The Emerging Role of Natural Killer Cells in Innate and Adaptive Immunity

  • In the early host defense system, effector function of natural killer (NK) cells results in natural killing against target cells such as microbe-infected, malignant, and certain allogenic cells without prior stimulation. NK cell cytotoxicity is selectively regulated by homeostatic prevalence between a repertoire of both activating and inhibitory receptors, and the discrimination of untransformed cells is achieved by recognition of major histocompatibility complex (MHC) class I alleles through inhibitory signals. Although it is well known that the bipotential T/NK progenitors are derived from the common precusor, functional mechanisms in terms of the development of NK cells remain to be further investigated. NK cells are mainly involved in innate immunity, but recent studies have been reported that they also play a critical role in adaptive immune responses through interaction with dendritic cells (DC). This interaction will provide effector functions and development of NK cells, and elucidation of its precise mechanism may lead to therapeutic strategies for effective treatment of several immune diseases.

  • High Voltage High Current Pulsed Electric Fields (HVHC-PEF) is a promising technology for the non-thermal pasteurization of foods and a sound complement or replacement to traditional thermal pasteurization, which inactivates bacteria and other microorganisms harmful to humans, but also degrades color, flavor, texture and nutrients. Foods can be pasteurized with pulsed electric fields at ambient or refrigerated temperatures for a short treatment time of seconds or less and the fresh-like quality of food is preserved. Although successful in laboratory tests, applying HVHC-PEF to food pasteurization on a large scale presents many unresolved engineering problems. In this paper the design considerations for 25kV 1kA class HVHC-PEF pasteurization equipment are analyzed and experimental results are discussed.

A Study on the WFS Co-mixtures by Small Scale Retaining Wall Test (모형옹벽실험을 이용한 폐주물사 혼합재의 지반공학 적용성 연구)

  • The purpose of this study is to present the application of WFS co-mixtures for retaining wall as flowable backfill. The fly ash, generated at the Tae-An thermoelectric power plant, was used in this research and was classified as Class F. Green Sand, Furane Sand, and Coated Sand, which had been used at a foundry located in Pusan, were used. Couple of laboratory tests and small scale retaining wall tests were performed to obtain the physical properties of the WFS co-mixtures and the possibility of backfill materials of retaining wall. The range of permeability for all the co-mixtures was from 3.0${\times}$10$\^$-3/ cm/s to 6.0${\times}$10$\^$-5/ cm/s. The unconfined strength of the 28-day cured specimens reached around 550kPa. Results of the consolidated-undrained triaxial test showed that the internal friction angle is between 33.5$^{\circ}$ and 41.8$^{\circ}$. The lateral earth pressure against wall decreased up to 80% of initial pressure within a 12 hours and the total lateral earth pressure is less than that of typical granular soil. It was enough to construct the backfill for the standard retaining of 6m with just two steps, like fill the co-mixtures for half of retaining wall, and then fill the others after 1 day. The stability of retaining wall for overturning and sliding increased as the curing time elapsed.

  • This report describes the case of an edentulous patient with an atrophic maxilla and severe class III malocclusion. Prosthetic rehabilitation was performed using CAD/CAM techniques for manufacturing an implant-supported overdenture with horizontal insertion. A vestibulo-lingual insertion overdenture is a precision prosthesis with a fixation system affording a good fit between the primary and secondary structure. Both structures exhibit passive horizontal adjustment. This treatment option requires the same number of implants as implant-supported fixed dentures. The horizontal assembly system prevents the prosthesis from loosening or moving in response to axial and non-axial forces. The technique was used to rehabilitate a patient presenting an atrophic upper maxilla, with the insertion of 8 implants. No complications were reported at follow-up 3, 6 and 12 months after fitting of the prosthesis. This system offers solutions to the clinical and laboratory complications associated with hybrid prostheses, concealing emergence of the chimneys and improving implant-prosthesis hygiene.

MicroRNA let-7c inhibits Bcl-xl expression and regulates ox-LDL-induced endothelial apoptosis

  • Endothelial cells (ECs) apoptosis induced by oxidized low-density lipoprotein (ox-LDL) is thought to play a critical role in atherosclerosis. MicroRNAs (miRNAs) are a class of noncoding RNAs that posttranscriptionally regulate the expression of genes involved in diverse cell functions, including differentiation, growth, proliferation, and apoptosis. MiRNA let-7 family is known to be involved in the regulation of cell apoptosis. However, the function of let-7 in ox-LDL induced ECs apoptosis and atherosclerosis is still unknown. Here, we show that let-7c expression was markedly up-regulated in ox-LDL induced apoptotic human umbilical cord vein endothelial cells (HUVECs). Let-7c over-expression enhanced apoptosis in ECs whereas inhibition of let-7c could partly alleviate apoptotic cell death mediated by ox-LDL. Searching for how let-7c affected apoptosis, we discovered that antiapoptotic protein Bcl-xl was a direct target of let-7c in ECs. Our data suggest that let-7c contributes to endothelial apoptosis through suppression of Bcl-xl.

Two Synthetic Ligands for Peroxisome Proliferator- Activated Receptor $\gamma$

  • The peroxisome proliferator-activated receptor $\gamma$ (PPAR${\gamma}$) is the molecular target for a class of drugs, the antidiabetic thiazolidnediones (TZDs). The heterodimer of PPAR${\gamma}$ with retinoid X receptor (RXR) plays a central role in the regulation of adipogenesis and insulin sensitization. We synthesized two chemicals, DANA87 and DANA88, sharing structural characteristics with TZDs. Given this structural similarity, it was hypothesized that DANA87 and DANA88 may act as PPAR$\gamma$ ligands. In transient transfection assays, DANA87 and DANA88 caused slight increases in the endogenous expression of a luciferase reporter gene containing the PPAR responsive element in 3T3-L1 preadipocytes. However, DANA87 and DANA88 significantly inhibited troglitazone-induced reporter gene activation when cells were treated with a combination of DANA87 or DANA 88 and troglitazone, one of the TZDs that activate PPAR$\gamma$. These results suggest that DANA87 and DANA88 are not only weak agonists of PPAR${\gamma}$ transactivation, but also competitively antagonize troglitazone-induced PPAR$\gamma$ reporter activity.

Effects of Fenofibrate on Adipogenesis in Female C57BL/6J Mice

  • Fibrates are a class of hypolipidemic agents whose effects are mediated by activation of a specific transcription factor called the peroxisome proliferator-activated receptor $\alpha\;(PPAR\alpha).\;PPAR\alpha$ regulates the pathways of lipid catabolism such as fatty acid oxidation and the triglyceride metabolism, resulting in the treatment of hyperlipidemia. The decreased levels of plasma triglycerides by fibrates are responsible for hypertrophy and hyperpalsia of adipose cells. To determine whether fenofibrate regulates adipogenesis in female C57BL/6J mice, we measured the effects of fenofibrate on not only body weight, adipose tissue mass and serum triglycerides, but also the histology of adipose tissue and the expression of adipocyte marker genes. Fenofibrate did not inhibit high fat diet-induced increases in body weight, adipose tissue mass and serum triglycerides. Furthermore, fenofibrate did not cause the changes in the size and number of adipocytes and the expression of adipocyte-specific genes such as leptin and $TNF\alpha$. Therefore, this study demonstrates that fenofibrate does not affect adipogenesis in female mice.

  • A compact and handhold near infrared (NIR) system using microspectrometer was developed. This system was suitable not only in the laboratory, but also in the field or in the process. This system was first applied for classification of geographical origin of herbal medicine such as ginseng and sesame. To identify the origin of ginseng on site, the portable NIR system is more suitable for real field application. For this study, using the compact NIR system, soft independent modeling of class analogies (SIMCA) with 1100-1750 nm NIR spectra was utilized for classification of geographical origin (Korea and China) of both ginseng and sesame. The accuracy of results is more than 90%. Quantitative analysis for petroleum such as toluene, benzene, tri-methyl benzene, and ethyl benzene was performed with partial least squares (PLS) regression with NIR 1100-1750 nm spectra. This study showed that the NIR method and gas chromatography (GC), which is a standard method, have good correlations. Furthermore, the ash content of Cornu Cervi Parvum was analyzed and the accuracy was confirmed by the developed compact NIR system.

  • Communication Subsystem of HAUSAT-2 which is 25kg class nanosatellite is designed, manufactured and tested for its performance verification at the Space System Research Laboratory (SSRL). HAUSAT-2 Communication Subsystem provides communication link for commands receiving, mission and state of health data transmission with high reliability. This paper describes design, manufacturing, test results of Engineering Model of HAUSAT-2.

  • This paper presents a mooring design procedure of a floating offshore wind turbine. The environment data of south offshore area of Jeju collected from Korea Hydrographic and Oceanographic Administration(KHOA) are used for hydrodynamic analyses as environmental conditions. We considered a semi-submersible type floating wind turbine based on Offshore Code Comparison Collaborative Continuation(OC4) DeepCWind platform and National Renewable Energy Laboratory(NREL) 5 MW class wind turbine. Catenary mooring with studless chain is chosen as the mooring system. Important design decisions such as how large the nomial sizes are, how long the mooring lines are, how far the anchor points are located, are demonstrated in detail. Considering ultimate limit state and fatigue limit state based on 100-year return period and 50-year design life, respectively, longterm predictions of breaking strength and fatigue are proposed.
