Glyphosate Toxicity: II. EPSP-synthase Activity in Cell Suspension Culture of Corydalis Sempervirens and Lycopersicon Esculentum (Glyphosate 독성(毒性): II. corydalis Sempervirens와 토마토의 세포배양체(細胞培養體)에서 EPSP-synthase의 활성(活性)에 미치는 영향(影響))

  • Glyphosate (N-[phosphonomethyl]glycine) applied to the assimilate-exporting leaves or sprayed to the whole plants of tomato(Lycopersicon esculentum Mil var. Moneymaker) induced the rapid inhibition of 5-enolpyruvyl skimic acid 3-phosphate synthase(EPSP-synthase). It shows that EPSP-synthase activity precedes chlorophyll loss. There is no difference in EPSP-synthase activity between in vivo tomato meristem and cell suspension culture if glyphosate is not applied. The EPSP-synthase activity is in a range of 4 to 6 nkat per mg protein. The inhibition of EPSP-synthase action is induced within 36 h after glyphosate application while the Chl contents were reduced 48 h after the application. In cell suspension culture of tomato and Corydalis (Corydalis sempervirens), a sublethal concentration of glyphosate retards the fresh weight increase and prolonged lag phase. The fresh weight is reached maximal about 14 days after the subculture in the presence of glyphosate. The inhibitory effect of glyphosate on EPSP-synthase is remarkably induced in lag phase.

Morphological Characterization of 'Ailsa Craig' Tomato Callus and Effect on Plant Regeneration (`Ailsa Craig` 토마토 캘러스의 형태학적 특성과 식물체 재분화에 미치는 영향)

  • In an attempt to optimize the in vitro-regeneration conditions necessary for the genetic manipulation of tomato species, we examined 'Ailsa Graig' cultivar of Lycopersicon for regeneration ability. The basal medium used for callus formation and shoot regeneration was MS (MS + vitamin) supplemented with six combinations of zeatin 2 mg/l, zeatin 2 mg/l + IAA 0.1 mg/l, zeatin 2 mg/l + IAA 0.5 mg/l, zeatin 4 mg/l, zeatin 4 mg/l + IAA 0.1 mg/l and zeatin 4 mg/l + IAA 0.5 mg/l. When all conditions tested were considered, however, only zeatin 2 mg/l was shown to be the best in shoot regeneration. The morphological characterization from in vitro-cultured callus of Lycopersicon esculentum L. var. 'Ailsa Craig' was investigated with scanning electron microscope (SEM). The surfaces of in vitro-cultured callus had well-defined epidermal cell in condition of zeatin 2 mg/l, but those of different treatments were twisted. These results suggested that shape of callus was involved in efficiency of shoot regeneration in tomato 'Ailsa Craig'.

Effect of Inorganic sale, Growth Regulators, and Thiamine on the Callus Growth and Plant Regeneration from long-term Cultured Solanum and Lycopenicon Genotypes (무기염류, 생장조절물질 및 타이아민이 장기간 계대배양한 Solanum과 Lycopnicon종의 캘러스 생장과 식물체분화에 미치는 영향)

  • Callus growth and shoot regeneration of Solanum and Lycopersicon depended on genotype, growth regulators, and thiamine concentration. L. peruvianum LA 1277 and L. peruvianum LA 1373 and PI 251301 had the greatest callus growth while L hirsutum LA 1777, L.esculentum 'Diego'and 'Red Plum' had the least callus growth. Lycopersicon penvianum genotypes were superior to L. esculentur genotypes in regenerating plants. MG medium was more effective in regenerating shoots than MS medium. A low level of IAA (0.2mg/L) and high level of BA (2 mg/L) resulted in the greatest shoot regeneration. Shoot growth varied depending on thiamine concentration and genotype. Shoot proliferation of Solanum ptycathum, Solanum nigrum, and L. peruvianum PI 199380 was best on medium with 20 mg/L thiamine. Regeneration of L. peruvianum PI 251301 and PI 128652 was better on medium with 30 and 10mg/L thiamine, respectively.

  • This study was carried out to investigate the influence of different types of nutrient solutions and culture media on the growth and quality of cucumber(Cucumis sativus L.) and tomato(Lycopersicon esculentum Mill.). The results are summarized as follows : 1. The growth and yield of cucumber and tomato were best in rockwool culture with Cooper solution. 2. In sand-sack culture, growth and yield of cucumber and tomato were higher with compound fertillzer solution. 3. Growth and yield of cucumber and tomato were more effective in rockwool culture than in soil culture in early growth stages, and vice versa in later growth stages. 4. Vitamin C contents of cucumber and tomato showed no differences between soil cultures and hydroponics, except rockwool culture with compound fertilizer solution. 5. The dry weight, total-N, and mineral content of cucumber and tomato showed no differences among all types of hydroponics.

Transient and stable expression of hepatitis B surface antigen in tomato (Lycopersicon esculentum L.)

  • Cotyledonary leaves of tomato cv. Megha were transformed with the hepatitis B virus 's' gene, which encodes surface antigen. Six plant expression cassettes (pHBS, pHER, pEFEHBS, pEFEHER, pSHER and pEFESHER) were used to assay the possible expression levels by agroinfiltration. The maximum transient expression level of 489.5 ng/g D.W. was noted in pEFEHER-infiltrated cotyledonary leaves. Transgenic tomato plants with pEFEHBS and pEFEHER expression cassettes were regenerated and characterized by molecular analysis. The expression of the antigen in the fruits was confirmed by RT-PCR and ELISA analysis. This is the first report on the expression of hepatitis B surface antigen in tomato.

  • Objectives: The objective of this study was to teach patients clinically useful external application of acne. Methods: We investigated the effects and manufacturing methods of the medical herbs published in the woman's monthly magazines according to "Zhong-Yao-Da-Ci-Dian(中藥大辭典)". Results: We investigated that Saururus chinensis Baill.(三白草), Houttuynia cordata Thunb.(魚腥草), Coix lachrymajobi L.(薏苡仁) were discovered 3 times, Brassica oleracea L.(甘藍), Raphanus sativus L.(래복), Daucus carota L. var. Sativa DC.(홍래복), Oryza sativa L.(米皮糠), Spinacia oleracea L.(파래), Cucumis sativus L.(黃爪) 2 times, and Glycyrrhiza uralensis Fisch.(甘草), Gallus gallus domesticus Braisson.(?子白), Aloe vera L.(노회), Phascolus radiatus L.(綠豆), Prunus persica Batsch.(桃葉), Lycopersicon esculentum Mill.(番茄), Taraxacum mongolicum Hand.-Mazz.(蒲公英), Prunus armeniaca L.(杏仁), Musa paradisiaca L. var. Sapientum O. ktze.(香蕉) and Malus pumila Mill.(평과) were discovered once in woman's monthly magazine during last two years. According to the effects, we classificated four groups which were defervescence(淸熱), subsidence of edema(消腫), pyorrhea(排膿) and gloss(滋潤) group. Effect of 1-Methylcyclopropene (1-MCP) Treatment on the Quality Characteristics and Pigmentation of Tomato Fruit (Lycopersicon Esculentum Mill.) (1-Methylcyclopropane 처리가 토마토 선도유지 효과 및 색소 변화에 미치는 영향)

  • The quality attributes of tomato fruit ($Lycopersicon$ $Esculentum$ Mill.) to treatments with 1-methylcyclopropene (1-MCP) were studied. Harvested tomato fruit was treated one time at the initiation of storage or once-a-day during storage with $1{\mu}L/L$ 1-MCP at different storage temperatures, 12, 17, and $23^{\circ}C$. The results showed that both lower temperature and duration of 1-MCP treatment played an important role in ripening of tomato fruit. The once-a-day 1-MCP treatment was presented to be very effective in delaying quality changes of tomato fruit. The amount of chlorophyll and lycopene were measured to assess the impact of 1-MCP and temperature treatments on ripening, using a specific extinction coefficient absorbance technique. Storing tomato fruit at $12^{\circ}C$ resulted in a longer ripening period (color change) than tomato fruit stored at $17^{\circ}C$ and $23^{\circ}C$. 1-MCP treatment was very effective in retarding chlorophyll degradation and lycopene formation in the pericarp tissue of the tomato fruit at the different storage temperatures. The 1-MCP treatments affected the total chlorophyll content in different fruit tissues of the pericarp and placenta. Exposure of tomato fruit to 1-MCP gas at $12^{\circ}C$, using the once-a-day treatment, was highly effective in delaying pigment and color change.

  • We surveyed on the host plants of Fungus gnat, Bradysia app. and found 21 species in the greenhouse and field. These are as follows: Cucumis sativus L., Cucumis melo L., Citrullus lanatus T., Cucurbita moschata F., Lycopersicon esculentum M., Capsicum annuum L (Pepper), Capsicum annuum L (Paprika), Lillium longiflorum T., Dianthus caryophyllus L., Rosa hybrida H., Gerbera jamesonii B., Chrysanthemum morifolium R, Phalaenopsis schilleriane R., Gladiolus grandiflours H., Zingiber officinale R., Cnidium officinale M., Canavalia gladiata DC., Angelica utilis M., Polygonatum odoratum D., Pinus densiflora S., and Pinus thunbergii P. Fungus gnat larvae cause damages to the root and promote decay and wilt by feeding on the roots and burrowing in plant tissue.

Effect of cultivar and ascorbic acid on in vitro shoot regeneration and development of bombardment-mediated plastid transformation of tomato (Lycopersicon esculentum) (토마토 재분화 효율 향상 및 엽록체 형질전환 조건)

  • Eighteen cultivars of tomato were tested for their regeneration response. Lycopersicon esculentum cv. 2001-58 showed a very high frequency of regeneration (93%). We evaluated the effect of two compounds with known antioxidant activity (ascorbic acid and cystein). The use of ascorbic acid ($200\;-\;300\;{\mu}M/L$) has a positive effect on shoot regeneration. To develope a system for plastid transformation in tomato via homologous recombination, we constructed the tomato plastid expression vector (pKRT22-AG) harboring 2.2 kb flanking sequences cloned from intact plastid genome and gfp gene. To investigate the factors affecting the delivery system of the pKRT22-AG into chloroplast using bombardment, We assessed the optimal DNA concentration, gold particle volume and target distance. Expression of the GFP protein was observed within chloroplast on protoplast of cotyledon explant by confocal laser scanning microscopy, which indicates that the protocol developed in this study be useful for the production of plastid transgenic plants in tomato.

Effects of Cultivation Season and Method on Growth and Quality of Tomato (토마토의 재배시기(栽培時期)와 재배방식(栽培方式)이 생육(生育) 및 품질(品質)에 미치는 영향(影響))

  • Tomato (Lycopersicon esculentum) fruits were grown by the hydroponic cultivation system using perlite and soil cultivation system. The growth, yield, and fruit quality were compared between two growing seasons of spring and summer and two cultivation systems, respectively. In tomato, little difference was observed in growth and yield between two cultivation systems, but plant growth and total yield of fruits were much more in summer than in spring. The contents of glucose and fructose were more in spring-grown fruit than in summer-grown one, and in hydroponically cultivated fruit than in soilcultivated, respectively. The weight loss during room temperature storage was lower in hydroponically cultivated fruit than in soil-cultivated one, and in summer-grown fruit than in spring-grown one, respectively. In terms of color change, the shelf-life of tomato fruit was shorter by one day in summer-grown one fruit than in spring-grown one, and in hydroponcically cultivated one fruit than in soil-cultivated, respectively.
