A movie is a fiction made on a basis of an author's and a writer's imagination, but all sorts of properties mixed with each other and most realistically expresses the era which becomes the background of a movie and acts as a carrier that connects designers with consumers. Thus, this study was carried out to review how the fashion products that designer's intention and commercial value added are expressed in collections by comparing and analysing the costumes in the movie "The Great Gatsby" that described the life of America's upper-class in 1920s and the 04 S/S Y&Kei collection which were proceeding after getting inspiration from this movie. For this, literature materials were inspected in order to make a theoretical review on social and cultural background and costumes history background in 1920s and the photo materials on movie costume were collected and analysed using DVD video captures, as well as the photo materials on 04 S/S Y&Kei were collected and analyzed through the institute providing domestic fashion information. The following conclusion was deduced through this study. First, in 1920s which becomes the background of this study, the slim shape of Flapper which looks like a young and boy became an ideal figure condition and the straight silhouette with low waist line and the short skirt that rose to knee was popular. Second, as a result of analysing movie costume by classifying it in silhouette, colors, and materials, straight silhouette of low waistline with a near colored - tone seen in the pastel series, including white, beige, pink, and gray was mainly constituted and the metal colors like silver and gold were used. As a material, chiffon, satin, velvet, flower patterned prints, and beads were used, which represented luxurious life of women in the upper classes. Third, as a result of comparing and analysing, it turned out that there was a similarity. However, in dress collection for a heroine, some dissimilarity differentiated from a movie costumes was found out in that the dresses in collection expressed moderate beauty and modernism and elegant beauty at the same time by matching a variety of materials and using black color.
Circadian rhythm is controlled by hormonal oscillations governing the physiology of all living organisms. In mammals, the main function of the pineal gland is to transform the circadian rhythm generated in the hypothalamic suprachiasmatic nucleus into rhythmic signals of circulating melatonin characterized by a largely nocturnal increase that closely reflects the duration of night time. The pineal gland has lost direct photosensitivity, but responds to light via multi-synaptic pathways that include a subset of retinal ganglion cells. Rhythmic control is achieved through a tight coupling between environmental lighting and arylalkylamine-N-acetyltransferase (AANAT) expression, which is the rhythm-controlling enzyme in melatonin synthesis. Previous studies on the nocturnal expression of AANAT protein have described transcriptional, post-transcriptional, and post-translational regulatory mechanisms. Molecular mechanisms for dependent AANAT expression provide novel aspects for melatonin's circadian rhythmicity. Extensive animal research has linked pineal melatonin for the expression of seasonal rhythmicity in many mammalian species to the modulation of circadian rhythms and to sleep regulation. It has value in treating various circadian rhythm disorders, such as jet lag or shift-work sleep disorders. Melatonin, also, in a broad range of effects with a significant regulation influences many of the body's physiological functions. In addition, this hormone is known to influence reproductive, cardiovascular, and immunological regulation as well as psychiatric disorders.
In this study, the effect on the free amino acid content and anti-oxidative activity of Arctium lappa samples by heat treatment were compared to obtain data on the conditions required for properly processing the root as a raw material for tea. The results of DPPH and ABTS radical scavenging activity assays showed that 180℃-4 min and 200℃-3 min treatments retained relatively high activity. All heat treatment conditions showed increased polyphenol content compared to the initial sample value of 42.72 mg/l, and the 180℃-4 min and 200-3 min treatments were higher than the others at 60.09 mg/l and 62.74 mg/l, respectively. In the free amino acid analysis of the A. lappa root tea, the tendency of the initial content was confirmed to decrease as temperature and time increased. Changes in phenylalanine, histidine, leucine, and isoleucine were found to be high, while aspartic acid and proline changes were relatively low. Overall, amino acid content decreased significantly under heating conditions above 180℃-3 min and 200℃-2 min. From these results, heat treatment of A. lappa root at 180℃-4 min was deemed optimal based on anti-oxidative activity and free amino acid content.
The objective of this study was to determine the shelf-life effects and residual nitrite contents of mugwort and pine needle extracts addition in emulsified sausage during cold storage. The sausages were of five types: no mugwort and pine needle extracts added (Control), mugwort water extract added (T1), mugwort ethanol extract added (T2), pine needle water extract added (T3), and pine needle ethanol extract added (T4). Each sausage type was tested in triplicate and assigned to one of four storage periods: 0, 10, 20 and 30 days. As storage time increased, the presence of mugwort and pine needle extracts resulted in decreased pH, CIE $L^*$, and residual nitrite value, but increased TBARS (thiobarbituric acid reactive substance) values, and total plate counts (TPC). Values for pH, TBARS, residual nitrite contents and total plate counts were significantly decreased by the addition of mugwort and pine needle extracts compared to the control (P<0.05). Among all treatments, T4 was more (P<0.05) effective in delaying lipid oxidation compared to other treatment groups. In conclusion, this study demonstrates that addition of pine needle ethanol extract to emulsified sausages tended to improve antioxidant and antimicrobial effects and residual nitrite contents during storage than other treatment groups.
The coastal regions of Yeosu, the South Sea of Korea, has occurred annually the red tide which is caused by potentially ichthyotoxic dinoflagellate C. polykrikoides, with a wide avenue for exchange with oceanic waters and freshwater runoff from Sumjin river. We attempted to examine the variability in response to vertical migration and concentration of extracted DNA/RNA of C. polykrikoides exposed to salinity-stratified waters. The experimental aquarium of the 60 liter was employed to culture C. polykrikoides. One aquarium was supplied with only sea water, the other was consisted of sea water and freshwater. Experiment was conducted for 5 days. In experimental column (mixture of freshwater and sea water), salinity was maintained to 20 at upper and approximately 30 at bottom during the period of this study. The fluctuation with related to dissolved oxygen and pH was similar pattern to both columns. Chlorophyll a was significantly higher value at upper than bottom. During 24h, chlorphyll a on experimental column was extremely high on the top as soon as lighting, compared with control. With elapsed time, the gap between experimental and control columns was a little. In darkness, chlorophyll a was not significantly different between upper and bottom, most cells appeared to randomly distribute on column regardless of water layer. Fluctuation with related to concentration of extracted DNA and RNA based on ratio of absorbance of 260 and 280 nm in experimental column was higher at final day or diel migration than control. These results implied that a large volume of freshwater could be associated with influence of concentration of DNA and RNA, in particular, rapid upward movement caused massive fish kills as soon as sunset.
This study was conducted to establish the optimal medium condition for the animal probiotic Lactobacillus plantarum by using onion juice. Cell yield and antioxidant activity increased in proportion to high additive levels of onion juice in medium. Onion juice, sucrose and yeast extract were selected as media ingredient factors and the effects of their mixed ratio in medium were evaluated. The full factorial design consisted of 24 experimental runs and was employed to estimate the main effects of the factors and their interactions. Significant positive effects on cell yield and antioxidant activity was shown with yeast extract and onion juice, respectively. Significant interaction was found only between sucrose and yeast extract in antioxidant activity. Finally, we selected an optimal medium that was composed of (g/l) onion juice, 600; sucrose, 15; yeast extract, 5. The efficiency of this optimum medium was estimated by using a 5 l jar fermenter. As a result, the maximum cell yield was $9.7\;{\log}_{10}$ (CFU/ml) at 12 hr. Cell yield at the end of incubation (20 hr) was $8.9\;{\log}_{10}$ (CFU/ml) and it was very similar with the predicted value, $9.0\;{\log}_{10}$ (CFU/ml). Antioxidant activity of culture was maintained at about 60~65% during all incubation time, resulting in a higher-than-predicted activity of 47.1%.

The immature fruit of Rubus coreanus Miquel (bokbunja in Korean) is mainly consumed as a fruit wine due to its sour taste and low sugar content. The lees (LBW) remaining after the production of bokbunja wine are discarded as they have no specific usage. The aim of this study was to develop high-value-added biomaterials for functional foods and beauty/health products by investigating the anti-microbial, anti-oxidant, and anti-thrombosis activities of LBW using ethanol and hot water extracts and their subsequent organic solvent fractions. The ethyl acetate (EA) fraction of LBW extracts has a high polyphenol content (413–459 mg/g), and showed strong anti-microbial activity against gram-positive bacteria. The EA fraction also showed excellent radical-scavenging activity against DPPH anion, ABTS cation, and nitrite, with strong reducing power. The polyphenol-enriched EA fraction strongly inhibited thrombin, prothrombin, and blood coagulation factors. The butanol fraction showed a specific inhibition of coagulation factors, as measured in activated partial thromboplastin time assay, which is linked to intrinsic blood coagulation. The butanol fraction also showed strong inhibition of platelet aggregation, at levels comparable to aspirin. The residue of the hot-water extract, which is produced by sequential solvent fractionation of the LBW extract, showed superior inhibition against platelet aggregation when compared to aspirin. Our results suggest that the LBW, which are currently discarded, are a promising source of novel functional foods and beauty/health products.

An antibacterial substance to the Streptococcus mutans, a causative bacterium for decayed teeth, was isolated from the dried sea mustard, Undaria pinnatifida, and identified by GC and GC/MS. Acetone extract from the sea mustard (10.4 kg), was evaporated and partitioned to 4 fractions such as hexane, chloroform, butanol and water. The most active chloroform fraction were further purified through basic alumina, silicic acid and ODS column, successively, and finally, 3 antibacterial substances were isolated on the HPLC attached ODS column by using 95% MeOH and guided with UV detector (254 nm). Antibacterial substances (total 160mg, yield $1.5\times10^{-3}$%) had the same Rf value (0.42) on the TLC developed hexane diethyl ether acetic acid (80:30:1) and those methyl esters moved to 0.95. They were identified as the same unsaturated fatty acid, $C_{18:4,\;n-3}$ (3,6,9,12-octadecatetraenoic acid, stearidonic acid) compared relative retention times (15.5 min) with authentic fatty acid on the GC chromatogram. It was further confirmed unambiguously on the GC/MS giving molecular ion peak at m/z 290 which coincided with its methyl ester.
This work was conducted to identify sweet potato [(Ipomoea batatas (L.) Lam.)] cultivars showing high yield and processing suitability in the Jeonbuk region of Korea. Shoot-slips (30 cm in length) of 14 cultivars were planted with a planting density of $75{\times}25cm$ on May 30, and cultivated with black film mulching. Vine elongation at 30 days after planting was significantly increased (> 90 cm) in 'Dahomi' and 'Sinhwangmi', and was excellent (> 250 cm) 120 days after planting in 'Sinhwangmi', 'Shinyulmi', 'Daeyumi', 'Jinhongmi', and 'Yeonhwangmi'. The branch number per plant showed a dramatic increase in 'Shinzami' 30 and 120 days after planting. The leaf number per plant showed the highest increase in 'Shinzami' 30 days and in 'Sinhwangmi' 120 days after planting. Total weight, mean weight, and number of marketable tubers per plant were significantly higher 150 days after planting than that 120 days after planting. Marketable tuber yield at 120 days was high, over 2.5 ton/10a in 'Daeyumi', 'Dahomi', and 'Jeonmi', and was considerably increased, from 3.2 ton/10a to 3.5 ton/10a, 150 days after planting. Total weight of marketable tubers per plant and marketable tuber yield 120 and 150 days after planting had significant positive correlations with vine length and number of nodes 30 days after planting. Starch value was significantly higher 150 days after planting than that 120 days after planting. 'Daeyumi' and 'Jeonmi' had good growth vigor, high yield, and high starch content; 'Dahomi' also exhibited beneficial traits such as good growth vigor, high yield, and bright orange-colored flesh.
Chitin, a $\beta$-1,4 polymer of N-acetyl-D-glucosamine, is one of the most abundant organic compounds in nature. Chitinase (EC 3.2.1.14) is an enzyme that degrades chitin to chito-oligosaccharides, diacetyl rhitobiose and N-acetyl-D-glucosamine. An extracellular chitinase-producing bacterial strain was isolated from soil and named to as Bacillus subtilis JK-56. Optimum culture condition of B. subtilis JK-56 for the production of chitinase was 1% chitin, 0.5% polypepton, 0.1% KCl, 0.05% MnS $O_4$.4$H_2O$, 37$^{\circ}C$, initial pH 7.0 and 40 hour culture time. When B. subtilis JK-56 was grown in the optimum medium, one major active band and two minor active bands were detected by native-PAGE and active staining of the gel. Among them, the major band was purified from the culture supernatant by 70% ammonium sulfate precipitation and native-PAGE with BIO-RAD Model 491 Prep-Cell and named as Chi-56A. Its molecular weight was estimated to be 53kDa monomer and the isoelectric point (pI) was pH 4.3. The pH and temperature for the optimum activity of Chi-56A were pH 6.0 and $65^{\circ}C$, respectively. Chi-56A was stable up to $65^{\circ}C$ and in alkaline region. Its $K_{m}$ value for colloidal chitin was 17.33g/L. HPLC analysis of the reaction products confirmed that Chi-56A was an exo type chitinase.e.
