Saponin Composition of Liriope platyphylla and Ophiopogon japonicus (한국산 맥문동과 중국산 소엽맥문동의 사포닌 성분)

  • Saponin is one of major conponents in oriental medicine, which':s present in Ophiopogoneae. Saponin composition on ethyl acetate fractions from the butanolic extracts of two species, Liriope platyphylla and Ophiopogon japonicus was measured using high performance liquid chromatography (HPLC). HPLC analysis revealed that spicatoside A and B were identified in the tuber of L. platyphylla. However, spicatoside A was not identified in the tuber of O. japonicus. Related to that, it may be possible to discriminate between Liriopis and Ophiopogonis Tuber using HPLC analysis.

  • Nerve growth factor (NGF) is a protein plays a major role in the development and maintenance of central and peripheral nervous system. Recent data suggest that reduced availability of NGF may play a significant role in the pathogenesis of diabetic $polyneuropathy.^{1)}$ In our previous study, steroidal saponin from Liriope platyphylla showed neurotrophic effect by stimulation of NGF synthesis and activation of tyrosine kinase $signaling.^{2)}$ In this study, we examined the neurotrophic effect of Liriope platyphylla (LP); which was from Mylyang(MYL) and Cheongyang(CHE), and Ophiopogon japonicus (CHI) on in vitro and in vivo model for the comparison of their NGF induction. We quantitatively analyzed spicatoside A in the LP and CHI by HPLC. And we investigated the correlation between the contents of spicatoside A and NGF induction efficacy on PC 12 cells and mouse serum. These results suggest that spicatoside A may enhance NGF induction in animal model.

First Report of Larval Damage to Liriope spicata by Mahasena aurea (Lepidoptera: Psychidae) in Korea with a Redescription of External Morphology of the Larva and Adult (개맥문동의 새로운 해충 검정주머니나방 (나비목: 주머니나방과) 유충의 발견 및 형태특징 재기재)

  • In this study, one species of the bag moth, Mahasena aurea (Butler) (Lepidoptera: Psychidae), was found for the first time in Korea as a new insect pest on leaves of Liriope spicata (Thunb.) Lour. Also, the female larvae and adults of the species were first collected in this country. Larvae, pupae, and adults of both sexes are redescribed with illustrations of all external characteristics including the genitalia of the male.

New Record of Anthracnose Caused by Colletotrichum liriopes on Broadleaf Liriope in Korea

  • In 2015, the leaves of broadleaf liriope, Liriope muscari, showing anthracnose symptoms were found in a field of Korea. The fungus was isolated and identified as Colletotrichum liriopes based on the morphological characteristics and nucleotide sequence data of the glyceraldehyde-3-phosphate dehydrogenase, internal transcribed spacer and actin genes. To the best of our knowledge, this is the first report of C. liriopes isolated from L. muscari in Korea.

The Quality Characteristics of Baechukimchi added with Broadleaf Liriope (Liriope platyphylla) (맥문동을 첨가한 배추김치의 품질 특성)

  • The purpose of the study is to examine the convergence effects of the addition of Broadleaf liriope (Liriope platy phylla) on the taste and fermentation characteristics of kimchi. Based on the weight of the total materials used for making kimchi, the added rates of Liriope platy phylla were 0, 3, 5. 7, 10%, respectively. Kimchi was fermented for 28 days at $5^{\circ}C$ to investigate pH, acidity, total and lactic acid bacteria and sensory evaluation was performed. The control in pH showed a tendency to get rapidly lower, and the treatment slowly lower until 14 days, and it maintained the optimal taste. In the acidity change, the control rapidly rose after 14 days of fermentation, and the treatment maintained a regular level after 21 days. In the change of the total bacteria, the control increased after 21 days of maturing and the treatment progressed maturing without a big change after 14 days, and the lactic acid bacteria increased and had almost no change after 21 days. The sensory test, overall acceptability, flavor, taste, sour taste, palatability and texture were significantly high, and in the flavor, palatability and overall acceptability, the Liriope platy phylla 5% added treatment was the highest. The Liriope platy phylla 5% added treatment is expected to improve the taste and hypotonicity of kimchi.

Effects of Water Extract of Liriope platyphylla on the mRNA Expression and Protein Secretion of Nerve Growth Factors (맥문동 유수추출물이 NGF의 mRNA발현과 단백질 분비에 미치는 영향)

  • Liriope platyphylla has been though as an useful medical plant to improve the cough, sputum, neurodegenerative disorders, obesity, and diabetes in Korea and China from old times. In order to investigate the effects of Liriope platyphylla on expression and secretion of nerve growth factor (NGF), the mRNA expression and protein secretion were detected in the neuronal cell (B35) and neuroglial cell (C6) cultured with three differences concentration (5%, 10%, 15%) of Liriope platyphylla. In MTT assay and FACS anslysis, the some death of some B35 and C6 cells were observed in 15% extract-treated group, while other groups did not induce the death. Also, the mRNA expression of NGF were significantly increased in 5% and 10% extracts treated-group. Furthermore, the NGF protein concentration in supernatant collected from cultured cells showed the very similar pattern with mRNA expression. In order to verify the activity of secreted NGF, the culture supernatant collected from B35 and C6 cells cultured with Liriope platyphylla extracts for 24 hrs were treated into undifferentiated PC12 cells, and the differentiation level of PC12 cell were also observed with microscopes. The differentiation level of PC12 cell were significantly increased depend on the dose of extract. Therefore, these results suggested that the water extracts of Liriope platyphylla may contribute the regulation of NGF expression and secretion in the neuronal cell and be considered as an excellent candidate for a neurodegenerative disease-therapeutic drug.

Evaluation of Growth of Groundcovers as Affected by Planting Densities under the Roadside Trees (가로수 하부의 식재밀도에 따른 지피초화류의 생육 평가)

  • This field experiment aimed to identify the optimal planting density for establishing a management plan for ground-cover plants under roadside trees. Liriope platyphylla and Hosta longipes both widely used for planting under trees were selected as the plant materials and planted under Prunus serrulata var. spontanea at different planting densities. Based on the distance between each plant, 4 planting densities were used: 11%, 25%, 49%, and 83% with three replications. To estimate plant growth, plant height, number of leaves and tillers, fresh and dry weight, and visual quality were investigated. Liriope platyphylla exhibited relatively better growth at the highest planting density of 83%. For Hosta longipes, however, the lower growth was positively correlated with high planting density. Therefore, the optimal planting density for Liriope platyphylla is 83% and for Hosta longipes is 11%.

An Analysis of Saengmaegsan's Ingredients and a Comparison Study on Anti-Oxidation Effects According to Kinds of Extract (생맥산(生脈散)과 구성 약물의 성분 분석 및 항산화(抗酸化) 효능 비교)

  • Objectives: The experiment was to estimate the value of Saengmaegsan as a therapeutic anti-aging agent with an analysis of ingredients and their bio-activating effects by enzymologic methods. Methods: We analyzed anti-aging effects of the ingredients of Saengmaegsan being Liriope Platyphylla, Schizandrae Chinensis, and Schizandrae Chinensis. Results: In Liriope Platyphylla and Schizandrae Chinensis, reducing sugar concentration was shown at the highest level. Of mineral contents, K+ and Na+ were the highest in Saengmaegsan and each ingredient also, and then Ca++ and Al. The amount of polyphenol showed was in order Schizandrae Chinensis > Saengmaegsan > Panax Ginseng > Liriope Platyphylla. The electron donating ability of Schizandrae Chinensis doubled that of Saengmaegsan and Panax Ginseng, at the same time that one per 1 ml in Saengmaegsan was significantly found the greatest level as compared to each single ingredient. SOD-like activity was high in Saengmaegsan and Schizandrae Chinensis. SOD-like activity of Saengmaegsan was higher than in single components. Nitric oxide inhibition in pH 1.2 was high in Saengmaegsan and Schizandrae Chinensis, and for per 1 ml it showed the same pattern as above. In pH 3.0 the result was not different from in pH 1.2. Xanthine oxidase inhibition was high in Schizandrae Chinensis and for per 1 ml showed the greatest activity in Liriope Platyphylla. Tyrosinase inhibition in Omija was the most noticeable, and for per 1 ml was Liriope Platyphylla more than any others. Conclusions: With this analysis of ingredients, it is proven that Saengmaegsan and each component fosters antioxidation. On the whole, the composite prescription Saengmaegsan was superior to each individual component.

Regulation of Gastric Acid Secretion of Liriope platyphylla Extract in Gastroesophageal Reflux Disease

  • Objectives: The purpose of this study was to confirm the effects of Liriope platyphylla extract on relieving Gastroesophageal reflux disease (GERD) through regulation of acid secretion. Methods: 8-week-old ICR mice were divided into untreated control group (Ctrl), GERD elecitation group (GERDE), Omeprazole administrate group before GERD elicitation (OMA), and Liriope platyphylla extract administrate group before GERD elicitation (LPA). After inducing GERD, gross observation and histological examination were performed and ATP6V1B1 (ATPase H+ Transporting V1 Subunit B1), GRPR (Gastrin-releasing peptide receptor), COX-1 (Cyclooxygenase 1), 8-OHdG (8-hydroxy-2'-deoxyguanosine), Cathelicidin, p-JNK (phospho c-Jun N-terminal kinase) were observed to confirm the damage defense effect of the esophageal mucosa, acid secretion regulation, antioxidant, anti-inflammatory, mucosal protection, and apoptosis regulation Results: OMA and LPA showed lower levels of damage compared to GERDE in gross observation and histological examination. ATP6V1B1, GRPR, and 8-OHdG showed lower positive reactions in OMA and LPA than in GERDE. COX-1 were less positive in GERDE and OMA than in Ctrl, but showed higher secretion in LPA than in Ctrl. Cathelicidin showed a decreased positive reaction in GERDE, OMA and LPA compared to Ctrl, but the decrease in positive reaction was smaller in OMA and LPA compared to GERDE. p-JNK showed increased positive reaction in GERDE, OMA and LPA than in Ctrl, but the increase in the positive reaction was smaller in the OMA and LPA compared to GERDE. Conclusions: The effects of Liriope platyphylla extract on esophageal mucosal damage protection, acid secretion regulation, antioxidant, anti-inflammatory, mucosal protection and apoptosis regulation were confirmed.

Cosmetics Preservation and Moisturizing Effect by Methanol Extracts of Scutellaria baicalensis George and Liriope platyphylla (황금(黃芩)과 맥문동 추출물에 의한 화장품 방부 및 보습효과)

  • Methanol extract (1.0 wt%) of Scutellaria baicalensis George satisfied preservation standard of CTFA (The Cosmetics, Toiletry, and Fragrance Association) against Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans and Escherichia coli which were inoculated in skin lotion. Even at a low concentration (0.01 wt%) the extract showed high superoxide dismutase-like activity. The CTFA standard could also be satisfied for the above four strains when methanol extract of Liriope platyphylla were combined with the 1.0 wt% extract of S. baicalensis George. This result suggests that extract of L. platyphylla does not hinder the preservation activity of S. baicalensis George. Moreover, skin moisture content could be maintained for 6 hours after the application of a cream containing the extract of L. platyphylla.
