A study on the prevalence and causes of sub-clinical mastitis was conducted on ten smallholder and large-scale dairy farms in Morogoro urban and peri-urban areas. A total of 65 lactating cows were screened using the California Mastitis Test (CMT). Confirmatory tests used included; the direct microscopic somatic cell count (DMSCC), culture, bacteriological and biochemical tests. Structured questionnaires were used to collect information on management aspects. Results showed 62% and 4% cows as sub- clinical and clinical mastitis cases respectively. Levels of infection were higher on smallholder farms (75%) than on large-scale farms (25%). All tested cows had high cell counts (>500,000) per ml of milk. Incidences of mastitis were significantly (p<0.05) related to milking practices. The dominant bacterial isolates in the same order were Staphylococcus aureus, Streptococcus spp, and Escherichia coli. Other organisms isolated included Pseudomonas spp and Klebsiella spp. It was concluded that the high rates of sub-clinical mastitis in the research area were mainly due to poor management and unhygienic milking practices.
A large-scale hot water production system using a natural circulation loop was developed. A computer simulation program was developed to design and evaluate thermal performance of the natural circulation system for hot water production. An experimental apparatus was set up and was tested against various conditions to exhibit a stable operating region of the natural circulation loop. When the system was a stable state for heating rate (695 ㎾) and feed water was circulated at 0.3 $\ell$/s constantly. A unstable state was checked by experiment and that time flow rate was oscillated with 0.4∼0.6 $\ell$/s. The result showed that the program can predict the thermal performance of the large-scale hot water system using the natural circulation loop and can be utilized to design the system.

During the past two decades, China has seen her great progress in plant biotechnology. Since the Chinese market of herb medicine is huge, while the plant resources are shrinking, particular emphasis has been placed in plant tissue and cell cultures of medicinal plants, this includes fast propagation, protoplast isolation and regeneration, cell suspension cultures and large scale fermentation. To optimize culture conditions for producing secondary compounds in vitro, various media, additives and elicitors have been tested. Successful examples of large scale culture for the secondary metabolite biosynthesis are quite limited : Lithospermum ery throrhizon and Arnebia euchroma for shikonin derivatives, Panax ginseng, P. notoginseng, P. quinquefolium for saponins, and a few other medicinal plants. Recent development of genetic transformation systems of plant cells offered a new approach to in vitro production of secondary compounds. Hairy root induction and cultures, by using Ri-plasmid, have been reported from a number of medicinal plant species, such as Artemisia annua that produces little artemisinin in normal cultured cells, and from Glycyrrhiza uralensis. In the coming five years, Chinese scientists will continue their work on large scale cell cultures of a few of selected plant species, including Taxus spp. and A. annua, for the production of secondary metabolites with medicinal interests, one or two groups of scientists will be engaged in molecular cloning of the key enzymes in plant secondary metabolism.
Ginsenoside Rg3 (G-Rg3) contained only in red ginseng has been found to show various pharmacological effects such as an anticancer, antiangiogenetic, antimetastastic, liver protective, neuroprotective immunomodulating, vasorelaxative, antidiabetic, insulin secretion promoting and antioxidant activities. It is well known that G-Rg3 could be divided into 20(R)-Rg3 and 20(S)-Rg3 according to the hydroxyl group attached to C-20 of aglycone, whose structural characteristics show different pharmacological activities. It has been reported that G-Rg3 is metabolized to G-Rh2 and protopanaxadiol by the conditions of the gastric acid or intestinal bacteria, thereby these metabolites could be absorbed, suggesting its absolute bioavailability (2.63%) to be very low. Therefore, we reviewed the chemical, physical and biological transformation methods for the production on a large scale of G-Rg3 with various pharmacological effects. We also examined the influence of acid and heat treatment-induced potentials on for the preparation method of higher G-Rg3 content in ginseng and ginseng products. Futhermore, the microbial and enzymatic bio-conversion technologies could be more efficient in terms of high selectivity, efficiency and productivity. The present review discusses the available technologies for G-Rg3 production on a large scale using chemical and biological transformation.
As a preliminary study of cost estimates for nuclear hydrogen systems, the hydrogen production costs of the nuclear energy sources benchmarking GT-MHR are estimated in the necessary input data on a Korean specific basis. G4-ECONS developed by EMWG of GIF in 2008 was appropriately modified to calculate the cost for hydrogen production of SI process with VHTR as a thermal energy source rather than the LUEC. The estimated costs presented in this paper show that hydrogen production by the VHTR could be competitive with current techniques of hydrogen production from fossil fuels if $CO_2$ capture and sequestration is required. Nuclear production of hydrogen would allow large-scale production of hydrogen at economic prices while avoiding the release of $CO_2$. Nuclear production of hydrogen could thus become the enabling technology for the hydrogen economy. The major factors that would affect the cost of hydrogen were also discussed.

In a turbulent free shear flow, the large scale motion is characterized by the intermittent flow which arises from the interaction between the turbulent fluid and the irrotational fluid of the environment through the mean velocity gradient. This large scale motion causes a bulk convection whose effect is similar to the spatial diffusion process. In this paper, the total diffusion process is proposed to be approximated by weighted sum of the bulk convection due to the large scale motion and the usual gradient diffusion due to small scale motion. The diffusion term in conventional .kappa.-.epsilon. model requires on more equation of the intermittency transport equation. A production term of this equation means mass entrainment from the irrotational fluid to the turbulent one. In order to test the validity of the proposed model, a plane jet is predicted by this method. Numerical results of this model is found to yield better agreement with experiment than the standard .kappa.-.epsilon. model and Byggstoyl & Kollmann's model(1986). Present hybrid diffusion model requires further tests for the check of universality of model and for the model constant fix.
Photoautotrophic micropropagation systems do not include sugar in the culture media. This characteristic provides advantages to scale up the micropropagation systems comparing photomixotrophic micropropagation systems. A closed, large-scale photoautotrophic micro-propagation for transplant production system has been developed at Chiba University, Japan. New concepts and technologies were adapted to produce high quality transplants at minimum usage of resources, and as scheduled. Newly developed software for production management was used to enhance the efficiency of the transplant production system. Currently, virus-free transplants of sweetpotato (Ipomoea batatas (L.) Lam.) are vegetatively propagated and produced under sterilized conditions in this system. This system can also be used for production of transplants of any other species including horticultural and woody plants with a minimum of modification.

In order to improve the competitiveness of Korean large purse seine fishery, the purpose of this study is to find out the problems and present the improvement of the production structure of Korean large purse seine fishery by comparing to the production structure of Norwegian purse seine fishery. Norwegian purse seine fishery has been operating labor and energy-saving systems through a single wire operation and avoiding fishing competition through the introduction of IVQ system with different types of fishing vessel and obtaining benefits from Leading fishery management integrated with fish pumps and cooling water systems. In contrast, our country has a large purse seine fishery issues such as high cost and low efficiency of production structures, the volatility of mackerel resources, the lack of leading fishery management after catching fishes, the reduction of fishing ground in accordance with the North East Asia EEZ system. To solve these problems, initiatives to improve the structure of our large purse seine fishery are as follows: First, Implementing the promotion of the effective immature fishes management and establishing Korea-China-Japan resource management system. Second, the promotion of reduction of fleet size, the review of possible single fishing operation, leading out to strengthen its competitiveness by switching to energy-efficient production systems through enhanced efforts at fisheries catches steps. Third, it is necessary to realize corporate through the diversification of business areas and the large-scale commercialization of production structure.
The optimization of the production of a fusion protein containing the antigenic domain 1 (AD-1) is of a great importance, considering its use in diagnostic tests. The fusion protein is produced by the fermentation of a recombinant strain of Escherichia coli containing the plasmid Mbg58, which expresses the AD-1 (aa 484-650) of human cytomegalovirus glycoprotein B as a fusion protein together with aa 1-375 of ${\beta}-galactosidase$. An important characteristic of promoters (lac and derivatives) used in recombinant protein production in E. coli is their inducibility. Induction by IPTG is widely used for basic research; however, its use in large-scale production is undesirable because of its high cost and toxicity. In this work, studies using different inducers and carbon sources for the production of a fusion protein containing the AD-l were performed. The results showed that lactose could be used as an inducer in the fermentation process for the production of this protein, and that expression levels could exceed those achieved with IPTG. The use of lactose for protein expression in E. coli should be extremely useful for the inexpensive, large-scale production of heterologous proteins in E. coli. Addition of sucrose to the fermentation medium improved the yield of recombinant protein, whereas addition of fructose or trehalose decreased the yield.

Mulberry leaves containing 1-deoxynojirimycin (DNJ) known to be a strong inhibitory effect for ${\alpha}-glucosidase$. Thus, DNJ has been recognized as a potentially important source for prevent or treat hyperglycemia. More effective method for the DNJ high-production is needed because DNJ content of natural mulberry leaf are as low as 0.1%. Many researchers have studied for the DNJ high-production in mulberry leaves such as the harvest season, fermentation using microorganisms, optimal culture conditions, and optimal extraction conditions. In order to provide for useful data that is anticipated at the level of industrial scale, we investigated ${\alpha}-glucosidase$ inhibitory activity, pH value and DNJ content in large-scale based on the optimal culture conditions for mulberry leaf fermentation of small-scale in our previous study. The ${\alpha}-glucosidase$ inhibitory activity, pH value, and DNJ content in this study were measured from the mulberry leaf fermentation broth for 7 days. During mulberry leaf fermentation, the ${\alpha}-glucosidase$ inhibitory activity and DNJ content was increased until 2 to 4 days, but after 4 day was decreased. The pH value showed a decreasing trend up to 2 day, and little changes in 2 to 4 days. However, the pH was started to increase after 4 days.
