Effect of corn gluten and its hydrolysate consumptions on weight reduction in rats fed a high-fat diet

  • This study examined the effects of com gluten (CG) and its hydrolysate consumptions on weight reduction in rats fed a high-fat diet. Eight-month-old male Sprague-Dawley rats (n=40) were fed a high-fat diet (40% calorie as fat) for 4 weeks. They were then randomly divided into four groups and fed the isocaloric diets with different protein sources for 8 weeks. The protein sources were casein (control group), intact CG (CG group), CG hydrolysate A (CGHA group, 30% of protein as peptides and 70% as free amino acids) and CG hydrolysate P (CGHP group, 93% of protein as peptides and 7% as free amino acids). Body weight gain, adipose tissue weights, nitrogen balance, absorptions of energy, protein and fat, lipid profiles in plasma, liver and feces and hepatic activities of camitine palmitoyl transferase (CPT), fatty acid synthase (FAS), malic enzyme (ME) and glucose-6-phosphate dehydrogenase (G6PDH) were assessed. The CGHA diet had the highest amount of BCAAs, especially leucine, and most of them existed as free amino acid forms. The CGHA group showed significant weight reduction and negative nitrogen balance. Protein absorption and apparent protein digestibility in the CGHA group were significantly lower than those in other groups. Adipose tissue weights were the lowest in the CGHA group. Activity of CPT tended to be higher in the CGHA group than in other groups and those of FAS, ME and G6PDH were significantly lower in the CGHA group than in other groups. In conclusion, the CGHA diet which had relatively high amounts of free amino acids and BCAAs, especially leucine, had a weight reduction effect by lowering adipose tissue weight and the activities of FAS, ME and G6PDH in experimental animals, but it seemed to be a negative result induced by lowering protein absorption, increasing urinary nitrogen excretion and protein catabolism.

고지혈증 흰쥐에 있어서 버섯 분말의 콜레스테롤 저하효과 (Cholesterol-lowering Effect of Mushrooms Powder in Hyperlipidemic Rats)

  • 본 실험에서는 0.5% 콜레스테롤과 0.25% 콜산나트륨을 첨가한 콜레스테롤 식이와 콜레스테롤 식이에 4.0% 수준의 버섯분말(Lentinus edodes, Ganoderma lucidum, Pleurotus ostreatus; 5 3:2, w/w/w) 첨가 식이를 female Sprague-Dawley 흰쥐에 4주동안 섭취시켜 지질농도에 미치는 영향을 조사하였다. 혈청 콜레스테롤 농도는 버섯분말 첨가 식이군에서 57% 감소하고, 혈청 HDL-콜레스테롤 농도는 230% 증가하여 동맥경화 지수가 68.4% 감소함으로써 버섯분말에 항동맥경화 효과가 있는 것으로 사료되었다. 간장 콜레스테롤 농도는 콜레스테롤 식이군에 비해 버섯분말 첨가식에서 24.7% 감소하였으나, 중성지질 농도는 50%의 유의한 증가를 나타내었다. 본 실험의 결과 콜레스테롤 식이에 버섯분말의 첨가에 의해 혈증 콜레스테롤 농도 저하효과와 동시에 항동백경화 효과가 인정되어 버섯분말을 소재로 새로운 생리활성을 가지는 기능성식품개발의 가능성이 시사되었다.

직장인의 만성질환 위험 요소별로 구성된 영양 교육의 효과 비교 연구 (Effects of Workplace Nutrition Education Program Tailored for the Individual Chronic Disease Risks)

  • The incidence of chronic disease is continuously increasing in Korea. Especially, office workers have higher risk of chronic disease because of their dietary habit and lifestyle. The study aimed to investigate the effect of tailored nutrition counseling on improving chronic disease risk factors. Ninety-nine male workers (age $46.9{\pm}7.0$ yrs) volunteered for 12 weeks of nutrition program containing dietary intake and physical activity adjustment. Five individualized programs were performed with the main theme of weight loss (WL, n = 16), blood pressure lowering (BL, n = 34), normalizing blood glucose (GL, n = 21), lipid lowering (LL, n = 13) and reducing MS risk factors (ML, n = 15). Anthropometric data, blood-pressure, self-reported questionnaire, blood profiles were measured before and after 12weeks of nutrition education. The education program included 5 times of 1:1 interview. Compared to 0 week, anthropometric data (weight, BMI, fat, visceral fat, waist, SBP, DBP) were significantly decreased after 12 weeks (p < 0.001). Fasting blood glucose and total cholesterol were decreased (p < 0.05). The primary outcomes with individually tailored programs showed to be more effective than one general nutrition program.

김천산 자두가 Streptozotocin 당뇨 유발 쥐에 미치는 영향 (Effect of Plums Produced in Gimcheon on Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study examined the effects of plums produced in Gimcheon area on the glucose and lipid metabolism in streptozotocin-induced diabetic rats. Male Sprague-Dawley diabetic rats were divided into four groups: control, diabetic control, Diabetes-low plum (containing 10% plum powder), and Diabetes-high plum (containing 20% plum powder). The animals were fed isocaloric experimental diets based on an AIN-76 diet for 6 weeks. Feed efficiency ratio (FER) of the diabetic groups were significantly lower than that of the control. On the other hand, among the diabetic groups, the FER of the high plum intake group was higher than that of the diabetic control. The liver weight per 100 g body weight of each group was similar but the liver weights tended to decrease as the amount of plum intake was increased. Kidney weight per 100 g body weight of the plum intake groups were significantly different compared to that of the diabetic control. The supplementation of plums lowered the fasting blood glucose level of the diabetic groups and improved the glucose tolerance, thereby lowering the glycosylated hemoglobin index. In addition, the supplementation of plum was lowered the blood total cholesterol concentration and increased the HDL-C/TC (%) significantly, thereby lowering the atherosclerotic index (AI) and hepatic peroxide level. A steady diet of plums produced in Gimcheon may be effective in controlling the blood glucose level and preventing chronic diabetes mellitus.

  • Rice(Oryza stiva L.) is a major cereal food providing nourishment to over half of the world's populations and was considered only as a source of energy. However, The recent many researches have been made to suggest that rice may relate to prevention chronic disease and health-promoting properties such as postprandial glucose response, hypocholesterolemic effect and blood pressure-lowering effect. There has been numerous observation supporting that rice has hypocholesterolemic effect. Rice, rice bran, rice bran oil and unsaponifiable matter of rice bran oil reduced plasma cholesterol in rat, hamster as well as human. Components of rice showing hypocholesterolemic effect include dietary fiber(hemicellulose, phytic acid). protein, ${\gamma}$-oryzanol, $\beta$-sitosterol, and tocotrienols. Crapo et al has been studied that the effect of various of starchy foods on the postprandial blood glucose and insulin responses in healthy and diabetic humans. The results showed that rice had lower blood glucose and insulin responses compared to potato, bread and dextrose. The different physical forms in the same starch also produce the different postprandial glucose and insulin responses. In recent years, several studies have shown that some components of rice have potent antioxidant activity against Fe$^{2+}$ -ascorbate induced lipid peroxidation in rat liver microsomal membranes. Cell culture and animal studies have shown that some components of rice have inhibitory effect on the growth and proliferation of several types of human cancer cell. It was also reported that the methanol extract of brown rice has antimutagenic activity against various mutagens. In addition, the pepsine hydrolysate from rice protein is reported to inhibit angiotensin converting enzyme activity. GABA (${\gamma}$ - aminobutyric acid) and GABA enriched rice germ is also effective for lowering blood pressure and triglyceride levels.s.

김치의 용매획분이 고콜레스테롤 식이를 섭취한 토끼의 혈중지질 농도 변화에 미치는 영향 (The Effects of Solvent Fractions of Kimchi on Plasma Lipid Concentration of Rabbit Fed High Cholesterol Diet)

  • The antioxidative effects of solvent fractions of kimchi on LDL oxidatiojn in vitro as well as hypolipidemic effects of these fractions in rabbit fed atherogenic diet were studied. Methanol extract of deffated kimchi was fractionated sequentially with dichloromethane, ethylacetate, butanol and water. All solvent fractions of kimchi inhibited Cu2+-induced LDL oxidation. Among these fractions, the dicholoromethane fraction at the concentration of 25$\mu\textrm{g}$/mL showed the highest antioxidant effects against LDL oxidation in the aspect of inhibiting TBARS production by 28.03% or prolonged lag phase duration 2-fold compared to those of control. Based on the results from in vitro study, New Zealand White Rabbits grouped six each were fed for 8 weeks either basal diet containing 1% cholesterol or experimental diet containing dichloromethane, ethylacetate or water fraciton added to the basal diet. The amount of solvent fraction of kimchi added to the experimental diet was equivalent to 5% of freeze-dried kimchi. The hypolipidemic effects was observed from all experimental gropus, especially from dichloromethane fraction added group. The plasma and LDL cholesterol levels of this group were decreased by 49% and 47%, respectively while that of HDL increased by 91% compared to those of control. The calculated atherogenic index for the dichloromethane group was the lowest among groups. However, TG lowering effect of experimental group was not observed since solbent fraction of kimchi was used instead of freeze-dried kimchi. The TBARS concentration of LDL isolated from rabbit fed dichloromethane fraction was decreased 21% than that of control. These results indicate that active principles responsible for inhibiting LDL oxidation and lowering plasma cholesterol may present abundantly in dichloromethane fraction of kimchi.

산사청혈탕(山楂淸血湯)이 Triton WR1339로 유발된 흰쥐의 고지혈증(高脂血症)에 미치는 영향 (Effects of Sansachunghyul-tang (SCT) on blood cholesterol levels in Triton WR1339-induced hyperlipidemic rats)

  • Objectives : Sansachunghyul-tang (山楂淸血湯) is a new formula consist of some herbs to treatment of hyperlipidemia. The lipid-lowering effects of the water extract of Sansachunghyul-tang (SCT) were examined using Triton WR1339-induced hyperlipidemic rats. Methods : Hyperlipidemia was induced by intravenous injection of Triton WR1339 (tyloxapol) at 200 mg/kg body weight. Four groups of experimental animals were used: normal, control, and two sample groups. Sample groups were injected with SCT (100 and 1000 mg/kg) for 7 days. Results : SCT produced statistically significant lowering effects on levels of total cholesterol (81.8 and 83.1%, p < 0.01, respectively), triglyceride (88.9 and 93.4%, p < 0.01, respectively), and phospholipid (86.7 and 94.3%, p < 0.01, respectively) than the control group. Taken together, SCT improved parameters of the HDL-cholesterol and LDL-cholesterol levels. Conclusions : In conclusion, These results suggest that SCT could act as a potent antihyperlipidemic in therapeutics for hyperlipidemia.

청국장 점질성 중합체의 첨가 식이가 Streptozotocin 유발 당뇨쥐의 혈당 및 혈청지질 감소에 미치는 영향 (The Effects of Chungkookjang Biopolymer on Blood Glucose and Serum Lipid Lowering in Streptozotocin-Induced Diabetic Rats)

  • 본 연구는 청국장 점질성 중합체의 첨가식이가가 STZ로 유발된 당뇨쥐의 혈당과 지질대사에 미치는 영향을 알아보고자 하였다. 실험동물은 정상군(N), 당뇨 유발군(D) 및 청국장 점질성 중합체 첨가식이 당뇨 유발군(DCB)으로 4주간 실험식이를 공급하였다. 체중증가량과 식이효율은 D군이 N군에 비해 유의적으로 낮았다. 간무게는 D군이 N군에 비해 유의적으로 높았으나 DCB군의 간무게는 D군에 비해 유의적으로 낮았다. D군의 혈당은 4주 동안 비슷한 범위를 유지하였으나 DCB군의 혈당은 실험 4주째에 D군에 비해 유의적으로 낮았다. 혈중 중성지방은 D군이 N군에 비해 유의적으로 높았다. 그러나 DCB군은 D군에 비해 유의적으로 낮았다. DCB군의 혈중 총콜레스테롤은 DM군에 비해 낮은 경향이었으나 유의적인 차이는 없었다. 혈중 HDL-콜레스테롤은 N군과 D군이 유의적인 차이가 없었으나 DCB군은 두 군에 비해 유의적으로 높았다. 혈중 LDL-콜레스테롤은 DCB군이 D군에 비해 낮은 경향이었으나 유의적인 차이는 없었다. 간의 중성지방 함량과 콜레스테롤 함량은 D군이 N군에 비해 유의적으로 높았으나 DCB군은 D군에 비해 낮은 경향이었다. 따라서 청국장 점질성 중합체의 첨가 식이는 STZ로 유발된 당뇨쥐의 혈당을 유의적으로 감소시켰으며 당뇨의 대표적인 합병증인 지질대사 이상 중 증가된 혈중 중성지방을 유의적으로 감소시켰으며 감소된 HDL-콜레스테롤을 유의적으로 증가시켰으므로 당뇨의 혈당조절과 고지혈증 개선에 유익할 것으로 사료된다.

흰쥐에서 키토올리고당의 장 생태와 지질 상태에 미치는 용량에 따른 효과 (Dose-Response of Chitooligosaccharide on Gut Ecology and Lipid Status in Rats)

  • 본 연구는 흰쥐에서 저분자량(1~3 kDa) 키토올리고당(chitooligosaccharide, COS)이 장내 미생물에 미치는 항균활성과 지질 개선에 관한 용량-반응 효과를 조사하기 위하여 수행되었다. 4주령 된 Sprague-Dawley 수컷 32마리를 1주일 동안 사육 환경에 적응시킨 후 군당 8마리씩 4군으로 나누어서 고지방식이에 키토올리고당을 0%(대조식이), 0.5%, 2%, 5% 수준으로 첨가한 식이를 5주 동안 자유롭게 급여하였다. 실험식이 5주째에 장 통과시간을 측정하였고, 신선한 분변을 취하여 장내 균수를 측정하였으며, 희생 후 혈장 지질, 간 지질, GPT 활성, 분변 지질을 측정하였다. 체중 증가량과 식이섭취량은 COS 5% 첨가군에서 유의하게 낮았다. 분변 중 bifidobacteria, lactobacilli, bacteroides, total anaerobes, total aerobes 수는 대조군, COS 0.5% 첨가군과 COS 2% 첨가군 사이에 차이가 없었으나, COS 5% 첨가군에서는 모두 유의하게 감소하였다. 혈장 총 콜레스테롤, HDL-C 및 LDL-C 농도는 군간 차이가 없었으나, 중성지방 농도는 COS 2% 첨가군과 COS 5% 첨가군에서 유의하게 낮았으며, 키토올리고당의 용량이 높아질수록 낮았다. 간 조직의 중성지방 농도는 COS 5% 첨가군에서 낮았으며, 반면에 분변의 콜레스테롤 및 중성지방 농도는 COS 5% 첨가군에서 높았으나 담즙산은 차이가 없었다. 결론적으로 저분자량(1~3 kDa) 키토올리고당을 5% 첨가한 식이는 흰쥐의 식이섭취량, 체중증가량, 혈장 중성지방을 낮추어 지질 개선효과를 보였고, 분변의 혐기성균 중에 bacteroides의 균수를 크게 감소시켰으며 bifidobacteria와 lactobacilli 균수도 억제함으로써 장 미생물생태 변화를 초래하였다.

보리 및 보리 ${\beta}-glucan$ 농축획분 섭취에 의한 흰쥐의 간 콜레스테롤 저하효과 (Cholesterol-Lowering Effects in Rat Liver Fed Barley and ${\beta}-Glucan-enriched$ Barley Fraction with Cholesterol)

  • 총 식이섬유 함량과 에너지 수준을 동일하게 조정한 보리첨가식이 및 ${\beta}-glucan$ 함량을 높인 농축획분 첨가식이가 간에서의 지질대사에 미치는 영향을 시험하였다. 체중을 고려한 간무게 결과는 콜레스테롤 투여로 증가되는 것으로 나타났으며 보리 및 ${\beta}-glucan$ 투여에 의한 차이는 없었다. Control군과 fiber-free군의 간에서는 normal(cholesterol-free)군의 간과는 달리 콜레스테롤 첨가로 인하여 심한 지질 침착이 관찰되었다. 반면 보리첨가군의 경우 동량의 콜레스테롤을 섭취하였으나 지질 침착이 거의 없거나 미약하여 normal 간과 유사한 형태였다. 간의 단위무게당 콜레스테롤과 중성지방 농도는 보리 ${\beta}-glucan$ 농축획분을 첨가한 N glucan(5%)와 W glucan(5%)군에서 각각 대조군의 50%와 60%이하 수준으로 현저하게 감소하였다. 식이섬유 함량이 10%가 되도록 찰보리 농축획분의 양을 증가시킨 W glucan(10%)군의 간 콜레스테롤 저하효과는 더욱 현저하였다. 따라서 0.5%의 콜레스테롤을 첨가하고 5%와 10%의 식이섬유를 보리로부터 공급했을 때 변으로 배설된 콜레스테롤과 중성지방을 보상하기 위한 간에서의 생합성 증가는 일어나지 않고 오히려 간의 콜레스테롤과 중성지방 함량이 현저히 낮아지고 간에 지질 침착이 일어나지 않는 것으로 나타났다.
