Comparative Genome Analysis Reveals Natural Variations in the Genomes of Erwinia pyrifoliae, a Black Shoot Blight Pathogen in Apple and Pear

  • Erwinia pyrifoliae is a Gram-negative bacterial plant pathogen that causes black shoot blight in apple and pear. Although earlier studies reported the genome comparison of Erwinia species, E. pyrifoliae strains for such analysis were isolated in 1996. In 2014, the strain E. pyrifoliae EpK1/15 was newly isolated in the apple tree showing black shoot blight in South Korea. This study aimed to better understand the similarities and differences caused by natural variations at the genomic level between newly isolated E. pyrifoliae EpK1/15 and the strain Ep1/96, which were isolated almost 20 years apart. Several comparative genomic analyses were conducted, and Clusters of Orthologous Groups of proteins (COG) database was used to classify functional annotation for each strain. E. pyrifoliae EpK1/15 had similarities with the Ep1/96 strain in stress-related genes, Tn3 transposase of insertion sequences, type III secretion systems, and small RNAs. The most remarkable difference to emerge from this comparison was that although the draft genome of E. pyrifoliae EpK1/15 was almost conserved, Epk1/15 strain had at least three sorts of structural variations in functional annotation according to COG database; chromosome inversion, translocation, and duplication. These results indicate that E. pyrifoliae species has gone natural variations within almost 20 years at the genomic level, and we can trace their similarities and differences with comparative genomic analysis.

A Study on dental hygienist's humor (치과위생사의 유머에 관한 연구)

  • The purpose of this study was to verify the reliability of the instrument and to analyze the contents of the humor. This study was conducted from December 16th, 2002 to January 13th, 2003, centering on Busan Dental Hospital & public health service. A total of 193 questionnaires was distributed for the survey. The result were as followings: 1. There were significant differences in humor values concerning age(F=1l.44, p=0.000), marriage(t= -3.556, p=0.000), education(F=14.83, p=0.000), clinical carrier(F=9.99, p=0.000), status(F=5.83, p=0.001), working place(F=7.39, p=0.000), and living parents(F=3.65, p=0.014). Humor values were higher for over-forties, married dental hygienists, public health service worker and no living parents. 2. There were no Significant correlation between stress and the dental hygienist's humor values. 3. There were significant differences in humor values concerning sources of the dental hygienist's humor. Data related "book/mass media"(t=-6.32, p=0.000), "conversation" (t=-12.05, p=0.000) and "daily life"(t=-10.33, p=0.000) are examples of these sources. 4. There were no significant differences in humor values concerning the dental hygienist's humor degree related frequency of used humor. 5. There were significant differences in humor values concerning the type of humor used, "word humor"(t=-7.00, p=0.000), "imitation"(t=2.68, p=0.008), "adequate situation" (t=-8.03, p=0.000), "technical terms"(t=6.65, p=0.000) pertain to this. 6. There were Significant differences in humor values concerning the time of using humor expression, "loose situation"(t=-3.75, p=0.000), "tired situation"(t=4.01, p=0.000), "tense situation"(t=5.37, p=0.000), "adequate situation"(t=-16.03, p=0.000) pertain to this.

Expression of Flagellin Proteins of Campylobacter jejuni within Microaerobic and Aerobic Exposures

  • Campylobacter, one of the emerging foodborne pathogens, is highly adaptable to the external environments by changing its morphology. In the present study, a question of whether the whole-cell antibody would still be effective for its detection even though the morphology of C. jejuni was changed was examined. When microaerophilic C. jejuni was exposed to aerobic conditions for 48 h, its morphological change was detected by confocal laser scanning microscope: Its morphology was confirmed as a spiral-bacilli form in microaerobic condition, however, as a coccoid form with a little spiral-bacilli form, when exposed to aerobic conditions. Also, the expressions of the whole-cell proteins of C. jejuni, and the suppression or induction of newly synthesized proteins in both aerobic and microaerobic conditions were analyzed by two dimensional gel electrophoresis. Additionally, immunoblotting assay with the whole cell antibody for the proteins expressed under the two conditions was performed. It was confirmed that the commercial whole-cell antibody of C. jejuni raised in rabbit was reactive. When analyzed with MALDI- TOF MS, the expressed proteins were confirmed as flagellins. Therefore, even though the morphology changed in aerobic condition, these flagellins were expressed and worked as the eitope proteins, thus making it possible to utilize for the development of an immunosensor for real-time detection of any kind of C. jejuni cell.

Lack of O-Polysaccharide Renders Bradyrhizobium japonicum More Resistant to Organic Acid Stress

  • In previous studies, we isolated an isogenic LPS mutant of Bradyrhizobium japonicum 61A101C, which was completely devoid of O-polysaccharide and had altered cell surface characteristics. Subsequently, the mutated gene was identified, cloned, and used to complement the LPS mutant strain JS314 to restore the phenotype. Since it has been reported that in Escherichia coli LPS O-polysaccharide is involved in resistance to an organic acid such as acetic acid under low pH (Barna et al., Molecular Microbiology 43: 629-640, 2002), we compared the organic acid resistance of the three B. japonicum strains; wild-type 61A101C, the LPS mutant JS314, and the complemented strain to determine whether the role of O-polysaccharide in the resistance to organic acid could be generalized. Growth of all three strains was inhibited by the presence of 3 mM acetic acid under acidic condition (pH 5.5). To our surprise, however, in the presence of 2 mM acetic acid, wild-type and the complemented strains did not grow while the $LPS^-$ mutant showed a significant growth. Therefore, unlike in E. coli, the lack of O­polysaccharide of LPS appears to render B. japonicum more resistant to organic acid.

  • The purpose of this study was to look into the views of marriage of gold miss' who participate in extreme sports. Two women, the participants of this study, have a specialized job and they take part in risky sports. Collecting data has been performed from December, 2012 to March, 2013 by conducting in-depth interviews and participatory observation. The interview was conducted for a couple of hours with flexibility. These were the conclusions of this study. The participation motivation in extreme sports was to cure their diseases such as arrhythmia or inflammation, and this activities caused a stress reduction, sense of competent, way to escape from melancholy daily life, happiness, and satisfaction. Also, unhappy family matter, economic independence, and sensation-seeking were found as the main factors that affect their views of marriage.

DESIGN OF A SINGLE MODE VARIABLE BRIDGE TYPE SPLIT-POWERED CVT WITH AN INNER-SPHERICAL CONTINUOUSLY VARIABLE UNIT

  • One method for improving the torque capacity of the CVT is to use a split-powered CVT(SPCVT) to reduce the power transmitted into a continuously variable unit(CVU). A variable bridge SPCVT with two planetary gear units(PGUs), which are composed of a sun gear, a ring gear, and carrier and planetary gears, can minimize the power to the CVU. However, a SPCVT with a conventional CVT should possess a dual mode, which would allow the conventional CVT to be used at high speeds and an additional gear train to be used at low speeds. The inner-spherical CVU(ISCVU) with an inner and outer spherical contact mechanism developed in this study can cover the range from low to high speeds. The rated power and the overall speed ratios were 100 kW and $0.09{\sim}0.36$, respectively. Power efficiency was numerically calculated to be over 90% over the speed ratio range of $0.1{\sim}0.29$. The maximum shear stress at the two contact areas of the rotor pairs, the minimum life and the overall size were estimated to be 700 MPa, 276 kh and $350{\times}350{\times}400mm^3$, respectively. This study shows that an ISCVU and a variable bridge type PGU can realize the SPCVT with a single mode for a vehicle.

  • 1. King Hyeonjong(1641-1674) mainly suffered from eye disease and abscess. He specially took a hot spring bath for cure of eye and skin problems. He probably died of septicemia and the following gastro-intestinal infection at the age of 34, quite early for his age. 2. King Sukjong(1661-1720) was not very well all through his life, but lived quite longer than other Kings in the Joseon Dynasty. He suffered from various diseases like heart-based heat, abscess, edema, upper respiratory infections, etc. He frequently took the treatments of acupuncture and moxibustion. He presumably died of dysfunction of liver and kidney at the age of 60. 3. King Gyeongjong(1688-1724) suffered from political problems from birth to death, so he may have had excessive mental stress for his poor health. He mainly suffered from heart-based heat and abscess. It is quite not sure why he died in only one month from the onset of his symptoms, so many people thought that he was poisoned to death. He probably died of septicemia and the following gastro-intestinal infection at the age of 37. 4. King Yeongjo(1694-1776) lived for 83 years, which is the longest of all the Kings in the Joseon Dynasty. He mainly suffered from hypofunction of gastro-intestinal system with cold symptoms, coughs, uneasiness. He took various kinds of herbal medicine, of which he took ginseng the most. He is supposed to be dead due to his old age at 83.

Analysis of Published Papers and their Keywords in the Korean Journal of Women Health Nursing (2003-2006) (여성건강간호학회지의 최근 3년간의 논문분석(2004-2006년))

  • Purpose: This research was aimed to explore the major subjects and research theme and to classify characteristics of the key words in recent Korean Journal of Women Health Nursing. Methods: With survey design, whole 103 published papers during 2003-2006 were analyzed with structured analysis form; research subjects, research methodology, data analysis, and key words of the published papers were classified and extracted. Various theme was founded and classified to 9 domains. Results: Most of the research subjects were women. Eighty-eight percent of papers conducted quantitative research; 83% chose convenience sampling and 69% used survey design, while experimental design was 29%. Key words were categorized 9 domains: demographic, health related concept, health behavior(intervention), sex-related, life event, disease, et al. Among 9 domains, health related concept, especially psycho-social topics such as depression, anxiety, stress were mostly studied and then physiologic topics such as labor pain, fatigue, and menstrual symptoms. Conclusion: Most of the subjects were women and the most common domain studied was health related concepts in Korean Journal of Women Health Nursing. And key words and topics were on women's health issues. We can conclude that Korean Journal of Women Health Nursing has published by her own philosophy.

Analysis of Published Papers by Keywords and Research Methods in the Korean Journal of Women Health Nursing (2007~2009) (여성건강간호학회지의 최근 3년간의 논문분석(2007~2009년))

  • Purpose: This study s aim was to analyze literature using the identified key words of published literature within the Korean Journal of Womens Health Nursing from 2007~2009. The study explored the major subjects and research methodology used and evaluated the quality of this Journal articles. Methods: Survey design using a structured analysis format was used for consistent evaluation of research subjects, type of research, research methodology, data analysis, along with comparing intervention research, theoretical framework, ethical consideration, and the implications. The frequency of the use of each keyword was classified by 8 domains. Results: Ninety eight papers were analyzed altogether. Most of the subjects in these papers were women with 86.7% being quantitative research. In these studies, 76.5% used convenience sampling; 70.6% used a survey design whereas 23.5% used experimental design. Health related concepts (n=159) was mentioned the most among the 8 domains. The key words frequently used were women, depression, knowledge, quality of life, attitude, elderly, anxiety and stress in descending order. Conclusion: It can be concluded that most papers in this Journal during this time period addressed recent women's health issues. Also it showed this Journal and its literature has reached the indexing for Medline.

The Antifibrotic and Antioxidant Activities of Hot Water Extract of Adventitious Root Culture of Panax ginseng (ARCP)

  • The anti-fibrotic effects of hot water extract of adventitious root culture of Panax ginseng (ARCP) and the possible mechanisms were investigated on $CCl_4-induced$ hepatotoxicity model mice. Fibrosis was induced by a mild treatment of $CCl_4$. Then silymarin as a positive control drug and ARCP or carrier alone as a negative control were treated. Serum GPT, GOT and ALP activity levels were lowered by 25, 21 and 11% for silymarin treated group and by 48, 39 and 14% for ARCP treated group compared to carrier treated alone. Hepatic collagen for ARCP treatment group was reduced by 18% and MDA contents decreased a little more. Pro-fibrotic gene ($TGF-{\beta}1$, TIMP-1 and ${\alpha}-SMA$) expression increased following the $CCl_4$ treatment, but both the silymarin and the ARCP treatments decreased the expressions of these genes by 20% to 50%. The antioxidant effect of ARCP was studied by DPPH free radical scavenging activity. In addition, a generation of reactive oxygen species (ROS) was also reduced in $H_2O_2-treated$ HepG2 cells upon the ARCP treatment. In summary, ARCP has antioxidant property, and can have some protection against oxidative stress; more importantly, ARCP can efficiently protect mice against $CCl_4-induced$ fibrosis.
