Thermal Inactivation of Salmonella enteritidis, Salmonella typhimurium and E. coli O111 in Liquid Cultures During Microwave Radiation (Microwave 조사에 의한 Salmonella enteritidis, Salmonella typhimurium 과 E. coli의 불활성에 관한 연구)

  • The purpose of this study was to determine the thermal inactivation of Salmonella enteritidis, Salmonella typhimurium and E. coli O111 in liquid cultures treated with microwave energy. Furthermore, this study was to introduce new methodologies for studying nonthermal microwave effects on microorganisms, using controlled microwave energy and specially designed apparatuses. For the automatic temperature control during microwave heating, the real time data acquisition and computation system is designed with BASIC routine. The automatic temperature control system used in the experiments perform relatively stable control at the experiment temperature of 45, 50, 55 60$^{\circ}C$ and 65$^{\circ}C$ for 30 minutes. The effects of microwave heating on liquid cultures was compared with that of conventional heating, still reduces effectively the number of pathogenic bacteria in liquid cultures. While no particular differences between microwave heating and conventional heating was observed in the activation of E. coli at 45$^{\circ}C$ test, the activation of Sal. enteritidis and Sal. typhimurium was slightly reduced during the microwave treatments.

  • Irradiation with high doses of gamma rays induced the reduction of mycelial weight and anaylase activity, and increased relative amylase activity in surface cultures. Biphase in growth curves was shown in aeration-agitation cultures but the behavior of the first phase of growth could be eliminated by replacing the amylasehydrolysed starch substrates, so that enzyme production was shortened ca. 40 hours and relative amylase activity was increased about 3 times higher before onset of autolysis. In the effect of gibberellin on amylase production, the positive stimulation was appeared to only surface culturs of the liquid medium and the negative effect to shake-cultures in a mutant. Trials of various continuous culture were resulted not only the approalch to the value of amylase activity in surface cultures of liquid medium, but also higher productivity than in batch cultures. The culture-degeneration was observed in two-stage continuous culture, but did not appear in continuous elevation culture.

  • We researched effects of growth on initial inoculum size, liquid volume, and medium feeding rate etc. Cell suspension inoculated at low cell concentrations showed a typical growth reduction, whereas root cultures showed an improvement in growth. In this paper, Hairy roots showed high growth rate at 0.4 % inoculum size and 100 mL liquid volume in 250 mL flask cultures.

Shoot multiplication kinetics and hyperhydric status of regenerated shoots of gladiolus in agar-solidified and matrix-supported liquid cultures

  • In vitro shoot regeneration of gladiolus in three different culture systems, viz., semi-solid agar (AS), membrane raft (MR), and duroplast foam liquid (DF) cultures was evaluated following the kinetics of shoot multiplication and hyperhydricity at optimized growth regulator combinations. Compared to the AS system, matrixsupported liquid cultures enhanced shoot multiplication. The peak of shoot multiplication rate was attained at 18 days of incubation in the MR and DF systems, whereas the maximum rate in the AS system was attained at 21 days. An early decline in acceleration trend was observed in liquid cultures than the AS culture. The hyperhydric status of the regenerated shoots in the different culture systems was assessed in terms of stomatal attributes and antioxidative status. Stomatal behavior appeared to be normal in the AS and MR systems. However, structural anomaly of stomata such as large, round shaped guard cells with damage in bordering regions of stomatal pores was pronounced in the DF system along with a relatively higher $K^+$ ion concentration than in the AS and MR systems. Antioxidative status of regenerated shoots was comparable in the AS and MR systems, while a higher incidence of oxidative damages of lipid membrane as evidenced from malondialdehyde and ascorbate content was observed in the DF system. Higher oxidative stress in the DF system was also apparent by elevated activities of superoxide dismutase, ascorbate peroxidase, and catalase. Among the three culture systems, liquid culture with MR resulted in maximum shoot multiplication with little or no symptoms of hyperhydricity. Shoots in the DF system were more prone to hyperhydricity than those in the AS and MR systems. The use of matrix support such as membrane raft as an interface between liquid medium and propagating tissue could be an effective means for rapid and efficient mass propagation with little or no symptoms of hyperhydricity.

Ultrasound-Assisted Liquid-Liquid Extraction for Recovery of Paclitaxel from Plant Cell Cultures (식물세포배양으로부터 파클리탁셀 회수를 위한 초음파를 이용한 액-액 추출)

  • In this study, an efficient ultrasound-assisted liquid-liquid extraction process was developed for recovering of paclitaxel from plant cell cultures. The optimal ultrasonic power and operating time were 250 W and 15 min at fixed ratio of bottom phase, methylene chloride to top phase, MeOH (25%, v/v). Under the optimal conditions developed in the present method, most of the paclitaxel (~92%) was recovered from crude extract by a single extraction step. Due to the synergistic effect of ultrasound by the addition of inorganic salt, an appropriate inorganic salt concentration and the ultrasonic power were found to be required for the effective recovery of paclitaxel using ultrasound-assisted liquid-liquid extraction.

  • We established hairy root cultures of F. esculentum transformed with A. rhizogenes for in vitro rutin production. Additionally, we describe the effects of different media and plant growth regulators on growth and rutin biosynthesis in buckwheat hairy root cultures. Excised leaves of P. tinctorium from 10-day-old seedlings were used as the explant material for co-cultivation with A. rhizogenes 15834. The hairy culture of Fagopyrum esculentum Moench. was established by infecting leaf explants with Agrobacterium rhizogenes 15834. About four to five weeks after co-cultivation with A. rhizogenes, 10 hairy roots were excised from the necrotic explant tissues. After repeated transfer to fresh medium for three months, ten clones were transferred to MS liquid culture medium. The growth and rutin production of each clone differently response to the MS liquid medium. Among these clones, H8, which had exhibited good growth rate and one of the highest rutin productivity, was selected for the following experimment.(중략)

Detection of Bacteriochlorophyll-c Containing Species of Green Sulfur Photosynthetic Bacterium Chlorobium vibrioforme

  • Bacteriochlorophyll(BChl)-c containing species of green sulfur photosynthetic bacterium Chlorobium (ChI.) vibrioforme, which has BChl-d mainly, was detected. We obtained colonies on agar plates by spreading the liquid culture of ChI. vibrioforme f. sp. thiosulfatophilum strain NCIB 8327 which contained the high ratio of BChl-c/BChl-d, and transferred each colony into a new liquid medium. These cultures after growing were found to be classified into two categories. One possessed BChl-d as a light-harvesting pigment and the other did BChl-c. No colonies examined here contained both BChls-d and c. Therefore, the presence of both BChls-d and c in our cultures of ChI. vibrioforme was ascribed to the coexistence of two different cells which had BChl-d and c as the chlorosomal pigment, respectively. The change of pigment composition observed in our liquid cultures can be thus explained by the difference of growth rates between two kinds of cells.

Effect of Cadium Ions on the Activity of Fungal Laccase and Its Decolorization of Dye, RBBR

  • The effect of cadmium ions on ligninolytic and decolourizing activities in cultures of two white-rot fungi, Cerrena unicolor and Trametes versicolor, were examined. Cadmium was added to the shallow stationary cultures growing on a liquid mineral medium. Both examined strains sorbed Cd ions in the first 24 hr of incubation. An appreciable stimulation of the activity of extracellular laccase (LAC) and inhibition of the extracellular manganese-dependent peroxidase (MnP) were simultaneously observed when 25 mgL-1 and 50 mgL-1 of cadmium ions were added to the cultures. On the other hand, the addition of cadmium ions also resulted in stimulating the decolorization activity of C. unicolor to decolorize Remazol Brilliant Blue R (RBBR) in the cultures, but decreasing it in the culture of T. versicolor, which is compared to the inhibition of MnP activity in this fungus. Our data indicate that the presence of Cd(II) ions can affect the ligninolytic activity of white-rot fungi. It was found that C. unicolor is a strain resistant to the presence of Cd ions in the liquid culture media, and has a potential to use this strain for bioremediation of sites contaminated with both heavy metals and aromatic pollutants.

Liquid-Liquid Extraction for Recovery of Paclitaxel from Plant Cell Cultures by Adding Inorganic Salts (식물세포배양으로부터 파클리탁셀 회수를 위한 무기염이 첨가된 액-액 추출)

  • We developed a liquid-liquid extraction method using an inorganic salt to dramatically improve the recovery efficiency of the anticancer agent paclitaxel from plant cell cultures. As a result of liquid-liquid extraction using a diverse types of inorganic salt (NaCl, KCl, $K_2HPO_4$, $NaH_2PO_4$, $NaH_2PO_4{\cdot}2H_2O$), NaCl gave the highest yield (~96%) and lowest partition coefficient (0.053) of paclitaxel. The optimal NaCl/solvent ratio, methylene chloride/MeOH ratio, and pure paclitaxel content for liquid-liquid extraction using NaCl were 1% (w/v), 26% (v/v), and 0.066% (w/v), respectively. Under the optimal conditions developed in the present method, most of the paclitaxel (~96%) was recovered from biomass by a single extraction step. In addition, this method facilitated 3-fold higher recovery efficiency of paclitaxel in a shorter extraction number than the conventional liquid-liquid extraction method.

  • The isoprenoid quinone composition of Leclercia adecarboxylata KCTC $1036^T$ was determined by using high-performance liquid chromatography. L. adecarboxylata KCTC $1036^T$ are characterized by their production of both ubiquinone-7, ubiquinone-8 and menaquinone-8 as major quinones. It is clear that the analysis of isoprenoid quinone profiles provides a new criterion of great promise for identifying Leclercia strains.
