The Characteristics of Biodegradation for VOCs in Unsaturated Soil by Bio-filter (Bio-filter에 의한 토양중의 VOCs 분해특성)

  • The objective of this study was to develop a mechanistically based mathematical model that would consider the interdependence of VOCs transport, microbial activity, and sorptive interactions in a moist, unsaturated soil. Because the focus of the model was on description of natural attenuation, the advective VOCs transport that is induced in engineered remediation processes such as vapor extraction was not considered. The utility of the model was assessed through its ability to describe experimental observations from diffusion experiments using toluene as a representative VOCs in well-defined soil columns that contained a toluene degrading bacterium, Pseudomonas putida G7 md Fl, as the sole active microbial species. The gas-liquid mass-transfer was found to be a key parameter controlling the ability of bacteria to degrade VOCs. This finding indicates that soil size and geometry are likely to be important parameters in assessing the possible success of natural attenuation of VOCs in contaminated unsaturated soils. A Fast and Sensitive Method for the Simultaneous Determination and Quantification of Six Anionic Surfactants in Surface Water using HILIC-ESI-MS Technique

  • The hydrophobic hydrocarbon chain and the polar sulfate group confer surfactant properties and enable them to be used as anionic surfactants. Anionic surfactants (AS) are known for their adverse impact on environment, particularly on aquatic ecosystem. In the present study a fast, sensitive and selective method for the determination and subsequent quantification of six anionic surfactants was developed using hydrophilic interactive liquid chromatography (HILIC) coupled to a electrospray ionization (ESI) mass spectrometer (MS), in the concentration range 15-20 ${\mu}g/L$. The capability of the method was established using regression analysis and ANOVA. The method performance was evaluated by analyzing real time surface water spiked with 1-dodecyl hydrogen sulfate at 15 ${\mu}g/L$. Combined efficiency of solid phase extraction and MS detection established recovery of 89% in presence of natural matrix. These results point out that HILIC coupled to multistage MS procedures can be a powerful technique for environmental applications concerning the screening of polar contaminants.

A study on the Pharmaceutical & Chemical Characteristics and Analysis of Natural Omija Extract (천연 오미자 추출물의 약리, 화학적 특성 및 분석)

  • Natural Omija belongs to magnoliaceae was known to possess natural odor, taste, color, and various pharmaceutical & chemical characteristics. Omija extraction was extracted using ethanol as a solvent. Omija extract showed a light red-violet color of viscous liquid state. Some conclusions from the result of characteristic experiment were obtained as follows. From the result of antimicrobial experiment, occurrence of staphylococuss aureus and aspergillus niger as microbes was decreased according to the passage of time. This phenomenon could know that Omija component is affected to antimicrobial effect. From the result of dye experiment, fiber dyeing showed with some ivory color after dyed to cotton and silk. This phenomenon could know that Omija component is affected to dyeing effect from observation of scanning electron microscope(SEM). From the result of instrument analysis, inorganic components of K(107.30ppm), Na(2.110ppm), Ca(0.935ppm), Mg(0.891ppm), Li(0.270ppm) etcs from Omija were detected with ICP/OES, and aromatic components of benzene(10.808), a-pinene(13.996), phenol(14.183), ${\beta}$-terpene(15.840), a-terpinolene(17.616) etcs from Omija were also detected with GC/MSD.

Applications and Prospects of Ionic Liquids in Microbiology and Biochemical Engineering (이온성액체의 미생물.생명화학공학에의 응용과 전망)

  • Ionic liquids (ILs) have been widely recognized as environmentally benign solvents. Their unique properties, including negligible vapor pressure, non-flammability, a wide liquid range and their tunable physicochemical properties by proper selection of cations and anions, make them attractive green solvents in a variety of fields such as organic synthesis/catalysis, extraction/ separation, and electrochemistry, amongst others. In this paper, the recent technological developments and their prospects in the application of ILs in microbiology and biochemical engineering, including enzymatic reactions, protein folding/refolding and biomass dissolution, are discussed.

Studies on the Possibility of the Utilization of Macol Meal as Feedstuffs for Cattle (젖소 사료로서의 맥콜박 이용 가능성에 관한 연구)

  • This experiment was conducted to evaluate the feed value of Macol meal, which is one of the byproducts from the extraction of liquid syrup from barley grain, on lactating dairy cows. All of the lactating dairy cows feeding in eight dairy farms were used in feeding experiment and fed concentrates plus Macol meal 5Kg as wet basis. The milk yields and milk fit during the periods of 10 months were investigated and blood picture, blood chemistry and diseases occuring frequency before and after feeding Macol meal were analyzed. The results obtained were as follows ; 1. In the proximate compositions, the contents of moisture, crude protein, crude fat, crude fiber, crude as and nitrogen free extracts was 68.8%, 7.43%, 1.04%, 7.08%, 1.56% and 14.09% respectively. On the other hand, the contents of NDF, ADF and cellulose was 36.68%, 35.71% and 10.85%, as dry matter basis, respectively. 2. The daily milk yield per head was 22.84kg during two months before feeding Macol meal and 23.89 kg during the periods of ten months after feeding Macol meal. 3. The blood picture and blood chemistry were not affected by feeding Macol meal. 4. The diseases occuring frequency were not significantly affected by feeding Macol meal. from the results of this experiment it could be concluded that Macol meal can be used in lactating dairy cows rations without any adverse effects.

Dissolution of Tc(IV) Oxides in Aqueous Solutions

  • The long-lived fission product $^{99}Tc$ is present in large quantities in nuclear wastes and its chemical behavior in aqueous solution is of considerable interest. Under oxidizing conditions technetium exists as the anionic species $TcO_4^-$ whereas under the reducing conditions it is generally predicted that technetium will be present as $TcO_2{\cdot}nH_2O$. Technetium oxide was prepared by reduction of a technetate solution with $Sn^{2+}$. The concentration of total technetium and Tc(IV) species in the solutions were periodically determined by separating the oxidized and reduced technetium species using a solvent extraction procedure and counting the beta activity of the $^{99}Tc$ with a liquid scintillation counter. The experimental results show that the rate of oxidation of Tc(IV) in simulated groundwater and redistilled water is about $(1.49{\~}1.86){\times}10^{-9} mol/(L{\cdot}d$) under aerobic conditions, but Tc(IV) in simulated groundwater and redistilled water is not oxidized under anaerobic conditions. Under aerobic or anaerobic conditions the solubility of Tc(IV) oxide in simulated groundwater and redistilled water is equal on the whole.

  • Abnormal operating sounds radiated from a moving transfer robot in LCD (liquid crystal display) product lines have been used for the fault detection line of a robot instead of other source signals such as vibrations, acoustic emissions, and electrical signals. Its advantage as a source signal makes it possible to monitor the status of multiple faults by using only a microphone, despite a relatively low sensitivity. The wavelet packet transform for feature extraction and the artificial neural network for fault classification are employed. It can be observed that the abnormal operating sound is sufficiently useful as a source signal for the fault diagnosis of mechanical components as well as other source signals.

Tocopherol Content and Composition in Peanut

  • The content and composition of tocopherol were analyzed in peanuts produced in Korea. The tocopherol assay was consisted of an extraction with n-hex-ane:isopropanol mixture followed by high pressure liquid chromatography with fluorescence detection. Tocopherol content was higher in leaves and seeds but lower in seed coats and shells. $\alpha$-/${\gamma}$- Tocopherol ratio was as low as 0.53 in seeds and as high as 22.5 in shells. Tocopherol content in seeds of nine varieties ranged from 142 to 220 ng/mg dry weight, and the $\alpha$-/${\gamma}$-tocopherol ratio from 0.40 to 0.75. Tocopherol content decreased by 18% but $\alpha$-/${\gamma}$-tocopherol ratio increased by 44% in roasted seeds. The results indicate that the level of tocopherol in Korean peanut varieties is moderate and the ratio of $\alpha$- to ${\gamma}$-tocopherol is low.

Changes in Carotenoids Contents in Pureed and Cooked Carrot and Spinach during Storage (가열처리 및 저장조건에 따른 당근과 시금치퓨레의 Carotenoids 함량변화와 이성질화 형성에 관한 연구)

  • Investigations were conducted on the changes in carotenoids content, and quantification of cis-trans-${\beta}$-carotene Isomers in pureed and cooked carrot and spinach during storage. The isomerization and degradation of carotenoids were monitored by high-performance liquid chromatography on a C$\_$30/ reversed-phase column with diode-array detection. The results showed that lutein, ail-trans-${\beta}$-carotene, ${\alpha}$-carotene, 9-cis-${\beta}$-carotene and 13-cis-${\beta}$-carotene were present in carrot and spinach. Zeaxanthin and cryptoxanthin were present in raw spinach. The contents of lutein, zeaxanthin, cryptoxanthin, ${\alpha}$-carotene and all-trans-${\beta}$-carotene in pureed and cooked carrot and spinach decreased with increasing storage period. The 9-cis and 13-cis carotenoid isomers were the major types formed in cooked carrot during storage. Cooking was not found to alter the carotenoid profile of the sample, but increased the total amount of carotenoids compared with pured ones. This increase could be explained that cooking itself increased the extraction efficiency and inactivated the enzymes degradating carotenoids.

Measurement Uncertainty for Analytical Method of Hyaluronic Acid Used as a Dietary Supplement (식이보충제로 사용되는 히알루론산의 분석법에 대한 불확도 산정)

  • This study aimed to estimate the measurement uncertainty related to determination of hyaluronic acid used as a dietary supplement by high-performance liquid chromatography. According to the guidelines of the Association of Official Analytical Chemists, the analytical processes for determination of hyaluronic acid were performed. And the measurement uncertainty obtained during the analytical processes were expressed in accordance with mathematical/ statistical guidances of GUM (Guide to the Expression of Uncertainty in Measurement) & EURACHEM (Focus for Analytical Chemistry in Europe) for the analytical operations. For the uncertainty in measurement produced based on this analytical method, the expanded uncertainty was calculated by using the relative standard uncertainty between analytical results and sources of uncertainty in measurement (sample weight, final volume, extraction volume, standard solution, matrix and instrument etc). In the results of 95% confidence interval, it was calculated that the uncertainty in measurement was $57.75{\pm}8.76{\mu}g/kg$ (k=2.0). Therefore, it showed that the measurement uncertainty obtained by this analytical method influences on 15.2% of the contents of hyaluronic acid as the analytical results.
