치매음이 해마손상 백서의 기억기능회복에 미치는 영향 (Improving Effects of Chimae-eum on Learning and Memory Function in the Hippocampal Damaged Rat)

  • In order to make an efficient prescription and cope with dementia, learning and memory functions of Sprague-Dawley model rats were tested with Morris water maze. And to evaluate the effect of the sample drug(CHM) on choline acetyltranferase and acetylcholine esterase, immunoreactive measurement and enzymatic activity measuring were carried out. Rats were injected with ibotenic acid through hippocampus CA1 and CA3 area. The results are as following. CHM improves the learning ability in the acquisition test and memory function in the retention test significantly. And CHM increases the level of AChE which is resolving acetylcholine. Though it doesn't increase the level of ChAT significantly which is synthesizing acetylcholine, but it shows the tendency of increase. So these results show that CHM improve the cholinergic catabolism and anabolism, and the increment of metabolic activity of cholinergic system. Thus it can be concluded that CHM will be helpful to cholinergic brain disease induced by primary or senile reduction of acetylcholine secretive activity.

기억력 감퇴모델에서 산사의 기억력 개선 효과에 관한 연구 (The Fruits of Crataegus pinnatifida Bunge ameliorates Learning and Memory Impairments Induced by Scopolamine)

  • Objectives : In the present study, we assessed the effects of the ethanolic extract of Crataegus pinnatifida Bunge on the learning and memory impairments induced by scopolamine using the passive avoidance and the Morris water maze tasks in mice. Methods : The cognition-enhancing effect of C. pinnatifida was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The ethanolic extract of C. pinnatifida (100, and 200 mg/kg) significantly reversed the scopolamine-induced cognitive impairments in the passive avoidance test (p < 0.05). Moreover, C. pinnatifida (200 mg/kg) also improved escape latencies in training trials and increased swimming times and distances within the target zone of the Morris water maze (p < 0.05). On the Y-maze test, C. pinnatifida (100, and 200 mg/kg) also significantly reversed scopolamine- induced cognitive impairments in mice (p < 0.05). Conclusions : The ethanolic extract of Crataegus pinnatifida dramatically possesses the anti-amnestic and cognitive-enhancing activities related to the memory processes, and these activities were parallel to treatment duration and dependent on the learning models.

사육탕가미방(四六湯加味方)이 흰쥐의 학습(學習) 및 기억(記憶)에 미치는 영향(影響) (An exprimental Study on the Effects of saruktanggamibangbangbang on Memory and Learning of Rats)

  • Objective: This study was conducted to find out the effect of saruktanggamibang(SRT) on memory and learning of rats. Methods: The mon·is water maze was used. It was performed Acquisition test, Retention test of water maze. After Behavioral test, it was investigated AChE cell numbers of CAI and CA3 on hippocampus. Results: of water maze revealed that acqusitive abilities of SAP+SRT group significantly improved on 3, 4, 5th day compared with SAP+Saline group. Retention test of water maze didn't reveal that retentive abilities of SAP+SRT group significantly improved comparing with another group. ChAT cell numbers of medial septum of SAP+SRT group significantly increased compared with SAP+Saline group. AChE cell numbers of CAl and CA3 on hippocampus of Sham+SRT group significantly increased compared with Sham group. And AChE cell numbers of CAl and CA3 on hippocampus of SAP+SRT group significantly increased compared with SAP+Saline group. Conclusion: Saruktanggamibang(SRT) has an effect on memory and learning of rats.

사향소합원(麝香蘇合元)이 흰쥐의 방사형 미로 학습과 기억에 미치는 영향 (The Effects of Sahyangsohapwon on Learning and Memory of Rats in the Radial Arm Maze Task)

  • Purpose: This study was conducted to find out the effects od Sahyangsohapwon on learning and memory of rats.Method: In the experiment, rats were divided two groups. One was control group which was adminstered Sahyangsohapwon and the other was sample group administered placebo. Numbers of each group were 13 rats. 8-arm radial maze task was used in it, and working memory test and retention(reference memory) test were done.Before the beginning of the test, the rats were deprived of water for 24hrs.In the frist test, each of eight arm was baited with water and a rat was permitted to remain on the maze until all eight arms were entered. A working memory error was defined as revisit of any previously entered arm. When a rat made an error not exceeding one time in consecutive 3 days-performance, it was regarded as learning criteria and the test was ended. The reference memory was evaluated with total days which it took rats to pass the learning crtirtia.The second test was performed after 24 hours when the first test was over. When a rat entered 4 arms, the entrance of arm was cut off during 30 seconds.Here the number if errors which was produced during a rat find remaining 4 tracks was regarded as the index of memory.This experiment compared the number of error at the control group with that of the sample group.Result: 1. In the first test, it was shown that the sample group took 7.69${\pm}$1.11 days and the control group 9.31${\pm}$1.97 days to pass the learning criteria.There was statistically significant reference mernory development at the sample group.2. In the second test, the frequency of errors made by the two groups were 0.92${\pm}$1.32 times for the control group and 1.23${\pm}$1.59 times for the Sahyangsohapwon group. There was no difference between the groups in frequency of errors.Conclusion: It is suggested that Sahyangsohapwon has effects on the improvement of learning and memory.

Impairments of Learning and Memory Following Intracerebroventricular Administration of AF64A in Rats

  • Three types of learning and memory tests (Morris water maze, active and passive avoidance) were performed in rats following intracerebroventricular infusion of ethylcholine aziridium (AF64A). In Morris water maze, AF64A-treated rats showed the delayed latencies to find the platform iron 6th day after the infusion. In pretrained rats, AF64A caused the significant delay of latency at 7th days but not 8th day. In the active avoidance for the pretrained rats, the escape latency was significantly delayed in AF64A-treatment. The percentages of avoidance in AF64A-treated rats were less increased than those in the control. Especially, the percentage of no response in the AF64A-treated rats was markedly increased in the first half trials. In the passive avoidance, AF64A-treated rats shortened the latency 1.5 h after the electronic shock, but not 24 h. AF64A also caused the pretrained rats to shorten the latency 7th day after the infusion, but not 8th day. These results indicate that AF64A might impair the learning and memory. However, these results indicate that the disturbed memory by AF64A might rapidly recover after the first retrain. Furthermore, these results suggest that AF64A may be a useful agent for the animal model of learning for Spatial cognition .

합환피의 인지 및 기억력 개선 효과에 대한 연구 (Effect of Albizziae Cortex Water Extract on Cognition and Memory Impairments)

  • The aim of this study was to investigate the effects of the water extract of Albizziae Cortex (AC) on the learning and memory impairments. AC was administered to normal mouse and scopolamine-injected amnesia mouse model. Passive avoidance test, Y-maze test, and Morris water maze test were conducted to confirm the cognitive-enhancing activities of AC. Acetylcholinesterase (AChE) activity and acetylcholine (Ach) content were measured after oral administration of AC. On the passive avoidance test, AC (200 mg/kg) significantly increased latency time and recovered scopolamine-impaired learning and memory in mice. In addition, AC (200 mg/kg) reduced Exploration time in target quadrant and reversed the scopolamine-induced cognitive impairments in the Y-maze test. Moreover, AC (200 mg/kg) increased exploration time in target quadrant and improved scopolamine-reduced escape latencies in the Morris water maze test. These effects were presented by regulatory effects of AC on AChE activity and Ach content. Taken together, AC increases cognitive-enhancing activities and ameliorates scopolamine-induced learning and memory impairment. AC might be a potential agent for prevention and treatment of amnesia and dementia.

기억력이 낮은 수학부진아를 위한 마인드맵 활용방안 (Teaching Mathematics by Mindmap Activities for Low Achievers in Mathematics learning who have a serious Problem in Memory)

  • 본 연구는 수학학습부진아를 원인에 관계없이 일률적으로 지도하는 기존의 교수법에 문제의식을 가지고, 특히 낮은 기억력으로 인해 수학학습부진을 보이는 아동을 지도하기 위한 방안을 마련하고자 하였다. 이를 위해 최근 여러 교과에서 시도되고 있는 마인드맵을 수학부진아 지도에 활용하는 방안을 강구하였다. 본 연구 대상자는 수학부진아 가운데, 기억력 테스트에서 낮은 점수를 기록한 아동 4명이며, 학습소재는 정의나 성질, 공식 등이 많아 비교적 기억부담이 크다고 판단되는 도형 및 측정 영역을 선정하였다. 연구 결과, 마인드맵 활동이 기억력이 낮은 수학부진 아들에게 수학적 개념을 형성하고 기억하는 것뿐만 아니라 수학 문제해결력 및 수학에 대한 태도에도 긍정적인 영향을 주는 것으로 나타났다.

태봉음이 콜린성 신경세포손상 백서의 학습 및 기억에 미치는 영향 (Effects of Taebong-eum on Learning and Memory Function in the Cholinergic Cell Damaged Rat)

  • This research was done to make the effective prescription and cope with various senile dementia. So Sprague-Dawley rats were injected with ibotenate to make a damage on learning and memory functions. At first acquisition test and retention rest were done in the Morris water maze. And to evaluate the effects of the sample drug(TBM) on choline acetyltranferase and acetylcholine esterase, immunoreactive measurement and enzymatic activity measuring were carried out. The ibotenic acid were injected to hippocampus CA1 and CA3 area. The results were as following. TBM improved the learning ability in the acquisition test and memory function in the retention test significantly. And TBM increased the level of ChAT which is synthesizing acetylcholine in CA3 area, and at the same time it increased the level of AChE which is resolving acetylcholine. These results show that T8M improved the cholinergic catabolism and anabolism, and the increment of metabolic activity of cholinergic system. In other words, it contributes to the recovery of damaged learning and memory function by ibotenic acid. So it can be concluded that TBM will be helpful to cholinergic brain damage induced by primary or senile reduction of acetylcholine secretive activity.

Ginsenoside Rg3 Alleviates Lipopolysaccharide-Induced Learning and Memory Impairments by Anti-Inflammatory Activity in Rats

  • The purpose of this study was to examine whether ginsenoside Rg3 (GRg3) could improve learning and memory impairments and inflammatory reactions induced by injecting lipopolysaccharide (LPS) into the brains of rats. The effects of GRg3 on proinflammatory mediators in the hippocampus and the underlying mechanisms of these effects were also investigated. Injection of LPS into the lateral ventricle caused chronic inflammation and produced deficits in learning in a memory-impairment animal model. Daily administration of GRg3 (10, 20, and 50 mg/kg, i.p.) for 21 consecutive days markedly improved the LPS-induced learning and memory disabilities demonstrated on the step-through passive avoidance test and Morris water maze test. GRg3 administration significantly decreased expression of pro-inflammatory mediators such as tumor necrosis factor-${\alpha}$, interleukin-1${\beta}$, and cyclooxygenase-2 in the hippocampus, as assessed by reverse transcription-polymerase chain reaction analysis and immunohistochemistry. Together, these findings suggest that GRg3 significantly attenuated LPS-induced cognitive impairment by inhibiting the expression of pro-inflammatory mediators in the rat brain. These results suggest that GRg3 may be effective for preventing or slowing the development of neurological disorders, including Alzheimer's disease, by improving cognitive and memory functions due to its anti-inflammatory activity in the brain.

스코폴라민으로 유도된 Mice에서 유근피(楡根皮)(Ulmi Cortex)의 기억력 개선 효과 (Ulmi Cortex Ameliorates Scopolamine-induced Memory Impairments in Mice.)

  • Objectives : In the present study, we assessed the effects of water extract of Ulmus davidiana(UED) on the learning and memory impairments induced by scopolamine in mice through its favorable acetylcholinesterase (AChE) activity and antioxidant effect. Methods : The memory and cognitive enhancing effect of the UDE was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. In addition, to examine the mechanism of UDE using acetylcholinesterase (AChE) and antioxidant activity. Results : The water extract of UDE (100, and 200 mg/kg) significantly reversed the scopolamine-induced cognitive impairments in the passive avoidance test (P < 0.05). Moreover, UDE (100, and 200 mg/kg) also improved escape latencies in training trials and increased swimming times and distances within the target zone of the Morris water maze (P < 0.05). On the Y-maze test, UDE (100, and 200 mg/kg) also significantly reversed scopolamine-induced cognitive impairments in mice (P < 0.05). In an in vitro study, UDE was found to inhibit acetylcholinesterase, changes in neurotrophic factor (CREB), and antioxidant activity in a dose-dependent manner. Conclusions : The water extract of UDE dramatically possesses the anti-amnestic and cognitive-enhancing activities related to the memory processes, and these activities were parallel to treatment duration and dependent on the learning models. These results suggest that the administration of UDE enhances learning and memory, and that this effect is partially mediated by ERK-CREB-BDNF signaling and the survival of immature neurons.
