Modification in the Responsiveness of Cat Dorsal Horn Cells during Carrageenin-Induced Inflammation (피부염에 의해 유발된 척수후각세포의 Activity 변동에 관한 연구)

  • The present study was undertaken to investigate modification in electrophysiological characteristics of cat dorsal horn cells resulting from carrageenin-induced inflammation. The followings were studied; 1) the time-course of changes in responses of the WDR (wide dynamic range) cell 1-3h after subcutaneous injection of carrageenin in its receptive field; 2) the responses of the same dorsal hern cells before and after induction of inflammation; 3) the effect of inflammation on the responsiveness of dorsal horn neurons to algogens (bradykinin & potassium); and 4) the effect of inflammation on the activity of WDR cell following administration of indomethacin and clonidine. Though responses of WDR neuron were increased dramatically during first 1h, the maximal enhancement was observed 3h after induction of inflammation especially by repetitive light tactile stimulus. Following carrageenin injection the majority of WDR neurons (10/15 units) showed enhanced responses to all the mechanical stimuli while in 3 cases responsiveness were intensified during activation by one tactile stimulus (brush or pressure). One cell was unaffected by inflammation and in another case the response was enhanced only to noxious stimulus. Five of 9 cells that could initially be driven by noxious stimulus were activated more strongly by same stimulus and even by tactile stimulus (pressure) following inflammation. In 2 cases neurons were sensitized only to noxious stimulus whereas in another 2 cells that did not show enhanced responses to noxious stimulus responses to light tactile stimulus (pressure) appeared after inflammation. Of 16 LT cells tested 6 responded to squeeze while 4 showed the characteristics of WDR cell following inflammation. No modification in responsiveness was recognized in 3 cells whereas response to only brush was enhanced in another 3 neurons. Following carrageenin injection responses of LT cell to bradykinin or $K^{+}$ were not altered whereas those of WOR neurons to bradykinin or $K^{+}$ were suppressed in 22.2% and 33.3% of cases, respectively. In two of 8 activity of HT cells were inhibited by bradykinin while in five of 8 responsiveness to $K^{+}$ were rather enhanced by inflammation. In the rest inflammation was ineffective. In inflammation-induced animal the receptive field of LT cell was not changed whereas those of WDR cell and HT cell were tremendously expanded. The enhanced responses of WDR neurons to mechanical stimuli resulted from inflammation were suppressed by intravenously injected indomethacin and clonidine suggesting that postaglandin is involved in inflammation-induced sensitization of these cells. The involvement of peripheral and central mechanisms in the modification in responsiveness of dorsal horn cells in the carrageenin-induced inflammation was discussed.

Estimation of Visual Stimulus Intensity From Retinal Ganglion Cell Spike Trains Using Optimal Linear Filter (최적선형필터를 이용한 망막신경절세포 Spike Train으로부터의 시각자극 세기 변화 추정)

  • As a preliminary study for the development of electrical stimulation strategy of artificial retina, we set up a method fur the reconstruction of input intensity variation from retinal ganglion cell(RGC) responses. In order to estimate light intensity variation, we used an optimal linear filter trained from given stimulus intensity variation and multiple single unit spike trains from RGCs. By applying ON/OFF stimulation(ON duration: 2 sec, OFF duration: 5 sec) repetitively, we identified three functional types of ganglion cells according to when they respond to the ON/OFF stimulus actively: ON cell, OFF cell, and ON-OFF cell. Experiments were also performed using a Gaussian random stimulus and a binary random stimulus. The input intensity was updated once every 90 msec(i. e. 11 Hz) to present the stimulus. The result of reconstructing 11 Hz Gaussian and binary random stimulus was not satisfactory and showed low correlation between the original and reconstructed stimulus. In the case of ON/OFF stimulus in which temporal variation is slow, successful reconstruction was achieved and the correlation coefficient was as high as 0.8.

Effects of d-Amphetamine and Chlorpromazine on the Orienting Response and General Behavioral Activity in Rats (d-Amphetamine 및 Chlorpromazine이 흰쥐의 정위반응(定位反應) 및 일반행동에 미치는 영향)

  • To see if the treatments of d-amphetamine and chlorpromazine have any influence upon the orienting response and general behavioral activity, 3 groups of male Holtzman rats were prepared, namely d-amphetamine animals (1.0 mg/kg.i.p.), chlorpromazine rats (1.0 mg/kg.i.p.) and the physiological saline control animals. The general behavioral activity was examined by visual scanning using the time-sample method in the adaptation period of orienting response. The occurence of orienting response and its rate of habituation were evaluated by observing cessation of ongoing activity in response to a sound stimulus (1,000 Hz, 70 db & 0.1 sec), or turning of head toward the source of stimulus in 20 trials. Attention shift from sound to light stimulus was also tested in 10 trials. The results obtained were as follows; 1. The general behavioral activity of d-amphetamine group was significantly greater than that of control, however, the chlorpromazine animals showed the tendency to decrease in activity. 2. The d-amphetamine group showed the occurence of orienting response to sound significantly more often than that of placebo controls. However, the chlorpromazine group exhibited significantly fewer orienting response than the placebo group did. 3. The d-amphetamine group displayed no clear out habituation to the orienting response following the repetition of trials, though the placebo and the chlorpromazine groups demonstrated apparent habituation to the response. 4. The three animal groups did not differ significantly from each other with regard to shift of attention from sound to light stimulus. It is inferred that the d-amphetamine tends to increase general activity has a definite facilitative action of orienting response and a inhibitory influence upon the habituation of the latter response. On the contrary, the chlorpromazine tends to decrease general activity, has a inhibitory action of orienting response and facilitatory action of habituation of the response.

Nonpharmacological Treatment of Insomnia (불면증의 비약물학적 치료)

  • Several nonpharmacological treatment methods of insomnia and their effects were reviewed. A long-term use of most hypnotics may produce tolerance, dependence, cognitive and psychomotor impairments at daytime, shallow sleep, and rebound insomnia on drug withdrawal. To reduce hypnotic abuse, nonpharmacological strategies have been developed to correct disordered behavioral and cognitive factors. These treatments aim at modifying maladaptive sleep habits, lowering physiological and cognitive arousal levels, and correcting dysfuctional beliefs and attitudes about sleep. These non-pharmacological or cognitive behavior treatments include stimulus control, sleep restriction, relaxation training, sleep hygiene education, cognitive therapy, and light therapy. Among them the stimulus control therapy has been demonstrated most effective as a single treatment or in combination with other treatments. Through nonpharmacological treatments, sleep latency was most significantly reduced and wake time after sleep onset was also reduced. About 50% of insomniacs reported clinical improvements in terms of nearly normalized sleep latency, awakening time, sleep efficiency, and reduction of hypnotic use. Compared to the hypnotic therapy, nonpharmacological treatments are more cost-effective and more readily accepted by patients, and their effects last longer.

Development of Retinal Prosthesis Module for Fully Implantable Retinal Prosthesis (완전삽입형 인공망막 구현을 위한 인공망막모듈 개발)

  • To restore visual sensation of blind patients, we have proposed a fully implantable retinal prosthesis comprising an three dimensionally (3D) stacked retinal chip for transforming optical signal to electrical signal, a flexible cable with stimulus electrode array for stimulating retina cells, and coupling coils for power transmission. The 3D stacked retinal chip is consisted of several LSI chips such as photodetector, signal processing circuit, and stimulus current generator. They are vertically stacked and electrically connected using 3D integration technology. Our retinal prosthesis has a small size and lightweight with high resolution, therefore it could increase the patients` quality of life (QOL). For realizing the fully implantable retinal prosthesis, we developed a retinal prosthesis module comprising a retinal prosthesis chip and a flexible cable with stimulus electrode array for generating optimal stimulus current. In this study, we used a 2D retinal chip as a prototype retinal prosthesis chip. We fabricated the polymide-based flexible cable of $20{\mu}m$ thickness where 16 channels Pt stimulus electrode array was formed in the cable. Pt electrode has an impedance of $9.9k{\Omega}$ at 400Hz frequency. The retinal prosthesis chip was mounted on the flexible cable by an epoxy and electrically connected by Au wire. The retinal prosthesis chip was cappted by a silicone to pretect from corrosive environments in an eyeball. Then, the fabricated retinal prosthesis module was implanted into an eyeball of a rabbit. We successfully recorded electrically evoked potential (EEP) elicited from the rabbit brain by the current stimulation supplied from the implanted retinal prosthesis module. EEP amplitude was increased linearly with illumination intensity and irradiation time of incident light. The retinal prosthesis chip was well functioned after implanting into the eyeball of the rabbit.

  • This study was conducted to investigate visual threshold of nigro (Cichlasoma nigrofasciatum) on white LED light. The visual threshold was obtained by analyzing electrocardiogram (ECG) of the nigro. 5 individuals (body weight: 15.62~45.49 g; TL: 8.9~12.4 cm) were trained for lights by an electric stimulus. And then the heart rate (beats/10s) before and after switching on the light were compared. Light intensity range was from 0.00 to 226.4 lux. Average heart rate was 10.36 beats/10s in the normal condition. When the fish perceived the light, the heart rate was decreased. Visual threshold of the fish was 2.59 lux.

The Effects of Stimulus Velocity and Skill Levels on Anticipation Timing Performance of Passing (자극의 가속 및 감속 조건에 따른 숙련도별 농구 패스의 예측 타이밍 수행의 차이)

  • The study was to investigate the effects of stimulus velocity and passer's skill level on anticipation timing performance. Fourteen subjects(seven novices and seven experts) were required to make a total 12 passes in coincidence with an experimentally manipulated moving light signal in randomly presented three different conditions(4m/s, $3m/s{\rightarrow}5m/s$, $5m/s{\rightarrow}3m/s$). Results of analyses showed that absolute error(AE) and constant error(CE) were greater in constant acceleration of the moving stimulus. In addition, experts were more accuracy and consistency than novices on absolute, constant and variable error(VE). These findings indicated that stimulus velocity served as the major determination of anticipation timing performance of passing.

Orienting Response of Rats Following Damage to the Mammillary Bodies (유두체가 제거된 흰쥐의 정위반응)

  • A study was planned to see if the mammillary bodies have any influence upon orienting response Thirty-nine male rats were divided into two groups. One group of animals received electrolytic lesion of the mammillary bodies through stereotaxically implanted electrodes (mammillary body group), and the other group received the same treatment short of electrolytic lesion(operated control group). All animals were tested for orienting response 3 weeks after surgery. Orienting response was regarded as elicited when an animal in alertness ceased ongoing activity upon 1,000 Hz tone presentation and/or turned head toward the source of stimulus. Weak (momentary and inconspicuous) and strong (marked and more persistent) responses were discriminated. Occurrence and duration of orienting response were measured, and the rate of habituation was estimated in 20 trials. Shift of attention from sound to light stimulus (10 trials) as well as orienting response of thirsty animals to sound stimulus while drinking (5 trials) and while at rest (5 trials) were also tested. 1. Simple orienting respons to sound stimulus tended to occur more often and its duration tended to be shorter in the mammillary body group than in the operated control group. 2. With repetition of trials, the occurrence of orienting response in each trial decreased progressively in both groups (habituation). However, the rate of habituation was significantly less prominent in the mammillary body group than in the operated control group. 3. Under attention·shift situation, orienting response tended to occur less often and its duration tended to be shorter in the mammillary body group than in the operated control group. 4. Orienting response to sound stimulus while drinking to quench thirst tended to occur more often and its duration tended to be shorter in the mammillary body group than in the operated control group. In quiet state, however, there existed no group difference with regard to the occurrence and duration of the orienting response. Although no obvious inference can be drawn from the above results, they may suggest the mammillary bodies being involved in the orienting response as a control mechanism inhibitory to the occurrence and facilitatory to the habituation of the response.

Vital Signal Monitoring Simulation System by Various Visual Stimulus (다양한 시각자극에 따른 생체신호 모니터링 시뮬레이션 시스템)

  • With the development of next IT convergence technology and the construction of infrastructure for personalized services, the importance of services based on user's sensibility status and vital signal is being spotlighted. It is the most crucial factor for the strategy of sensibility engineering to investigate student's vital signal according to the sensibility. In this paper, we proposed the vital signal monitoring simulation system by the various visual stimulus. The proposed method obtained the student's vital signal by the various visual stimulus. And the power spectrum analysis using the fast fourier transform is evaluated the activity level between the sympathetic and the parasympathetic. The analyzed result is monitoring the sensibility by displaying the heath signal light. To evaluate the sensibility between the high school's students and the university's students, we conducted SD profile and paired T-tests so as to verify usefulness. This evaluation found that the difference of sensibility activity by positive and negative stimulus was statistically meaningful. Ultimately, this paper suggests empirical application to verify the adequacy and the validity.
