  • Brillouin time-domain analysis in optical fibres is a novel technique making possible a distributed measurement of temperature and strain over long distance and will deeply modify our view about monitoring large structures, such as dams, bridges, tunnels and pipelines, Optical fibre sensing will certainly be a decisive tool for securing dangerous installations and detecting environmental and industrial threats.

  • A continuous enrichment culture procedure was used to isolate bacteria from various soil sources capable of suppressing large patch disease of turfgrass. Six isolates consistently suppressed large patch in turfgrass, and ranged in the spectrum of extracellular enzymes that they expressed. The best disease- suppressing isolate, TBM912, expressed protease, CMCase, and pectinase activity and inhibited the growth of Rhizectonin solani and Betrytis cinerea in vitro. Here we show that this strain also produces an antibiotic that was identified by TLC, SDS-PACE and HPLC analysis as lipopeptide.

  • The present paper illustrates the outcome of the monitoring of the consolidation behavior of a soft foundation soil under a large submerged sand embankment. Measurements of settlements and excess pore water pressures showed a good agreement with predictions evaluated using the large strain consolidation theory. Soft soil improvement by means of deep mixing has been optimized. Moreover, the principles and developments of underwater geosynthetics applications are discussed.

  • In this paper numerical algorithm for the continuum large crack model is proposed based on the return-mapping formulation. The numerical test results show that the present algorithm works appropriately under cyclic loading. It is also shown that in continuum damage models a large crack model to prevent excessive tensile plastic strain should be used to have realistic cyclic loading simulation results.

  • The recent advancements in sensing technologies allow us to record measurements from target structures at multiple locations and with relatively high spatial resolution. Such measurements can be used to develop data-driven methodologies for condition assessment, control, and health monitoring of target structures. One of the state-of-the-art technologies, Fiber Optic Strain Sensors (FOSS), is developed at NASA Armstrong Flight Research Center, and is based on Fiber Bragg Grating (FBG) sensors. These strain sensors are accurate, lightweight, and can provide almost continuous strain-field measurements along the length of the fiber. The strain measurements can then be used for real-time shape-sensing and operational load-estimation of complex structural systems. While several works have demonstrated the successful implementation of FOSS on large-scale real-life aerospace structures (i.e., airplane wings), there is paucity of studies in the literature that have investigated the potential of extending the application of FOSS into civil structures (e.g., tall buildings, bridges, etc.). This work assesses the feasibility of using FOSS to predict operational loads (e.g., wind loads) on chain-like structures. A thorough investigation is performed using analytical, computational, and experimental models of a 4-story steel building test specimen, developed at the University of Southern California. This study provides guidelines on the implementation of the FOSS technology on building-like structures, addresses the associated technical challenges, and suggests potential modifications to a load-estimation algorithm, to achieve a robust methodology for predicting operational loads using strain-field measurements.

Rot Deformation Behavior of AISI 316 Stainless Steel (AISI 316 스테인리스강의 고온 변형특성에 관한 연구)

  • The dynamic softening mechanisms of AISI 316, AISI 304 and AISI 430 stainless steels were studied with torsion test in the temperature range of $900 - 1200^{\circ}C$ and the strain rate range of $5.0x10^{-2}-5.0x10^0/sec$. The austenitic stainless steels, such as AISI 316 and AISI 304 were softened by dynamic recrystallization (DRX) during hot deformation. Also, the evolutions of flow stress and microstructure of AISI 430 ferritic stainless steel show the characteristics of continuous dynamic recrystallization (CDRX). To establish the quantitative equations for DRX of AISI 316 stainless steel, the evolution of flow stress curve with strain was analyzed. The critical strain (${\varepsilon}_c$) and strain for maximum softening rate (${\varepsilon}^{*}$) could be confirmed by the analysis of work hardening rate ($d{\sigma}/d{\varepsilon}={\theta}$). The volume fraction of dynamic recrystallization ($X_{DRX}$) as a function of processing variables, such as strain rate ( $\varepsilon$ ), temperature (T), and strain ( $\varepsilon$ ) were established using the ${\epsilon}_c$ and ${\varepsilon}^{*}$. For the exact prediction the ${\varepsilon}_c,\;{\varepsilon}^{*}$ and Avrami' exponent (m') were quantitatively expressed by dimensionless parameter, Z/A, respectively. It was found that the calculated results were agreed with the experimental data for the steels at my deformation conditions. Also, we can reasonably conclude that the DRX, CDRX and grain refinement of stainless steels can be achieved by large strain deformation at high Z parameter condition.

A Plant Growth-Promoting Pseudomonas fluorescens GL20: Mechanism for Disease Suppression, Outer Membrane Receptors for Ferric Siderophore, and Genetic Improvement for Increased Biocontrol Efficacy

  • Pseudomonas fluorescens GL20 is a plant growth-promoting rhizobacterium that produces a large amount of hydroxamate siderophore under iron-limited conditions. The strain GL20 considerably inhibited the spore germination and hyphal growth of a plant pathogenic fungus, Fusarium solani, when iron was limited, significantly suppressed the root-rot disease on beans caused by F. solani, and enhanced the plant growth. The mechanism for the beneficial effect of strain GL20 on the disease suppression was due to the siderophore production, evidenced by mutant strains derived from the strain. Analysis of the outer membrane protein profile revealed that the growth of strain GL20 induced the synthesis of specific iron-regulated outer membrane proteins with molecular masses of 85- and 90 kDa as the high-affinity receptors for the ferric siderophore. In addition, a cross-feeding assay revealed the presence of multiple inducible receptors for heterologous siderophores in the strain. In order to induce increased efficacy and potential in biological control of plant disease, a siderophore-overproducing mutant, GL20-S207, was prepared by NTG mutagenesis. The mutant GL20-S207 produced nearly 2.3 times more siderophore than the parent strain. In pot trials of beans with F. solani, the mutant increased plant growth up to 1.5 times compared with that of the parent strain. These results suggest that the plant growth-promoting P. fluorescens GL20 and the genetically bred P. fluorescens GL20-S207 can play an important role in the biological control of soil-borne plant diseases in the rhizosphere.

  • For the simultaneous measurement of strain and damage signal a fiber Bragg grating sensor system with a dual demodulator was proposed. The dual demodulator is composed of a demodulator using a tunable Fabry-Perot filter measuring the low-frequency signal with large magnitude such as strain and the other using a passive Mach-Zehnder interferometer detecting the high-frequency signal with small amplitude such as impact or damage signal. Using the proposed fiber Bragg grating sensor system, both the strain and damage signals of a cross-ply laminated composite beam under tensile loading were simultaneously measured. The strain and damage signals detected by single fiber Bragg grating sensor showed that sudden strain shifts were accompanied with vibration at a maximum frequency of several hundreds of kilohertz at the instant of matrix crack propagation in the 90 degree layer in composite beam.

  • A new hydrogen-oxidizing bacterium, Aeromonas sp. strain JS-1, that can fix $CO_2$ via the reductive pentose phosphate cycle (Calvin-Benson cycle) under chemoautotrophic conditions but not photoautotrophic conditions was isolated from fresh water. Strain JS-1 showed considerable $CO_2$ fixation ability during continuous cultivation even at high $CO_2$ concentration. Strain JS-1 used $H_2$ and $CO_2$ fixation as energy and carbon sources, respectively. Carbon dioxide fixation is carried out through the Calvin-Benson cycle, in which ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO) is the key enzyme. Hydrogen-oxidizing chemoautotrophic Aeromonas sp. strain JS-1 exhibited remarkedly strong RubisCO [EC 4.1.1.39] activity. RubisCO was purified as an $L_8S_8$-type hexadecamer with molecular mass of 560 kDa by gel filtration. The enzyme consisted of two different subunits eight large (56 kDa) and eight small (15 kDa), as demonstrated by SDS-PAGE. The specific activity of the purified enzyme was about 3.31 unit/mg and stable up to $45^{\circ}C$. The $K_m$ values for RuBP, $CO_2$, and $Mg^{2+}$ were estimated to be 0.25 mM, 5.2 mM and 0.91 mM, respectively.

Stress-strain behaviour of reinforced dredged sediment and expanded polystyrenes mixture under cyclic loading

  • Reinforced soil and Expanded Polystyrenes (EPS) mixture (RSEM) is a geomaterial which has many merits, such as light weight, wide strength range, easy for construction, and economic feasibility. It has been widely applied to improve soft ground, solve bridge head jump, fill cavity in pipeline and widen highway. Reutilizing dredged sediment to produce RSEM as earthfill can not only consume a large amount of waste sediment but also significantly reduce the construction cost. Therefore, there is an urgent need understand the basic stress-strain characteristics of reinforced dredged sediment-EPS mixture (RDSEM). A series of cyclic triaxial tests were then carried out on the RDSEM and control clay. The effects of cement content, EPS beads content and confining pressure on the cyclic stress-strain behaviour of RDSEM were analyzed. It is found that the three stages of dynamic stress-strain relationship of ordinary soil, vibration compaction stage, vibration shear stage and vibration failure stage are also applicative for RDSEM. The cyclic stress-strain curves of RDSEM are lower than that of control clay in the vibration compaction stage because of its high moisture content. The slopes of backbone curves of RDSEMs in the vibration shear stage are larger than that of control clay, indicating that the existence of EPS beads provides plastic resistance. With the increase of cement content, the cyclic stress-strain relationship tends to be steeper. Increasing cement content and confining pressure could improve the cyclic strength and cyclic stiffness of RDSEM.
