Postmortem Proteolysis of Breast and Leg Muscles from Taiwan Colored Chickens and Silkie Bantams

  • Postmortem proteolysis of breast (BM) and leg (LM) muscles from Taiwan colored chickens (TCC) and silkie bantams (SB) at $5^{\circ}C$ were compared. Myofibrils were prepared from BM and LM samples that were randomly taken from the carcasses of SB and TCC after 0, 1, 3, 7 and 14 days of storage at $5^{\circ}C$. pH of samples was determined, and degradation of myofibrillar proteins was analyzed by the SDS-PAGE and western blots. The results showed that pH was lower in BM than in LM samples from both avian strains. Appearance of 30 kDa components and disappearance of titin and nebulin were more rapidly as seen on SDS-PAGE in BM than in LM samples. Western blots labeled with a monoclonal antibody to desmin also demonstrated that desmin degraded more quickly in BM samples. Our data might suggest that postmortem proteolysis occurred more rapidly in BM than in LM from both TCC and SB.

  • The bioactivity-guided fractionation of the methylene chloride extract of the sclerotium of Poria cocos led to the isolation of (S)-(+)-turmerone (1), ergosterol peroxide (2), polyporenic acid C (3), dehydropachymic acid (4), pachymic acid (5), and tumulosic acid (6). Compounds 4-6 exhibited moderate cytotoxicities, with $IC_{50}$ values of 20.5, 29.1, and $10.4{\;}\mu\textrm{m}$, respectively, against a human colon carcinoma cell line. However, 3-6 not only showed inhibitory activities as potent as etoposide used as a positive control on DNA topoisomerase II (36.1, 36.2, 43.9 and 66.7% inhibition at a concentration of $20{\;}\mu\textrm{m}$, respectively), but also inhibition of DNA topoisomerase I (55.8, 60.7, 43.5, and 83.3% inhibition at a concentration of $100{\;}\mu\textrm{m}$, respec-tively).

  • Electrochemical behaviors of acetaminophen at a muti-wall carbon nano-tube composite film modified glassy carbon electrode were investigated by cyclic voltammetry, linear sweep voltammetry and chronocoulometry. Compared with that obtained at the unmodified electrode, the peak currents were enhanced significantly, and the oxidation peak shifted towards more negative potential with the reduction peak shifted positively. The peak-to-peak separation turned narrow, and suggested that the reversibility was improved greatly. Experimental parameters, such as scan rate, pH and accumulation conditions were optimized. It was found that a maximum current response can be obtained at pH = 5.0 after accumulation at -0.50 V for 80 s. The oxidation peak current was found to be linearly related to acetaminophen concentration over the range of $5.0{\times}10^{-7}\;\sim\;1.0{\times}10^{-4}$ mol $L^{-1}$ with a detection limit of $5.0{\times}10^{-8} $mol $L^{-1}$. A convenient and sensitive electrochemical method was developed for the determination of acetaminophen in a commercial paracetamol oral solution. Its practical application demonstrated that it has good selectivity and high sensitivity.

  • Dynamic characteristics of separation in membrane filtration system of tangential flow was investigated to find the functional relationship among the filtrate flux, transmembrane pressure, inulin concentration and recirculation rate. In case that NMWL is 1000, tracts-membrane pressure $0.4kgf/\;{\textrm{cm}^2}$ to $3.2kgf/\;{\textrm{cm}^2}$, inulin concentration lwt% to 5wt%, and recirculation rate 4ml / sec, mathematical model for the function among filtrate flux, transmembrane pressure, and inulin concentration was deduced and expressed as follows.Jv = (0.0022p + 0.0003) ln \frac{3p\;+\;2.1}{C_b}$ The values calculated by the above equation and those measured were compared to find to be nicely in accord with each other. Especially the agreement was enhanced in the region of higher concentration of inulin.

  • Epidemiological study on strongyloidiasis in humans is currently lacking in Malaysia. Thus, a cross-sectional study was carried out to determine the prevalence of Strongyloides stercoralis infection among the inhabitants of longhouse indigenous communities in Sarawak. A single stool and blood sample were collected from each participant and subjected to microscopy, serological and molecular techniques. Five species of intestinal parasites were identified by stool microscopy. None of the stool samples were positive for S. stercoralis. However, 11% of 236 serum samples were seropositive for strongyloidiasis. Further confirmation using molecular technique on stool samples of the seropositive individuals successfully amplified 5 samples, suggesting current active infections. The prevalence was significantly higher in adult males and tended to increase with age. S. stercoralis should no longer be neglected in any intestinal parasitic survey. Combination of more than 1 diagnostic technique is necessary to increase the likelihood of estimating the 'true' prevalence of S. stercoralis.

Wind-induced vibration fragility of outer-attached tower crane to super-tall buildings: A case study

  • To gain insight into the wind-induced safety concerns associated with attached tower cranes during the construction of super-tall buildings, a 606 m level frame-core tube super-tall building is selected to investigate the wind-induced vibration response and fragility of an outer-attached tower crane at all stages of construction. The wind velocity time history samples are artificially generated and used to perform dynamic response analyses of the crane to observe the effects of wind velocity and wind direction under its working and non-working resting state. The adverse effects of the relative displacement response at different connection supports are also identified. The wind-resistant fragility curves of the crane are obtained by introducing the concept of incremental dynamic analysis. The results from the investigation indicate that a large relative displacement between the supports can substantially amplify the response of the crane at high levels. Such an effect becomes more serious when the lifting arm is perpendicular to the plane of the connection supports. The flexibility of super-tall buildings should be considered in the design of outer-attached tower cranes, especially for anchorage systems. Fragility analysis can be used to specify the maximum appropriate height of the tower crane for each performance level.

  • The paper deals with the Kalman stochastic filtering problem for linear continuous-time systems with both instantaneous and time-delayed measurements. Different from the standard linear system, the system state is corrupted by multiplicative white noise, and the instantaneous measurement and the delayed measurement are also corrupted by multiplicative white noise. A new approach to the problem is presented by using projection formulation and reorganized innovation analysis. More importantly, the proposed approach in the paper can be applied to solve many complicated problems such as stochastic $H_{\infty}$ estimation, $H_{\infty}$ control stochastic system with preview and so on.

Novel Filtering Power Divider with External Isolation Resistors

  • In this paper, a novel filtering power divider with external isolation resistors is presented. The proposed power divider can be considered as an integration of a bandpass filter and a Gysel power divider. Based on the circuit topology, a high-order filtering power divider can be easily realized. Odd- and even-mode models are employed to analyze the filtering and power splitting functions. For demonstration, a third-order filtering power divider operating at 1.5 GHz is designed and implemented. The measured results exhibit an isolation between the output ports that is better than 20 dB at around the center frequency.

  • The data of crown width with 4 directions, DBH, tree height, and coordinate for sample trees were collected from 30 permanent sample plots in secondary fore st of the Maoershan Experimental Forestry Farm, Northeast China. In this paper, the competition of individual trees in stand were discussed for secondary forest by using iterative Hegyi competition index and crown overlap index that represented the competitive and cooperative interactions among neighboring trees. Active competitors of subject tree in the competition zone were selected to calculate the iterative competition index. Using the results of crown classification based on the equal crown projection area, a new distance dependent competition index called crown overlap index (COI) was developed for secondary forest. The COI performed well in describing the crown competition rather than crown competition factor (CCF). The individual-based competition index discussed in this paper will provide more precise for developing individual tree growth models for secondary forest and it can also use to adjust the stand structure for spatial optimal management.
