set percentage, fruit weight, marketable fruit weight and yield, and the highest fruit set observed as 62.5% in combined spray of $GA_3$ and 2,4-D at each $5mg{\cdot}L^{-1}$. Fruit size and total soluble solids significantly varied with the concentrations of $GA_3$ sprayed. The result indicates that the spray of $10mg{\cdot}L^{-1}$ $GA_3$ and $5mg{\cdot}L^{-1}$ 2,4-D can be more economic and effective to increase the fruit set, and yield in tomato under high temperature condition in plastic house.

Mineral- and Tissue-Specific Metabolic Changes in Tomato (Lycopersicon esculentum L.) Plants Grown under NPK-Starved Conditions

  • Specific metabolic network responses to mineral starvation are not well-defined. We examined a detailed broad-scale identification of metabolic responses of tomato leaf and root to N, P or K starvation. Tomato plants were grown hydroponically under optimal (5 mM N, 0.5 mM P, or 5 mM K) and starved (0.5 mM N, 0.05 mM P, or 0.5 mM K) conditions and metabolites were measured by LC-MS and GC-MS. Overall, the levels of metabolites (lipids, nucleotides, peptides and secondary metabolites) presented in this paper largely showed mineral- and tissue-specific responses. Most strikingly, G3P (glycerol-3-P), GPC (glycerol-P-choline) and choline phosphate responded differently to a type of mineral; an increase in N or K starvation and a decrease in P starvation. A dramatic increase in the levels of secondary metabolites, in particular, rutin and chlorogenate in both tomato tissues during N starvation were observed. Based on these data, it is necessary to clearly elucidate an unknown event taking place in a variety of abiotic impacts, and we are now studying to expand our knowledge on metabolic- and proteomic-responses using GS-MS and LC-MS.
