The Association between the T102C Polymorphism of the HTR2A Serotonin Receptor Gene and HDL Cholesterol Level in Koreans

  • 5-HT2A is one of major serotonin receptor that is involved in the action of serotonin-targeting drugs. Previous clinical studies have shown an unexpected association between lower cholesterol level and psychiatric diseases, in which T102C polymorphism of HTR2A, gene of 5-HT2A serotonin receptor, might be involved. Therefore, we hypothesized a potential association between lower cholesterol level and T102C polymorphism. The effect of the T102C polymorphism on the serum lipid profiles of 646 subjects without specific psychiatric disease was investigated. Genotype was determined by polymerase chain reaction and restriction fragment length polymorphism analysis. There were significantly lower levels of total cholesterol ($193.6{\pm}35.0$ versus $202.1{\pm}45.5\;mg/dl$, p = 0.016) and HDL-cholesterol ($42.7{\pm}11.6$ versus $46.3{\pm}12.7\;mg/dl$, p = 0.004) in CC genotype than non-CC genotypes. Moreover, multivariate analysis showed that the CC genotype is a strong predictor of a lower HDL-cholesterol level (p < 0.001). In conclusion, this study shows that the CC genotype of the HTR2A gene is related to lower HDL-cholesterol level in Koreans. This is the first demonstration showing the potential genetic relationship between the serotonin receptor gene polymorphism and the HDL-cholesterol level.

Studies on the Required Amount and Safe Level of Polyunsaturated Fat -1. Effects of different levels of perilla seed oil on Blood and Liver of Rats- (불포화 지방의 필요량과 안전성에 관한 연구 -들깨유의 fat level차이에 따른 rat의 혈액과 간에 미치는 영향-)

  • This study was carried out using rats weighing $40{\sim}50\;g$ which were devided into seven groups with various diet compositions emphasizing fat levels of perilla seed oil for the period of 41/2 weeks. The levels of fat in the diet were 5%, 10%, 15% and animals were fed ad libitum. The results are as follows : 1) Yellow pjgmentation of both neck and tail was clearly observed in groups fed 10% and 15% level perilla seed oil without vitamin I supplementation (IV and VII). 2) The growth rate in groups fed 15% level perilla seed oil was reduced as compared to that in groups fed 5% or 10% level perilla seed oil. 3) The mean hematocrit values of 15% level perilla seed oil groups tended to be lower than those of control group, tut the differences were not significant. 4) The serum vitamin I concentration showed different value in various groups, the values of control group were significantly higher than those of perilla seed oil groups-15% level with or without vitamin E supplementation (VI and VII) and 10% level without vitamin E supplementation (IV). According to the results, 10% level-perilla seed oil in the diet can be considered safe if vitamin E is not omitted from the vitamin mixture ana the group fed 15% fat with P/S ratio of 1 appeared to be safe during the experimental period. Finally the long-term studios have to be persued in many aspects by using perilla seed oil in the experimental diet. Because rats are known t4 be quite resistant to the experimental diets, comparative studies using various animal species have to he conducted.

Evaluation of the protective efficacy of trivalent Salmonella inactivated vaccine including Chlorhexidine-inactivated Salmonella Enteritidis, S. Typhimurium and S. Gallinarum in poultry (산란계에서 Chlorhexidine-inactivated Salmonella Enteritidis, S. Typhimurium 및 S. Gallinarum 3가 백신의 효능평가)

  • Protective efficacy of trivalent Salmonella inactivated vaccine containing Chlorhexidine-inactivated S. Enterltidis (SE), S. Typhimurium (ST), and S. Gallinarum (SG) strains, was evaluated in this study. A total of 70 brown nick layers were divided into 7 groups, A to G, containing 10 hens per group. All hens in groups B to D were intramuscularly immunized with approximately 7×108 cells (3×108 cells of SE+1×108 SE+1×108 cells of ST+3×108 cells of SG) of the trivalent vaccine in 0.5 mL of PBS. All chickens in groups E to G were injected with sterile PBS. All hens of groups B and E, groups C and F, and groups D and G were orally challenged with approximately 2 ×109 CFU of wild-type SE, ST, and SG, respectively. Serum IgG titers and CD3+CD4+ T-cells, and CD3+CD8+ T-cells levels of groups B to D significantly higher than those of group A. In addition, all animals in groups A to C, E and F showed no clinical symptoms and survived after the virulent challenges, whereas one chicken in group D died and all chickens in group G died following the challenge. The protection against wild-type SE and ST in liver, spleen, cecum, and cloaca of groups B and C chickens was significant effective as compared with those in groups E and F. These indicate that the trivalent inactivated vaccine can be an effective tool for prevention of Salmonella infections by inducing robustly protective immune responses and cellular immune response in chickens.

Comparative evaluation of the biological properties of fibrin for bone regeneration

  • Fibrin is a natural provisional matrix found in wound healing, while type I collagen is a major organic component of bone matrix. Despite the frequent use of fibrin and type I collagen in bone regenerative approaches, their comparative efficacies have not yet been evaluated. In the present study, we compared the effects of fibrin and collagen on the proliferation and differentiation of osteoblasts and protein adsorption. Compared to collagen, fibrin adsorbed approximately 6.7 times more serum fibronectin. Moreover, fibrin allowed the proliferation of larger MC3T3-E1 pre-osteoblasts, especially at a low cell density. Fibrin promoted osteoblast differentiation at higher levels than collagen, as confirmed by Runx2 expression and transcriptional activity, alkaline phosphatase activity, and calcium deposition. The results of the present study suggest that fibrin is superior to collagen in the support of bone regeneration.

Centrobin/Nip2 Expression In Vivo Suggests Its Involvement in Cell Proliferation

  • Centrobin/Nip2 was initially identified as a centrosome protein that is critical for centrosome duplication and spindle assembly. In the present study, we determined the expression and subcellular localization of centrobin in selected mouse tissues. Immunoblot analysis revealed that the centrobin-specific band of 100 kDa was detected in all tissues tested but most abundantly in the thymus, spleen and testis. In the testis, centrobin was localized at the centrosomes of spermatocytes and early round spermatids, but no specific signal was detected in late round spermatids and elongated spermatids. Our results also revealed that the centrosome duplication occurs at interphase of the second meiotic division of the mouse male germ cells. The centrobin protein was more abundant in the mitotically active ovarian follicular cells and thymic cortex cells than in non-proliferating corpus luteal cells and thymic medullary cells. The expression pattern of centrobin suggests that the biological functions of centrobin are related to cell proliferation. Consistent with the proposal, we observed reduction of the centrobin levels when NIH3T3 became quiescent in the serum-starved culture conditions. However, a residual amount of centrobin was also detected at the centrosomes of the resting cells, suggesting its role for maintaining integrity of the centrosome, especially of the daughter centriole in the cells.

Deficiency of calpain-6 inhibits primary ciliogenesis

  • The primary cilium is a microtubule-based structure projecting from a cell. Although the primary cilium shows no motility, it can recognize environmental stimuli. Thus, ciliary defects cause severe abnormalities called ciliopathies. Ciliogenesis is a very complex process and involves a myriad of components and regulators. In order to excavate the novel positive regulators of ciliogenesis, we performed mRNA microarray using starved NIH/3T3 cells. We selected 62 murine genes with corresponding human orthologs, with significantly upregulated expression at 24 h after serum withdrawal. Finally, calpain-6 was selected as a positive regulator of ciliogenesis. We found that calpain-6 deficiency reduced the percentage of ciliated cells and impaired sonic hedgehog signaling. It has been speculated that this defect might be associated with decreased levels of ${\alpha}-tubulin$ acetylation at lysine 40. This is the first study to report a novel role of calpain-6 in the formation of primary cilia.

Changes of Hormonal Level and Blood Biochemistry Following Superovulation Treatments of Jeju Black Cow (제주 흑우에서 다배란 처리 후 호르몬 수준과 혈액 생화학치의 변화)

  • The objective of this study was to determine changes in serum hormone concentrations, blood chemical values and recovery rate of in vivo embryos during the estrous cycle following super-ovulation treatments in Jeju black cows. Superovulation was induced by subcutaneous administration of FSH twice a day for 4 days. Serum hormones were assayed by radioimmunoassay (RIA) and blood chemical values were analyzed by blood analyser system. Embryos were collected from all treated black cows using nonsurgical technique on day 7 after artificial insemination (AI). The results of this study were summarized as follows: 1. The progesterone concentrations were $7.2{\pm}3.8ng/ml$ at day -11 and $0.3{\pm}0.1ng/mL$ at day 0 (Day 0 is the first day of AI). The estradiol concentrations were $10.6{\pm}4.48pg/ml$ at day -11 and $15.0{\pm}2.2pg/ml$ at day 0. The lowest level of progesterone was measured at day 0. The highest levels of estradiol was measured at day 0. 2. The blood chemical values of treated black cows were no significant differences in normal cow values. 3. Sixty two embryos were collected in 12 black cows. Among the collected embryos, 37 embryos (59.7%) could be transferred into recipients. These results would be used as the basic informations for changing patterns of hormonal level and blood biochemistry in Jeju black cow with superovulation.

Effect of Soyangin-Hyeongbangpaedok-san on Anti-CD3 Stimulated Mouse T Cells In Vivo and In Vitro (소양인 형방패독산(荊防敗毒散)의 마우스 T 세포 활성에 대한 실험적 연구)

  • Soyangin-Hyeongbangpaedok-san(SHBPDS) is a herbal formula used for the common cold or upper respiratory illness. In order to investigate the effect of SHBPDS, mice were orally administered with SHBPDS alcohol extract for 7 days followed by intravenous anti-CD3 injection. In addition, splenocytes and CD4 T cells were cultured with SHBPDS in response to anti-CD3 in vitro and cytokines and transcription factors were evaluated. In vivo treatment with SHBPDS significantly augmented the expressions of the percentage of CD4 T cells and CD 69, an indicator of early T cell activation. Serum levels of IL-4 were significantly increased but those of IFN-${\gamma}$ and IL-2 did not reach statistical significance. The expressions of IFN-${\gamma}$ and T-bet mRNA were significantly downregulated in SHBPDS treated mice while those of IL-4 and C-Maf were significantly upregulated. In vitro stimulation of splenocytes and CD4 T cells by SHBPDS resulted in a reduction in IFN-${\gamma}$ secretion and STAT4 activity. The IL-4 releases from both cells were slightly reduced, but STAT6 activity was rather increased. In conclusion, SHBPDS exerted an inhibition in the expression of IFN-${\gamma}$, T-bet and STAT4 while IL-4, C-Maf and STAT6 were increased. Further studies are required to examine its pharmacological effects using more appropriate animal experiments.

Effects of vitamin D supplementation on metabolic indices and hs-CRP levels in gestational diabetes mellitus patients: a randomized, double-blinded, placebo-controlled clinical trial

  • BACKGROUND/OBJECTIVES: Vitamin D plays an important role in the etiology of gestational diabetes mellitus (GDM). This study evaluated the effect of vitamin D supplementation on metabolic indices and hs-C-reactive protein (CRP) levels in GDM patients. SUBJECTS/METHODS: The study was a randomized, placebo-controlled, double-blinded clinical trial. Seventy-six pregnant women with GDM and gestational age between 24-28 weeks were assigned to receive four oral treatments consisting of 50,000 IU of vitamin $D_3$ (n = 38) or placebo (n = 38) once every 2 weeks for 2 months. Fasting blood glucose (FG), insulin, HbA1c, 25-hydroxyvitamin D, lipid profile, hs-CRP, and homeostasis model assessment-insulin resistance (HOMA-IR) were measured before and after treatment. Independent and paired t-tests were used to determine intra- and intergroup differences, respectively. ANCOVA was used to assess the effects of vitamin D supplementation on biochemical parameters. RESULTS: Compared with the placebo group, in the vitamin D group, the serum level of 25-hydroxyvitamin D increased (19.15 vs. -0.40 ng/ml; P < 0.01) and that of FG (-4.72 vs. 5.27 mg/dl; P = 0.01) as well as HbA1c (-0.18% vs. 0.17%; P = 0.02) decreased. Improvements in the lipid profiles were observed in the vitamin D group, but without statistical significance. Significant increases in concentrations of hs-CRP, FG, HbA1c, total cholesterol, and LDL cholesterol were observed in the placebo group. No significant change in fasting insulin and HOMA-IR was observed in either group. CONCLUSIONS: In GDM patients, vitamin D supplementation improved FG and HbA1c but had no significant effects on lipid profile or hs-CRP.

The Variation of Serum Thyroglobulin Measured after Thyrogen Administration (Thyrogen 투여 후 측정된 Thyroglobulin 농도변화)

  • Purpose Assessment of Serum Thyroglobulin (sTg) value in total thyroidectomy patients having an ablation dose of radioactive iodine indicates remaining cancer or metastasis. Especially, sTg in patients on withdrawal thyroxine or thyrogen administration for radioiodine ablation is an important indicator to determine the direction of further treatment and prognosis. Current guidelines suggest measurement of sTg is performed at 72 hours after the last injection of thyrogen. and assumes that sTg reaches maximum serum levels at that time. The purpose of this study is to evaluate the variation of sTg measured after thyrogen administration. Materials and Methods We compared with sTg performed at 24hours(D0) and 72hours(D2) after the last injection of thyrogen. We reviewed D0 and D2 from 276 patients were divided them into three groups according to ablation dose of radioactive iodine, 5mCi(A group), 30~80mCi(B group) and 100~200mCi(C group). We used T-test for comparison between D0 and D2. sTg was measured in serum using immunoradiometric assay (Tg-plus RIA; BRAHMS, Berlin, Germany). Results There is no critical variation between D0 and D2 in A group(n=100)(P=0.32), The case of increase(D2>D0) is 45, no change(D2=D0) is 23, decrease(D2D0 is 91, D2=D0 is 28, D2D0 is 19, D2=D0 is 2. The biggest increase is 143.6 ng/mL from 98.4 to 242. Conclusion There was a significant difference in the group over 30mCi. and the case of D2>D0 is 45%, 58.7%, 90.5% for each group. therefore, D2 increased as the dose of radioactive iodine increased. Furthermore, the most sTg values of D0 and D2 are variation under 2.0 ng/mL, so reproducibility as well as sensitivity of sTg will be important at values below 2ng/mL.
