A Study on the Effects of Various Disk Shape of Hydrant on the Pressure Drop (옥외소화전의 디스크 형상을 고려한 압력손실에 관한 연구)

  • In this study, the effects of various disk shapes of hydrant on the pressure drop are experimentally and numerically analyzed. The test methods for measuring pressure drop of hydrant are comply with standard of Underwriters Laboratory (UL). The hydrant as used in this study has one inlet, diameter 150 mm, and three outlet, 114.3 mm diameter for one outlet and 63.5 mm diameter for the others. The pressure of the hydrant are measured in the range 760 L/min~2,270 L/min for 63.5 mm outlet and 3,030 L/min~6,060 L/min for 114.3 mm outlet. Also, the numerical results of pressure drop are compared with the experiments to verify the accuracy and to analyze the of various valve shape of hydrant on the pressure drop. The engineering parameters, flow coefficients, are reduced from 181.57 to 136.35 ($L/min/kPa^{0.5}$) with inclined angle of disk from $0^{\circ}$ to $45^{\circ}$. These results are able to practical use for design hydrant to minimize pressure drop.

Effect of Reduction in the Adipose Accumulation of Akkermansia muciniphila in Mature 3T3-L1 Adipocytes (성숙한 3T3-L1 지방세포에서 Akkermansia muciniphila의 지방축적 감소 효과)

  • Objectives: The aim of this study was to observe the reduction of lipid accumulation by treatment with Akkermansia muciniphila extract on 3T3-L1 adipocytes. Methods: After treating pasteurized Akk. muciniphila strains in HT-29 colorectal cancer cell, the relative expression of interleukin (IL)-8, tumor necrosis factor-α, IL-6, and IL-1β mRNA was analyzed by real time polymerase chain reaction, respectively. 27 strains of Akk. muciniphila which have anti-inflammatory effects were selected. 3T3-L1 pre-adipocytes were treated with Akk. muciniphila for 24 hr and then measured the toxicity using water soluble tetrazolium salt assay. The cells were incubated for 4 days and then differentiated into adipocytes using the medium including adipogenic reagents for 10 days. The Akk. muciniphila was treated when the medium was exchanged for differentiation medium at 4th day and insulin medium at 6th day. To observe the lipid accumulation, the cells were stained with Oil red O dye and were measured using a spectrophotometer. Results: In the cytotoxicity test, the cell viability of 3T3-L1 pre-adipocytes was significantly increased compared to the control group which untreated with Akk. muciniphila, and there was no cytotoxicity of Akk. muciniphila at 1×107 CFU/mL. The results on Oil red O staining and absorbance measurements were showed a significant decrease in lipid accumulation in the group which was treated with Akk. muciniphila compared to the control group. Conclusions: In our results, Akk. muciniphila has the inhibitory effect of lipid accumulation in 3T3-L1 adipocytes. This suggests that Akk. muciniphila could be help to improve obesity.

Determination of Silymarin and Silybin Diastereomers in Korean Milk Thistle using HPLC/UV Analysis (HPLC 분석을 통한 한국산 밀크씨슬 중 실리마린과 실리빈의 정량)

  • Silymarin (SM) and silybin diastereomers (SD) in milk thistle (Silybum marianum) were determined using high-performance liquid chromatography and quantified using a reverse-phase column in a gradient elution system. UV detection was performed at 288 nm. The content of SM and SD in milk thistle was 3.236 and 0.553 mg/g DW, respectively. Determining the presence and quantifying the content of SM and SD in milk thistle are vital for the pharmaceutical industry to identify optimal sources for developing health supplements or therapeutics.

  • Phytochemical study on the aerial parts of Solanum lyratum (Solanaceae) was carried out. On the basis of phytochemical and spectroscopic evidences, compound I was identified as mixtures of hexadecanoic acid methyl ester, 2,6,10,15-tetramethyl heptadecane, tricosane, tetracosane, pentacosane, docosanoic acid methyl ester, docosane, tricosanoic acid methyl ester, 8-hexyl pentadecane, tetracosanoic acid methyl ester, pentatriacontane, hexatriacontane, eicosane, hexacosane, hentriacontane and stigmasta-5,23-dien $3-{\beta}-ol$, and compound II, III, IV and V were identified as hexacosanoic acid methyl ester, ${\beta}-sitosterol-{\beta}-{_D}-glucoside$, $3-O-{\alpha}-{_L}-rhamnopyranosyl-(1->2)-{\beta}-{_D}-glucuronopyranosyl$ diosgenin and $3-O-{\alpha}-{_L}-rhamnopyranosyl-(1->6)-{\beta}-{_D}-glucopyranosyl$ quercetin (rutin), respectively. Rutin was identified as one of the active principles having antioxidative effect from S. lyratum.

  • The reactions of mercury(II) chloride with O3S2-donor macrocyclic ligands with (L1) and without (L2) dibenzosubunit afforded respective exo- (1) and endo-coordinated (2) complexes depending on the ring rigidity of the ligands. From the X-ray crystal structures and comparative NMR studies for the complexes 1 and 2, it is confirmed that the resulting species with different coordination modes exist not only in solid state but also in solution state.

Neuroprotective Effects of Banryong-hwan in Primary Rat Mesencephalic Dopaminergic Neurons (반룡환의 흰쥐태아중뇌에서의 도파민세포 보호효과)

  • Objectives : Oxidative stress has a critical role in neurodegenerative diseases. In this study, we investigated the antioxidant and neuroprotective effects of the ethanolic extract of Banryong-hwan (BRHE) in SH-SY5Y cells and primary rat mesencephalic dopaminergic neurons. Methods : To assess the antioxidant effects, we carried out 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical scavenging assay, 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)(ABTS) radical cation decolorization assay, and determination of total polyphenolic content. We evaluated the effect of BRHE treatment on neuroprotection against 6-hydroxydopamine(6-OHDA) toxicity using thiazolyl blue tetrazolium bromide(MTT) assay, nitric oxide(NO) assay, reactive oxygen species(ROS) assay in SH-SY5Y cells and tyrosine hydroxylase(TH) immunocytochemistry in primary rat mesencephalic dopaminergic neurons. Results : BRHE showed IC50 values of 328.10 ${\mu}g/mL$ and 43.12 ${\mu}g/mL$ in DPPH assay and in ABTS assay, respectively. Total polyphenolic content was 180.76 ${\mu}g/mL$. In SH-SY5Y cells, BRHE significantly attenuated the toxicity induced by 6-OHDA at the concentrations of 25-100 ${\mu}g/mL$ pre- and post- treatment in MTT assay. While 6-OHDA increased the NO and ROS contents, BRHE decreased them in a dose dependent manner. Moreover, in primary dopaminergic neuron culture, BRHE significantly protect-ed the dopaminergic cell loss against 6-OHDA toxicity up to 136% at the concentration of 75 ${\mu}g/mL$. Conclusions : These results demonstrate that BRHE has neuroprotective effect against 6-OHDA induced neurotoxicity through decreasing NO and ROS generation.

Growth Characteristics of Korean Native Kentucky Bluegrass(Poa pratensis L.) Ecotypes (국내에서 수집된 주요 왕포아풀(Poa pratensis L.)의 생육 특성)

  • The purpose of this study is to analyze the growth characteristics of native Kentucky bluegrass (Poa pratensis L.) collected in Korea. Out of Kentucky bluegrasses collected in 2000 and 2008, eight ecotypes ("Pyengchang," "Gongju," "Inje," "Donggang," "Hwasun," "Pureundle," "Cheongsong," and "Geumsan") were planted in experimental fields and pots on May 11, 2008 and May 3, 2009, respectively, for analyzing their growth characteristics. Data such as visual quality (1-9), visual density (1-9), visual color (1-9), plant height, leaf length and leaf width were analyzed. The visual quality of "Pyengchang," "Inje" and "Gongju" were enhanced compared to the rest native ecotypes of Kentucky bluegrass. "Pyengchang" showed the best visual density while "Gongju" and "Inje" showed almost evenly favorable density. However, "Pureundle," "Cheongsong," and "Geumsan" represented loose density due to the damage from the summer climate of high temperature and humidity in 2009. As for visual color, "Pyengchang" was also ranked first, maintaining high values of at least 7.6 even in the summer climate of high temperature and humidity. "Donggang" showed good color immediately after planting in the experimental field in 2008, however, undesirable color due to the damage from the summer climate of high temperature and humidity in 2009.

Salicylate-Selective Electrodes Based on Tripodal Tris-thiourea Derivatives

  • A new highly salicylate-selective PVC membrane electrode based on tripodal tris-thiourea derivatives, L1 and L2, as neutral carriers is described. The electrodes display an excellent potentiometric response to salicylate ions and an anti-Hofmeister selectivity sequence in the following order: Salicylate? > ClO4 ? > Benzoate? > I? >NO3? > NO2? > Maleate? > Acetate? > Lactate? > Fumarate?. It also exhibited a near-Nernstian potential in a linear range of 5.0 × 10?5 - 1.0 × 10?1 M with a detection limit of 9.0 × 10?5 mol/L and a slope of ?59.9 mV/decade at a pH of 7.0 in a saline buffer solution at 25 oC. The stability constant (log KS) of the anionsionophore complex was also determined at 25 oC by a conductometric titration in DMSO solution.

Effects of Taurine Supplementation on Mitochondrial Function in Chronic Ethanol Administered Rats

  • The present investigation was undertaken in vivo to determine whether the functional alterations of hepatic mitochondria induced by ethanol might be prevented by taurine. We examined the effects of supplementation of taurine on hepatic mitochondrial oxidative phosphorylation in the chronic ethanol-administered rats. Isolated hepatic mitochondria from three groups of rats were functionally tested by an analysis of $\beta-hydroxbutyrate-supported$ respiration and the coupling of this process to ATP synthesis in the presence of ADP. The three groups were control group(CO), ethanol(60g/L) administered group (AL), and ethanol (60g/L) + taurine (5g/L) supplemented group (AT). Ethanol and/or taurine were given in drinking water for 10 weeks. The mitochondria from AL group had lower state 4 respiratory rate, respiratory control (RC) ratio and ADP : O(P/O) ratio than those from CO and AT group. It showed that the ethanol administered rats were less coupled and thus less efficient with respect to mitochondrial ATP synthesis than both control rats and ethanol + taurine supplemented rats. It suggests that taurine supplementation might improve the impaired oxidative phosphorylation efficiency in mitochondrial dysfunction that is recognized as a cause of liver diseases in chronic ethanol consumption.

Occurrence of Leptosphaerulina Leaf Blight on Kentucky Bluegrass Caused by Leptosphaerulina trifolii (Leptosphaerulina trifolii에 의한 Kentucky Bluegrass의 Leptosphaerulina 잎마름병 발생)

  • In May of 2004 through 2007, Leptosphaerulina leaf blight caused by Leptosphaerulina trifolii occurred on Kentucky bluegrass (Poa pratensis) at golf courses in Gangwon Province, Korea. Symptoms on the turfgrass caused by L. trifolii were leaf blights, dying from the leaf tip downwards to the crown, which appeared patches in the field because of local pockets of severely infected (blighted) grass. Perithecia were produced on old or weak leaves, including club-shaped asci, each of which contained 8 pale brown muriform ascospores with cross and longitudinal septa. Ascospores of the fungus isolated from the diseased leaf tissue and cultured on potato-dextrose agar (PDA) were muriform multicellular (composed of 3-6 cells) and $23.4-40.5{\times}7.8-15.6{\mu}m$ in size with 3-4 transverse and 0-3 longitudinal septa, which were morphologically identical to L. trifolii reported previously. DNA sequences of ribosomal RNA gene (internal transcribed spacer) of the fungus were homologous with similarity of 99% to those of L. trifolii isolates in GenBank database, confirming the identity of the causal agent of the disease. Pathogenicity of the fungus was also confirmed on the creeping bentgrass by Koch's postulates. This is first report of Leptosphaerulina leaf blight on turfgrass caused by L. trifolii in Korea.
