Effects of Candida norvegensis Live Cells on In vitro Oat Straw Rumen Fermentation

  • This study evaluated the effect of Candida norvegensis (C. norvegensis) viable yeast culture on in vitro ruminal fermentation of oat straw. Ruminal fluid was mixed with buffer solution (1:2) and anaerobically incubated with or without yeast at $39^{\circ}C$ for 0, 4, 8, 16, and 24 h. A fully randomized design was used. There was a decrease in lactic acid (quadratic, p = 0.01), pH, (quadratic, p = 0.02), and yeasts counts (linear, p<0.01) across fermentation times. However, in vitro dry matter disappearance (IVDMD) and ammonia-N increased across fermentation times (quadratic; p<0.01 and p<0.02, respectively). Addition of yeast cells caused a decrease in pH values compared over all fermentation times (p<0.01), and lactic acid decreased at 12 h (p = 0.05). Meanwhile, yeast counts increased (p = 0.01) at 12 h. C. norvegensis increased ammonia-N at 4, 8, 12, and 24 h (p<0.01), and IVDMD of oat straw increased at 8, 12, and 24 h (p<0.01) of fermentation. Yeast cells increased acetate (p<0.01), propionate (p<0.03), and butyrate (p<0.03) at 8 h, while valeriate and isovaleriate increased at 8, 12, and 24 h (p<0.01). The yeast did not affect cellulolytic bacteria (p = 0.05), but cellulolytic fungi increased at 4 and 8 h (p<0.01), whereas production of methane decreased (p<0.01) at 8 h. It is concluded that addition of C. norvegensis to in vitro oat straw fermentation increased ruminal fermentation parameters as well as microbial growth with reduction of methane production. Additionally, yeast inoculum also improved IVDMD.

Effects of Dietary Synbiotics from Anaerobic Microflora on Growth Performance, Noxious Gas Emission and Fecal Pathogenic Bacteria Population in Weaning Pigs

  • Synbiotics is the term used for a mixture of probiotics (live microbial feed additives that beneficially affects the host animal) and prebiotics (non-digestible food ingredients that beneficially affect the organism). This study investigated the effect of probiotics from anaerobic microflora with prebiotics on growth performance, nutrient digestibility, noxious gas emission and fecal microbial population in weaning pigs. 150 pigs with an initial BW of 6.80${\pm}$0.32 kg (20 d of age) were randomly assigned to 5 dietary treatments as follows: i) US, basal diet+0.15% antibiotics (0.05% oxytetracycline 200 and 0.10% tiamulin 38 g), ii) BS, basal diet+0.2% synbiotics (probiotics from bacteria), iii) YS, basal diet+0.2% synbiotics (probiotics from yeast), iv) MS, basal diet+0.2% synbiotics (probiotics from mold), v) CS, basal diet+0.2% synbiotics (from compounds of bacteria, yeast and mold). The probiotics were contained in $10^{9}$ cfu/ml, $10^{5}$ cfu/ml and $10^{3}$ tfu/ml of bacteria, yeast and molds, respectively. The same prebiotics (mannan oligosaccharide, lactose, sodium acetate and ammonium citrate) was used for all the synbiotics. Pigs were housed individually for a 16-day experimental period. Growth performance showed no significant difference between antibiotic treatments and synbiotics-added treatments. The BS treatment showed higher (p<0.05) dry matter (DM) and nitrogen digestibility while ether extract and crude fiber digestibility were not affected by the dietary treatment. Also, the BS treatment decreased (p<0.05) fecal ammonia and amine gas emissions. Hydrogen sulfide concentration was also decreased (p<0.05) in BS, YS and MS treatments compared to other treatments. Moreover, all the synbioticsadded treatments increased fecal acetic acid concentration while the CS treatment had lower propionic acid concentration than the US treatment (p<0.05) gas emissions but decreased in fecal propionate gas emissions. Total fecal bacteria and Escherichia coli populations did not differ significantly among the treatments, while the Shigella counts were decreased (p<0.05) in synbiotics-included treatment. Fecal bacteria population was higher in the YS treatment than other treatments (p<0.05). The BS treatment had higher yeast concentration than YS, MS and CS treatments, while US treatment had higher mold concentrations than MS treatment (p<0.05). Therefore, the results of the present study suggest that synbiotics are as effective as antibiotics on growth performance, nutrient digestibility and fecal microflora composition in weaning pigs. Additionally, synbiotics from anaerobic microflora can decrease fecal noxious gas emission and synbiotics can substitute for antibiotics in weaning pigs.

  • The cyprinid loach, Misgurnus mizolepis $G\ddot{u}nther$, a freshwater teleost, is a member of the family Cobitidae and is known to distribute in Korea and China. This fish is considered to be an excellent food fish in Korea. This report is a description of the gross morphology of the embryos and larvae up to the development of juvenile period of life. The eggs were obtained by injecting gonadotropin and fertilized by wet method in the laboratory on July 10 in 1986. The inseminated eggs were rinsed with tap water and incubated at $25^{\circ}C$. Live, freshly laid eggs were separative, demersal, adhesive, light yellow and average 1.1mm in diameter. The hatching took place in about yellow and average 1.1mm in diameter. The hatching took place in about 24 hours after fertilization. The newly hatched larvae were average 2.7 mm in total length possessing yolk sac and 35-36 myotomes. Ten hours after hatching, the prelarvae attained 3.5 mm in total length. The first heart pulse and melanophores at the side of the body were observed at this stage. Yolk sac was completely reserved at four days after hatching. The larvae that was cultured 24 days after hatching, 15.4-16.4 mm in total length, entered the juvenile period of life.   • Probiotics are live organisms that when administered in adequate amounts confer a health benefit on hosts. The administration of direct-fed microbials (DFM) such as lactobacilli and bacillus, may be a more direct approach to beneficially alter gastrointestinal microflora than altering dietary ingredients or supplementing with growth-promoting levels of antibiotics. It is apparent that microbes have an important influence on immune development and resistance to infection; that microbes are not static colonizers of our bodies, but are dynamic, symbiotic coresidents. And it can improve the surrounding environments; decrease the malodor caused by degrade the excrement. Recently, new paradigm such as environment protection and safe food have been settled. In domestic farm house, there is a great demand for probiotics as a substitute of antibiotics for the improvement of environmental quality and the production of a competitive goods. Probiotics circulated in a country have three categories: an animal medicine permitted by national veterinary research quarantine service (NVRQS), a support feed registered in city or country house, and not-registered goods. However, lots of unqualified goods were produced and circulated. And thus, it is in urgent need of evaluating the present situation and effect of probiotics. This study was conducted to evaluate the system of a probiotics as a livestock-environment improving agents for the alternation of antibiotics and quality control of it.

Antilisterial activity of fresh cheese fermented by Lactobacillus paracasei BK57 (Lactobacillus paracasei BK57 균주로 발효시킨 프레쉬 치즈의 항리스테리아 활성)

  • This study is focused on establishing the optimal conditions to enhance the production of antilisterial substances by Lactobacillus paracasei BK57 isolated from Baikkimchi. In addition, the growth and in situ lactic acid and bacteriocin production of this strain were investigated during the manufacture of fresh cheese. And then the efficacy of using Lactobacillus starter as a protective culture to improve the safety of fresh cheese against Listeria monocytogenes KCTC 3569 was estimated. Maximum growth rate and activity of antibacterial substances were obtained in Lactobacilli MRS broth at $37^{\circ}C$ with controlled pH 6.0 after 30 h of incubation under aerobic condition. However, the growth rate and antimicrobial activity of bacteriocin produced in whole milk supplemented with yeast extract (2.0%) as a substrate were lower than those obtained in MRS broth. Live cells and cell-free culture supernatant of BK57 strain were effective in the suppression of L. monocytogenes in milk, whereas the inhibitory of the bacteriocin obtained from BK57 strain was higher in BHI broth than in milk. During storage at $4^{\circ}C$ and $15^{\circ}C$ for 6 days, no significant difference was found in the cell viability and antimicrobial activity of BK 57 strain in fresh cheese. In samples held at two temperatures, there was at least a 15% reduction in the numbers of the pathogen in fresh cheese artificially contaminated with approximately $10^5CFU/ml$ of L. monocytogenes within 6 days. Our results demonstrated the usefulness of L. paracasei BK57 having antilisterial activity as a biopreservative in the cheese making process.

Artificial Spawning, Larval and Spat Developments of the Bay Scallop, Argopecten irradians (해만가리비, Argopecten irradians의 인공산란과 유생 및 치패의 발생)

  • Artificial spawning, larval and spat developments of the bay scallop, Argopecten irradians, which was transplanted from China on 16 August 1996, were investigated monthly until August 1997 in the Deukyang Bay, Jangheung-gun, Jeollanam-do, Korea. Sufficient amount of cultured microalgae supplemented seawater were supplied as food (6 kinds of phytoplankton) for fully grown adult individuals at 17.1-23.2$^{\circ}C$ for 44 days. A total of 45,320,000 eggs were spawned by way of 2 times of artificial spawning inductions such as exposure stimulus to the air and thermal shock (with water temperature) on 29 January and 31 January in 1997. Artificially fertilized eggs were developed to D-shaped larvae (77.5 ${\times}\;63.8\;{\mu}m)$ and metamorphosed to larvae (191.8 ${\times}\;181.2\;{\mu}m)$ 181.2 m) in the attached larval stage on the collectors. A total of 110,000 spats (average 3.04 mm in shell length) were produced at 22.8-26.3$^{\circ}C$ and 31.0-34.4 psu in the indoor rearing tank from 14 February through 7 May in 1997. In case of Argopecten irradians, if the attached larvae in the attachment stage are detached from the collector, they could not live. Accordingly, it is assumed that survival (%) of the attached larvae of A. irridians showed very low because of weak power attached to the collector due to the small number of the byssuses of the attached larva, not the short attachment period by the byssus as seen in other scallops such as Argopecten balloti.

Dietary Value of Three Benthic Diatom Species on Haliotis discus hannai Larvae (북방전복 Haliotis discus hannai 유생에 대한 3종 부착 규조류의 먹이효율)

  • Although the method of seedling production of Haliotis discus hannai is well known, the optimum benthic diatom species as a live food at early larval stage are not fully developed. In this study three Pennales diatom species, Caloneis schroederi, Rhaphoneis sp., and Cocconeis californica were examined on settlement, metamorphosis, survival, and growth of Haliotis discus hannai larvae. The larvae fed Raphoneis sp. or C. californica showed high settlement rate with 80-82% within 48 hrs, which was significantly higher than those fed C. schroederi or mixed diets with three diatom species. The larvae fed the former microalgal species also showed higher metamorphosis rate with 32-34% than the latter species with 10-12% within 4 days. With regard to survival and growth of the larvae, single diet with Rhaphoneis sp. or C. californica had better dietary value than the mixed diets for the early larvae of H. discus hannai.

Selection of Optimum Species of Tetraselmis for Mass Culture (대량배양에 적합한 Tetraselmis종의 선택)

  • Tetraselmis is widely used as a live food because of its easy handling, high nutrient, large size and wide tolerant range of temperature and salinity. In order to find the optimum Tetraselmis species for mass culture in Korea, five species of this microalgae were examined on size, optimum culture condition ${\textperthousand}$s, $^{\circ}C.$) and nutrient composition. The results obtained were as follows: Among five species of Tetraselmis, T. sp.(Haeundae) was the largest(major axis $17.6{\pm}1.87^{\mu}$m, mean cell volume 727${\mu}$m), and T. sp. (China) the smallest (major axis $14.6{\pm}1.46^{\mu}$m, mean cell volume 625m). Tetraselmis was very eurythermal and euryhaline species. But optimum temperature and salinity for growth were 24~$30^{\circ}C.$ and 27~30${\textperthousand}$, respectively. Among five species of Tetraselmis, T. sp. (China) seemed to be the most tolerant of high temperature over $30^{\circ}C.$, and T. tetrathele of low temperature below $6^{\circ}C.$. In culture density, T. suecica showed the highest growth rate among the among the five species. The cell density of this microalgae attained to $141{\times}10^4$cells/ml at $24^{\circ}C.$ and 30${\textperthousand}$ within 7 days. In chemical composition, crude protein amount was the highest in T. suecica (44.50%), and crude lipid amount it T. sp. (Haeundae, 7.13%). Total essential amino acid amount was the highest in T. sp. (Haeundae, 50.4%) and total polyunsaturated amount in T. sp. (China, 11.7%) The results on growth and chemical composition of five species of Tetraselmis indicated that T. suecica seemed to be the most suitable species for mass culture in Korea.

  • This study investigates the impact of the attributes of the eco-friendly agricultural products traceability system on perceived value and behavioral intention. Empirical subjects are those who live in Seoul and Gyeonggi province, over the age of 20 and have experience of buying eco-friendly agricultural products though department stores, discount stores and specialized eco-friendly product stores. After distributing 550 copies of questionnaire from April 10th, 2013 to May 9th, 2013, 470 copies(85.5%) are used for a final statistical analysis of the survey after excluding the copies with biased opinions or missing values. The results of this study show sufficient theoretical base for future research by verifying a causal relationships between the attributes of eco-friendly agricultural products traceability system such as reliability, safety and innovation, and consumers' perceived value and behavioral intention. Therefore, producers should correctly record producing history base so that consumers can continue to use the system. The traceability system can not only satisfy consumers by enhancing transparent management and producing systems for eco-friendly products but also guarantee us to eat all products we get without worries.

Comparison of Gene Expression in Larval Fat Body of Helicoverpa assulta in Different Temperature Conditions (온도변화에 따른 담배나방 유충 지방체의 유전자 발현 비교 분석)

  • Insects are known to live at wide range of temperature, but can not survive when they are exposed to over $40^{\circ}C$ or below supercooling point. The larvae of Helicoverpa assulta have been reared at high ($35^{\circ}C$), low (3 to $10^{\circ}C$), and room temperature ($25^{\circ}C$; control). To identify stress-related genes, the transcriptomes of fat body have been analyzed. Genes such as cuticular proteins, fatty acyl ${\Delta}9$ desaturase and glycerol 3 phosphate dehydrogenase were up-regulated whereas chitin synthase, catalase, and UDP-glycosyltransferase were down-regulated at low temperature. Superoxide dismutase, metallothionein 2, phosphoenolpyruvate carboxykinase and trehalose transporter have been up-regulated at high temperature. In addition, expressions of heat shock protein and glutathione peroxidase were increased at high temperature, but decreased at low temperature. These temperature-specific expressed genes can be available as markers for climate change of insect pests.
