목 적 : 재태연령 34주 이상의 적정체중아(appropriate for gestational age, AGA)에서 제대혈의 ghrelin과 leptin의 농도를 측정하고 출생체중, 체질량지수(body mass index, BMI) 등의 신체계측치와의 연관성 및 IGF-I, IGFBP-3와 같은 성장과 관련된 호르몬과의 관계에 대해 알아보았다. 방 법 : 재태연령 34주에서 42주 미만의 신생아 중 재태기간과 비교해 적정체중을 가진 건강한 남아 31명과 여아 29명을 대상으로 하였다. 대상 신생아의 출생체중, 체질량지수를 계측하고 제대정맥혈을 채취하여 ghrelin, leptin, IGF-I 및 IGFBP-3 농도를 측정하여 분석하였다. 결 과 : 대상 신생아들의 제대혈에서 ghrelin 농도($528.3{\pm}307.3pg/mL$)는 신생아의 출생체중(r=-0.29, P<0.05)과 재태연령(r=-0.28, P<0.05)과 반비례하였다. 한편, 대상 신생아 제대혈의 leptin 농도($4.43{\pm}3.66ng/mL$)는 신생아의 출생체중(r=0.44, P<0.01), 재태연령(r=0.36, P<0.01), 체질량지수(r=0.28, P<0.05) 등과 비례하였다. 제대혈의 ghrelin 농도는 성별에 따른 차이가 없었으나 leptin 농도는 여아($5.45{\pm}4.32ng/mL$)에서가 남아($3.48{\pm}2.64ng/mL$)에서보다 유의하게 높았으며(P<0.05), 특히 재태연령 37주 이상인 군에서 성별에 따라 뚜렷한 차이를 보였다(P<0.01). 제대혈의 ghrelin과 leptin 농도 사이에서는 의미있는 상관관계가 없었으며, ghrelin 농도와 제대혈의 IGF-I 및 IGFBP-3 사이에서도 역시 의미있는 상관관계가 없었다. 결 론 : 적정체중을 가진 신생아의 제대혈 ghrelin 농도는 출생체중 및 재태연령에 반비례하였다. 제대혈의 leptin 농도는 신생아의 출생체중, 재태연령 및 체질량지수 등의 신체계측치에 비례하였으며 여아에서 남아에서보다 더 높은 농도를 나타냈다. 제대혈의 ghrelin과 leptin 농도 사이에는 특별한 상관관계가 없었다. 앞으로 이들 물질이 태아 성장에 생리적, 병리적으로 상호 어떤 역할을 하는지 규명하기 위해 더 많은 연구가 필요할 것으로 생각된다.
목 적 : Ghrelin은 위에서 주로 분비되는 펩타이드로, 성장호르몬의 분비를 촉진시키고, 식욕을 증가시켜 에너지 균형을 조절한다고 알려져 있으며, leptin은 지방세포에서 분비되어 식욕을 억제시키는 작용을 통해 체중 및 에너지 대사에 관여한다. 저자들은 소아 종양으로 진단받고 항암 요법중인 환아의 ghrelin과 leptin 농도를 정상아와 비교해 보았다. 방 법 : 2003년 12월부터 2004년 12월까지 강남 성모병원과 성모병원에서 소아 종양을 진단 받고 방사선 조사나 항암 화학요법을 받은 환아들 중 43명(남아 31명, 여아 12명)을 대상으로 하였으며, 환아군과 성별, 나이, 체중 및 신장이 비슷한 건강한 소아 45명(남아 26명, 여아 19명)을 대조군으로 하였다. 대상아들의 혈중 leptin과 ghrelin 농도를 방사면역측정법으로 측정하였다. 결 과 : 환아군과 대조군에서 BMI와 leptin 농도는 환아군에서 더 높았으며, ghrelin은 대조군에서 더 높았다. 또한 leptin 농도는 환아군(r=0.61, P<0.05)과 대조군(r=0.40, P<0.05) 모두에서 BMI와 양의 상관관계가 있었다. ghrelin 농도는 정상군에서는 연령, 신장, 체중 및 BMI와 음의 상관관계를 보였으나 환아군에서는 의미있는 상관관계는 없었다. 한편, 방사선요법을 받은 군과 화학요법만을 받은 군으로 나누어 비교했을 때에는 두 군간의 의미있는 차이는 없었으며, 급성 골수성 백혈병군과 급성 림프구성 백혈병군으로 나누었을 때 ghrelin의 농도가 골수성 백혈병군에서 훨씬 높았다. 결 론 : 소아 종양으로 항암 요법을 받고 있는 환아는 정상아에 비해 BMI가 더 높고, leptin 농도도 더 높았으나, ghrelin 농도는 더 낮았다. 그러나 본 연구가 한 시점에서의 단기적이고 단편적인 연구이므로 추후 오랜 기간에 걸친 전향적인 연구가 더 필요하다고 생각된다.
The adipocyte-derived hormone leptin regulates appetite and bone mass. Recent research demonstrates that reciprocally, osteoblasts have a role in controlling energy metabolism. Several genes expressed in osteoblasts are involved in this process, and one of them is the Esp gene. The remaining genes regulate Esp gene expression. OST-PTP, the protein name of Esp, regulates the carboxylation of osteocalcin secreted from osteoblasts, thus affecting insulin sensitivity and insulin secretion. This review provides evidence for a novel interpretation of the connection between bone and energy metabolism and expands our understanding of the novel physiology of bone beyond its classical functions.
The somatotropic (GH-IGF-I) axis consists of many hormonal and nutritional factors that control GH release from the somatotrophs in the anterior pituitary. The GH-releasing substances are GHRH and GHS (GHRP or ghrelin), while the GH release-inhibiting substances are somatostatin (SRIF), insulin-like growth factor-I (IGF-I), leptin and glucocorticoids. However, there is evidence showing that nutrition is involved in the control of the somatotropic axis. In addition, weaning is a drastic event for neonates because their alimentary and endocrine circumstances are changed due to the switch, even if gradual, from a liquid milk diet to one composed of such solids as hay and grains. The biological role of ghrelin is one of the hormonal factors that have been focused on ever since ghrelin was discovered at the end of the last century. A 27-amino acid peptide that is mainly synthesized and released from the abomasum epithelium, ghrelin has not been fully evaluated in relation to the somatotropic axis of the ruminant. It has also proven difficult even to investigate the cellular mechanisms of ghrelin action, because this hormone exerts animal-species-dependent actions via a complex set of intracellular signaling pathways. This is also the case for the action of leptin. Another substance, IGF-I, shows a partial inhibitory action on GH secretion in the ruminant. The effect of nutrition is also different among animal species. This is evident by the fact that undernutrition suppresses the circulating GH levels in rodents, but increases it in ruminants and humans. Recently, weaning has been shown to change the postprandial GH responses in ruminants; milk feeding increases, but hay and concentrate feeding suppress, the postprandial circulating GH levels. Even if the postprandial GH level is increased, the ghrelin level is decreased by milk feeding. Macronutrients also possess stimulatory and inhibitory actions on GH secretion in vivo and in vitro. These findings indicate the complexity of the control mechanisms of the somatotropic axis. In the present review, we summarize recent findings on the factors controlling the axis of the ruminant.

Dyglomera® is an aqueous ethanol extract derived from the fruit and pods of Dichrostachys glomerata. A previous study has revealed that Dyglomera regulates adipogenesis and lipolysis by modulating AMP-activated protein kinase (AMPK) phosphorylation and increased expression levels of lipolysis-related proteins in white adipose tissue of high fat diet-induced mice and 3T3-L1 adipocyte cells. To further investigate mechanisms of Dyglomera, additional studies were performed using 3T3-L1 cells. Results revealed that Dyglomera downregulated adipogenesis by inhibiting the protein kinase B/mammalian target of rapamycin signaling pathway and reconfirmed that it downregulated gene expression levels of proliferator-activated receptor (PPAR)-γ, CCAAT enhancer binding protein α, sterol-regulation element-binding protein-1c. Dyglomera also reduced adipokines such as tumor necrosis factor alpha, interleukin-1β, and interleukin 6 by regulating leptin expression. Moreover, Dyglomera promoted beige-and-brown adipocyte-related phenotypes and regulated metabolism by increasing mitochondrial number and expression levels of genes such as T-box protein 1, transmembrane protein 26, PR domain 16, and cluster of differentiation 40 as well as thermogenic factors such as uncoupling protein 1, proliferator-activated receptor-gamma co-activator-1α, Sirtuin 1, and PPARα through AMPK activation. Thus, Dyglomera not only can inhibit adipogenesis, but also can promote lipolysis and thermogenesis and regulate metabolism by affecting adipokine secretion from 3T3-L1 adipocytes.
이 연구는 지방조직에서 분비되어 비만의 진행 조절에 관여하는 adiponectin과 leptin의 복부지방두께와의 관련성을 보고자 하였다. 연구대상자는 남성 근로자 138명으로 초음파를 이용하여 피하 및 내장지방두께를 측정하고 ELISA kit을 이용하여 adiponectin, BDNF 및 leptin의 농도를 측정하였다. 그 결과, 대상자의 평균피하지방과 내장지방두께는 $1.58{\pm}0.51$과 $4.52{\pm}1.44cm$이었다. Adiponectin, BDNF 및 leptine의 평균농도는 각각 $3.14{\pm}3.52$ ng/ml, $24.11{\pm}8.52$ pg/ml와 $4.27{\pm}2.38$ ng/ml이었다. 내장지방두께는 총 콜레스테롤, LDL-콜레스테롤, 중성지방 및 인슐린 농도와 양의 상관관계를 보였으나, HDL-콜레스테롤과는 음의 상관관계를 보였다. Adiponectin 농도는 HDL-콜레스테롤의 농도와 양의 상관관계를 보였고 총 콜레스테롤, LDL-콜레스테롤, 중성지방, 피하 및 내장지방두께와는 음의 상관관계를 보였다. 그러나 leptin의 농도는 adiponectin과 상반되는 결과를 보였다. Adiponectin의 농도는 비만하지 않은 대상자(BMI<25 $kg/m^2$)에서 유의하게 높았으나 leptin의 농도는 비만한 대상자((BMI>25 $kg/m^2$)에서 높았다. 비만과 adiponectin, BDNF 및 leptin의 관련성을 보기 위하여, 연령, 흡연 및 음주습관 등을 보정한 다음, 다중 로지스틱 회귀분석을 실시한 결과, 비만은 adiponectin(교차비=0.784, p=0.006)과 leptin(교차비=1.493, p=0.001)이 관련이 있는 것으로 나타났다. 이상의 결과는 adiponectin과 leptin의 농도가 복부지방에 영향을 주고, 이들 신경영양 물질들의 생성과 분비 기능저하는 궁극적으로 비만과 심혈관계 질환의 유발에 관여함을 보여준 결과라 판단된다.
랩틴은 지방세포의 비만유전자에서 분비되는 포식인자로써 음식섭취, 에너지대사, 체중, 번식생리 및 신경호르몬 분비를 조절하는 역할을 한다. 본 연구는 antisense 비만유전자를 발현하는 형질전환 생쥐를 생산하기 위하여 실시하였다. 먼저 랩틴을 분비하는 지방세포에서 RNA 를 추출한 후 역전사 PCR을 실시하여 303 bp의 anti I과 635 bp의 anti II cDNA 들을 합성하였다. 이러한 cDNA 들을 지방세포 특이적 발현 프로모터인 aP2 프로모터와 SV40 poly(A) 사이에 역방향으로 결합하여 미세주입용 유전자를 구축하였다. 생쥐의 수정란전핵에 antisense 비만유전자를 미세 주입하여 14 마리의 형질전환 생쥐를 생산하였으며, anti I 을 지닌 4 마리의 형질전환 생쥐와 anti II를 지닌 5마리의 형질전환 생쥐계통을 확립하였다. 그리고 형질전환 생쥐의 지방세포를 추출하여 RT -PCR을 실시한 결과 antisense 비만유전자 mRNA발현을 확인하였다. 따라서, 본 연구에서 생산된 형질전환생쥐는 생체 랩틴저하에 의해 비만을 일으키는 질환모텔동물로써의 사용가능성을 나타내었다.
Obesity is considered a mild inflammatory state, and the secretion of inflammation-related cytokines rises as adipose tissue expands. Inflammatory cytokines, including tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interlukin 6 (IL-6) and monocyte-chemoattractant protein 1 (MCP-1), are modulated by adipose tissue and known to play an important role in insulin resistance which is the common characteristics of obesity related disorders. In this study we analyzed the effects of Sasa borealis leaves extract on inflammatory cytokines and insulin resistance in diet induced obese C57/BL6J mice. The obese state was induced by a high fat diet for 20 weeks and then the mice were divided into two groups; obese control group (OBC, n = 7) and experimental group (OB-SBE, n = 7). The OBC group was fed a high fat diet and the OB-SBE group was fed a high fat diet containing 5% Sasa borealis leaves extract (SBE) for 12 weeks. We also used mice fed a standard diet as a normal control (NC, n = 7). The body weight and adipose tissue weight in the OB group were significantly higher than those in the NC group. The effects of the high fat diet were reduced by SBE treatments, and the body weight and adipose tissue deposition in the OB-SBE group were significantly decreased compared to the OBC group. The OBC group showed higher serum glucose and insulin levels which resulted in a significant increase of incremental area under the curve (IAUC) and HOMA-IR than the NC group. Also, serum leptin, TNF-${\alpha}$, and IL-6 levels were significantly higher in the OBC group than in the NC group. In contrast, the OB-SBE group showed a reversal in the metabolic defects, including a decrease in glucose, insulin, IAUC, HOMA-IR, TNF-${\alpha}$, IL-6 and leptin levels. These results suggest that BSE can suppress increased weight gain and/or fat deposition induced by a high fat diet and theses effects are accompanied by modulation of the inflammatory cytokines, TNF-${\alpha}$ and IL-6 secretion resulting in improved insulin resistance.
Sphingosine 1-phosphate is a metabolite of complex sphingolipids that acts as both a second messenger and as a high-affinity ligand for cell surface receptor. Since the possible involvement of sphingosine 1-phosphate has not been investigated in adipocyte, we examined the response of intracellular calcium ([Ca$^{2+}$]$_{i}$) and intracellular cAMP ([cAMP]$_{i}$ and the effect of sphingosine 1-phosphate on adipocyte function using rat primary adipocyte.(omitted)ted)
This study was conducted to evaluate the effects of Sunsik prepared by steaming -drying cycles- on blood glucose and inflammatory markers in high fat diet-induced obese rats. The experimental animals were divided into five groups and subjected to diets for 8 weeks; normal diet (control), high fat diet (HF), and high fat diet+10% supplementation of Sunsik (1HF, 3HF, 7HF, 9HF). The results showed body weight, food efficiency ratio, blood glucose and insulin levels of high fat diet groups were significantly higher than those of the control group. Increased glucose levels by high fat diet were reduced to normal levels by the four Sunsik supplementations. Insulin resistance (HOMA-IR) of the HF group was significantly higher than those of the control group, whereas supplementation with the four types of Sunsik reduced insulin resistance to similar levels as the control group. Increased insulin secretion and leptin levels of high fat diet-induced obese rats were significantly reduced by supplementation with the four types of Sunsik. C-reactive protein (CRP) of inflammation related components was significantly higher than the control group. However, increased CRP concentration by high fat diet was significantly reduced by supplementation with Sunsik.
