Comparison of Treatment Effect of Domestically Distributed Major Silage Inoculant

  • Silage inoculants, crucial in modern silage production, comprise beneficial microorganisms, primarily lactic acid bacteria (LAB), strategically applied to forage material during ensiling. This study aimed to compare the effectiveness of various inoculants produced by different companies. Five treatments were evaluated, including a control group: T1 (Lactobacillus plantarum), T2 (Lactobacillus plantarum + Pediococcus pentosaceus), T3 (Lactobacillus plantarum + Pediococcus pentosaceus + Lactobacillus buchneri), T4 (Lactobacillus plantarum + Lactobacillus acidophilus + Lactobacillus bulgaricus), and T5 (Lactobacillus plantarum + Pediococcus pentosaceus + Enterococcus faecium). Italian ryegrass was harvested at the heading stage and treated with these silage inoculants. Samples were collected over a 60-day ensiling period. Co-inoculation with L. plantarum and P. pentosaceus (T2) resulted in significantly higher CP compared to the control group co-inoculation exhibited with resulted in Lactobacillus plantarum and Pediococcus pentosaceus in the T2 treatment exhibited higher CP content of 106.35 g/kg dry matter (DM). The T3 treatment, which included heterofermentative bacterial strains such as Lactobacillus buchneri, exhibited an increase in acetic acid concentration (11.15 g/kg DM). In the T4 treatment group, which utilized a mixed culture of Lactobacillus acidophilus and Lactobacillus bulgaricus, the NH3-N/TN content was observed to be the lowest (20.52 g/kg DM). The T5 containing Enterococcus faecium had the highest RFV (123) after 60 days. Expanding upon these findings, the study underscores not only the beneficial effects of particular inoculant treatments on silage quality but also underscores the potential of customized inoculation strategies in maximizing nutrient retention and overall silage preservation.

Isolation and Identification of Newly Isolated Antagonistic Streptomyces sp. Strain AP19-2 Producing Chromomycins

  • A new antagonistic strain of actinomycete, designated AP19-2, was isolated from the feces of giant pandas inhabiting the Foping National Nature Reserve in China. Cultural characteristic studies strongly suggested that this strain is a member of the genus Streptomyces. The nucleotide sequence of the 16S rRNA gene of strain AP19-2 evidenced profound similarity (97-99 %) with other Streptomyces strains. Two pure active molecules were isolated from a fermentation broth of Streptomyces sp. strain AP19-2 via extraction, concentration, silica gel G column chromatography, and HPLC. The chemical structures of the two related compounds (referred to as chromomycin $A_2$ and chromomycin $A_3$) were established on the basis of their Infrared spectra (IR), High Resolution Electrospray Ionization Mass Spectrometry (HR-ESI-MS), and Nuclear Magnetic Resonance (NMR) data, and by comparison with published data.

Association of Polymorphisms in the Calpain I Gene with Meat Quality Traits in Yanbian Yellow Cattle of China

  • The calpain I (CAPN1) gene is an important marker for meat tenderness and marbling score in the bovine, but there were no studies to determine whether the CAPN1 gene had an association with other meat quality traits. In this study, we examined the relation between genetic polymorphisms of the CAPN1 gene and some meat quality traits in Yanbian Yellow Cattle of China. By PCRSSCP and gene sequencing in 321 unrelated Yanbian yellow cattle, twenty seven single nucleotide polymorphisms (SNPs) were detected in CAPN1, two existed SNPs in exon 8 and exon 17 resulted in the change of AA at F311S and M599V, respectively, and the otherpolymorphisms were at intron 7, 8, 14, 16 and 17. There were different preponderant genotypes at the corresponding gene locus and all genotypes were not associated with tenderness but other meat traits. This is the first study of the relationship between CAPN1 and meat quality besides tenderness in Yanbian yellow cattle of China.

Knockdown of a Proliferation-inducing Ligand (PRIL) Suppresses the Proliferation of Gastric Cancer Cells

  • Purpose: PRIL (proliferation-inducing ligand) is a newly identified member of the tumor necrosis factor (TNF) family and modulates death ligand-induced apoptosis. Here, we investigated the effect of PRIL on cellular characteristics relating to tumor progression in human gastric cancer. Method: Recombinant lentivirus containing PRIL siRNA was constructed and then infected MGC803 and SGC7901 gastric cancer cells. MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] colony formation and cell cycle analysis were used to study the effect of PRIL knockdown on gastric cancer cell proliferation. Results: PRIL expression in lentivirus infected cells was significantly reduced as evidenced by quantitative real-time PCR. Cell viability and colony formation of MGC803 and SGC7901 cells were significantly hampered in PRIL knock-down cells. Moreover, the cell cycle was arrested at G2/M phase, elucidating the mechanism underlying the inhibitory effect of siRNA on cell proliferation. Conclusions: Our study indicated that PRIL functions in promoting cell growth, and lentivirus-mediated PRIL gene knockdown might be a promising strategy in the treatment of gastric cancer.

Eccentric strength and design of RC columns strengthened with SCC filled steel tubes

  • Self-compacting Concrete Filled steel Tubes (SCFT), which combines the advantages of steel and concrete materials, can be applied to strengthen the RC columns. In order to investigate the eccentric loading behavior of the strengthened columns, this paper presents an experimental and numerical investigation on them. The experimental results showed that the use of SCFT is interesting since the ductility and the bearing capacity of the RC columns are greatly improved. And the performance of strengthened columns is significantly affected by four parameters: column section type (circular and square), wall thickness of the steel tube, designed strength grade of strengthening concrete and initial eccentricity. In the numerical program, a generic fiber element model which takes in account the effect of confinement is developed to predict the behavior of the strengthened columns subjected to eccentric loading. After the fiber element analysis was verified against experimental results, a simple design formula based on the model is proposed to calculate the ultimate eccentric strength. Calibration of the calculated results against the test results shows that the design formula closely estimates the ultimate capacities of the eccentrically compressed strengthened columns by 5%.

Downregulated MicroRNA-133a in Gastric Juice as a Clinicopathological Biomarker for Gastric Cancer Screening

  • Circulatory miR-133a is a marker shared by several types of cancer. In this study we evaluated the feasibility of using miR-133a levels in gastric juice to screen for gastric cancer. A total of 204 samples of gastric juice and mucosa from gastric cancer, atrophic gastritis, gastric ulcer, superficial gastritis and healthy cases were collected by gastroscopy. The results showed that miR-133a levels in gastric juice and carcinoma tissues of patients with gastric cancer were significantly downregulated and positively correlated. Moreover, miR-133a in gastric juice has high operability, high reliability, high sensitivity, high specificity and relative stability, fit for clinical diagnosis of gastric cancer.

Research on the Mechanism of Neutral-point Voltage Fluctuation and Capacitor Voltage Balancing Control Strategy of Three-phase Three-level T-type Inverter

  • In order to solve the neutral-point voltage fluctuation problem of three-phase three-level T-type inverters (TPTLTIs), the unbalance characteristics of capacitor voltages under different switching states and the mechanism of neutral-point voltage fluctuation are revealed. Based on the mathematical model of a TPTLTI, a feed-forward voltage balancing control strategy of DC-link capacitor voltages error is proposed. The strategy generates a DC bias voltage using a capacitor voltage loop with a proportional integral (PI) controller. The proposed strategy can suppress the neutral-point voltage fluctuation effectively and improve the quality of output currents. The correctness of the theoretical analysis is verified through simulations. An experimental prototype of a TPTLTI based on Digital Signal Processor (DSP) is built. The feasibility and effectiveness of the proposed strategy is verified through experiment. The results from simulations and experiment match very well.

Kaiso represses the expression of glucocorticoid receptor via a methylation-dependent mechanism and attenuates the anti-apoptotic activity of glucocorticoids in breast cancer cells

  • Kaiso is a Pox Virus and Zinc Finger (POZ-ZF) transcription factor with bi-modal DNA-binding specificity. Here, we demonstrated that Kaiso expression is inversely correlated with glucocorticoid receptor (GR) expression in breast carcinomas. Knockdown of Kaiso increased GR expression, while overexpression of Kaiso inhibited GR expression in breast cancer cells. Furthermore, Kaiso repressed GR proximal promoter-reporter activity in a dose-dependent manner. Remarkably, ChIP experiments demonstrated that endogenous Kaiso was associated with the GR promoter sequence in a methylation-dependent manner. Since glucocorticoids inhibit chemotherapyinduced apoptosis and have been widely used as a co-treatment of patients with breast cancer, we assessed the role of Kasio in GR-mediated anti-apoptotic effects. We found that overexpression of Kaiso attenuated the anti-apoptotic effects of glucocorticoids in breast cancer cells. Our findings suggest that GR is a putative target gene of Kaiso and suggest Kaiso to be a potential therapeutic target in GC-combination chemotherapy in breast cancer.

Towards Smart Card Based Mutual Authentication Schemes in Cloud Computing

  • In the cloud environment, users pay more attentions to their data security since all of them are stored in the cloud server. Researchers have proposed many mutual authentication schemes for the access control of the cloud server by using the smart card to protect the sensitive data. However, few of them can resist from the smart card lost problem and provide both of the forward security and the backward security. In this paper, we propose a novel authentication scheme for cloud computing which can address these problems and also provide the anonymity for the user. The trick we use is using the password, the smart card and the public key technique to protect the processes of the user's authentication and key exchange. Under the Elliptic Curve Diffie-Hellman (ECDH) assumption, it is provably secure in the random oracle model. Compared with the existing smart card based authentication schemes in the cloud computing, the proposed scheme can provide better security degree.

Kinetics of di-n-Butyl Phthalate Degradation by a Bacterium Isolated from Mangrove Sediment

  • Biodegradation of the endocrine-disrupting chemical di-n-butyl phthalate (DBP) was investigated using a bacterium, Pseudomonas fluorescens B-1, isolated from mangrove sediment. The effects of temperature, pH, salinity, and oxygen availability on DBP degradation were studied. Degradation of DBP was monitored by solid-phase extraction using reversed-phase HPLC and UV detection. The major metabolites of DBP degradation were identified as mono-n-butyl phthalate and phthalic acid by gas chromatography-mass spectrometry (GC-MS) and a pathway of degradation was proposed. Degradation by P. fluorescens B-1 conformed to first-order kinetics. Degradation of DBP was also tested in seawater by inoculating P. fluorescens B-1, and complete degradation of an initial concentration of $100{\mu}g/l$ was achieved in 144 h. These results suggest that DBP is readily degraded by bacteria in natural environments.
