Study on the Action Mechanism of Polycation in Cell Wall Formation of Suspension Cultured Cells of Daucus carota (당근 현탁배양세포의 세포벽 형성에 있어서 다가양이온의 작용기작에 관한 연구)

  • The aims of this sutdy was to investgate the action mechanism of polycation on the $\beta$-glucan synthetase II (GS II) related to cell wall synthesis in suspension cultured carrot cells. In the suspension cultured cells treated with poly-L-Iysine($12{\mu}M$) and poly-L-ornithine ($12{\mu}M$) having ploycationic nature, GS II activity increased about 40% and 50% than that of the control respectively. And similar response was observed when ATP and NaF were treated. On the other hand, ploy-L-lysine and ploy-L-ornithine did nor affect the membrane permeability. Phorbol-12-myrlstate-13-acetate (TPA), activator of protein klnase, increased about 35% and 1-(5-isoquinolinyl sulfonyl)-2-methyl-piperrazine (H-7), inhibitor of protein kinase, decreased about 30% of GSII activity than that of control. These results suggest that polycation plays a role in the cell wall synthesis by increasing GS II activity through phosphorylation.

Occurrence and Antibiotic Susceptibility of Listeria Species in Turkey Meats

  • The aims of this study were to investigate the occurrence of Listeria species in turkey meats and to check the antimicrobial susceptibility of the isolated strains. Hundred and fifteen raw turkey meat samples were randomly collected from the supermarkets, butchers and restaurants. Strain isolation and identification were made according to the ISO11290-1 method. Antimicrobial susceptibility was determined by the standard disc diffusion method. A total of 47 Listeria spp. were isolated from 115 (40.9%) raw turkey meat samples. The isolates were distributed between L. monocytogenes (25.53%), L. innocua (34.04%), L. grayi (31.91%) and L. welshimeri (8.51%). A total of 55.3 % of Listeria spp. isolates were multi-resistant to at least 3 of the antimicrobial agent tested. The level of multi-resistance was higher in L. monocytogenes strains (66.7%) than in L. innocua (62.5%) and L. grayi (53.3%). Listeria spp. isolates were highly resistant to ampicillin, cephalothin, penicillin, meticillin, oxacillin, and trimethoprime-sulfamethoxazole. The isolates particularly L. monocytogenes are increasingly resistant to one or more antibiotics and may represent a potential risk for public health because these antibiotics are common used in treatment of listeriosis. The correct and controlled use of antibiotics in veterinary medicine is important to the emergence of resistant strains.

  • The mammalian adenosine deaminase(ADA) gene was stably expressed in transgenic tobacco plane. The chimeric ADA gene 35S/35S/AMV/ADA/Tnos, has been constructed. This chimeric gene was introduced into the binary vector pRD400, which was thereafter mobilized into Agrobacterium tumefaiens strain MP90 harboring disarmed Ti-plasmid. The resulting strains were used to transform Nicofiana tabacum L. using the leaf disc. Incorporation of the chimeric gene into plant were confirmed by PCR and Northern blot analyses. Immunoblot analysis showed that ADA protein was successfully synthesized in the transgenic tobacco plants.

Antipathogenic Activity of Lactobacillus plantarum Isolated from Pickled Mulberry Leaf (뽕잎 장아찌로부터 분리된 Lactobacillus plantarum 균주의 유해균 증식 억제 활성)

  • Strains of lactic acid bacteria were isolated from a variety of fermented foods collected in Korea. The strain L2167 showed a strong antipathogenic activity against Bacillus cereus, Listeria monocytogenes, Salmonella Typhimurium, Staphylococcus aureus, and Staphylococcus epidermidis. L2167 was identified as Lactobacillus plantarum by sequence analysis of its 16S rRNA gene. Scanning electron microscopy revealed rough and wrinkled morphology of B. cereus, L. monocytogenes, S. Typhimurium, S. aureus, and S. epidermidis cell membranes after treatment with a crude cell extract of L. plantarum L2167, indicating that Lactobacillus plantarum L2167 might destroy the cell membrane of pathogenic bacteria. The optimal temperature and initial medium pH for Lactobacillus plantarum L2167 growth were 35℃ and 5.5, respectively. Lactobacillus plantarum L2167 was more sensitive to NaCl than Lactobacillus plantarum KCTC21004, used as a control strain. Lactobacillus plantarum L2167 is expected to be developed as a prominent starter strain for efficient inhibition of growth of pathogens.

Effect of Temperature and Bake-out on Formaldehyde Emission from UF Bonded Wood Composites

  • This study analysis the effect of various temperatures (20, 35 and $50^{\circ}C$) on the formaldehyde emission from wood composites, which were particleboard (PB), medium density fiberboard (MDF), high density fiberboard (HDF) and laminated HDF (L-HDF) by Japanese desiccator method. Also, to reduce formaldehyde emission by wood composites, it has been suggested that undergo a bake-out conditions. On average, the level of formaldehyde emission increased many times with a $15^{\circ}C$ increase in temperature from 20 to $35^{\circ}C$ for PB, MDF, HDF and L-HDF, respectively. Formaldehyde emissions from wood composites could be expected to increase with increasing ambient temperature. At $35$ for 28 days bake-out treatment of boards, the free formaldehyde emission reduced 67.8% (PB), 40.1% (MDF), 37.8% (HDF), and 35.2% (L-HDF). On the other hand, after the bake-out at $50^{\circ}C$ for 28 days, the formaldehyde concentration decreased by 88.2, 66.9, 62.2 and 59.3% of the concentration before the bake-out for PB, MDF, HDF and L-HDF, respectively. An interesting of the bake-out treatment at $50^{\circ}C$ after 14 days, formaldehyde emission grade of PB & MDF down $E_2$ to $E_1$, and HDF & L-HDF down $E_1$ to $E_0$.

Analysis Growth Performance and Ascorbic Acid Contents of Allium victorialis var. platyphyllum, Ligularia fischeri, and L. stenocephala Under Changing Light Intensity (광도 변화에 따른 산마늘, 곰취, 곤달비의 생장 및 Ascorbic acid 함량분석)

  • The present study was conducted to investigate growth performance and ascorbic acid contents of Allium victorialis var. platyphyllum, Ligularia fischeri, and L. stenocephala growing under four different light intensity regimes (full sun, and 64~73%, 35~42%, 9~16% of full sun). The leaf area and total biomass of Allium victorialis var. platyphyllum were highest in 35~42% of the full sun. However, the leaf area and total biomass were highest in 64~73% of the full sun in Ligularia fischeri, and L. stenocephala. The leaf thickness decreased with increasing shading in all three wild vegetables. The growth of root comparing to that of shoot decreased as shading increased, and as the result, the ratio of shoot biomass to root biomass increased. Ascorbic acid contents were higher in Allium victorialis var. platyphyllum than in Ligularia fischeri and L. stenocephala. Allium victorialis var. platyphyllum showed the highest contents of ascorbic acid in 35~42% of full sun. Whereas Ligularia fischeri and L. stenocephala showed the highest in 64~73% of full sun.

Cryopreservation of Bovine IVM/IVF/IVC Hatched Blastocysts (체외생산된 소 완전탈출 배반포기배의 동결보존)

  • This study was to test whether the viability of bovine hatched blastocysts (HBs) can be maintained after vitrification and thawing. The HBs were produced in vitro at Day 9 and Day 10 after IVF, and they were classified to small (S-HBs; ø$\leq$300 ${\mu}{\textrm}{m}$) and large(L-HBs; ø>300 ${\mu}{\textrm}{m}$) on the basis of embryo diameter using eyepiece micrometer. As freezing solution, we used EFS35 which consisted of 35% ethylene glycol (EG), 18% ficoll, 0.3 M sucrose and 10% FBS added in mDPBS. Vitrification was taken by two-step method, the HBs were equilibrated in 10% EG for 5 minutes and then shortly exposed in EFS35 and plunged into L$N_2$for 30~45 sec. After thawing, the survival rates were assessed by the re-expansion of the blastocoel during 2 h and 16 h of culture. The results obtained in these experiments were summarized as follows; 1) When the blastocysts(40.8%) recovered at Day 8 after IVF were further cultured for 24 h(Day 9 after IVF) and 48 h(Day 10 after IVF), the rates of HBs were 20.5% and 6.7%, respectively. Also, the total cell number of HBs on Day 9 was significantly higher than that of HBs on Day 10 (p<0.01). 2) When the effects of freezing solution to the survival of Day 9 L-HBs were examined, the rate of vitrified group (75.7%) was significantly lower than 100% of control and exposed group(p<0.05). 3) When the survival rates of vitrified HBs according to size and developmental age were examined, the data of L-HBs (75.5%) and S-HBs(63.6%) on Day 9 were slightly higher than those of L-HBs(64.3%) and S-HBs(60.7%) on Day 10. 4) Also, when the in vitro survival of Day 9 HBs was evaluated under different culture condition after thawing, the result in culture medium only (79.3%) was significantly higher than 43.2% in co-culture group (p<0.05). These results demonstrated that bovine HBs can be successfully cryopreserved by two-step vitrification method using EFS35.

활어수송시 수온, 염분 및 마취제에 의한 넙치(Paralichthys olivaceus)의 스트레스 반응

  • 어류의 양식생산 과정중에는 사육중인 어류에게 스트레스로 작용할 수 있는 여러 가지 요인들이 있다. 그 중에서도 생산된 종묘의 양식장 운송, 수확한 어류의 소비지 운반 등은 양식과정중 피할 수 없는 작업이다. 어류의 수송 후에는 혈장 corticosteroid, glucose, 전해질, 삼투질 농도, 적혈구수, hematocrit, hemoglobin 등이 변화하는 것으로 알려지고 있다(Chang et al. 2001; Hur et al., 2002, 2003). 스트레스 요인별 연구로는 염분, 수온, 밀도 및 마취제에 관한 것 등이며, 아울러 이들의 복합적인 요인에 대하여 스트레스 반응을 최소화시키려는 연구가 진행되고 있다. 어류종묘의 수송이나 수확된 어류가 대량으로 수송되고 있으므로, 넙치(Paralichthys olivaceus)와 같은 대량수송 어류의 수송에 따른 스트레스 반응에 대한 연구의 필요성이 제기된다. 그러므로 본 연구에서는 넙치를 사용하여 염분, 수온 및 마취제에 의한 수송이 스트레스 지표로 알려져 있는 혈액학적 요인, cortisol, glucose, lactic acid 및 삼투질 농도 둥에 나타나는 생리학적 반응을 조사하여, 활어수송 과정에서 나타나는 스트레스 반응에 대한 기초자료를 제공하고자 하였다. 실험어는 21.2 cm, 97.4 g인 양식 넙치를 사용하여, 수온은 20℃ (natural water temperature, NWT)와 15℃ (cooling water temperature, CWT), 염분은 해수(35‰)와 15‰해수, 마취제(anesthesia, Anes., MS-222)는 50 ppm의 조건으로 혼합한 실험구를 설정하였다. 실험구는 각각 NWT+35‰, CWT+35‰, NWT+15‰, NWT+15‰, NWT+35‰+Anes., CWT+35‰+Anes., NWT+15‰+Anes. 및 CWT+15‰+Anes.의 8개 실험구를 2반복으로 설정하여 경북울진∼부산까지 약 400 km (6시간)를 차량수송하였다. 수송용기는 스티로폼상자(66×42×20 cnn)로서, 여기에 해수 3 L와 액화산소를 넣은 비닐봉지에 넙치 8마리씩 수용하여 수송하였다. 혈액의 성상 및 분석항목은 수송전ㆍ후에 채혈하여 비교하였다. 수송전 hematocrit는 22.2±3.8%에서 수송후 NWT+35‰에서 15.3+3.9%, CWT+35‰은 16.7±3.0%, NWT+15‰구에서는 19.2±1.8%로 낮아졌으며, CWT+15‰구는 20.9±3.6%로 수송전과 차이가 없었다. 한편 NWT+15‰+Anes.구는 17.8±0.9%, CWT+15‰+Anes.구는 14.5±1.5%로 낮아졌다. Cortisol은 수송전 2.4±0.1 ng/ml로부터 CWT+35‰구는 16.7±12.8 ng/ml, NWT+35‰구는 47.9+19.8 ng/ml, NWT+15‰구는 43.5±13.9 ng/ml, CWT+15‰구는 26.1±8.3 ng/ml, NWT+15‰+Anes.구는 61.7±3.3 ng/ml, CWT+15‰+Anes.구는 86.1±19.0 ng/ml로 높아졌다. Glucose는 수송전 74.2±32.6 mg/dl로부터 NWT+35‰구는 197.9±27.5 mg/dl, CWT+35‰구도 272.1±29.9 mg/dl로 유의하게 높아졌다. Na/sup +/의 수송전 농도는 163.5±0.6 mEq/L로부터 NWT+35‰구와 CWT+35‰구는 각각 175.3±1.2 mEq/L, 190.0±5.0 mEq/L로 높아졌으며, 다른 실험구에서는 차이가 없었다. 본 연구 결과, cortisol과 glucose에서 수송전보다는 모든 실험구에서 높게 나타나 수온, 염분 및 마취제를 사용하여도 스트레스를 받고 있는 것으로 나타났다. 특히, cortisol의 경우, 수온과 염분만을 혼합한 실험구보다 마취제를 혼합한 실험구에서 높게 나타났다. 다른 혈액학적 항목에서는 염분과 마취제를 사용하지 않았던 실험구인 NWT+35‰구와 CWT+35‰구에서 다른 실험구에 비하여 glucose, Na/sup +/ 및 Cl/sup -/ 등에서는 높게 나타나는 경향을 보였다.

Inhibition of Escherichia coli 0157:H7 and Staphylococcus aureus 196E by allspice (Pimenta dioica L.) during cold storage (저온저장중 Allspice(Pimenta dioica L.)에 의한 Escherichia coli 0157:H7과 Staphylococcus aureus 196E의 생존억제)

  • The effect of low concentrations of allspice (Pimenta dioica L.) in culture broth as an antibacterial agent against Escherichia coli 0157:H7 and Staphylococcus aureus 196E was tested at 35,5 and -20$^{\circ}C$. Tryptic soy broth (TSB) containing 0∼2% (w/v) of allspice was inoculated with 10$\^$5/∼10$\^$6/ cells/$m\ell$ of E. coli and S. aureus and incubated at each temperature. The growth of E. coli was not inhibited at 0.1∼1.0% allspice and growth occured at 2% allspice but only after a prolonged lag period. Growth of S. aureus was inhibited with increasing concentration of allspice at 35$^{\circ}C$. Growth of S. aureus occured at the presence of 0.1∼0.3% allspice but the viability of S. aureus at 0.5∼2.0% allspice was decreased during storage at 35$^{\circ}C$. During refrigerated storage at 5$^{\circ}C$, inhibition of E. coli and S. aureus was increased with the progress of time and increasing spice concentration. During frozen storage at -20$^{\circ}C$, antibacterial activity of allspice against E. coli was increased with increasing storage time and spice concentration while that activity against S. aureus was effective during early period of storage. There was no major changes in population of S. aureus in TSB with different concentration of spice frozen at -20$^{\circ}C$. Viable counts of E. coli and S. aureus at 0.l% of allspice was less than that of control during frozen storage.

Injury and Recovery of Pathogenic Bacteria Isolated from Seafoods - Changes in the Viability of Staphylococcus aureus and Listeria monocytogenes in Some Fish Homogenates during Cold Storage - (해산물에서 분리된 식중독세균의 손상 및 회복 -생선 homogenate에서 Staphylococcus aureus와 Listeria monocytogenes의 저온저장중 세균수 변화 -)

  • The survival and growth of Staphylococcus aureus and Listeria monocytogenes in fish homogenates (flounder, shrimp and oyster homogenate) and tryptic soy broth (TSB) were tested during storage at simulated ambient (35$^{\circ}C$), refrigerated (5$^{\circ}C$) and frozen (-20$^{\circ}C$) temperature. A similar growth pattern of S. aureus at 35$^{\circ}C$ was observed in fish homogenates and TSB. Survival of S. aureus decreased at refrigerated or frozen temperature and that was greater at -20$^{\circ}C$ (0.3-1.2 log reduction/6 weeks) than at 5$^{\circ}C$ (1-1.6 log reduction/3 weeks). Viable cells of L. monocytogenes increased rapidly at 35$^{\circ}C$ in flounder homogenate, shrimp homogenate and TSB but after a prolonged lag period in oyster homogenate. During 3 weeks of storage at 5$^{\circ}C$, the levels of L. monocytogenes increased 3.8-5.0 log cycles in flounder homogenate, shrimp homogenate and TSB whereas levels increased 2.2 log cycles in oyster homogenate. Viable cells of L. monocytogenes during 6 weeks of frozen storage decreased 1.5-1.8 log cycles in flounder homogenate, shrimp homogenate and TSB while decreased 2.8 log cycles in oyster homogenate.
