The purpose of this study was to investigate the volatile flavor compounds of Ligularia stenocephala and Ligularia fischeri, edible wild plants. The volatile flavor compounds were isolated by the simultaneous distillation extraction method and analyzed by GC-MSD (gas chromatography-mass selective detector). Forty-eight volatile flavor compounds were identified in the extracts from L. stenocephala and the major compounds were sabinene, cis-ocimene, trans-caryophyllene, and β-elemene. Fifty-one volatile flavor compounds were identified in the extracts from L. fischeri and the major compounds were α-pinene, germacrene-D, transcaryophyllene, endo-1-bourbonanol and 1-limonene. The common volatile flavor compounds between two plants were transcaryophyllene, sabinene, β-elemene and β-cubebene etc. However, α-phellendrene and myrcene were identified in L. stenocephala, but were not identified in L. fischeri. However α-pinene, germacrene-D and limonene were identified in L. fischeri but were not identified in L. stenocephala.

Ligularia stenocephala leaves were subjected to extraction by using several solvents with different polarities, which were then investigated for their antioxidant and antimicrobial activities. Extraction yield of ethanol extract of Ligularia stenocephala was 21.36%. The water fraction showed the highest extraction yield of 60.35%, followed by the n-butanol, n-hexane, and ethyl acetate fractions. Total polyphenolic content was the highest (254.00 mg/g GAE) in the ethyl acetate fraction of Ligularia stenocephala extract. The ethyl acetate fraction had $IC_{50}$ values of 0.28 mg/mL for DPPH radical scavenging, and 96.$67{\pm}0.09%$ for ABTS radical scavenging activity. Further, FRAP value was significantly higher in the ethyl acetate fraction. The ethyl acetate fraction showed antimicrobial activities against B. cereus, and the diameter of the zone of inhibition was 10.2 mm at 5 mg/disc. These results suggest that the ethyl acetate fraction of Ligularia stenocephala possesses the antimicrobial activities against B. cereus, and its high antioxidant activity could be applicable to food additives and as a natural cosmetic ingredient.
The leaves and roots of Ligularia stenocephala, which are widely used as a food in Korea, were investigated for their antioxidant activities and cytotoxicity in vitro, and their hepatoprotective effect, alcohol detoxicant efficacy, and memory-enhancing property were investigated in vivo. The unique odor of the leaves was analyzed by GC-MS. Lipid peroxidation, superoxide anion formation, and DPPH radicals were inhibited remarkably by the extracts of the leaves and roots. The leaves of this edible plant significantly protected the hepatotoxicity induced by carbon tetrachloride and further diminished the blood alcohol content in mice. While the roots of this plant exhibited adequate cytotoxicity against four human tumor cell lines, especially against melanoma, the leaves revealed relatively weak activity. Both the leaves and the roots exerted an excellent ameliorating property on scopolamine-induced memory impairment in the passive avoidance task using an animal model. The hexane fraction of the leaves was analyzed by GC-MS, suggesting that a series of terpenoids may be odorous compounds in this plant.
Two edible plants, Allium victorialis var. platyphyllum and Ligularia stenocephala, and one medicinal plant, Panax ginseng, were cultivated in the understory of an artificial Larix kaempferi plantation for ten years. Growth characteristics (number of leaves and flower stalks per plant, and leaf length and width), survival rate, and yield (fresh weight of plants) per unit area (1 ㎡) were investigated one year after planting, and six and ten years following cultivation. P. ginseng and L. stenocephala survived at a high percentage for two years after planting. Results showed that P. ginseng had longer and thicker roots when aged; however, a large number of plants died and the yield was low. In contrast, almost half of A. victorialis var. platyphyllum died within two years of planting. The surviving plants grew well for ten years and the yield was increased. The leaf length and width of L. stenocephala increased; however, the survival rate and the number of leaves per plant decreased as the period of cultivation was extended. In contrast, A. victorialis var. platyphyllum survived at a lower rate (50%) than the two other crops (98% for L. stenocephala and 83% for P. ginseng) during the early cultivation period, with little change in the survival rate over an extended time; however, the yields increased. This species showed an increase in the number of flower stalks and leaves, and as a result, the larger leaves increased the yield. We evaluated the understory cultivation of three crops in a L. kaempferi plantation under natural conditions, with no irrigation or fertilization, and Allium victorialis var. platyphyllum showed the greatest growth potential among the three tested crops.
The leaves of Ligularia stenocephala (Compositae) are used as a vegetable food to treat anxiety, weakness and menstrual disorder and are said to be effective against diabetic disease in Korea. This research was aimed to develop this vegetable food as a functional food to treat diabetic disease. Oral administration of MeOH extract exhibited significant hypoglycemic and hypolipidemic effects in streptozotocin (STZ)-induced diabetic rats at 100 and 200 mg/kg dose. This extract inhibited the increase of body weight, water consumption, food intake, blood glucose, lowered atherogenic index, and overcome glucose intolerance in the STZ-treated rat. These results suggest that the leaves of Ligularia stenocephala could be developed as a functional food with anti-diabetic effect.
Screening was conducted using 200 kinds of plant extracts to explore herbicide-activated components of plant origin. We separated and purified active substances and elucidated chemical structures using Ligularia stenocephala M., which has strong activity and has not yet been studied. When the solvent fractions of the leaves of Ligularia stenocephala M. were tested for their herbicidal activity, ethyl acetate and chloroform layer showed an inhibition rate of 95.2% and 94.1%, respectively. In particular, the chloroform layer exerted more than 50% herbicidal activity at 10 ppm. From the chloroform layer with the highest herbicidal activity, we isolated three herbicidal active compounds using stepwise chromatography, specifically silica gel or octadecyl silica (ODS) column chromatography, Sep-pak cartridges, and high performance liquid chromatography (HPLC). Based on the analysis of the active compounds using electron ionization mass spectroscopy (EI-MS), 1H-NMR, and 13C-NMR, we identified the active compounds as euparin, 5,6-dimethoxy-2-isopropenylbenzofuran, and liguhodgsonal. When the herbicidal activity of the identified compounds was tested, euparin showed selective herbicidal activity for lettuce at 10-3 M, and both liguhogsonal and 5,6-dimethoxy-2-isoprophenylbenzofuran exerted selective activity for rice and Echinochloa crus-galli.

This experiment was carrid out to develop a culture method by sequential highland-lowland cultivation for year-round production of fresh leaves in Ligularia stenocephala. Rootstocks of Ligularia stenocephala cultivated in Daegwallyeong openfield were digged up on late October 2007 and then stored in $-2^{\circ}C$ refrigerator. The rootstocks were monthly transplanted in protected greenhouse in Gangneung from November 2007 to June 2008 for lowland cultivation, and also they were monthly transplanted in rainshelter in Daegwallyeong from June to August 2008 for highland cultivation. The early growth of Ligularia stenocephala transplanted on January 2008 was faster. Most plant of Ligularia stenocephala regardless of transplanting time of rootstock grew over 30cm at the time of first harvesting of leaves till May, while the yield decreased during the summer season. The average plant height of Ligularia stenocephala transplanted on July and August in Daegwallyeong highland was lower than that cultivated in Gangneung lowland during spring season. Using of rootstock stored made it possible to produce in highland from Apr. to Oct. during summer season, and also to produce in lowland from Nov. to May next year. Therefore, the results indicates that sequential highland-lowland cutivation by using of stored rootstock will be capable of yearround production of fresh leaves in Ligularia stenocephala.
The leaf extract of Ligularia stenocephala showed the highest anti-platelet aggregating activity in large numbers of edible and herbal plants. The active fraction fractionationed from L. stenocephala extract by using preparative HPLC inhibited the platelet aggregation up to above 80% and its blood coagulating time (PT and APTT) also showed similar effect to aspirin (0.2 ${\mu}g/mL$), known as an anti-thrombus compound. Two antithrombus active compounds were purified and identified as 3,5-dicaffeoylquinic acid and 3,4-dicaffeoylquinic acid, respectively, on the basis of NMR and FAB-MS spectroscopic data. Two active compounds has not only antiplatelet aggregating activity, but also has anticoagulating activity.
High performance liquid chromatography (HPLC) analysis on the MeOH extract of Ligularia stenocephala leaves identified six caffeoylquinic acids, viz. 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoyl-mucoquinic acid, 3,5-di-O-caffeoylquinic acid, 4,5-di-O-caffeoylquinic acid, 5-O-caffeoylquinic acid, and 3-Ocaffeoylquinic acid using standard compounds, and determined the quantity of each extract. Percentage of total caffeoylquinic acids of the MeOH extract and its BuOH fraction were calculated as $67.83{\pm}3.79%$ and $94.52{\pm}1.84%$, respectively. Since the caffeoylquinic acid-rich MeOH extract exhibited a potent peroxynitrite-scavenging effect in vitro ($IC_{50}=0.87{\pm}0.33\;{\mu}g/ml$ (mean $\pm$ SEM)), the experiment was designed to identify whether or not that extract has an anti-obesity effect on rat obesity induced by high fat diet. Oral administration of the MeOH extract and its BuOH fraction abundant in caffeoylquinic acid decreased the rat body weight to the level of untreated group and decreased abdominal fat pad weight. The atherogenic index and thiobarbituric acid-reactive substance (TBARS) values were restored by treatment, indicating that the caffeoylquinic acid-rich extract probably inhibited hyperlipidemia and oxidative stress caused by high fat diet. These results suggest that L. stenocephala in vegetable form or its caffeoylquinic acid-rich fraction (BuOH fraction) as an agent can be used for treatment or prevention of obesity.
This study was carried out to establish a proper cultivation condition and diagnose the drought-tolerance of three wild edible greens belonging to genus of Ligularia by using pressure-volume curves methods. The result of the original bulk osmotic pressure at maximum turgor ${\Psi}_{\circ}{^{sat}}$ was -0.8 MPa in Ligularia fischeri and L. stenocephala, which was somewhat lower than the value, -0.7 MPa, in L. fischeri var. spiciformis. In addition, the values of the osmotic pressure at incipient plasmolysis ${\Psi}_{\circ}{^{tlp}}$ in L. fischeri and L. stenocephala, both of which were -0.9 MPa, These were slightly lower than that of -0.8 MPa in L. fischeri var. spiciformis. On the other hand, it appeared that the values of maximum bulk modulus of elasticity $E_{max}$ of L. fischeri and L. stenocephala were approximately two times higher than that of L. fischeri var. spiciformis. However, There was a distinct difference between the values of the relative water contents in these three species. Therefore, Ligularia spp, occurring incipient plasmolysis in the high water contents, have a relatively low property of drought-tolerance, suggesting that growth of those Ligularia spp. are appropriate for relative moisture forest.
