First Report of Macrophomina phaseolina Causing Charcoal Rot in Bottle Gourd in Korea

  • Stem blight symptoms were observed in greenhouse-grown bottle gourds (Lagenaria siceraria) in Wanju, Korea. Infected plants showed leaf chlorosis and wilting; the plants eventually died with the infected leaves remaining attached, and black sclerotia formed on the diseased stem. Based on the morphological characteristics and pathogenicity results and sequence analyses of the internal transcribed spacer, translation elongation factor 1-α, β-tubulin, and calmodulin regions, the isolated fungus was Macrophomina phaseolina. This is the first report of charcoal rot in Lagenaria siceraria caused by Macrophomina phaseolina in Korea.

Seed Treatment to Improve Germinability of Gourd (Lagenaria siceraria Standl.) (종자처리가 수박대목용 박 종자의 발아성에 미치는 영향)

  • Various methods for seed treatments have been devised to improve the rate and uniformity of seed germination of gourd (Lagenaria siceraria Standl.). Hydropriming and priming, the imbibition of seeds in distilled water and osmotic solutions respectively, are useful techniques for early establishment of uniform seedlings. In this study, the optimum conditions for hydropriming and priming were determined in gourd (Lagenaria siceraria Standl.) seeds. Effect of hydropriming was compared with those of osmotic priming. Seeds, hydroprimed or primed with 50 mM solution of $KH_2PO_4$ and $KNO_3$ at $30^{\circ}C$ for 2 days, showed shorter $T_{50}$ (number of days to 50% of the final germination percentage) and MDG (mean number of days to germination) than those unprimed. The relative effectiveness of hydropriming and priming in improving the germinability of gourd seeds was more evident when germinated at $20^{\circ}C$ than at $25^{\circ}C$ and $35^{\circ}C$.

Fe-Nanoparticle Amalgamation Using Lagenaria siceraria Leaf Aqueous Extract with Focus on Dye Removal and Antibacterial Efficacy

  • Iron nanoparticles (Fe-NPs) were synthesized employing Lagenaria siceraria (LS) leaf aqueous extract as a reducing and capping medium to remove methylene blue (MB) dye and have antibacterial properties against G-negative (Escherichia coli) and G-positive bacteria (Staphylococcus aureus). The formation of LS-Fe-NPs (Lagenaria-siceraria-iron-nanoparticles) was confirmed by a change in color from pale yellow to dark brown. Characterization techniques, such as particle size analysis (PSA), transmission electron microscopy (TEM) and scanning electron microscopy (SEM), were employed to prove nano spherical particles of size range between 80-100 nm. Phytochemicals and the presence of iron in LS-Fe-NPs nanoparticles were proved by UV-visible spectrophotometry. Further, Fourier transform infrared spectroscopy (FTIR) analysis results confirmed the existence of bioactive molecules in the plants. The magnetic property was analyzed using a vibrating sample magnetometer (VSM), which displayed that the synthesized nanoparticles were superparamagnetic and exhibiting a saturation magnetization of 12.5 emu/g. Synthesized magnetic nanoparticles were used in methylene blue (MB) dye removal through adsorption. About 83% of 100 mg/L MB dye was removed within 120 min at pH 6 with a maximum adsorption capacity of 246.8 mg/g. Antibacterial efficacy of LS-Fe-NPs was screened against G-negative (Escherichia coli) and G-positive bacteria (Staphylococcus aureus), respectively, and found that LS-Fe-NPs were effective against Staphylococcus aureus.

Control of Fusarium Wilt of Watermelon with the Root-Stock Grafting of Sicyos angulatus L. (안동오이 대목을 이용한 수박 덩굴쪼김병 방제)

  • Watermelon plants grafted with the root-stock of wild-cucumber (Sicyos angulatus) were not infected by Fusarium oxysporum f.sp. niveum in pot inoculation and infected fields tests. Controlling efficacy of the root-stock grafting with S. angulatus on Fusarium wilt of watermelon was more excellent than that of the root-stock grafting with Lagenaria siceraria. The isolates of Fusarium oxysprum from cucurbitaceae plants had a certain host-specific pathogenicity, but they did not express the absolute one forma specialis-one host-plant phenomenon by the root dipping inoculation. The pathogenic isolates of Fusarium oxysproum from cucurbitaceae crops did not infect the root-stock plant such as S. angulatus, L. siceraria and Cucurbita ficifolia. The fast-wilting of watermelon caused by uncertain agents was observed in watermelon plant grafted with L. siceraria in the continuously cropping fields, but it was not observed in watermelon plants grafted with S. angulatus in the same fields.

Cultivation and use of bottle gourd (Lagenaria siceraria) in ancient Korea (한반도 선·역사시대 박의 재배와 이용)

  • Although the bottle gourd (Lagenaria siceraria) is a crop with a cultivation history of about 10,000 years in the Old and New Worlds, archaeological considerations on the cultivation and use of bottle gourds on the Korean Peninsula are extremely rare. Accordingly, we reviewed previous reports on bottle gourds and examined the morphological characteristics of seeds and rind fragments from the Korean Peninsula. The investigation yielded several conclusions. First, bottle gourd cultivation likely began during the Bronze Age alongside the introduction of so-called southern crops. Evidence suggests that bottle gourd remains were more prevalent during the Three-Kingdoms period, indicating its significance as a crop during the historical era. Second, bottle gourd seeds from the Three-Kingdoms period exhibit characteristics of both African and Asian subspecies, showcasing a high level of morphological diversity. Third, rind thickness indicates that bottle gourds found at the Bongseon-ri site were of varieties with large fruits. Taken together, it is concluded that the bottle gourd was introduced to the Korean Peninsula during the Bronze Age, and people cultivated a range of bottle gourd varieties during the Three-Kingdoms period.

The bottle grourd was the most important plant before the invention of pottery in many areas of Asia, New Guinea, Polynesia, America, and Africa. The dug-out finds in Japan proved to be about 95,000 years old according to the /sup 14/C analysis. The bottle grourd was the most important plant before the invention of pottery in many areas of Asia, New Guinea, Polynesia, America, and Africa. I would like to suggest that there should be "The Bottle Gourd Age" prior to the Pottery Age. Bottle gourds are also used for various purposes such as food, masks, pipes, musical instruments, medicine, symbols, artistic products and also as penis-sheaths of men's attire. Their purposes number more than 220 including 70 varienties of containers or bottles. I consider that its utilization should be called a culture, as it were, "The gourd culture." The shape and the size of the fruit of bottle gourd have a larger variety than those of any other plant. As for the size, it is reported that they range from those that are shorter than 3cm to those that are ovoid and longer than 60cm in diameter. With regard to the shape of its fruit, the bottle gourd can be classified into 7 groups and even more than 30 races, considering the difference of the size. The seeds are so variable without two horn-like projections, with smooth surface or longitudinalines, white or brown, with smooth corky margin. Generally, it seems that there is no correlation between seem shape and fruit shape. My study shows that the seeds of gourd. My study shows that the seeds of gourd in Asia are so simple in shape and in color except for the size. But the seeds of those in Africa are various and seem to be beyond the confines. Explaining the great diversity of the seeds of those in Africa therefore, they appear to have no correlation among the types of fruit of the bottle gourd with African origin. It might be supposed that another 4 wild Lagenaria spp. distribute only in Africa. The intraspecies hybrid is confirmed between the bottle gourd, Lagenaria siceraria, and wild L. spehaerica. And Fl hybrid is sterile: it has no fertility. However, even though the shapes of the fruit and the seeds are different, they can cross with each other. Moreover, their hybrids grow up to be extremely vigorous and have normal fertility. Fruit shapes of Fl plants are similar to those of their parents, if they have the same character. Whereas, the fruit shape often became different from each other when the cross occurs between those with different fruit shapes. It is shown that dumbbell shapes, HYOTAN in Japanese, is recessive while its bitterness is dominant. Thus the character of F2 hybrids Is segregative. I'll give further particulars of the heredity of the groud in my lecture.
