흰쥐에서 녹차추출물의 트랜스 지방 소장 흡수 억제 작용 (Green Tea Extract Decreases the Lymphatic Absorption of Trans Fat in Rats)

  • 트랜스 지방 과다섭취는 심혈관계 질환의 위험인자이다. 이 연구는 녹차의 섭취가 대표적인 트랜스 지방인 elaidic acid의 소장 흡수율에 미치는 영향을 조사하기 위해서 설계하였다. 소장 지방 흡수를 in vivo 상태에서 측정하기 위해서 흰쥐의 '소장지방흡수모델(mesenteric lymph duct cannulated rat model)'을 이용하여 십이지장주입관(intraduodenal catether)과 림프채취관(lymph duct cannula)을 각각 십이지장과 소장 림프관에 설치하였다. 십이지장주입관으로 주입된 지질유화액은 $180.0{\mu}mol$ elaidic acid, $400.0{\mu}mol$ triolein, $20.7{\mu}mol$ cholesterol, $3.1{\mu}mol$ a-tocopherol, $396{\mu}mol$ sodium taurocholate 그리고 24 mL PBS 용액을 기본성분으로 했고 이 지질유화액만 공급받은 동물을 대조군(control), 녹차추출물이 추가된 지질유화액을 공급받은 동물을 녹차군으로 하였다. 지질유화액은 시간당 3 mL씩 8시간 동안 주입하였고 동시에 소장 림프관에 설치된 림프채취관으로 분비되는 림프 시료를 매시간 8시간 동안 채취하여 분석하였다. 결과적으로, 8시간 동안 소장 림프채취관으로 분비된 림프의 양은 녹차에 의해서 유의적으로 감소하였고 elaidic acid의 흡수율(분비율) 또한 녹차에 의해서 유의적으로 감소하였다. 콜레스테롤, 올레산, 인지질의 흡수율도 녹차에 의해서 유의적으로 감소하였다. 이상의 결과들은 녹차가 elaidic acid 뿐만 아니라 기타 주요 식이성 지방들의 소장흡수를 억제하는데 효과적인 수단이 될 수 있다는 것을 증명한 연구라 판단된다.

솔잎 및 녹차추출물의 항산화성 및 아질산염 소거작용 (Antioxidative and Nitrite Scavenging Activity of Pine Needle and Green Tea Extracts)

  • 솔잎과 녹차의 열수와 에탄올 추출물을 이용하여 free radical 반응 및 지방산화에 미치는 영향과 아질산염 소거작용을 검토하였다. 각종 항산화제와 항산화력 비교에서 추출물 농도별 지방산화 억제능력은 0.1%보다는 0.3%가 낮은 TBARS값을 나타내었고, iron sources에 대한 binding 능력은 열수추출물이 우수하였다. Deoxyribose상에서의 hydroxyl radical 포집능력은 솔잎과 녹차 에탄올추출물이 대조구 보다 낮은 TBARS값을 나타내었다. 추출물의 iron 함량은 솔잎 에탄올 추출물이 가장 높은 함량을 나타내었으며, ascorbic acid 함량은 녹차의 열수와 에탄올 추출물에서 높은 값을 나타내었다. 전자공여능은 열수추출보다는 에탄올 추출물이 높은 활성을 나타내었다. 또한 솔잎과 녹차추출물은 각종 항산화제에 비하여 높은 전자공여능을 나타내었다. 아질산염 소거작용은 pH의 감소에 따라 높게 나타났으며, 0.3%의 솔잎과 녹차 추출물은 pH 1.2와 pH 3.0에서 95% 이상의 소거능을 나타내었으며, ascorbic acid 자체보다도 높은 소거능을 나타내었다. 이상의 결과로 보아 솔잎과 녹차추출물은 각종 육제품 제조시 첨가물로서 산화방지 효과 및 아질산염 소거제로서의 사용을 시사하는 것이다.

Evaluation of Antioxidative Activity of Various Levels of Ethanol Extracted Tomato Powder and Application to Pork Patties

  • This study was performed to evaluate antioxidant activity of tomato powder extracted by various concentrations of ethanol (0, 25, 50, 75, 100%) and to evaluate the physicochemical properties and antioxidant activities of pork patties with ethanol extracted tomato (EET) powder. No differences in the contents of total of individual phenolic compounds including gallic acid and catechin, were observed among the treatments (p>0.05). Among the various concentrations, 50% and 75% EET powder showed the highest free radical scavenging and iron chelating activities (p<0.05). Lipid peroxidation was retarded in linoleic acid emulsion with the addition of 50% and 75% EET powder (0.1%). Based on the model study, five pork patties were actually manufactured; control patty, reference patty with 0.01% of butylated hydroxytoluene, patty with 1% of water extracted tomato (WET), and patties with 0.5 and 1.0% of EET. Addition of 1% WET and EET decreased pH value, and increased redness values of pork patties, as compared to the control (p<0.05). Pork patties with WET (1.0%) and EET (0.5% and 1.0%) had lower 2-thiobarbituric acid reactive substances values compared with control patties after 7 d of storage (p<0.05). Pork patties containing EET powder showed lower total bacterial and Enterobacteriaceae counts than control patties (p<0.05). In conclusion, WET and EET (50%) could be used as a natural antioxidant and antimicrobial agent in meat products.

스피루리나 첨가 샐러드 드레싱의 품질 특성 (Quality Characteristics of Spirulina-Added Salad Dressing)

  • Six kinds of spirulina added dressing $(0\~1.09\%)$ were prepared and their rheological, physical and sensory characteristics were evaluated The hardness and adhesiveness of spirulina-added salad dressing were increased with the added amounts of spirulina. Viscosity of spirulina-added salad dressing was not significantly different up to $0.28\%$ whereas that of dressing added more than $0.55\%$ spirulina significantly increased, compared with that without spirulina. Emulsion stability of all of spirulina-added salad dressings was $40\%$ Antioxidant activities of spirulina-added salad dressing increased with spirulina increased: $IC_{50}$ values of DPPH radical scavenging activity and lipid peroxidation inhibition activity of $0.28\%$ added-dressing were 104.98 mg/mL and $6.71{\mu}g/mL$ of TBARS, respectively, which were higher than those of mayonnaise. The fat globule size of $0.28\%$ spirulina-added salad dressing was distributed within $0.5\~4.0{\mu}m$, of which $85.6\%$ of total fat globules were consisted of the size of less than $1.5\{\mu}m$. Total microbial number of salad dressing was 6.2log(CFU/mL), but E coli was not detected Sensory preference test of spirulina-added salad dressing showed that scores of appearance, flavor, viscosity and over-all preference for $0.28\%$ added-dressing were the highest with 7.83, 7.50, 5.33 and 7.97, respectively. Based on these results, spirulina-added salad dressing might have heath promoting effect showing antioxidant activity, and the most appropriate concentration of spirulina for salad dressing was $0.28\%$.

멸치를 이용한 식품가공용 중간소재의 동결저장안정성 (Frozen Storage Stability of the Frozen Seasoned Anchovy Meat Products)

  • 냉동멸치조미육제품의 저장안정성을 살피보기 위해 본 시제품을 동결저장하면서 저장 중 품질안정성에 대하여 실험하였다. 동결저장 중 멸치조미육제품의 pH 및 휘발성염기질소량은 점차 약간씩 증가하는 반면, 생균수는 $0.8{\sim}2.6{\times}10^5/g$ 범위로 저장 중 감소하는 경향이었다. Histamine 함량은 $70.6{\sim}76.7mg/100g$ 범위로 식품위생학적 면에서 안전하였다. 동결저장 중 고도 불포화지방산의 조성비는 점차 감소하는 반면 포화산, 모노엔산은 상대적으로 약간씩 증가하였다. 고도불포화 지방산의 산화분해 정도, 유지특가, 색조의 변화 및 단백질의 법동변성 정도를 측정한 결과 유화커어드 및 sodium erythorbate를 첨가함으로써 제품의 지방산패, 단백질변성을 다소 억제시킬 수 있었다. 이상의 결과에서 본 시제품(A)는 동결저장 150일 동안 품질이 안정하게 유지되었으며 식품가공용 중간소계로서 충분히 이용가능하다는 결론을 얻었다.

Effects of Drying Temperature on Antioxidant Activities of Tomato Powder and Storage Stability of Pork Patties

  • This study was performed to evaluate the antioxidant activity of oven-dried tomato powder (OTP) as affected by drying temperature and the effect of OTP on the product quality of pork patties. Three OTP products were obtained by drying of fresh tomato at 60, 80 and 100℃ oven until constant weight was obtained. Total phenolic content of three kinds of OTPs ranged from 1.95 to 5.94 g/100 g. The highest amount of total phenolic compound was observed in OTP dried at 100℃. Antioxidant activity of three kinds of OTPs was measured by 1,1-diphenyl-2-pycrylhydrazyl (DPPH)-radical scavenging activity, iron chelating ability, reducing power and measurement of lipid peroxide in linoleic acid emulsion system. In all parameters, OTP at 100℃ showed the higher antioxidant activity than other temperatures (p<0.05). Based on the model study, the physicochemical properties, and antioxidant and antimicrobial activities of pork patties containing 1% OTP were measured. Redness of pork patties were increased with the addition of OTPs (p<0.05). Thiobarbituric acid reactive substances (TBARS) values of raw pork patties containing OTPs were lower than those of control (CTL) until 7 d of storage, regardless of drying temperatures (p<0.05). Peroxide values of pork patties made with OTP (1%) were lower than those of CTL until the end of storage time (p<0.05). However, no antimicrobial activities were observed among the treatments (p>0.05). Therefore, OTPs could be used as a natural antioxidant in meat products.

초미세분쇄/공기분급을 이용한 탈지대두박 분획물의 특성과 응용 (Characteristics and Application of Defatted Soybean Meal fractions Obtained by Microparticulation/Air-Classification)

  • 분급휠속도(CWS, sut-off wheel speed) 9,000 rpm에서 초미세분쇄한 탈지대두박 분말을 공기분급휠속도 (ACWS, air classifying wheel speed) 21,000 rpm에서 9,000 rpm까지 3,000 rpm 간격으로 단계적으로 공기분급한 결과 ACWS가 감소함에 따라 단백질과 회분은 증가하는 반면 탄수화물, 지방 및 식이섬유는 감소하였다. 아미노산 조성과 함량은 원료 대두와 비슷하였고, 주요 아미노산은 aspartic acid와 glutamic acid였다. 수율은 ACWS의 감소에 따라 증가하였고, 입자의 경우 $4.9{\mu}m$에서 $14.2{\mu}m$의 범위로 ACWS의 감소에 따라 증가 경향이있었으며, 모서리가 있는 타원형의 형태를 보였다. 보수력, 보유력 및 유화력은 낮은 ACWS에서 더 낮은 값이었고, 탈지대두박을 이용한 커어드는 사용한 응고제와 전지대두분의 대체 비율에 따라 다른 특성을 나타내었다. 또 초미세분쇄한 탈지대두박 분말을 케익에 첨가하였을 때 콩 특유의 비린냄새가 강하게 나타나지 않았으며, 10%까지는 대체 가능함을 알 수 있었다.

Complexation of Adiponectin-encoding Plasmid DNA with Rosiglitazone-loaded Cationic Liposomes

  • To enhance therapeutic effects of insulin-sensitizing adipokine, ADN gene and potent agonists, rosiglitazone for the $PPAR{\gamma}$, cationic liposomes as non-viral vectors were formulated. The particle size and zeta potential of drug loaded and unloaded cationic liposomes were investigated. The complex formation between cationic liposomes and negatively charged plasmid DNA was confirmed and the protection from DNase was observed. In vitro transfection was investigated in HepG2, HeLa, and HEK293 cells by mRNA expression of ADN. Encapsulation efficacy of rosiglitazone-loaded liposomes was determined by UV detection. Particle sizes of cationic liposomes were in the range of 110-170 nm and those of rosiglitazone-loaded cationic liposomes were in the range of 130-180 nm, respectively. Gel retardation of complexes indicated that the complex was formed at weight ratios of cationic lipid to plasmid DNA higher than 20:1. Both complexes protected plasmid DNA from DNase either drug free or drug loading. Encapsulation efficiency of rosiglitazone-loaded emulsion was increased by drug dose. The mRNA expression levels of ADN were dose-dependently increased in cells transfected with plasmid DNA. Therefore, cationic liposomes could be potential co-delivery system for drug and gene.

Growth of flounder larvae, Paralichthys olivaceus using enriched rotifer fed with artificial microparticle diets

  • Three types of artificial microparticle diets were developed for rotifer (Brachionus plicatilis) enrichment. The efficacies of enrichment with the artificial diets were evaluated and compared to those with commercial enrichment diets on the growth and survival of flounder larvae. Total lipid content was highest in the rotifer enriched with oil capsule (40.5% in dry weight). The n-3 highly unsaturated fatty acid (n-3 HUFA) content was also highest in the rotifer fed with oil capsule (7.08% in dry weight). The flounder larvae fed on the rotifer enriched with oil capsule showed the highest growth compared to those fed on any other enriched rotifer (P<0.05). The survival ratio of flounder larvae fed on the rotifers enriched with oil capsule and emulsion oil were higher than those fed on any other enriched rotifer (P<0.05). From the feeding study, the growth and survival of flounder larvae were enhanced by feeding rotifer enriched with oil capsule compared to rotifer enriched with any other diets. The rotifer fed on oil capsule containing high contents of n-3 HUFA. Therefore, a significant relationship between the growth and survival of flounder larvae and the n-3 HUFA content of rotifer could be obtained.

A Case of Isoniazid Intoxication in a Dog

  • A seven-month-old castrated male Chihuahua weighing 1.6 kg presented with generalized tonic-clonic seizure following ingestion of isoniazid. Emergency treatment with three doses of diazepam (total 1.5 mg/kg, intravenous [IV]) and phenobarbital (15 mg/kg IV) was administered. The seizure stopped after administration of propofol (constant rate infusion [CRI]; 0.2 mg/kg/min). Blood analyses showed mildly increased serum blood glucose concentration, hyperkalemia, and hyperphosphatemia. On suspicion of isoniazid toxicity, activated charcoal (1 g/kg, orally), lipid emulsion (CRI; 9 mL/hr), and pyridoxine hydrochloride (70 mg/kg IV) were added to the treatment regimen. Twelve hours after presentation, the dog showed increased serum liver enzyme activities, serum blood urea nitrogen, and creatinine concentrations indicating hepatic and renal failure. Twenty-two hours after presentation, blood analysis still revealed increased liver enzyme activities, blood urea nitrogen, and creatinine concentrations with low blood glucose concentration. Twenty-six hours after presentation, the dog's vital signs deteriorated and the owner elected for the dog to be euthanized. This is the first report of the clinical course of isoniazid toxicosis in a dog in South Korea. Furthermore, to our best knowledge, this is the first report where secondary multiple organ failure was observed due to isoniazid toxicosis. Clinicians should be aware of the possibility of isoniazid toxicosis in dogs. Rapid initiation of treatment after clinical recognition is warranted in such cases.
