Effect of Dietary Hot Red Pepper Powder on Humoral Immune Response in Rat (고춧가루 첨가 식이가 흰주의 체액성 면역기능에 미치는 영향)

  • Hot red pepper(Capsicum annum L.) has been extensively used as a spicy food additive and preservative in Korea. In this study, we investigated the effect of dietary hot red pepper powder on humoral immune response in rats. Sprague-Dawley rats were divided into 4 groups and fed experimental diets containing 0, 2, 5, 10% hot red pepper powder for 27 days. All groups were immunized with sheep red blood cells. In order to measure the immune response, plaque-forming cell number, agglutination titer, and serum antibody level were measured. Tissue ascorbic acid contents were also determined by high-performance liquid chromatogrphy. There was an increased plaque-forming cell number, agglutination titer, and serum IgG level in the groups supplemented with hot red pepper powder as compared to control. Tissure ascorbic acid contents in the hot pepper powder supplemented group were higher than those of control. The results suggest that the dietary hot red pepper powder enhances humoral immune response in rats, indicating that the hot pepper contains biological response modifier.

  • Pregnant rats were fed liquid diet to determine the influence of maternal ethanol intake on maternal and pup liver copper when dietary copper was low. The diets, which contained either 0.75(low) or 3.75(control)mg copper/1 with or without 30% of kcal from ethanol, were fed throughout gestation and the first 15 days of lactation. maternal calorie intake and body weight were unaffected by dietary treatment. Ethanol intake depressed maternal liver copper concentration only when diet copper was low(interactive effect P<0.05). Although ethanol intake depressed total pup liver copper concentration regardless of dietary copper level, the interactive effect observed in maternal liver was reflected incopper content of the pup liver metallothionein fraction eluted from a Sephadex G-75 column. The zinc content of metallothionein was inversely related to copper content of metallothionein. Results suggest that pregnancy and lactation is a special period to develop a copper deficiency when low copper intake and ethanol ingestion are combined not only in mothers but also in their offspring.

Clinical Study on Prenatal care, and Dietary Intakes for Pregnant Women and new Mothers (임산부의 산전관리와 산욕기 영양실태에 관한 연구)

  • This study was projected to provide basic data on prenatal care for future direction in maternity and child care, and also to investigate the diet of women during pregnancy and the period directly afterwards in order to offer to mothers appropriate advice for the improvement of nutritional standards. A clinical study on prenatal care was based on 1054 delivery cases. A nutritional survey was performed on 174 mothers admitted to the department of obstetrics at St. Mary's Hospital during the period of March, 1975 to February, 1976. The results obtained are summarized as follows; I. Clinical study on prenatal care 1) The age distribution showed 59.4% of the mothers were between the ages of 25 to 29 years old. 2) The gestational period was highest between the 37th and 40th gestational weeks. 33.7% of the mothers were primigravidae and 31.8% of them primiparae. 3) 41.3% of the mothers had not received prenatal care or had only received it once before. 4) Induced deliveries were 61.8% and spontantaneous deliveries 38.2%. 61.9% of the mothers had received prenatal care, while those without prenatal care accounted for 61.6% of the total induced deliveries. 5) Low birth weights were 7.7% and 5.0% of the mothers had received prenatal care, while 11.5% had no prenatal care. 6) There were 1.13% of still births, 0.32% of the mothers had prenatal care and the remainder did not have prenatal care. 7) Of those receiving prenatal care, 2.1% showed in the $0{\sim}3$ Apgar score group, 6.3% in the $4{\sim}6$ Apgar score group, and 91.6% in the $7{\sim}10$ Apgar score group. Among the non-prenatally cared for group 5.0% of the newborns were in the $0{\sim}3$ Apgar score group, 9.7% were in $4{\sim}6$ Apgar score group and 85.3% were in the $7{\sim}10$ Apgar score group. 8) Obstetrical complications were developed in 11.86% of the pregnant women when they were hospitalized. Among the group receiving the prenatal care 8.1% of the mothers had obstetrical complications. In the group without prenatal care 17.16% of the mothers had obstetrical complications. The most common obstetrical complication was malpresentation. 9) The first prenatal care was received between the 37th and 40th gestationl weeks. II. Food intake during pregnancy The following are the results from the questionnaires of the mothers concerning diets during pregnancy; 1) Main meals and snacks In 32.2% of the cases, their main meals during the diet amounted to more than was usually eaten at other times. In 67.8% of the cases, their main meals during the diet were the same as that usually eaten. In 22.4% of the cases, snacks during the diet amounted to more than usually eaten at other times. In 77.6% of the cases, snacks during the diet were the same as usually eaten. 2) Itemized list The mothers made a special effort to include certain items in their diets, the following is a breakdown of those items; a. egg, meat, fish 33.3% b. fruit, vegetables 32.2%. c. milk, fruit juice 18.4% d. cake, bread 2.9% e. nothing special 13.2% 3) Milk 44.8% of the mothers had at least one cup of milk everyday. 33.4% of the mothers had at least one cup of milk on occasion. 15.5% of the mothers did not have any milk. 4) Vitamins 39.7% of the mothers had vitamins everyday. 24.7% of the mothers had vitamins occasionally. 35.6% of the mothers did not have any vitamins. 5) Anemic symptoms 9.2% of the mothers very often had anemic symptoms during pregnancy. 39.1% of the mothers often had anemic symptoms during pregnancy. 51.7% of the mothers did not have anemic symptoms at all. 6) Taboos on food 23% of the mothers recognized 'taboos' on food during pregnancy 27% of the mothers displayed on uncertainty about the 'taboos' on food during pregnancy 50% of the mothers displayed indifference toward the taboos. III. Nutritional survey on the new mothers diet. 1) The diets for new mothers can be divided into four categories, such as general diet, low sodium diet, soft diet and liquid diet. 2) Cooked rice and seaweed soup were the main foods for the new mothers as has been the traditional diet for Korean mothers. 3) The average diet contained 1,783g. And the average consumption of the basic food groups per capita per day was 1,265g for cereals and grains, 456g for meats and legumes, 58g for fruits and vegetables, 0g for milk and fish and 4g for fats and oils. 4) In addition to the 1,783g of food in the main diet there was also 142.8g of food taken as snacks. 5) The average daily consumption of calories and nutrients was 2,697 Kcal and 123.4g for proteins, 44.9g for fats, 718.2mg for calcium, 14mg for iron, 2,101.4 I.U. for vitamin A, 0.43mg for thiamine, 1.02mg for riboflavin, 15.88mg for niacin, 5.26mg for ascorbic acid. When these figures are compared with the recommended allowances for new mothers in Korea, the calories and nutrients taken in were satisfactory. But the intake of minerals and vitamins was below the recommended allowance.

Evaluation on Feed-Nutritional Change of Food Waste According to Different Processing Methods and Trouble-shooting Strategy (음식물쓰레기의 가공처리방법별 사료영양소 함량 변화 평가 및 문제점 개선 방안)

  • This study was conducted to introduce recycling procedures of food waste(FW) as feed according to the dehydration, semi-dehydration fermentation and liquid fermentation methods through the on-site survey of companies related, to trace physico-chemical components and nutritional losses depending upon the processing stage for each method and finally to suggest more desirable methodology for the efficient utilization of FW as animal feed. For the dehydration method, dewatering of FW alone reduced(P<0.05) moisture(approximately 10%) and ether extract contents and increased(P<0.05) fiber contents. Dewatering and subsequent dehydration of FW decreased(P<0.05) contents of ether extract, limiting amino acids such as lysine, methionine and histidine, pepsin digestibility of protein by half, and NaCl content by 40%, increased(P<0.05) contents of fiber, crude ash, Ca and P, and did not alter(P>0.05) pH. The semi-dehydration fermentation method of FW did not affect(P>0.05) the chemical components, pepsin digestibility of protein, pH and NaCl content. For the liquid fermentation method, pasteurization and fermentation of FW decreased(P<0.05) contents of dry matter, ether extract, crude fiber, lysine and NaCl; however, it did not affect(P>0.05) other chemical components, pepsin digestibility of protein and pH. Among the processing methods, nutrient losses were highest for the dehydration method(25% of metabolizable energy loss, 12% of organic matter loss) and little for the semi-dehydration and liquid fermentation methods. The on-site survey of companies related revealed that the existence of foreign materials in FW products were problematic for all the three companies surveyed, thus it was necessary to develop a more efficient screener. Before feeding FW-containing diets to pigs, high quality of protein and energy feedstuffs needed to be fortified for the dehydration method. For the semi-dehydration fermentation method, the scientific diet formulation technology was required at the initial mixing stage. For the liquid fermentation method, possibly most energetic and proteinaceous feeds needed to be supplemented for the normal animal growth.

Effects of Fermented Diets Including Liquid By-products on Nutrient Digestibility and Nitrogen Balance in Growing Pigs (착즙부산물을 이용한 발효사료가 육성돈의 영양소 소화율 및 질소균형에 미치는 영향)

  • This study was conducted to evaluate the effects of fermented diets including liquid by-products on nutrient digestibility and nitrogen balance in growing pigs. Treatments were 1) CON (basal diet), 2) F (fermented diet with basal diet), 3) KF (fermented diet with basal diet including 30% kale pomace), 4) AF (fermented diet with basal diet including 30% angelica keiskei pomace), 5) CF (fermented diet with basal diet including 30% carrot pomace) and 6) OF (fermented diet with basal diet including 30% grape pomace). A total of 24 pigs (41.74kg average initial body weight, Landrace $\times$ Yorkshire $\times$ Duroc), were assigned to 6 treatments, 4 replicates and 1 pig per metabolic cage in a randomized complete block (RCB) design. Pigs were housed in $0.5\times1.3m$ metabolic cage in a 17d digestibility trial. During the entire experimental period, Digestibility of dry matter (p<0.05) of treatment CON, F and CF were higher than other treatments. In crude protein digestibility, treatment F was higher than treatment AF and GF (p<0.05). Treatment GF showed the lowest digestibility of crude fiber among all treatments (p<0.05). In ether extract digestibility, treatment AF and CF showed higher than other treatments (p<0.05) except KF treatment. CF treatment showed the best digestibility of ash among all treatments (p<0.05). Whereas, For Ca and P digestibility, CF and OF treatments were improved than other treatments (p<0.05). Energy digestibility (p<0.05) of CON, F and CF treatments were higher than KF, AF and GF treatments. In total essential amino acid digestibility, F treatment was improved than AF, CF and GF treatments (p<0.05). In total non-essential amino acid digestibility, F treatment was higher than CON, AF and GF treatments (p<0.05). In total amino acid digestibility, F treatment was higher than AF and CF treatments (p<0.05) and GF treatment showed the lowest digestibility (p<0.05). In fecal nitrogen excretion ratio, GF treatment was greatest among all treatments (p<0.05) and F treatment was decreased than other treatments (p<0.05). In urinary nitrogen excretion ratio, CON and GF treatments showed the lowest among all treatments (p<0.05). In nitrogen retention ratio, CON treatment showed the high and KF treatment showed the lost among all treatments (p<0.05). Therefore, this experiment suggested that fermented diet could improve nutrient and amino acid digestibilities of growing pigs.

Production of Lignan-Rich Eggs as Functional Food by Supplementing Schisandra chinensis By-Product in Laying Hens (사료에 오미자 가공부산물 분말의 첨가 급여가 계란의 리그난 함량에 미치는 영향)

  • Laying hens are known to be able to 'bio-accumulate' the health-promoting ingredients of their diet into eggs. The purpose of this study was to characterize lignan-rich eggs as functional food fed with Schisandra fruit by-product (SCP). Experimental diets were formulated using yellow corn, rice bran, soybean meal, fish meal, meat bone meal, poultry meal, vitamin premix, mineral premix, CaCO3, and supplemented Schisandra chinensis by-product. This experiment conducted a completely randomized design with 5 treatments for 5 laying hens. Levels of SCP were fed control diet or each formulated diet containing 1%, 3%, 5% and 7% SCP powder. The weight of eggs and the lignan content in white and yolk of egg were investigated every 7 days. Egg production and egg weight were not affected by diet at less than 5% SCP in the diet, but were significantly reduced when the diet was supplemented with a high concentration of 7% SCP after 3 weeks. Yolks and white in eggs were analyzed by using a high performance liquid chromatography (HPLC) to determine the lignans profile. Higher dietary SCP supplementation significantly increased gomisin N and schisandrin C in Acetonitrile (p<0.05). Gomisin N in egg white increased in a concentration-dependent manner, but shisandrin C not detected. These results indicated that the use of SCP powder in layering diets was effective in egg quality and for the production of lignans fortified eggs. In conclusion, dietary supplementation of Schisandra by-product with less than 5% can produce lignans-enrich eggs used as functional foods.

Simultaneous determination of illegal galactagogue adulterants in supplement diets by LC-MS/MS

  • Recently, for successful lactation, many breastfeeding mothers seek various products, including herbal medicine, dietary supplements, and prescribed medicines, to improve milk production. As demand for galactogogues grows, it is highly possible that pharmaceutical galactogogues may be adulterated with illegal products to maximize their efficacy. For continuous control and supervision of illegal products, we developed and validated a simple and sensitive LC-MS/MS method capable of simultaneously determining five galactogogues. Chromatographic separation was conducted using an Agilent Poroshell $120SB-C_{18}$ column with a mobile phase consisting of 20 mM ammonium formate (pH 5.4) and 100 % acetonitrile. The total run time was 13 min per analyte. The proposed method was performed according to the guidelines of the International Conference of Harmonization and it produced reliable results. This method showed high sensitivity and specificity, with a limit of detection (LOD) and limit of quantitation (LOQ) of 0.01-0.82 ng/mL and 0.02-2.45 ng/mL, respectively, for the solid- and liquid-type samples. Specificity was evaluated by analyzing matrix-blank samples spiked with the target compounds at LOQ levels, which provided a good separation of all peaks without interference. Additionally, the repeatability and intermediate precision were typically <15 %, whereas the recovery was 80-120 % of the values obtained using blank samples. Thus, we concluded that this method could be used for the identification and quantification of galactogogues in food or herbal products.

The Metabolism of Lipids in Adipose Pads and Superficial Pectoral Muscle of Chicks (닭의 지방조직(脂肪組織) 및 천흉근(淺胸筋)에서의 지질(脂質)의 대사(代謝))

  • In order to investigate the effect of dietary fatty acids on lipids metabolism in adipose pads and superficial pectoral muscle of chicks, the present, experiment was carried out using signle comb White Leghorn male chicks fed a fat-free diet and diets containing margaric, stearic or linoleic acids. Total lipids of tissues were extracted with a chlorofrom-methanol solution. The lipid components were fractionated by thin layer chromatography and the fatty acid distribution of lipid fractions was determined by gas liquid chromatography. The neck adipose pads contained 34-62% total lipids, in which triglycerides, were dominant, being 97-98%. Margaric, stearic and linoleic acids were distributed at a relatively high rate in the adipose tissue when the corresponding acids were fed, and margaric acid feeding lowered palmitoleic acid distribution and linoleic acid feeding elevated stearic acid distribution. The wet superficial pectoral muscle contained 1.3-1.7% total lipids, of which 77-79% was phospholipids and 11-13% was free cholesterol. When margaric acid was fed, margaric and heptadecenoic acids appeared in the muscle lipids. When linoleic acid was fed, this acid was significantly highly distributed in every muscle lipid fractions, whereas, when stearic acid fed no elevation of stearic acid distribution was observed. In the muscle phospholipids, oleic acid was significantly highly distributed in the stearic acid fed chicks, and the linoleic acid feeding signigicantly lowered the distribution value of palmitic and oleic acids, but elevated the distribution value of stearic acid. And the linoleic acid feeding lowered the distribution value of eicosatrienoic acid and elevated the distribution value of arachidonic acid.

The Role of Fatty Acid Binding Protein in the Fatty Liver Induced by Alcohol or High Cholesterol Diet in Rats (알코올 및 고콜레스테롤 식이로 유도된 흰쥐의 지방간에서 지방산 결합단백질의 역할 및 특성)

  • There is a marked increase in geriatric disease, especially liver disease, due to the continuous increase in alcohol and fat consumption. Since the fatty liver, induced by alcohol or fat, is basically from abnormalities in the lipid metabolism, it is possible that fatty acid binding protein(FABP) which is related to the fatty acid metabolism may also be abnormal in these livers. FABP is a small molecular weight protein family present in cytosol in high concentration. It has been proposed as a fatty acid transfer protein and as a binding protein responsible for controlling intracellular free fatty acid concentration. In this research, we have examined the relationship between liver FABP and fatty liver induced by alcohol or high cholesterol diet. Rats were fed one of either semipurified liquid diets; control diet containing 65% carbohydrate, 20% protein, and 15% fat or high cholesterol diet containing 1%(w/w) cholesterol or alcohol diet containing 37% of alcohol instead of carbohydrate. After 5 weeks of feeding period, all rats received commercial chow diet for 5 weeks to examine recovery effect. Liver and blood samples were collected at 0, 1, 3, 5 and 10 weeks to analyze lipid compositions. FABP was purified from liver cytosol and injected to rabbit to obtain antiserum. Liver FABP amount was determined by SDS-PAGE and western blotting methods. Fatty acid binding capacity was determined by binding of 14Cpalmitate with the delipidated liver cytosol. Consumption of alcohol increased serum cholesterol, triglyceride concentration and decreased HDL-cholesterol concentration after 5 weeks. Serum apolipoprotein B concentration increased after 3 weeks and LDL-cholesterol and apolipoprotein A concentration changed after 1 week. Liver cholesterol and triglyceride concentration increased after 3 weeks. Consumption of high cholesterol diet changed liver and serum lipid composition after 3 weeks. Swiching to normal diet for 5 weeks did not normalize most of lipid composition in serum and liver except serum and liver except serum cholesterol, triglyceride and liver cholesterol. Liver cytosol FABP content and the fatty acid binding capacity decreased dramatically after 1 week with alcohol consumption. This results indicate that FABP content changes before the changes before the changes of blood or liver lipid composition, suggesting changes of FABP may cause development of the fatty liver induced by alcohol and can be used as an index of detecting a early development of fatty liver.

Utilization of Fermentable Carbohydrates in Feed Manufacturing and in Enzyme of Poultry Feed (사료 제조에서 발효 가능한 탄수화물 이용과 가금 사료에서 효소의 처리에 관한 연구)

  • Improvements in understanding the effects of dietary fermentable carbohydrates and their interaction with supplemental feed enzymes and the feed manufacturing process may lead to reductions in volatile organic compound (VOC) emissions from poultry manure. Starch digestibility has been improved by replacing ground wheat or barley with whole wheat or barley, but there was no consistent effect of cereal species or feed form on the pH value of the gizzard contents. Pelleting results in improvements in feed conversion from 0 to 12%. Starch digestibility has been reported to account for up to 35 % of the improvement in available metabolic energy as a result of xylase supplementation. Factors which affect starch utilization and non-starch polysaccharide (NSP) absorption include the presence of anti-nutrient facto. (ANF) in grains, the nature of grain starch, NSP and the digestive capacity of animals. Improvements in feed production technology have been made in enzyme stabilization, allowing some dry enzyme products to be pelleted after conditioning at up to $87.69^{\circ}C$ and liquid enzymes to be stored in the feed mill for up to low months prior to use. The soluble NSP, arabinokylans and beta-glucans are partially degraded into smaller fragments by enzymes. With fragmentation, the water holding capacity is decreased, which leads to a reduction in digesta moisture, wet feces, and dirty eggs from hens fed diets containing viscosity-inducing ingredients.
