Isolation and Chemical Analysis of Potent Anti-Complementary Polysaccharides from Fruiting Bodies of the Fomes fomentarius (말굽버섯 자실체에서 분리한 항보체 활성 다당체의 화학적 분석)

  • The five anti-complementary polysaccharides (MFKF-NP, MFKF-AP1${\alpha}$, ${\beta}$, and MFKF-AP2${\alpha}$, ${\beta}$) were separated from hot water extracts of fruiting bodies of Fomes fomentarius by two subsequent column chromatography using DEAE-sepharose FF and Concanavalin A-sepharose 4B. The order of anti-complementary activity was MFKF-AP1${\beta}$ > MFKF-AP1${\alpha}$ > MFKF-AP2${\alpha}$ > MFKF-AP2${\beta}$ > MFKF-NP > Polysaccharide Krestine (PSK). Especially, MFKF-AP1${\beta}$ among those showed the most excellent anti-complementary activity (70% of ITCH50 value at $20{\mu}g/ml$). The monosaccharide composition analysis by gas chromatography indicates that MFKF-AP1${\alpha}$ and ${\beta}$ are a kind of homoxylan consisted mainly of xylose above 97%. Molecular weight of MFKF-AP1${\beta}$, major anti-complementary polysaccharide, was estimated to be about 12,000 by high performance liquid chromatography (HPLC). After the incubation of the serum with MFKF-AP1${\beta}$ in the presence or absence of $Mg^{++}$ and $Ca^{++}$ ions, its anti-complementary activity was investigated. This result indicated that MFKF-AP1${\beta}$ seems to be activator both on the classical and the alternative pathway of complement activation.

Effects of 2-Mercaptoethanol on the Protoplast Formation and Osmotic Stabilizers on the Protoplast Reversion of Pyricularia oryzae Cavara (벼 도열병균(稻熱病菌)의 원형질체(原形質體) 생성(生成)에 미치는 2-Mercaptoethanol과 복귀(復歸)에 미치는 삼투압 안정제(安定劑)의 영향(影響))

  • The mycelia of Pyricularia oryzae were treated with enzyme solution mixture consisting of Driselase, ${\beta}-Glucuronidase$, Cellulase, and Macerozyme R-10 for the production of protoplasts. More protoplasts were formed from mycelia of race KJ 101 of P. oryzae than that of race KI 315a in the enzyme mixtures. The number of protoplasts was decreased in the untreated control three hrs after the enzyme treatment, whereas the number was increased in the treatments with 10, 50 and 100 mM 2-mercaptoethanol, respectively. The number of protoplasts increased to reach maximum at five hrs after treatment of 10 mM 2-mercaptoethanol, but the least was found in 200 mM. The protoplasts of P. oryzae in a liquid medium containing 2.5% yeast extract, and 2% dextrose reverted to the mycelia after five hrs shaking incubation at $27^{\circ}C$. Some protoplasts produced yeast like buds and the buds were developed to irregularly shaped chains of cell protruded a germ tube like hypha from the distal cell. Once in a while a germ tube like hypha protruded directly from the protoplasts. Except in the first type of reversion, other protoplasts reverted to the normal mycelia. The reversion frequency was highest on PDA with stabilizer of 0.6 M KCl. No reversion of protoplasts occurred on water agar regaardless oftreaatments.

Estimation of Surface Solar Radiation using Ground-based Remote Sensing Data on the Seoul Metropolitan Area (수도권지역의 지상기반 원격탐사자료를 이용한 지표면 태양에너지 산출)

  • Solar energy is calculated using meteorological (14 station), ceilometer (2 station) and microwave radiometer (MWR, 7 station)) data observed from the Weather Information Service Engine (WISE) on the Seoul metropolitan area. The cloud optical thickness and the cloud fraction are calculated using the back-scattering coefficient (BSC) of the ceilometer and liquid water path of the MWR. The solar energy on the surface is calculated using solar radiation model with cloud fraction from the ceilometer and the MWR. The estimated solar energy is underestimated compared to observations both at Jungnang and Gwanghwamun stations. In linear regression analysis, the slope is less than 0.8 and the bias is negative which is less than $-20W/m^2$. The estimated solar energy using MWR is more improved (i.e., deterministic coefficient (average $R^2=0.8$) and Root Mean Square Error (average $RMSE=110W/m^2$)) than when using ceilometer. The monthly cloud fraction and solar energy calculated by ceilometer is greater than 0.09 and lower than $50W/m^2$ compared to MWR. While there is a difference depending on the locations, RMSE of estimated solar radiation is large over $50W/m^2$ in July and September compared to other months. As a result, the estimation of a daily accumulated solar radiation shows the highest correlation at Gwanghwamun ($R^2=0.80$, RMSE=2.87 MJ/day) station and the lowest correlation at Gooro ($R^2=0.63$, RMSE=4.77 MJ/day) station.

Antioxidant Activity of Hwangki and Beni-Koji Extracts and Mixture (황기와 홍국추출물 및 혼합물의 항산화 활성)

  • This study was conducted to evaluate the antioxidant activity of liquid beni-koji (LBK), 70% ethanol extracts of beni-koji (EEB) and water extract of Hwangki (WEH). The yields of freeze dried powder of LBK, EEB and WEH were 32.17 g/L, 23.61 g/kg and 196.33 g/kg, respectively. Electron donating ability at 1% (w/v) of LBK, EEB and WEH were 82.67%, 15.71% and 8.60%; reducing power (OD700) were 2.06, 1.64 and 0.45, respectively. SOD-like activities were 24.32%, 11.11%, and 17.94%; nitrite scavenging activities were 74.92%, 72.31% and 31.83%, respectively. TBARS (%) were in order of LBK (69.65%)> EEB (67.32%)> WEH (4.42%). Electron donating ability at 1% (w/v) of EEB : WEH (1:1, w/w. EW), LBK : WEH (1:1, w/w. LW), EEB : LBK: WEH (1:1:1, w/w. ELW) were 14.58%, 60.66% and 20.42%; reducing power ($OD_{700}$) were 1.06, 2.01 and 1.71; SOD-like activities were 18.50%, 26.94% and 18.25%, respectively. While nitrite scavenging activities and TBARS (%) of ELW was higher than those of other materials. Total polyphenol content of LBK, EEB, WEH, EW, LW, ELW were 3.98%, 3.61%, 3.02%, 3.23%, 3.46% and 3.38%; total flavonoid content were 0.89%, 3.91%, 0.30%, 2.59%, 0.46% and 2.33%, respectively. In conclusion, this study provides experimental evidence that mixture of LBK, EEB and WEH could be used as a source of antioxidant ingredients in the food industry.

Reaction Mechanism and Curing Characteristics of Chicken Feather-Based Adhesives and Adhesive Properties of Medium-Density Fiberboard Bonded with the Adhesive Resins (닭털로 제조한 접착제의 반응기작 및 경화 특성과 이를 이용하여 제조한 중밀도섬유판의 접착 특성)

  • In this study, reaction mechanism and curing characteristics of adhesives formulated with NaOH- and $H_2SO_4$-hydrolyzed chicken feather (CF) and formaldehyde-based crosslinkers were investigated by FT-IR and DSC. In addition, adhesive properties and formaldehyde emission of medium-density fiberboards (MDF) applied with the adhesives were measured. CF-based adhesives having a solid content of 40% and over were very viscous at $25^{\circ}C$, but the viscosity reduced to $300{\sim}660m{\cdot}Pa{\cdot}s$ at $50^{\circ}C$. Consequently, the adhesives could be used as a sprayable resin. Through the FT-IR spectra of liquid and cured CF-based adhesives, addition reaction of methylol group and condensation reaction between the functional groups with the use of formaldehyde-based crosslinkers were identified. From the analysis of DSC, it was elucidated for CF-based adhesives to require a higher pressing temperature or longer pressing time comparing to commercial urea-formaldehyde (C-UF) resin. MDF bonded with CF-based adhesives, which was formulated with 5% NaOH-hydrolyzed CF (CF-AK-5%) and PF of formaldehyde to phenol mole ratio of 2.5 (PF-2.5), and pressed for 8 min had higher MOR and IB than those with other CF-based adhesives. MOR and IB of MDF bonded with the CF-based adhesives regardless of formulation type and pressing time were higher than those with C-UF resin. When the values compared with the minimum requirements of KS standard, IB exceeded the KS standard in all formulations and pressing time, but MOR of only MDF bonded with CF-AK-5% and PF-2.5 and pressed for 8 min satisfied the KS standard. What was worse, 24-TS of MDF bonded with all CF-based adhesives did not satisfied the KS standard. However, MOR and 24-TS can be improved by increasing the target density of MDF or the amount of wax emulsion, which is added to improve the water resistance of MDF. Importantly, the use of CF-based adhesives decreased greatly the formaldehyde emission. Based on the results, we reached the conclusion that CF-based adhesives formulated under proper conditions had a potential as a sprayable resin for the production of wood panels.

Report on the Stability of Freeze-dried Standard Solution (동결건조표준액의 안정성에 관한 보고)

  • Purpose : Since standard solution is the one that knows its exact concentration, the curve of the dissolution has been determined according to the amount of the solution, compared to the amount of the unknown sample. Therefore, the antigen that makes up standard materials should be made in a pure form. The configuration of the standard substance solution in the kit we use is a freeze-dried material, or made and comes as a liquid. Lyophilized reference material is used after dissolving in usually D.W. (Distilled Water), and if the antigen to use is too sensitive, reagents should be freeze-dried. Furthermore, when freeze-dried reference has to be frozen again after being dissolved, it should be kept under $-20^{\circ}C$ until the expiration date according to the reports. Since it is not expressed in the experiment if it is safe or stable to reuse the solution which was dissolved a few times, thus, this time it is tested and evaluated that the changes of the standard solution by freezing and melting several times, and its results and the effectiveness of it were compared to the solution which was kept in a fridge. Materials and Methods : Among Vitro diagnostic kits on the market made by radioimmunoassay, parathyroid hormone (PTH), adrenocorticotropic hormone (ACTH), luteinizing hormone (LH) are made of freeze-dried standard solution and all composed of the same Lot.NO. These hormones melted in D.W. and were separated into three groups. In the first group, melting and freezing were repeated, and in the second group, The solution only for one time use was put into a test tube after melting and freeze it. The third group was kept in the refrigerator. This experiment has been conducted from January to February in 2012. January to 2012. PH test was employed because ph is prone to changing depending on the change of protein. Each group of the standard solution, cpm (counter per minute), and the patient relative concentration values were compared by date, and Through the correlation coefficient and Paired t-test, the significant level of each group was analyzed. Results : ACTH, PTH, LH pH values were too subtle denaturation rather than numerical changes in the protein. In addition, when the standard solution of ACTH, PTH, LH was refrigerated, after 3 days and 7 days, there was a significant difference observed between the solution being kept in a refrigerator and a freezer within a significance level. Conclusion : Standard solution should be kept in a freezer, and being kept in a fridge, it is recommended to use the solution as soon as possible.

Temporal and Spatial Variation of Microalgal Biomass and Community Structure in Seawater and Surface Sediment of the Gomso Bay as Determined by Chemotaxonomic Analysis (색소분석을 통한 곰소만 내 해수와 퇴적물 중 미세조류 생체량과 군집구조의 시공간적 변화)

  • To compare monthly variations of phytoplankton biomass and community composition between in seawater and sediment of the Gomso Bay (tidal flat: approximately 75%), the photosynthetic pigments were analyzed by HPLC every month in 1999 and every two months in 2000. Ambient physical and chemical parameters (temperature, salinity, nutrients, dissolved oxygen, and chemical oxygen demand) were also examined to find the environmental factors controlling structure of phytoplankton community. The temporal and spatial variations of chlorophyll a concentration in seawater were correlated well with the magnitude of freshwater discharge from land. The biomass of microphytobenthos at the surface sediments was lower than that in other regions of the world and 2-3 times lower than phytoplankton biomass integrated in the seawater column. Based on the results of HPLC pigment analysis, fucoxanthin, a marker pigment of diatoms, was the most prominent pigment and highly correlated with chlorophyll a in seawater and sediment of the Gomso Bay. These results suggest that diatoms are the predominant phytoplankton in seawater and sediment of the Gomso Bay. However, the monthly variation of chlorophyll a concentration in seawater at the subtidal zone was not a good correlation with that in sediment of the Gomso Bay. Although pelagic plankton was identified in seawater by microscopic examination, benthic algal species were not found in the seawater. These results suggest that contribution from the suspended microphytobenthos in the tidal flat to the subtidal zone of the Gomso Bay may be low as a food source to the primary consumer in the upper water column of the subtidal zone. Further study needs to elucidate the vertical and horizontal transport magnitude of the suspended microphytobenthos in the tidal flat to the subtidal zone.

  • The depletion of fossil fuels, ecological problems associated with $CO_2$ emissions climate change, growing world population, and future energy supplies are forcing the development of alternative resources for energy (heat and electricity), transport fuels and chemicals: the replacement of fossil resources with $CO_2$ neutral biomass. Several options exist to cover energy supplies of the future, including solar, wind, and water power; however, chemical carbon source can get from biomass only. When used in combination with environmental friend production and processing technology, the use of biomass can be seen as a sustainable alternative to conventional chemical feedstocks. The biorefinery concept is analogous to today's petroleum refinery, which produce multiple fuels and chemical products from petroleum. A biorefinery is a facility that integrates biomass conversion processes and equipment to produce fuels, power, and value-added chemicals from biomass. Biorefinery is the co-production of a spectrum of bio-based products (food, feed, materials, and chemicals) and energy (fuels, power, and heat) from biomass [definition IEA Bioenergy Task 42]. By producing multiple products, a biorefinery takes advantage of the various components in biomass and their intermediates therefore maximizing the value derived from the biomass feedstocks. A biorefinery could, for example, produce one or several low-volume, but high-value, chemical or nutraceutical products and a low-value, but high-volume liquid transportation fuel such as biodiesel or bioethanol. Future biorefinery may play a major role in producing chemicals and materials as a bridge between agriculture and chemistry that are traditionally produced from petroleum. Industrial biotechnology is expected to significantly complement or replace the current petroleum-based industry and to play an important role.

A Study on the dose distribution produced by $^{32}$ P source form in treatment for inhibiting restenosis of coronary artery (관상동맥 재협착 방지를 위한 치료에서 $^{32}$ P 핵종의 선원 형태에 따른 선량분포에 관한 연구)

  • In this study, the dose distributions of a $^{32}$ p uniform cylindrical volume source and a surface source, a pure $\beta$emitter, were calculated in order to obtain information relevant to the utilization of a balloon catheter and a radioactive stent. The dose distributions of $^{32}$ p were calculated by means of the EGS4 code system. The sources are considered to be distributed uniformly in the volume and on the surface in the form of a cylinder with a radius of 1.5 mm and length of 20 mm. The energy of $\beta$particles emitted is chosen at random in the $\beta$ energy spectrum evaluated by the solution of the Dirac equation for the Coulomb potential. Liquid water is used to simulate the particle transport in the human body. The dose rates in a target at a 0.5mm radial distance from the surface of cylindrical volume and surface source are 12.133 cGy/s per GBq (0.449 cGy/s per mCi, uncertainty: 1.51%) and 24.732 cGy/s per GBq (0.915 cGy/s per mCi, uncertainty: 1.01%), respectively. The dose rates in the two sources decrease with distance in both radial and axial direction. On the basis of the above results, the determined initial activities were 29.69 mCi and 1.2278 $\mu$Ci for the balloon catheter and the radioactive stent using $^{32}$ P isotope, respectively. The total absorbed dose for optimal therapeutic regimen is considered to be 20 Gy and the treatment time in the case of the balloon catheter is less than 3 min. Absorbed doses in targets placed in a radial direction for the two sources were also calculated when it expressed initial activity in a 1 mCi/ml volume activity density for the cylindrical volume source and a 0.1 mCi/cm$^2$ area activity density for the surface source. The absorbed dose distribution around the $^{32}$ P cylindrical source with different size can be easily calculated using our results when the volume activity density and area activity density for the source are known.

The evaluation of usefulness of Electronic Portal Imaging Device(EPID) (Electronic Portal Imaging Device(EPID)의 유용성 평가)

  • Purpose : To supply the information of EPID system and to analyze the possibility of substitution EPID for film dosimetry. Materials & Methods : With amorphous silicon(aSi) type EPID and liquid filled lonization chamber(LC) type EPID, the reproducibility according to focus detector distance(FDD) change and gantry rotation was analyzed, and also the possible range of image acquisition was analyzed with Alderson Rando phantom. The resolution and the contrast of aSi type EPID image were analyzed through Las Vegas phantom and water phantom. DMLC image was analyzed with X-Omat V film and EPID to see wether it could be applied to the qualify assurance(QA) of IMRT. Results : The reproducibility of FDD position was within 1mm, but the reproducibility of gantry rotation was ${\pm}2,\;{\pm}3mm$ respectively. The resolution and the contrast of EPID image were affected by dose rate, image acquisition time, image acquisition method and frame number. According to the possible range of image acquisition of EPID, it is verified that the EPID is easier to use than film. There is no difference between X-Omat V film and EPID images for the QA of IMRT. Conclusion : Through various evaluation, we could obtain lots of useful information about the EPID. Because the EPID has digital data, also we found that the EPID is more useful than film dosimerty for the periodical Qualify Assurance of IMRT. Especially when it is difficult to do point dose measurement with diode or ionization chamber, the EPID could be very useful substitute. And we found that the diode and ionization chamber are difficult to evaluate the sliding window images of IMRT, but the EPID was more useful to do it.
