Hydrolysis by Alcalase Improves Hypoallergenic Properties of Goat Milk Protein

  • Goat milk is highly nutritious and is consumed in many countries, but the development of functional foods from goat milk has been slow compared to that for other types of milk. The aim of this study was to develop a goat milk protein hydrolysate (GMPH) with enhanced digestibility and better hypoallergenic properties in comparison with other protein sources such as ovalbumin and soy protein. Goat milk protein was digested with four commercial food-grade proteases (separately) under various conditions to achieve the best hydrolysis of αs -casein and β-lactoglobulin. It was shown that treatment with alcalase (0.4%, 60℃ for 30 min) effectively degraded these two proteins, as determined by SDS-PAGE, measurement of nonprotein nitrogen content, and reverse-phase high-performance liquid chromatography. Hydrolysis with alcalase resulted in a significant decrease in β-lactoglobulin concentration (almost to nil) and a ~40% reduction in the level of αs-casein. Quantification of histamine and TNF-α released from HMC-1 cells (human mast cell line) showed that the GMPH did not induce an allergic response when compared to the control. Hence, the GMPH may be useful for development of novel foods for infants, the elderly, and convalescent patients, to replace cow milk.

Identification and Characterization of a Novel Antioxidant Peptide from Bovine Skim Milk Fermented by Lactococcus lactis SL6

  • A novel peptide having free radical scavenging activity was separated, using an on-line high-performance liquid chromatography (HPLC) - ABTS screening method, from bovine skim milk fermented by Lactococcus lactis SL6 (KCTC 11865BP). It was further purified using reverse phase-HPLC (RP-HPLC) and sequenced by RP-HPLC-tandem mass spectrometry. The amino acid sequence of the identified peptide was determined to be Phe-Ser-Asp-Ile-Pro-Asn-Pro-Ile-Gly-Ser-Glu-Asn-Ser-Glu-Lys-Thr-Thr-Met-Pro-Leu-Trp (2,362 Da), which is corresponding to the C-terminal fragment of bovine ${\alpha}_{s1}$-casein (f179-199). The hydroxyl radicals scavenging activity ($IC_{50}$ $28.25{\pm}0.96{\mu}M$) of the peptide chemically synthesized based on the MS/MS data showed a slightly lower than that of the natural antioxidant Trolox ($IC_{50}$ $15.37{\pm}0.52{\mu}M$). Furthermore, derivatives of the antioxidant peptide were synthesized. The antioxidative activity of the derivatives whose all three proline residues replaced by alanine significantly decreased, whereas replacement of two proline residues in N-terminal region did not affect its antioxidative activity, indicating that $3^{rd}$ proline in C-terminal region is critical for the antioxidative activity of the peptide identified in this study. In addition, N-terminal region of the antioxidant peptide did not show its activity, whereas C-terminal region maintained antioxidative activity, suggesting that C-terminal region of the peptide is important for antioxidative activity.

Identification of Cisplatin-Resistance Associated Genes through Proteomic Analysis of Human Ovarian Cancer Cells and a Cisplatin-resistant Subline

  • Chemoresistance to cancer therapy is a major obstacle to the effective treatment of human cancers with cisplatin (DDP), but the mechanisms of cisplatin-resistance are not clear. In this study, we established a cisplatin-resistant human ovarian cancer cell line (COC1/DDP) and identified differentially expressed proteins related to cisplatin resistance. The proteomic expression profiles in COC1 before and after DDP treatment were examined using 2-dimensional electrophoresis technology. Differentially expressed proteins were identified using matrix-assisted laser desorption/ ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and high performance liquid chromatography-electrospray tandem MS (NanoUPLC-ESI-MS/MS). 5 protein spots, for cytokeratin 9, keratin 1, deoxyuridine triphosphatase (dUTPase), aarF domain containing kinase 4 (ADCK 4) and cofilin1, were identified to be significantly changed in COC1/DDP compared with its parental cells. The expression of these five proteins was further validated by quantitative PCR and Western blotting, confirming the results of proteomic analysis. Further research on these proteins may help to identify novel resistant biomarkers or reveal the mechanism of cisplatin-resistance in human ovarian cancers.

Preparation of 5-fluorouracil-loaded Nanoparticles and Study of Interaction with Gastric Cancer Cells

  • Aims: To prepare 5-fluorouracil (5-Fu) nanoparticles with higher encapsulation efficiency and drug loading, and then investigate interaction with the SGC-7901 gastric cancer cell line. Materials and Methods: Prescription was optimized by orthogonal experiments, the encapsulation efficiency and loading capacity were tested by high-performance liquid chromatography, and inhibition of proliferation by 5-Fu nanoparticles and 5-Fu given to cells for 24, 48 and 72 hours was investigated by methyl thiazolyl tetrazolium assay (MTT). In addition, 5-Fu nanoparticles were labeled by fluorescein isothiocyanate (FITC), and absorption into cells was tested by flow cytometry. Results: The optimal conditions for preparation were concentrations of 5-Fu of 5mg/ml, of $CaCl_2$ of 60 mg/ml and of chitosan of 2 mg/ml. With a stirring speed of 1200rpm, encapsulation efficiency of 5-Fu nanoparticles was $55.4{\pm}1.10%$ and loading capacity was $4.22{\pm}0.14%$; gastric cancer cells were significantly inhibited by 5-Fu nanoparticles in a time and concentration dependent manner, and compared to 5-Fu with slower drug release, in a certain concentration range, inhibition with 5-Fu nanoparticles was stronger. 5-Fu nanoparticles were absorbed by the cells in line with the concentration. Conclusions: 5-Fu nanoparticles can inhibit growth of gastric cancer cells in vitro to a greater extent than with 5-Fu with good adsorption characteristics, supporting feasibility as a carrier.

액적 충돌 거동에 대한 표면 패턴의 영향에 관한 연구 (A Study on the Effects of Surface Patterns on Droplet Impingement Behaviors)

  • 본 논문에서는 나노임프린트 리소그래피 기술을 이용하여 다양한 발수 표면을 제작하였으며, 발수기능의 안정성을 향상시키기 위해 액적의 충돌 거동에 대하여 평탄한 단위면적에 대한 구조물의 면적비인 표면 변수의 영향에 대한 연구를 수행하였다. 액적의 충돌 거동을 연구하기 위해 액적 충돌 시험을 통하여 충돌한 액적이 되튐에서 분산으로 천이 되는 임계높이를 측정하였다. 높은 표면 변수에서는 낮은 임계높이가 관찰되었으나, 낮은 표면변수에서는 임계높이가 증가하는 경향을 관찰하였다. 그러나, 표면변수가 더욱 감소할 경우 임계높이가 다시 감소하는 경향을 보였다. 관찰된 결과는 높은 임계높이를 위해서는 최적의 표면 형상 설계가 요구됨을 제시하고 있다.

제올라이트와 Klebsiella pneumonia sp.을 이용한 화학-생물학적 액상 암모니아의 제거 효율 연구 (Investigation of Liquid Phase Ammonia Removal Efficiency by Chemo-biological Process of Zeolites and Klebsiella pneumonia sp.)

  • 암모니아는 현대 산업에서 빠질 수 없는 유용한 물질이다. 일반적으로 농업용 폐기물의 분해과정을 통해 배출되며 인체에 매우 해로운 독극물로 알려져 있다. 산업에서 흔하게 쓰이는 물질이기에 직접 누출이나 간접 누출로 인한 수질오염의 가능성이 있다. 이 경우 암모늄이온을 빠르게 제거하는 데는 제올라이트의 흡착능을 이용하는 것이 좋지만 흡착만으로는 충분히 제거할 수 없다. 본 논문에서는 상용 제올라이트의 흡착능을 통한 암모늄이온의 제거 효율과 미생물의 생물학적 메커니즘을 통한 제거효율을 비교하였다. 추가적으로 제올라이트에 미생물을 고정하여 화학적 흡착 및 생물학적 전환 기술을 병합하여 그 효율을 비교하였다. 그 결과 100 ppm 기준 상용 제올라이트의 경우 2-4 h 사이에 67-81%의 제거효율을 보이는데 반해 선정 미생물인 Klebsiella pneumoniae subsp. Pneumoniae를 이용한 경우 8 h 내에 최대 97%의 제거 효율을 나타냈었다. 그리고 미생물을 제올라이트에 고정시켰을 때 8 h 이내에 98.5%로 제거 효율 및 속도가 증가하는 것을 확인할 수 있었다.

하수 슬러지 농축 및 탈수성에 미치는 오존의 영향 (Effect of Ozone Injection on Dewaterability and Thickening of Sewage Sludge)

  • It is required to propose an alternatives for appropriate sludge treatment owing to persistent construction of sewage treatment plant and increase of sewage sludge quantity. In order to treat sludge more efficiently, the methods which reduce the cost of sludge treatment have been studied such as sludge reduction and conditioning. Especially ozone treatment reduces solid quantity and improves separation of solid-liquid at the same time. Therefore ozone treatment have a positive effect on reduction and stabilization of sludge. So, this study applied ozone to sewage sludge and induced cell destruction of sludge. By comparing with the correlation between thickening and dewatering, and evaluating moisture content and solubilization of cake, this study verificates the effect of process improvement for ozone pre-treatment. In J-STP case, according to ozone dose solid flux increased about 12 times from $1kg/m^2{\cdot}h$ to $12kg/m^2{\cdot}h$. Also this plant were capable to shorten thickening time from 40 minutes to 6~7 minutes. Thus it is expected to reduce volume and retention time of thickener. On pH effect factor, dewatering at pH4 was more than at pH11, $3.05{\times}10^{11}$ and $3.82{\times}10^{11}(m/kg)$. But effect of pH was analogous to ozone, $2.81{\times}10^{11}(m/kg)$. The effect of pH on thickening was similar to law sludge, $0.68(kg/m^2{\cdot}h)$, and the effect of ozone injection on thickening was the biggest, $3.45(kg/m^2{\cdot}h)$. The COD solubilization rate improved from about 5 to 30%. So it is judged that we are able to utilize most solubilized sludge to another sewage treatment plants.

Population-Based Cervical Screening Outcomes in Turkey over a Period of Approximately Nine and a Half Years with Emphasis on Results for Women Aged 30-34

  • Purpose: To appraise the frequency of cervical cytological abnormalities in a population at normal risk via analysing the archive records of cytology for the period of approximately 9,5 years, comparing them with patient demographic charecteristics, and discuss the results for women under age of 35. Materials and Methods: A total of 32,578 cases of Pap smears were retrieved and analysed from our archive included the Pap tests performed between January 2001 and April 2010 at the Early Cancer Screening, Diagnosing and Education Center by the consent of three pathologists via utilizing the Bethesda System Criteria 2001 and the results were compared with some demographical characteristics. Results: Our rate of the cervical cytological abnormality was 1.83%, with ASCUS in 1.18%, LSIL in 0.39, HSIL in 0.16%, AGUS in 0.07%, squamous cell carcinoma in 0.02%, and adenoarcinoma in 0.006%. Cytological abnormalities were detected mostly in those with higher age, lower parity, and premenopausal period whereas the smoking status was without influence. Bacterial vaginosis (5.6%) was the most frequent infectious finding (Candida albicans 2.7%; Actinomyces sp. 1.3%; and Trichomonas vaginalis 0.2%) detected on the smears. The rate of abnormal cervical cytology was 9.5% among the women aged between 30-34. Conclusions: Early detection of the cervical abnormalities by means of the regular cervical cancer screening programmes is useful to attenuate the incidence, mortality, and morbidity of cervical cancer. Our prevalence of the cytological abnormalities was much lower than the one in Western populations in general but very similar to those reported from other Islamic countries that may be explained by the conservative lifestyle and the lower prevalence of HPV in Turkey. A remarkable rate of abnormal cervical cytology of women aged 30-34 was pointed out in the present study.

Flow Near the Meniscus of a Pressure-Driven Water Slug in Microchannels

  • Micro-PIV system with a high speed CCD camera is used to measure the flow field near the advancing meniscus of a water slug in microchannels. Image shifting technique combined with meniscus detecting technique is proposed to measure the relative velocity of the liquid near the meniscus in a moving reference frame. The proposed method is applied to an advancing front of a slug in microchannels with rectangular cross section. In the case of hydrophilic channel, strong flow from the center to the side wall along the meniscus occurs, while in the case of the hydrophobic channel, the fluid flows in the opposite direction. Further, the velocity near the side wall is higher than the center region velocity, exhibiting the characteristics of a strong shear-driven flow. This phenomenon is explained to be due to the existence of small gaps between the slug and the channel wall at each capillary corner so that the gas flows through the gaps inducing high shear on the slug surface. Simulation of the shape of a static droplet inside a cubic cell obtained by using the Surface Evolver program is supportive of the existence of the gap at the rectangular capillary corners. The flow fields in the circular capillary, in which no such gap exists, are also measured. The results show that a similar flow pattern to that of the hydrophilic rectangular capillary (i.e., center-to-wall flow) is always exhibited regardless of the wettability of the channel wall, which is also indicative of the validity of the above-mentioned assertion.

Effect of Pioglitazone on Perihematomal Edema in Intracerebral Hemorrhage Mouse Model by Regulating NLRP3 Expression and Energy Metabolism

  • Objective : Cerebral edema is the predominant mechanism of secondary inflammation after intracerebral hemorrhage (ICH). Pioglitazone, peroxisome proliferator-activated receptor gamma agonist has been shown to play a role in regulation of central nervous system inflammation. Here, we examined the pharmacological effects of pioglitazone in an ICH mouse model and investigated its regulation on NLRP3 inflammasome and glucose metabolism. Methods : The ICH model was established in C57 BL/6 mice by the stereotactical inoculation of blood (30 µL) into the right frontal lobe. The treatment group was administered i.p. pioglitazone (20 mg/kg) for 1, 3, and 6 days. The control group was administered i.p. phosphate-buffered saline for 1, 3, and 6 days. We investigated brain water contents, NLRP3 expression, and changes in the metabolites in the ICH model using liquid chromatography-tandem mass spectrometry. Results : On day 3, brain edema in the mice treated with pioglitazone was decreased more than that in the control group. Expression levels of NLRP3 in the ICH model treated with pioglitazone were decreased more than those of the control mice on days 3 and 7. The pioglitazone group showed higher levels of glycolytic metabolites than those in the ICH mice. Lactate production was increased in the ICH mice treated with pioglitazone. Conclusion : Our results demonstrated less brain swelling following ICH in mice treated with pioglitazone. Pioglitazone decreased NLRP3-related brain edema and increased anaerobic glycolysis, resulting in the production of lactate in the ICH mice model. NLRP3 might be a therapeutic target for ICH recovery.
