Arabidopsis AHL Gene Encodes a 3'(2'),5'-Bisphosphate Nucleotidase Sensitive to Toxic Heavy Metal Ions

  • Arabidopsis AHL gene contains 4 exons encoding a putative protein highly homologous to the yeast salt-sensitive enzyme HAL2, a 3'(2'),5'-bisphosphate nucleotidase involving in reductive sulfate assimilation. AHL cDNA complemented yeast met22 (hal2) mutant. AHL fusion protein expressed in E. coli exhibited $Mg^{2+}$-dependent, 3'-phosphoadenosine 5'-phosphate (PAP)-specific phosphatase activity. $Li^+,\;Na^+,\;K^+$ and $Ca^{2+}$ ions inhibit the enzyme activity by competing with $Mg^{2+}$ for the active site of the enzyme. The enzyme activity was also sensitive to ${\mu}M$ concentrations of toxic heavy metal ions such as $Cd^{2+},\;Cu^{2+}$ and $Zn^{2+}$, but was not recovered by addition of more $Mg^{2+}$ ions, suggesting that these ions inactivate the enzyme with a mechanism other than competition with $Mg^{2+}$ ions. Inhibition of the AHL enzyme activity may result in accumulation of PAP, which is highly toxic to the cell. Thus, the AHL enzyme could be one of the intial targets of heavy metal toxicity in plants.

Effect of Linker for Immobilization of Glutathione on BSA-Assembled Controlled Pore Glass Beads

  • Controlled pore glass bead was modified with bovine serum albumin (BSA), and glutathione (GSH) was immobilized through three kinds of linkers on top of BSA. Bis(3-sulfo-N-hydroxysuccinimide suberate) sodium salt $(BS^3)$, N-hydroxysuccinimide 3-(2-pyridyldithio)propionate (SPDP), or N-hydroxysuccinimide 4-maleimidobutyrate (GMBS) was introduced into the BSA-bound matrix. Subsequently, GSH was immobilized by addition of thiol side chain into the maleimido moiety, replacing a disulfide group, or formation of an amide group upon releasing 3-sulfo-N-hydroxysuccimide group. It was observed that conjugation methodology played a critical role for activity of the immobilized GSH. SDS-PAGE chromatogram showed that the matrix of glutathione immobilized on BSA through GMBS manifested high selectivity towards glutathione-S-transferase (GST) in cell lysate.

Investigation on Structure and Properties of a Novel Designed Peptide with Half-Sequence Ionic Complement

  • Although the existing design principle of full-sequence ionic complement is convenient for the development of peptides, it greatly constrains the exploration of peptides with other possible assembly mechanisms and different yet essential functions. Herein, a novel designed half-sequence ionic complementary peptide (referred to as P9), AC-Pro-Ser-Phe-Asn-Phe-Lys-Phe-Glu-Pro-$NH_2$, is reported. When transferred from pure water to sodium chloride solution, P9 underwent a dramatic morphological transformation from globular aggregations to nanofibers. Moreover, the rheological experiment showed that the P9 could form a hydrogel with a storage modulus of about 30 Pa even at very low peptide concentration (0.5% (wt/vol)). The P9 hydrogel formed in salt solution could recover in a period of about 1,800 sec, which is faster than that in the pure water. The data suggestcd that the half-sequence, ionic complementary peptide might be worthy of further research for its special properties.

  • A novel acrylol borate was designed and synthesized by reacting acrylate monomer and boric acid. The obtained acrylol borate was used as both gelator and anion receptor for the liquid electrolyte in a lithium secondary battery. It was found that the ionic conductivity of the gel polymer electrolyte (GPE) was as high as that of the liquid electrolyte, and the thermal stability of GPE was increased when only 2 wt% acrylol borate was incorporated into the liquid electrolyte. These results suggest that acrylol borate can be used as an effective additive to enhance the thermal stability of the electrolyte without adversely affecting its conductivity. It is believed that the strong complex formation between boron in the gelator and the anion of the salt is responsible for the enhanced thermal stability of the electrolyte solution and the increased ionic conductivity.

Flexible membranes with a hierarchical nanofiber/microsphere structure for oil adsorption and oil/water separation

  • Oil spill and oily wastewater have now become a serious threat to the freshwater and marine environments. Porous materials with super-hydrophobicity and super-oleophilicity are good candidates for the oil adsorption and oil/water separation. Here, flexible hybrid nanofibrous membrane (FHNM) containing $SiO_2$/polyvinylidene fluoride (PVDF) microspheres was prepared by simultaneous electrospinning and electrospraying. The obtained FHNM combined the flexibility of the nanofiber mat and super-hydrophobicity of the microspheres, which could not be achieved by either only electrospinning or only electrospraying. It was found that when the weight ratio between the $SiO_2$ and PVDF reached a critical value, the $SiO_2$ nanoparticles were present on the PVDF microsphere surface, significantly improving the surface roughness and hence the contact angle of the FHNM. Compared with the pure electrospun PVDF nanofiber mat, most of the FHNMs have a higher oil adsorption capacity. The FHNM could separate the oil with water quickly under the gravity and displayed a high efficiency and good reusability for the oil/water separation. More importantly, the FHNM could not only separate the oil with the pure water but also the corrosive solution including the salt, acid and alkali solution.

ASSESSMENT OF ACTIVITY-BASED PYROPROCESS COSTS FOR AN ENGINEERING-SCALE FACILITY IN KOREA

  • This study set the pyroprocess facility at an engineering scale as a cost object, and presented the cost consumed during the unit processes of the pyroprocess. For the cost calculation, the activity based costing (ABC) method was used instead of the engineering cost estimation method, which calculates the cost based on the conceptual design of the pyroprocess facility. The calculation results demonstrate that the pyroprocess facility's unit process cost is $194/kgHM for pretreatment, $298/kgHM for electrochemical reduction, $226/kgHM for electrorefining, and $299/kgHM for electrowinning. An analysis demonstrated that the share of each unit process cost among the total pyroprocess cost is as follows: 19% for pretreatment, 29% for electrochemical reduction, 22% for electrorefining, and 30% for electrowinning. The total unit cost of the pyroprocess was calculated at $1,017/kgHM. In the end, electrochemical reduction and the electrowinning process took up most of the cost, and the individual costs for these two processes was found to be similar. This is because significant raw material cost is required for the electrochemical reduction process, which uses platinum as an anode electrode. In addition, significant raw material costs are required, such as for $Li_3PO_4$, which is used a lot during the salt purification process.

  • Hydraulic concrete buildings in the northwest of China are often subject to the combined effects of low-temperature frost damage, during drying and wetting cycles, and salt erosion, so the study of concrete deterioration prediction is of major importance. The prediction model of the relative dynamic elastic modulus (RDEM) of four different kinds of modified concrete under the special environment in the northwest of China was established using Grey residual Markov theory. Based on the available test data, modified values of the dynamic elastic modulus were obtained based on the Grey GM(1,1) model and the residual GM(1,1) model, combined with the Markov sign correction, and the dynamic elastic modulus of concrete was predicted. The computational analysis showed that the maximum relative error of the corrected dynamic elastic modulus was significantly reduced, from 1.599% to 0.270% for the BS2 group. The analysis error showed that the model was more adjusted to the concrete mixed with fly ash and mineral powder, and its calculation error was significantly lower than that of the rest of the groups. The analysis of the data for each group proved that the model could predict the loss of dynamic elastic modulus of the deterioration of the concrete effectively, as well as the number of cycles when the concrete reached the damaged state.

Riboflavin and Bacillus subtilis effects on growth performance and woody-breast of Ross 708 broilers with or without Eimeria spp. challenge

  • This study was conducted to assess the effects of the dietary supplementation of riboflavin (as a bile salt hydrolase [BSH] inhibitor) and Bacillus subtilis on growth performance and woody breast of male broilers challenged with Eimeria spp. Intestinal bacteria, including supplemented probiotics, can produce BSH enzymes that deconjugate conjugated bile salts and reduce fat digestion. A 3 × 2 × 2 (riboflavin × Bacillus subtilis × Eimeria spp. challenge) factorial arrangement of treatments in randomized complete block design was used. On d 14, birds were gavaged with 20× doses of commercial cocci vaccine (CoccivacR -B52, Merck Animal Health, Omaha, NE). Dietary treatment of riboflavin and B. subtilis did not affect body weight (BW), body weight gain (BWG), and feed conversion (FCR) d 0 to 14 and overall d 0 to 41. Eimeria spp challenge reduced BWG, feed intake (FI), and increased FCR between d 14 to 28, but increased BWG and lowered FCR between d 28 to 35. There were no effects of the Eimeria spp. challenge on the overall d 0 to 41 FCR and FI, but BWG was reduced. Eimeria spp. challenge increased the abdominal fat pad weight and slight woody breast incidences on processed birds on d 42. Dietary inclusion of B. subtilis and riboflavin at tested levels did not help birds to mitigate the negative impact of Eimeria spp. challenge to enhance the growth performance.

Controlling interlayer spacing of GO membranes via the insertion of GN for high separation performance

  • Graphene oxide (GO) membranes have attracted extensive attention in water treatment and related fields. However, GO films are unstable and have low permeability, which have hindered their further development. In this paper, a simple and effective method was used in which GO and single-layer graphene (GN) were mixed, and the layer spacing was effectively controlled by accurately controlling the ratio of GO to GN. GO-GN composite membranes have excellent stability, salt rejection (95.4%), and water flux (26 L m-2 h-1 bar-1). This unique design structure can be used for precise and effective regulation of the layer spacing in GO, improving the rejection rate, and increasing water flux via the enhancement of low-friction capillary action. The rational development and use of this unique composite membrane provides a reference for the water treatment field.

Application of AC superimposed DC waveforms to bismuth electrorefining

  • Electrorefining in molten salts is used for purifying actinides. Optimizing electrorefining is key to minimizing processing time and radiological waste. One possible way of improving electrorefining efficiency is using an AC superimposed DC waveform. This waveform has demonstrated potential benefits in aqueous solutions but has never been utilized in a molten metal, molten salt application. This work investigates the effects of using an AC superimposed DC waveform on molten bismuth electrorefining in a molten LiCl-KCl-CaCl2 eutectic. Bismuth has been identified as a potential surrogate for plutonium electrorefining and a potential cathode in electrorefining used nuclear fuel (UNF). All electrorefining runs resulted in a high purity cathode ring and high yield with exception of the run using a low-frequency, high-amplitude superimposed AC waveform, which experienced some contamination and a lower yield. The other three AC superimposed DC runs experienced an average yield 6.7 % higher than the average yield of the DC runs. The electrorefining run using the high-frequency, high-amplitude superimposed AC signal had the highest yield. It is recommended in future studies to investigate the statistical variability of electrorefining yield and current efficiency and the impact of AC superimposed DC waveforms on solidified bismuth anodes.
