Avian leukosis virus subgroup J and reticuloendotheliosis virus coinfection induced TRIM62 regulation of the actin cytoskeleton

  • Background: Coinfection with avian leukosis virus subgroup J (ALV-J) and reticuloendotheliosis virus (REV) is common in chickens, and the molecular mechanism of the synergistic pathogenic effects of the coinfection is not clear. Exosomes have been identified as new players in the pathogenesis of retroviruses. The different functions of exosomes depend on their cargo components. Objectives: The aim of this study was to investigate the function of co-regulation differentially expressed proteins in exosomes on coinfection of ALV-J and REV. Methods: Here, viral replication in CEF cells infected with ALV-J, REV or both was detected by immunofluorescence microscopy. Then, we analyzed the exosomes isolated from supernatants of chicken embryo fibroblast (CEF) cells single infected and coinfected with ALV-J and REV by mass spectrometry. KEGG pathway enrichment analyzed the co-regulation differentially expressed proteins in exosomes. Next, we silenced and overexpressed tripartite motif containing 62 (TRIM62) to evaluate the effects of TRIM62 on viral replication and the expression levels of NCK-association proteins 1 (NCKAP1) and actin-related 2/3 complex subunit 5 (ARPC5) determined by quantitative reverse transcription polymerase chain reaction. Results: The results showed that coinfection of ALV-J and REV promoted the replication of each other. Thirty proteins, including TRIM62, NCK-association proteins 1 (NCKAP1, also known as Nap125), and Arp2/3-5, ARPC5, were identified. NCKAP1 and ARPC5 were involved in the actin cytoskeleton pathway. TRIM62 negatively regulated viral replication and that the inhibition of REV was more significant than that on ALV-J in CEF cells coinfected with TRIM62. In addition, TRIM62 decreased the expression of NCKAP1 and increased the expression of ARPC5 in coinfected CEF cells. Conclusions: Collectively, our results indicated that coinfection with ALV-J and REV competitively promoted each other's replication, the actin cytoskeleton played an important role in the coinfection mechanism, and TRIM62 regulated the actin cytoskeleton.

Prevalence of antibodies against bovine viral infectious diseases in farmed deer and wild water deer in Jeonbuk province (사육사슴 및 야생고라니의 소 바이러스성 전염병에 대한 혈청학적 연구)

  • Farmed deer could be susceptible carrier to bovine viral infectious disease. But unfortunately, there has not been an overall study over this subject in Korea so far. Therefore, a study was conducted to see serum antibodies to bovine leukosis, food and mouth disease, bovine viral diarrhea and infectious bovine rhinotracheitis in deer using the sera of farmed deer. As a result, two deer in a farms showed positive in bovine leukosis antibodies, using ELISA. For wild water deer, no antibodies were found for those diseases. As a result, it can be assumed that deer were relatively low rate of exposure to highly contagious disease such as viral bovine infectious disease in Korea. As this study was conducted over limited in number of subject and regions, continued study should be carried out in order to prevent and control the interspecies transmission in the future.

Prevalence of major legal communicable diseases in bovine and swine in Jeonbuk province (2004~2008) (전북지역에서 2004~2008년에 소와 돼지에서 법정전염병 발생 동향 분석)

  • Prevalence of major legal communicable diseases in bovine and swine had been monitored in Jeonbuk province from year 2004 to 2008. At least 1 communicable disease had been reported in 687 heads from 68 bovine farms and 17 farms (25.0%) of the 68 positive farms had 1~2 additional outbreaks during the surveillance. By disease, enzootic bovine leukosis, Johne's disease and Akabane disease were occurred in 53 farms (582 heads), 14 farms (100 heads) and 1 farm (5 heads), respectively. Swine communicable diseases were occurred in 4,466 heads from 63 swine farms and 18 farms (28.6%) of the 63 positive farms had 1~2 additional outbreaks during the surveillance. By disease, Aujeszky's disease (AD), porcine epidemic diarrhea (PED), classical swine fever (CSF), porcine reproductive and respiratory syndrome (PRRS), porcine transmissible gastroenteritis (TGE), atrophic rhinitis (AR) and Japanese encephalitis in swine (JE) were occurred in 20 farms (70 heads), 20 farms (2,817 heads), 12 farms (258 heads), 6 farms (1,257 heads), 1 farm (50 heads), 1 farm (2 heads) and 1 farm (10 heads), respectively. In total, 10 communicable diseases (1 species of first-class, 3 species of second-class, and 6 species of third-class) were reported. The first-class diseases were CSF. Johne's disease, and Aujeszky's disease. JE was the second-class and Akabane disease, enzootic bovine leukosis (EBL), PED, PRRS, TGE and AR were third-class diseases.

  • Hematological surveys using Bendixen's and Goetze's Key for bovine lymphosarcoma were made in dairy herds which have been composed of the cows imported from United States, Canada and Japan, and their calves in the area of Taegu and Chung-Nam. The results obtained were summarized as follows: 1. Of 521 cattle, 15(2.9%) were hematologically positive for lymphosarcoma; 28(5.4%) were suspect with Bendixen's Key, and 7 (1.3%) were positive; 26(5.%) were suspect with Goetze's Key. 2. Of 14 herds, 5 were involved in positive or suspect for lymphosarcoma with Bendixen's and Goetze's Key more than 5 per cent. Of the above 5 herds, 3 were involved in positive or suspect more than 30 per cent. These findings suggest that repeated hematological examinations and continuous clinical observations are required in the herds showed higher level of per cent of positive or suspect.

  • 닭의 전신장기에 림프종 발생이 특징인 마렉병(Marek's disease; MD)은 림프구성 백혈병(Lymphoid leukosis; LL)과 병리학적 소견이 매우 유사하여 감별이 요구된다. 마렉병 바이러스(Marek's disease virus; MDV)는 질병초기에서 후기에 이르기까지 모낭 상피세포에 세포용해성 감염을 지속적으로 일으킨다. 세포용해 성 감염이 있는 모낭상피세포는 변성내지 괴사되어 있고 핵내봉입체가 관찰된다. 또한 세포용해 성 감염이 있는 모낭상피세포 바로 밑의 진피와 깃털 수질(feather pulp)내의 혈관주위에 림프구 침윤이 관찰된다. 이러한 피부병 변은 MD의 특징적인 병 변들로서 LL과 감별할 수 있는 결정적인 단서이다. 본 고에서는 MD에 관한 전반적인 것과 특히 MD 진단을 위한 피부병 변의 유용 성에 대해서 자세히 논하고자 한다

  • 닭의 전신장기에 림프종 발생이 특정인 마렉병(Marek's disease; MD)은 림프구성 백혈병(Lymphoid leukosis; LL)과 병리학적 소견이 매우 유사하여 감별이 요구된다. 마렉병 바이러스(Marek's disease virus; MDV)는 질병초기에서 후기에 이르기까지 모낭상피세포에 세포용해성 감염을 지속적으로 일으킨다. 세포용해성 감염이 있는 모낭상피세포는 변성내지 괴사되어 있고 핵내봉입체가 관찰된다. 또한 세포용해성 감염이 있는 모낭상피세포 바로 밑의 진피와 깃털 수질(feather pulp)내의 혈관주위에 림프구 침윤이 관찰된다. 이러한 피부병변은 MD의 특징적인 병변들로서 LL과 감별할 수 있는 결정적인 단서이다. 본 고에서는 MD에 관한 전반적인 것과 특히 MD 진단을 위한 피부병변의 유용성에 대해서 자세히 논하고자 한다. 양계산업에 문제를 야기하고 있는 림프구 증식성 종양은 크게 마렉병(Marek’s disease; MD)과 백혈병(Lymphoid leukosis; LL)이 있다. 이들 질환의 원인체들이 분리되기 전까지는 이들 질병들은 발병부위에 따라 질병명을 붙였다. 즉, 내부장기냐 근육에 발생한 것은 visceral lymphomatosis, 피부에 발생한 것은 skin leukosis, 말초신경의 병변은 poly­n neuritis, neuritis, neurolymphomatosis gallinarum, range paralyis로 불리었다. 또한 눈에 발생한 것은 blindness, gray eye, iritis, uveitis, ocular lymphomatosis라고 불리었다. 1961년에 Biggs는 이러한 림프구 증식성 질병을 마렉병과 백혈병 으로 분류하자는 제안을 하였고, 드디어 1960년대 중반에 림프구 증식성 병변 중의 일부에서 herpesvirus가 분리되어서 Biggs가 제안한 병명인 마렉병을 본격적으로 사용하게 되었다. MD는 마렉병 바이러스(Marek’s disease virus; MDV)가 원 인제로서 닭에 전염성이 강한 염증성에서 종양성의 병변을 내부장기, 피부, 근육, 안구, 중추신경계, 말초신경계 등에 일 으킨다. MDV는 림프종을 유발하기 때문에 처음에는 사람에서 림프종을 유발하는 Epstein-Bar 바이러스와 관련이 있을 것으로 생각되어 gamma-herpesvirus로 분류되었지만, MDV의 게놈 구조와 세포배양에서 빠르게 성장한다는 점 때문에 지금은 alpha-herpesvirus로 재분류 되었다. MDV는 바이러스 중화시험과 한천 겔 침강법에 의해서 3개의 혈청형으로 분류 된다. 혈청형 1은 종양원성 바이러스와 종양원성 바이러스의 계대배양에 의한 약독주가 있다. 혈청형 2는 자연적으로 발 생하는 비종양원성 닭의 MDV이고, 혈정형 3은 바종양원성 칠면조 herpesvirus (HVT)이다. 림프종을 유발하는 MDV 감염은 4개의 과정, 즉 초기 세포용해성 감염, 잠복감염, 후기 세포용해성 감염, 마지막으로 종양화로 나눌 수 있다. 감염의 경로를 보면, 흡입된 MDV는 폐의 대식세포에 감염한 후 전선 장기로 MDV를 전파 시킨다. 특히, 흉선, F냥, 비장 등의 림프구에 초기 세포용해성 감염을 일으키는데, B 림프구가 주로 감염된다. 세포용해성 감염음 방어하기 위해 몰려든 T 림프구가 활성화가 되면, T 세포도 감염되게 된다. 잠복감염은 여러 가지 사이토카인 등 을 포함한 면역반응에 의해서 일어나며, 이 때 잠복감염된 세포는 특히 혈중의 T 세포라고 한다. 혈중 MDV 감염세포 는 피부 모낭상피세포, 선장 등 상피세포 유래의 조직에 MDV를 전파 시켜서 이들 조직에서 후기 세포용해성 감염이 일어나게 한다. 후기 세포용해성 감염을 유발하는 것은 이러한 MDV가 감염된 혈중 림프구라는 증거는 혈중의 세포 외에는 감염성 바이러스가 없기 때문이다. 후기 세포용해성 감염이 있는 후 육안적 혹은 현미경적으로 검출이 가능한 림프종이 여러 장기에서 관찰된다. MDV가 감염되면 병변 발생부위에 따라서 다양한 임상증상이 발생한다. 즉 내부장기에 병변이 있을 경우는 침울, 체중감소, 산란율 저하 등이 발생하며, 신경계는 발생부위별 신경증상이 일어난다. 이러한 장기나 조직에서는 육안적으로 백색에서 회백색의 종양성 병변이 관찰된다. 말초신경에 병변이 발생한 경우에는 특히 좌골신경 및 신경총에서 호발하는데, 이들 조직은 편측성 혹은 양측성으로 종창되어 있다. 안구는 간혹 육안적으로 식별이 가능한 홍채 퇴색 및 증식 병변이 관찰된다. 피부형 MD는 특히 육용계에서 문제가 되고 있으며, 산란계의 내장형 MD가 발생한 경우에도 피부를 자세히 설펴보면 피부형 MD를 간혹 관찰할 수 있다. 피부형 MD는 모낭주위에 원형의 결절형태로 발생하는데, 이들 병변이 커지면 바로 옆의 병변과 융합 되어 큰 결절을 형성하기도 한다. Clinical and Cytomorphological Studios on Bovine Leukosis (우백혈병의 임상 및 세포형태학적 연구)

  • Clinical and cytomorphological studies were carried out in 32 leukotic cattle from Tokachi and Kushiro districts in Hokkaido during the 12 year period from 1969 to 1980. The leukotic cattle were examined :and divided into four types(15 cases of the adult, 11 cases of the thymic, 4 casas of the calf and 2 cases of the skin types). The results obtained were as follows : 1. As for the frequency of the main clinical signs in each type, In the adult type, the main clinical signs (of decreasing order) are as follows: swelling of the superficial lymph nodes>depression and loss of weight>tachycardia>anorekia, anemia of the visible mucous membrane and tachypnea. Those of the thymic type were swelling of the thymus>swelling of the medial iliac lymph nodes> swelling of the superficial lymph nodes>tachypnea. Those of the calf type were swelling of the auperficial lymph nodes>depression and emaciation>tachypnea>anorexia, tachycardia, anemia of the visible mucous membrane and recumbency. Those of the skin type were generalized urticaria-like lesions in skin and swelling of superficial lymph nodes>and depression and loss of weight in the decreasing order of frequency. In addition, large tumor mass in the pelvic cavity and swelling of the medial iliac lymph nodes were detected through rectal palpation in 33.3% and 100% in the adult type cases, respectively. 2. As for the hematological findings, The frequency of occurrence of decreased erythrocyte counts in the decreasing order were as follows : adult>calf>thymic>and skin types. The increase in the total leukocyte count in the order of decreasing frequency were as follows: calf>thymic>adult>and skin types. The increase in the absolute lymphocyte counts was found to be at a low rate, 62.5% of all the cases examined. By contrast, the increase of 5% or more of abnormal lymphocyte rates was observed at a high rate, 96.9% of the total cases. 3. Abnormal lymphocytes were found in all cases examined for lymph nodes biopsied. 4. From the cytomorphological point of view, leukotic cells were divided into 3 types: reticulum cell, lymphoid cell and monocytic cell types. The adult type leukotic cattle were divided with reticulum cell type (66.7%), the lymphoid cell type(22.6%) and monocytic cell type(6.7%). The thymic type was lymphoip cell type(72.7%) and reticulum cell type(27.3%). In the calf type, all were lymphoid cell type while all of the skin type were reticulum cell type only. 5. The leukotic cattle had higher NP frequency in the blood and lymphoid tissue than non-1 eukotic cattle. Especially the adult type had the highest NP frequency. However, it was not recognized that NP were characteristic of leukotic cattle alone. 6. The above findings lead to the conclusion that the most effective diagnostic methods for bovine leukosis are the confirmation of swelling of the superficial and internal lymph nodes and thymus in addition to appearance of abnormal neoplastic cell in the peripheral blood and lymph nodes biopsied.

Prevalence of Lymphocyte Nuclear Pockets in Holstein-Friesian Dairy Cattle Infected with Bovine Leukemia Virus in Korea

  • The integral relationship between the occurrence of lymphocyte nuclear pockets (LNPs) and BLV-infection was examined in Holstein-Friesian dairy cattle in Korea. Transmission electron microscope (TEM) was used to detect LNP in peripheral blood lymphocytes. Morphologically, the membranes of LNP were composed of two layers of double nuclear membrane. The full thickness of LNP membranes including inner and outer nuclear membrane was 60 to 70 nm. LNP prevalence was different according to the bovine leukemia virus (BLV) infection status; in BLV-seropositive cattle, LNP prevalence was 48.4% and in BLV-seronegative cattle prevalence was 5.9%. Moreover, even in seropositive animals, leukemic group was the highest at 70% positive among the groups, followed by suspect group (42.4%) and aleukemic group (23.1%). Consequently, the numbers of LNP were increased in proportion to increase of the numbers of leukocytes among BLV-seropositive cattle. The numbers of LNP per lymphocyte were increased in BLVseropositive cattle compared with seronegative cattle. The mean numbers of LNP per 100-lymphocytes were 0.35, 0.77, 1.64 and 4.7 in BLV-seronegative, BLV-seropositive aleukemic, suspect and leukemic groups, respectively. Thus, it is reasonable that LNP test can be used as the one of the diagnostic criteria of BLV infection.

Genetic analysis of env and gag gene fragments of bovine leukemia virus identified in cattle from Korea

  • Bovine leukemia virus (BLV) is the causative agent of enzootic bovine leukosis. This study was conducted to clarify the molecular characteristics of BLVs obtained from a specific region in Korea. Proviral BLVs were detected in anti-BLV antibody-positive blood samples by PCR. Env and gag fragments were sequenced and compared to previously published reference sequences. Analysis of the env gene sequence revealed that the YI strain was highly similar to genotype 1, including United States and Japanese strains. The gag gene sequence had the highest degree of similarity with a Japanese strain.
