Pb 중독에 Fe, Cu가 흰쥐의 체내에 미치는 효과에 관한 연구 (The Effect of Fe and Cu on the Pb Toxicity in Rats)

  • This study was undertaken to investigate the effects of dietary Fe and Cu levels on Pb accumulation of Pb poisoned rats. 80 male Sprague weaning Dawley rate weighing 80-90g were divided into 8 groups(Pb groups: LFLCPb-low Fe, low Cu and Pb group, LFACPB-low Fe, adequate Cu and Pb group, AFLCPb-adequate Fe, low Cu and Pb group, AFACPb-adequate Fe, adequate Cu and Pb group, without Pb gorups: LFLC-low Fe, low Cu and without Pb grooup, LFAC-low Fe, adequate Cu and without Pb group, AFLC-adequate Fe, low Cu and without Pb group, AFAC-adequate Fe, adequate Cu and without Pb group) according to Pb administration (0, 500ppm in drinking water) and Fe and Cu levels(Fe :6ppm, 40ppm, Cu : 0.5ppm, 0.8ppm) for 12 weeks. The food intake, body weight, gain, and FER of Pb groups were significantly lower than those of without Pb groups(p<0.01, p<0.001, p<0.001). The weights of liver in Pb groups were significantly lower than that of adequate Fe and Cu group in without Pb group(AFAC) (p<0.001). The Cu contents of liver in Pb groups were significantly lower than that of AFAC (p<0.01). The liver Pb accumulation of LFLCPb was significantly higher than those of LFACPb, AFLCPb, AFACPb(p<0.05). The serum Pb content of LFLCPb was significantly higher than those of LFACPb, AFLCPb, AFACPb(p<0.05). The levels of Hb, and Hct, of 12 weeks in Pb poisoned rats were lower than those of 8 weeks, in other words, long term Pb poisoned rats were affected more serious on hematoopoiesis. The fecal excretions of Fe and Cu in Pb groups were significantly higher than those of without Pb groups(p<0.001, p<0.001). The urinary Fe and Cu excretions were significantly higher than those of AFAC(p<0.001, p<0.001). There were significant differences between Pb groups and without Pb groups according to dietary Fe and Cu levels. So, dietary levels of adequate Fe and Cu were effective to reduce Pb accumulation in rats. It were showed that the decrease of food intake, FER, body weight gain, serum level of Fe and Cu in Pb groups than those of without Pb groups. And the increase of liver Pb accumulation, serum Pb levels, fecal and urinary Fe and Cu excretions were showed in Pb groups than those of without Pb groups by long term lead poisoning. Otherwise, adequate Fe and Cu administrations in Pb groups have preventive effects on the Pb poisoned rats.

식이내 단백질과 철분수준이 흰쥐의 Fe, Cu 및 Zn 대사에 미치는 영향 (Effect of Different Levels of Dietary Protein and Iron on the Fe, Cu and Zn Metabolism in Rats)

  • This study was performed to investigate the effect of different levels of protein and iron in the diet upon Fe, Cu and Zn metabolism in rat during four weeks of growing period. Forty-five male weanling rats of Sprague-Dawley strain weighing $68.5{\pm}1.1g$ were divided into 9 groups and each group was given with one of the 9 different kinds of diets for four weeks. The three dietary protein levels used were 5, 20 and 40% and Fe levels 0, 35, and 350 ppms. The results obtained were summarized as following ; 1) Food intake and body weight gain in 20%(SP) and 40%(HP) dietary protein groups tended to be significantly higher than 5%(LP) protein groups. Protein efficiency ratio (PER) was higher in LP groups than in HP and SP groups. With dietary Fe levels, there were no significant differences among groups in food intake, body weight gain, and PER. 2) In LP groups, the Fe concentrations in liver, kidney, and hind limb muscle were higher than in SP and HP groups. Regarding with dietary protein levels, the liver Cu concentrations in LP groups were slightly higher, but the liver Zn concentrations were lower in LP groups. The Fe concentrations in liver and kidney tended to decrease with decrease in dietary Fe levels, but Cu and Zn concentrations showed no consistent tendency with dietary Fe levels. 3) The Fe, Cu and Zn concentrations in serum were not different from dietary treatments except that the serum Fe concentrations increased slightly in LP groups. 4) The Fe and Cu concentrations in urine tended to be higher in HP groups. Fecal Cu and Zn concentrations showed no significant differences in dietary protein or Fe levels, but the Fe concentrations tended to increase with increase in dietary Fe levels.

Effects of Dietary Copper Exposure on Accumulation and Histopathological Change in Liver of Juvenile Rockfish, Sebastes schlegeli

  • Experiments were carried out to investigate the accumulation and the histopathological changes in liver of juvenile rockfish, S. schlegeli, after sub-chronic dietary Cu (0, 50, 125, 250 and 500mg/kg) exposure for 60 days. Cu accumulation in liver was significantly increased with dietary exposure period and concentration for 60 days, and has a linear relation with dietary exposure days. After 60 days of Cu dietary exposure, the Cu concentration in the liver was $75.9\pm12.05,\;126.29\pm22.11\;and\;360.44\pm45.26\;{\mu}g/g$ dry weight and was approximately 11-fold, 18-fold and 51-fold higher than in the control diet group at 125, 250 and 500 mg/kg Cu diet group. The accumulation factors were increased with the dietary exposure period in liver of rockfish. In the primary exposed stage, the effect of hepatic tissue in the rockfish exposed to dietary Cu observed enlargement of hepatocytes nuclei, activity of hepatic cells and the swelling of hepatic cells. While exposed time and concentration were increased, the distinct granulation, irregular shape and necrosis of hepatic cells were observed. It was observed that granule degeneration and necrosis showed a part of cells in hepatic tissue after 60 days at 500 mg/kg.

유류에 오염된 아비류 (Loons)의 체내 조직 중 필수원소 농도 (Concentrations of Essential Metals in Tissues of Oiled Loons)

  • Concentrations of four essential metals (Fe, Cu, Mn and Zn) were determined in apparently oil-polluted dead or near-dead three loon species collected in the Busan coast, Korea. Fe, Cu, Mn and Zn concentrations were generally high in liver, and also, Mn and Zn were high in bone. There were significant differences in some tissue concentrations of Fe, Cu, Mn and Zn among three species, but did not show any consistent evidence of species-specific accumulation. Mean concentration of Fe was 11,635$\pm$4010 $\mu$g/g, Cu 64.3$\pm$30.7 $\mu$g/g, Mn 13.1$\pm$2.32 $\mu$g/g, and Zn 121$\pm$24.8 $\mu$g/g in liver of all three species examined. Mn and Zn levels in this study were similar to those of normal loons and other seabirds reported from different countries. However, Fe and Cu liver levels were extraordinarily higher than those of reported. This result suggests that unusual accumulations of essential metals such as Fe and Cu may partially arise from long-term starvation due to oil pollution.

사료 內 Cu 및 Zn-methionine chelates 첨가가 육계의 생산성에 미치는 영향

  • An experiment was conducted investigate the of supplemental Cu and Zn methionine chelates on the performance, nutrient digestibility, serum IgG level, gizzard erosion, and Cu and Zn contents in the liver and excretion of broiler chickens. One thousand hatched broiler chickens (Ross) of one day old were assigned to 4 treatments:control(T1), 100ppm of copper in the from of Cu-methionine chelate(Cu-Met, T2), 100ppm of zinc in the from of Zn-methionine chelate(Zn-Met, T3) and 100ppm of copper plus 100ppm of zinc in the from of methionine chelate(Cu-Zn-Met, T4). Each treatment had four replications of 50 bird each. Weight gain of chicks fed chelated products were significantly higher than that of chicks fed control(P<0.05).Combination of Cu and Zn chelates(Cu-Zn-Met) tended to show the best growth rate and feed conversion ratio. Nutrient digestibilities were not affected by dietary treatments. Serum IgG level of chicks fed Cu-Zn-Met was significantly higher than that of chicks fed control(P<0.05). Gizzard erosion index was not significantly different among treatments. Contents of Cu and Zn in liver were not significantly affected by dietary treatments, whereas excretions of these minerals were significantly affected by dietary treatments.

미량금속 폭로에 있어 생체시료의 특성(제1보) (Studies on the Characteristics of Biological Samples under the Exposure of Trace Metals (Report 1))

  • These studies were carried out to find out some characteristic responses of biological samples under the exposure of trace metals. In animal experiment, six rats per week were killed for 6 weeks under Cd exposures and the concentrations of Cd, Cu and Zn in their organs and blood were determined by flame atomic absorption spectrophotometry.The results were summarized as follows: In animal experiments, the concentrations of Cd in the liver and kidney of Cd exposure groups were increased, however, the values of hematocrit and hemoglobin in blood were decreased remarkably. The body weight of the Cd exposure groups showed lower trend than that of control group. The concentrations of Cd in heart, lung and testes were increased slowly during the first 2 weeks. The concentrations of Cd in their organs decreased while the rapid increase of the concentration of Cd in the liver and kidney was observed. While rapid increase of the concentra~ion of Cd in the liver and kidney was observed, the concentrations of Cd in the organs were decreased afterward and increased again while the low increase of the concentration of Cd in the liver and kidney was observed. The concentrations Cu and Zn were increased remarkably in all the organs for first 2 and 3 weeks. In the exposure groups of Cd+Cu and Cd+Zn, the liver and kidney didn't show lower concentration of Cd than that of Cd group.

Effects of Supplementary Copper Chelates in the Form of Methionine, Chitosan and Yeast on the Performance of Broilers

  • An experiment was conducted to investigate the effects of supplemental copper (Cu) chelates (methionine, chitosan and yeast) on the performance, nutrient digestibility, serum IgG level, gizzard erosion, Cu content in the liver and excreta and the level of total cholesterol in breast muscle and serum of broiler chickens. Two hundred and forty hatched broiler chickens (Ross$^{(R)}$ 208) were assigned to 4 treatments: control, 100 ppm Cu in methionine chelate (Met-Cu), 100 ppm Cu in chitosan chelate (Chitosan-Cu) and 100 ppm Cu in yeast chelate (Yeast-Cu). Each treatment had six replicates of 10 (5 males+5 females) birds each. Weight gain and feed intake tended to be higher in Cu chelate treatments than the control; weight gain was significantly higher in the Met-Cu chelate treatment and feed intake was significantly higher in the Yeast-Cu chelate treatment than the control (p<0.05). Feed/gain was significantly different between treatments in which Met-Cu was lowest followed by the control, Chitosan-Cu and Yeast-Cu. DM availability was increased by Cu chelates among which chitosan-Cu showed the highest DM availability. Cu chelates supplementation tended to increase gizzard erosion index, and Cu content in the liver was highest in the Met-Cu treatment. Supplementation of Cu chelates tended to decrease total cholesterol level in breast muscle and serum but tended to increase the level of HDL in serum. It was concluded that dietary supplementation of 100 ppm Cu in chelates increased weight gain, feed intake and DM availability. Met-Cu was more effective than Chitosan-Cu or Yeast-Cu in improving productivity of broiler chickens.

Influence of Dietary Zinc, Copper and Cadmium Levels on Rat Liver Aryl Sulfotransferase IV Activity

  • Aryl sulfotransferase (AST) IV is a liver enzyme involved in detoxication and has been shown to be susceptible to down regulation by a number of hepatotoxic xenobiotics. Studies presented here to investigate the ability of biological and non-biological divalent metal cations on AST IV activity showed that AST IV was strongly inhibited following in vitro or in vivo exposure to. Zn ( II ), Co ( II ) or Cd ( II ). It was found that $0.025\sim$2.5 uM of these metal ions were sufficient to cause $50\%$ of inhibition in vitro in purified AST IV and $0.25\sim$25 uM of these metal ions in liver cytosolic fractions. For the in vivo study, 1,000 mg Cu ( II )/kg, 2,000 mg Zn ( II )/kg or 250 mg Cd( II )/kg was added to individual diets and administered to three (3) group; of mts over a 7 week period The Co ( II )-supplemented diet produced no apparent change in rat growth rate and resulted in 30-fold increase in liver cytotolic Cu ( II ) levels, suggesting that elevated levels of Cu ( II ) ion in the liver were responsible for the loss of AST IV activity. In contrast, the Zn ( II )-supplemented diet caused a decrease in rat growth rates and resulted in zero increase in liver Zn ( II ) levels, which suggested an indirect inhibition mechanism was caused by Zn ( II ) in the liver. Rats were fed the Cd-supplemented diet also displayed a decrease in growth rate with little or no change in liver Cu ( II ) or Zn ( II ) levels. When the liver cytosols of mts from the metal ion diets were immunochemically analyzed for the AST IV and albumin contents, no significant changes were observed in albumin levels. However, AST IV contents in the cytosols of mts fed the Zn ( II )-supplemented diets showed a slight decrease in amount These results showed that AST IV activity in vitro and in vivo can be inhibited by Co ( II ), Zn ( II ), and Cd ( II ) by apparently different mechanisms. The immediate response to a Zn injection showed a decrease in AST IV activity but not in the AST IV content in liver cytosol. These mechanisms appeared to involve direct actions of the metal ion on AST IV activity and indirect actions affecting AST IV amount.

한국인 간장 및 신장피질에 함유된 카드뮴, 구리, 수은, 아연 함량과 메탈로치오나인과의 관계 (Relationships Between Cadmium, Copper, Mercury, Zinc Levels and Metallothionein in the Liver and Kidney Cortex of Korean)

  • In order to elucidate the relationships between cadmium, copper, mercury, zinc levels and metallothionein in the liver and kidney cortex of Korean, the levels of Cd, Zn, Hg, Cu and metallothionein (MT) were determined in the kidney cortex and liver of 50 subjects deceased in the period of January-November, 2001 in the area of Seoul and Gyeonggi Province of Korea. The mean age of the population studied was 36.3+/-12.3 years. The tissues were digested with microwave digestion system and the elements were determined by inductively coupled plasma atomic emission spectrometry. MT was determined by the Cd-hemoglobin affinity assay. The determined levels (mean+/-SD) were: 33.9+/-18.9 micrograms Cd/g wet weight; 47.5+/-12.6 micrograms Zn/g wet weight; 2.5+/-0.57 microgram Cu/g wet weight; 0.26+/-0.31 micrograms Hg/g wet weight, 4.0+/-3.1 mg MT/g wet weight in renal cortex and 2.5+/-1.9 micrograms Cd/g wet weight; 46.9+/-15.0 micrograms Zn/g wet weight; 6.2+/-2.5 micrograms Cu/g wet weight; 0.10+/-0.15 micrograms Hg/g wet weight, 0.92+/-0.57 mg MT/g wet weight in the liver. Positive relationships between Cd and MT, sum of four divalent metal and MT in the kidney cortex were observed. No other correlation was found between Cu and MT, Hg and MT, Zn and MT in either organs.

한반도 동해안 밍크고래 체내 미량금속 잔류특성 (Characteristics of Metal Concentration in the Organs of Minke Whale Entangled from the Eastern Coast of Korea)

  • 한반도 연안에 서식하는 고래의 조직 내 미량금속 농도 잔류특성을 알아보기 위하여 2009년 동해안에서 혼획된 밍크고래의 간, 창자, 근육, 표피, 지방 내 미량금속(Cu, Cd, Pb, As, Zn, Hg) 농도를 측정하였다. 조직 내 Cu, Cd, Zn, Hg은 간에서 가장 높고 지방에서 가장 낮았다. 이와 대조적으로 As는 지방에서 가장 높은 농도를 보였으며, 근육에서 가장 낮은 농도를 나타내었다. 전반적으로 밍크고래의 조직 내 미량금속은 Zn > Cu > As > Pb > Hg > Cd의 순으로 높은 농도를 나타내었다. 성별에 따른 미량금속은 수컷은 간과 표피에 Cu, As, Zn을, 암컷은 창자와 근육에 Cu와 Pb을 더 효율적으로 축적하고 있는 것으로 나타났다. 성숙도에 있어서는 간의 경우 Cd, 근육은 Hg, 표피와 지방은 As와 Hg, 창자는 Cd, As, Hg이 성숙한 개체에서 미성숙한 개체보다 상당히 높은 농도를 보였다.
